Description
従来、ヒト多能性幹細胞株は、フィーダー細胞または各種高分子などに接着させる平面培養によって増殖維持されてきた。しかしながら、高品質のヒト多能性幹細胞を、安定して大量に培養増殖させる技術は確立されていない。特に、実用化に必要となる大量の多能性幹細胞を調製するには、従来法である培養容器に接着させて継代により増殖させる方法では限界がある。例えば、ヒト多能性幹細胞用の接着基質材料として、品質面とコスト面で最適なものは開発されておらず、また、継代には複雑な多段階の操作が必要であり、通常は酵素処理など、安全面およびコスト面で不利なステップが含まれている。
[1] 以下の工程を繰り返すことを特徴とする、多能性幹細胞の維持増幅方法。
(i) 多能性幹細胞を、細胞塊の平均直径が約200-約300 μmとなるまで浮遊培養する工程
(ii) 工程(i)により得られた細胞塊を、平均直径が約80-約120 μmである均一な細胞塊に分割する工程
[2] 工程(i)において、多能性幹細胞を、細胞塊の平均直径が約250 μmとなるまで浮遊培養し、工程(ii)において、平均直径が約80 μmである均一な細胞塊に分割することを特徴とする、上記[1]記載の方法。
[3] 工程(ii)における分割が、細胞塊をメッシュに通すことにより行われることを特徴とする、上記[1]または[2]記載の方法。
[4] メッシュの孔径が約30-約70 μm、好ましくは約40-約60 μm、より好ましくは約50 μmである、上記[3]記載の方法。
[5] 工程(i)の培養が、細胞塊同士の接着を起こさない粘度を持つ水溶性高分子成分を含む培地中で行われる、上記[1]〜[4]のいずれかに記載の方法。
[6] 水溶性高分子が、多糖もしくはそのエーテル、合成ハイドロゲル高分子および生体高分子、並びにそれらを模倣した人工高分子から選ばれる、上記[5]記載の方法。
[7] 水溶性高分子がメチルセルロースまたは昇温型温度感受性ハイドロゲルである、上記[5]記載の方法。
[8] 多能性幹細胞がES細胞またはiPS細胞である、上記[1]〜[7]のいずれかに記載の方法。
[9] 多能性幹細胞がヒト由来である、上記[1]〜[8]のいずれかに記載の方法。
(i) 多能性幹細胞を、細胞塊の平均直径が約200-約300μmとなるまで浮遊培養する工程
(ii) 工程(i)により得られた細胞塊を、平均直径が約80-約120 μmである均一な細胞塊に分割する工程
ES細胞は、対象動物の受精卵の胚盤胞から内部細胞塊を取出し、内部細胞塊を線維芽細胞のフィーダー上で培養することによって樹立することができる。また、継代培養による細胞の維持は、白血病抑制因子(leukemia inhibitory factor (LIF))、塩基性線維芽細胞成長因子(basic fibroblast growth factor (bFGF))などの物質を添加した培養液を用いて行うことができる。ヒトおよびサルのES細胞の樹立と維持の方法については、例えばUSP5,843,780; Thomson JA, et al. 一方、DNAの形態の場合、例えば、ウイルス、プラスミド、人工染色体などのベクター、リポフェクション、リポソーム、マイクロインジェクションなどの手法によって体細胞内に導入することができる。ウイルスベクターとしては、レトロウイルスベクター、レンチウイルスベクター(以上、Cell, 126, pp.663-676, 2006; Cell, 131, pp.861-872, 2007; Science, 318, pp.1917-1920, 2007)、アデノウイルスベクター(Science, 322, 945-949, 2008)、アデノ随伴ウイルスベクター、センダイウイルスベクター(WO 2010/008054)などが例示される。また、人工染色体ベクターとしては、例えばヒト人工染色体(HAC)、酵母人工染色体(YAC)、細菌人工染色体(BAC、PAC)などが含まれる。プラスミドとしては、哺乳動物細胞用プラスミドを使用しうる(Science, 322:949-953, 2008)。ベクターには、核初期化物質が発現可能なように、プロモーター、エンハンサー、リボゾーム結合配列、ターミネーター、ポリアデニル化サイトなどの制御配列を含むことができるし、さらに、必要に応じて、薬剤耐性遺伝子(例えばカナマイシン耐性遺伝子、アンピシリン耐性遺伝子、ピューロマイシン耐性遺伝子など)、チミジンキナーゼ遺伝子、ジフテリアトキシン遺伝子などの選択マーカー配列、緑色蛍光タンパク質(GFP)、βグルクロニダーゼ(GUS)、FLAGなどのレポーター遺伝子配列などを含むことができる。また、上記ベクターには、体細胞への導入後、初期化因子をコードする遺伝子もしくはプロモーターとそれに結合する初期化因子をコードする遺伝子を共に切除するために、それらの前後にLoxP配列を有してもよい。
また、RNAの形態の場合、例えばリポフェクション、マイクロインジェクションなどの手法によって体細胞内に導入しても良く、分解を抑制するため、5-メチルシチジンおよびpseudouridine (TriLink Biotechnologies)を取り込ませたRNAを用いても良い(Warren L, (2010) Cell Stem Cell. 7:618-630)。
あるいは、37℃、5% CO2存在下にて、フィーダー細胞(たとえば、マイトマイシンC処理STO細胞、SNL細胞等)上で10%FBS含有DMEM培養液(これにはさらに、LIF、ペニシリン/ストレプトマイシン、ピューロマイシン、L-グルタミン、非必須アミノ酸類、β-メルカプトエタノールなどを適宜含むことができる。)で培養し、約25〜約30日又はそれ以上ののちにES様コロニーを生じさせることができる。望ましくは、フィーダー細胞の代わりに、初期化される体細胞そのものを用いる(Takahashi K, et al. (2009), PLoS One. 4:e8067またはWO2010/137746)、もしくは細胞外基質(例えば、Laminin(WO2009/123349)およびマトリゲル(BD社))を用いる方法が例示される。
この他にも、血清を含有しない培地を用いて培養する方法も例示される(Sun N, et al. (2009), Proc Natl Acad Sci U S A. 106:15720-15725)。さらに、樹立効率を上げるため、低酸素条件(0.1%以上、15%以下の酸素濃度)によりiPS細胞を樹立しても良い(Yoshida Y, et al. (2009), Cell Stem Cell. 5:237-241またはWO2010/013845)。
上記のようにして調製された多能性幹細胞を、細胞塊の平均直径が約200-約300μmとなるまで浮遊培養する。ここで「約」とは、±10%を許容する意味で用いる。細胞塊の直径が300 μmを超えると、細胞が分泌するサイトカイン等の影響で微小環境が形成され、分化が誘導されてしまう。また、細胞塊の中心部で壊死が起こるため、生細胞の回収率が悪くなるといった問題がある。一方、細胞塊の平均直径の下限は、浮遊培養開始時(継代後の浮遊培養では継代時)の細胞塊の平均直径より大きいサイズであれば特に制限はないが、多能性幹細胞の収率を考慮すれば約200 μm以上まで培養を続けることが好ましい。
工程(i)によって得られる、平均直径が約200-約300 μmである均一なサイズの多能性幹細胞の細胞塊は、次いで、平均直径が約80-約120 μmである均一な小細胞塊に分割され、継代される。ここで「約」とは±20%を許容する意味で用いる。細胞塊の平均直径が50 μm以下となるように分割すると、細胞がアポトーシス等の細胞死を起こしやすくなるので好ましくない。分割後の平均直径の上限は特に制限されないが、サイズが大きくなるほど次回の継代後の浮遊培養による増幅の効率が悪くなるので、約120 μm以下であることが好ましく、約80 μmとすることが特に好ましい。
基本培地及び培地添加剤ストック液
・mTeSR1 (既知組成培地; Stem Cell Technologyより購入, 型番05850)
・3% メチルセルロースストック溶液 (IMDM培地中)(R&Dより購入, 型番HSC001)
使用前にメチルセルロースを融解、2.0 ml容で再凍結し、直ちに-20℃で保存。
・10 mM Y-27632ストック溶液 (Ca, Mg不含PBS中)(Rock阻害剤; Sigmaより購入, 型番Y0503)
培養ディッシュ
・Ultra Low Cluster Plate, 6ウェル蓋付き (Corningより購入, 型番Costar 3471)
・35 mm ペトリディッシュ (BDより購入, 型番351008)
継代用メッシュ
・10, 20, 30及び50 μm孔径 CellTricsフィルター (PARTECより購入, 型番それぞれ06-04-004-2324, 06-04-004-2325, 06-04-004-2326及び06-04-004-2327)
・40 μm孔径 セルストレーナー(BDより購入, 型番352340)
培地1 (継代培養用; Rock阻害剤含有)
mTeSR1 10 ml
3% 滅菌メチルセルロース 1 ml (終濃度 0.28%)*
10 mM Y-27632 11 μl (終濃度 10 μM)
*実験によっては、終濃度0 (非添加), 0.25, 0.3及び0.5%の培地も使用した。
培地2 (培地交換用; Rock阻害剤不含)
mTeSR1 10 ml
3% 滅菌メチルセルロース 1 ml (終濃度0.28%)*
*実験によっては、終濃度0 (非添加), 0.25, 0.3及び0.5%の培地も使用した。
ヒト多能性幹 (hPS) 細胞株
・ヒト胚性幹細胞株 (hES細胞株): KhES-1 (京都大学再生医科学研究所より入手)
・ヒト人工多能性幹細胞株 (hiPS細胞株): IMR90-1 (WiCell, Wisconsin, USAより入手)
253G1 (RIKEN BIORESOURCE CENTER CELL BANKより入手)
浮遊スフェア培養及び継代培養
ウェル又はディッシュを円を描くように動かして、ウェル又はディッシュの中央にhPS細胞スフェアを集め、15 mlチューブAに移した。mTeSR 2.5 mlをウェル又はディッシュに添加し、残った凝集体を15 mlチューブAに集めた。チューブAを遠心分離して上清を除いた。スフェアを0.5 mlの培地1に再懸濁した。別の新しい15 mlチューブBを用意し、3 mlの培地1を添加した。スフェア浮遊液を200 μl ピペットを用いて穏やかに3回ピペッティングした後、必要量のスフェア浮遊液をチューブBにとった。継代時の分割比率は、継代初期(継代数1-2)には1:3-1:4、その後は1:8-1:12とした。
次いで、スフェア浮遊液を、滅菌したCellTricsフィルター又はセルストレーナー(5 mlチューブの先に装着)に通した。スフェア浮遊液を5 ml ピペットを用いて穏やかに3回ピペッティングした後、ウェル又はディッシュに播種した。ウェル又はディッシュをCO2 インキュベーター中、37℃, 5% CO2の通常の条件下で培養した。1日目及び3日目に、培地を、予め加温した培地2に交換した。
予備実験として、0.25%, 0.28%又は0.5%(w/v)のメチルセルロースを含む培地1及び培地2でhES細胞株KhES-1の培養を行った。0.28 w/v% メチルセルロースの濃度条件において、細胞スフェアの融合を最も効率よく防ぐことができたので(4回の計測で1.0%, 1.1%, 1.3%及び1.6%の融合率; 計722-978スフェア中8-13が融合スフェア)、以後の実験においては、特にことわらない限り、0.28 w/v% メチルセルロース含有培地を使用した。
継代時に使用するメッシュとして、50 μm孔径のCellTricsフィルターと40 μm孔径のセルストレーナーとを比較したところ、50 μm孔径のメッシュの方が優れた細胞増殖効果を示したので、以後の実験においては、特にことわらない限り、50 μm孔径のメッシュを使用した。
0.28 w/v% メチルセルロースを含有する培地1及び培地2、並びにメッシュとして50 μm孔径のCellTricsフィルターを用い、前記[方法]に記載した手順で、20継代までKhES-1株の浮遊スフェア培養を行った。結果を表1に示す。
尚、70 μm又は100 μmの孔径のメッシュも試験したが、開口が大きすぎてhES細胞スフェアを効率よく継代培養することはできなかった。
(1)hES細胞
継代4回目のKhES-1細胞のスフェアを、継代0, 1, 3及び5日後に、顕微鏡で観察した (図1)。スフェアのサイズは均一に増大していた。
11回継代後のhES細胞における多能性マーカー (SSEA4) の発現をFACS分析で調べた。hES細胞の96.3%がSSEA4陽性であった (図2)。
10回継代後のhES細胞スフェアの凍結切片を調製し、抗Oct3/4抗体で染色した。その結果、全ての細胞の核が染色された(図3)。
これらの結果は、hES細胞の未分化状態が10又は11回継代後であっても、よく維持されていることを示している。
次に、18回継代後のKhES-1細胞をフィーダー細胞上に播種し、付着培養した。その結果、典型的なhES細胞コロニーが形成された (図4)。これらのhES細胞コロニーの、多能性マーカー (Tra-1-60, Oct 3/4, SSEA-4) についての免疫細胞染色により、該hES細胞の多能性は18回継代後も維持されていることが明らかとなった (図5)。
さらに、17回継代後のKhES-1細胞の核型分析の結果、該hES細胞の染色体は、17回継代後でも正常であることが示された (図6)。
実施例3と同様の浮遊培養及び継代プロトコルにより培養したhiPS細胞(IMR90-1株及び253G1株)の特性解析を実施した。hES細胞と同様の結果が、hiPS細胞についても得られた。即ち、10回以上継代したIMR90-1細胞のスフェアを、継代直後及び継代5日後に、顕微鏡で観察した。スフェアのサイズは均一に増大していた (図7)。また、5回継代後のIMR90-1細胞をフィーダー細胞上に播種し、付着培養した。その結果、典型的なhiPS細胞コロニーが形成された (図7)。さらに、11回継代後のIMR90-1細胞における多能性マーカー (SSEA4) の発現をFACS分析で調べた。IMR90-1細胞の97%がSSEA4陽性であった (図7)。また、22回継代後の253G1細胞スフェアの凍結切片を調製し、未分化マーカー (OCT3/4, NANOG, SSEA4) の発現を免疫染色で調べた。その結果、全ての細胞で未分化性が確認できた(図8)。
これらの結果は、上記プロトコルがES細胞だけでなく、iPS細胞の維持・増幅にも利用できることを示すものである。
次に、長期継代培養後のhES細胞 (KhES-1株) 及びhiPS細胞 (253G1株) の多能性を調べた。まず、41回継代後のKhES-1細胞スフェアと、10回継代後の253G1細胞スフェアとを、I. Minami et al. (2012) Cell Reports, in pressに記載の方法に従って培養し、心筋細胞への分化を誘導した。その結果、253G1細胞スフェア由来のコロニーの89.1%、KhES-1細胞スフェア由来のコロニーの84.2%が拍動していた (図9A)。また、FACS解析の結果、心筋マーカーであるcTnT陽性の細胞は、いずれも約90%であった (図9B,C)。
さらに、25回継代後の253G1細胞スフェアを、K. Sakurai et al. (2010) Nucleic Acids Res., 38: e96に記載の方法に従って培養し、神経細胞への分化を誘導した。分化誘導32日後の細胞について、抗βIII-Tubulin抗体とDAPIを用いて二重染色を行った (図10)。その結果、神経細胞マーカーであるβIII-tubulin陽性のニューロン様の形態の細胞が確認された。253G1細胞スフェア由来のコロニーの89.1%、KhES-1細胞スフェア由来のコロニーの84.2%が拍動していた (図9A)。また、FACS解析の結果、心筋マーカーであるcTnT陽性の細胞は、いずれも約90%であった (図9B,C)。
(1)継代24回目の253G1細胞のスフェアを、継代1, 3, 5, 7及び9日後に、顕微鏡で観察した (図11)。継代後7日以降では、内部に不均一な構造体が現れたので、継代5日後に次の継代を行うこととした。
また通常、メッシュは細胞分散後に残存する細胞塊の除去に使われてきたので、それ自体で細胞塊を分割するというアイデアは全く予見不可能である。
さらに、培養液の粘性を高める方法は、従来は造血系細胞のような非付着性細胞において、培養液内の拡散や流れを抑止して細胞周囲の微細環境を形成する目的で使用されてきたが、細胞塊の接着融合防止の目的で使用されている例は少ない。実際、これまで10編程度の浮遊培養法に関する論文のほとんど全てにおいて、細胞塊同士の接着融合の問題が大きいので、これを抑制したいと記述しながら、このような方策を思いついた論文は皆無である。
本発明は、これらの従来技術からは予期し得ない複数の効果の発見に基づくものであり、これにより、多能性幹細胞の医療や創薬への実用化に不可欠な、大量培養装置、特に自動化培養装置の設計と組み込みにとって、従来法と比較して、極めてシンプルで有効であり、これら培養装置の開発に活用できる。
ここで述べられた特許及び特許出願明細書を含む全ての刊行物に記載された内容は、ここに引用されたことによって、その全てが明示されたと同程度に本明細書に組み込まれるものである。
Claims (9)
- 以下の工程を繰り返すことを特徴とする、多能性幹細胞の維持増幅方法。
(i) 多能性幹細胞を、細胞塊の平均直径が約200-約300 μmとなるまで浮遊培養する工程
(ii) 工程(i)により得られた細胞塊を、平均直径が約80-約120 μmである均一な細胞塊に分割する工程 - 工程(i)において、多能性幹細胞を、細胞塊の平均直径が約250 μmとなるまで浮遊培養し、工程(ii)において、平均直径が約80 μmである均一な細胞塊に分割することを特徴とする、請求項1記載の方法。
- 工程(ii)における分割が、細胞塊をメッシュに通すことにより行われることを特徴とする、請求項1または2記載の方法。
- メッシュの孔径が約30-約70 μm、好ましくは約40-約60 μm、より好ましくは約50 μmである、請求項3記載の方法。
- 工程(i)の培養が、細胞塊同士の接着を起こさない粘度を持つ水溶性高分子成分を含む培地中で行われる、請求項1〜4のいずれか1項に記載の方法。
- 水溶性高分子が、多糖もしくはそのエーテル、合成ハイドロゲル高分子および生体高分子、並びにそれらを模倣した人工高分子から選ばれる、請求項5記載の方法。
- 水溶性高分子がメチルセルロースまたは昇温型温度感受性ハイドロゲルである、請求項5記載の方法。
- 多能性幹細胞がES細胞またはiPS細胞である、請求項1〜7のいずれか1項に記載の方法。
- 多能性幹細胞がヒト由来である、請求項1〜8のいずれか1項に記載の方法。
