Description
(式中、Rは、S、O、N、SO、SO2から選ばれる1個以上のヘテロ原子又はヘテロ原子の群によって任意に割り込まれていてもよいC10-24不飽和炭化水素基であり、前記炭化水素基は、少なくとも4個の非共役二重結合を含み:
Lは、R基とカルボニルCOとの間に原子を1〜5個有する橋状結合を形成する連結基であり;
Xは、電子求引基である。)

(II)N原子を任意にメチル化すること;
(III)TFPOと反応させること;及び
(IV)形成された水酸基をケトンまで酸化すること。
(ii).疾患を阻害すること、つまり、疾患の発症、又はその再発、又はその臨床的若しくは亜臨床的な症状の少なくとも1つを停止させる、減少させる、又は遅延させること;あるいは
(iii).疾患の臨床的若しくは亜臨床的な症状の1つ以上を軽減させるか又は減退させること。
、又は制御放出性の用途のために投与することができる。
[試薬]
炎症誘発性のサイトカインの遺伝子発現及び産生がRA由来の滑膜の線維芽細胞様の細胞に似ていることから、コンフルエントの状態又はスフェロイドの状態のSW982モデルセルラインのセルカルチャー(Wada Y、2005)を用いた。
刺激実験のために、0.5mLの細胞懸濁液(5〜7×105細胞/mL)を48ウエル・プレートの内側のウエル内に播種した。外側のウエルは、0.5mLのPBSで満たした。ほとんどコンフルエントな細胞数とするために、細胞を1日間インキュベートした(37℃、5%CO2)。コンフルエント後の細胞を得るために、プレートを3日間インキュベーター中に置いた。このインキュベーション時間後、300μLの上清を取り除き、50μLの刺激用培地で置換した。
阻害は、刺激と同様な方法において実施した。播種、インキュベーション及び200μLの0.5%FBS含有DMEMで培地置換の後、50μLの個々の阻害剤溶液を添加した(表1)。37℃で2時間後、50μLの上清を取り除き、前記の3種の刺激物質で置換した。
化合物2及び化合物1は、DMEM/0.5%FBS中で50μMプレ希釈液を調製するのに先だって、96%エタノール中で1:50に希釈した。
PGE2の検出
サンプル及びコントロールを、標準希釈剤中でゆっくり溶かし、1:1と1:2500の間に希釈した。最大の希釈は、1段階で1:10とした。いくつかの中間希釈液を調製したのは、このためである。最初は、全ての値は2連で測定した。テクニカルエラーを最小限にした後は、サンプルは一個のものとして分析しただけである。いくつかの軽微な修正を除いて、これ以上の全てのステップは、EIAキットのマニュアルにある製造業者の推奨に従って実施した。結果を最適化するために、アルカリホスファターゼの基質のインキュベーション時間は、15分間延長した。インキュベーションの間、プレートは暗所に置いた。サンプルとコントロールの配置の例は、添付書類(appendix)において図示する。読み取りは、120rpmで10秒間振盪後、マルチスキャン・プレート・リーダー(Ascent Labsystems)を用いて波長414nm及び595nmで行った。データ取得のための対応するソフトウエアーは、Ascent software for Multiscan, Version 2.4.1を用いた。
非増殖性のSW982滑膜細胞における炎症誘発性メディエーターの誘導因子としてのTNF及びIL1の効果
増殖性のSW982細胞、コンフルエントなSW982細胞、及び分化型の(コンフルエントの3日後の)SW982細胞における炎症誘発性メディエーターの誘導因子として、2種のサイトカイン、すなわちTNF及びIL1、の効果を調べた(SW982は、本願の実施例を通して用いた滑膜細胞のセルラインである)。qPCRによって、マトリックスメタロプロテアーゼ(MMP1)、間質性接着分子(interstitial adhesion molecule)(ICAM)、IL6、IL8、TNF及び酵素COX2を、転写レベルについて評価した。結果を図1に示す。図1は、IL1と比べた場合TNFはより強力な誘導因子であるが、評価した炎症誘発性メディエーターの範囲では両サイトカインともに強い誘導因子であるという明らかな傾向を示している。従って、下記の実施例において用いたモデル系は、RAの治療のための臨床上の関連性を有している。
化合物2は、SW982細胞におけるアラキドン酸放出の強力阻害剤である。
化合物1及び化合物2によるTNF誘導性のPGE2誘導の制御
SW982細胞を、12〜48時間の種々の長さの時間、TNFで刺激した。TNF刺激は、PGE2の産生の増加を誘導し、これは図2に明らかに示されている。しかしながら、驚くべきことに、このPGE2の産生は、化合物1及び化合物2(2.5μMで>100%の阻害)によって、用量依存的に阻害することができる。図3は、TNF誘導性のPGE2産生に対する化合物1及び化合物2の阻害について、それぞれ、IC50値を〜630nM及び〜430nMと計算したことを示す。
化合物1によるTNF誘導性のIL8産生の阻害
IL8のような滑膜細胞の炎症誘発性メディエーターは、軟骨細胞、軟骨とオートクリンの方法の滑膜細胞の両方に関して、関節における炎症反応の重要な制御因子である。図4は、TNF(図4)によって刺激したSW982セルラインにおけるIL8レベル(ELISAでモニターした)を示す。図4は、化合物1がIL8産生を用量依存的に阻害することを示す。
本発明の化合物による、TNF誘導性のESE−1活性化(NF−kBにより制御される転写因子)の制御
ESE−1は、etsドメインと呼ばれる高度に保存されたca85アミノ酸のDNA結合領域によって定義されるets転写因子のファミリーに属する。ets転写因子は、細胞の増殖及び分化、細胞周期進行、血管新生並びに癌化を含む多様な機能に関与する遺伝子を制御することが示された。ESE−1は、RAの滑膜組織において、高度に上方制御されることが分かった。NF−kBが主要な制御因子であると考えられているにもかかわらず、ESE−1プロモーターは、多くの補助因子のための結合部位を含んでいる。
化合物2は、炎症及び関節破壊に関与する多くの遺伝子の転写制御に作用する。
プロスタグランジンE2合成は、化合物2によって大幅に減少する
化合物2がAAの放出を効率的に減少させたので、次に、我々は、阻害は、また、引き続いて起こるAAのPGE2への変換にも反映するかどうかを調べた(PGE2は、関節における炎症及び破壊の両プロセスの重要な制御因子である)。

- 式(I)の化合物又はその塩を有効成分として含むリウマチ性関節炎の治療用薬剤。

(式中、Rは、直鎖のC10〜24不飽和炭化水素基であり、前記炭化水素基は、少なくとも4個の非共役二重結合を含み;
Lは、R基とカルボニルCOとの間の−SCH 2 −であり;
Xは、CF 3 である)。 - Rが、5個の非共役二重結合を含む、請求項1に記載の薬剤。
- R基は、炭素原子を17〜19個有する請求項1または2に記載の薬剤。
