Description
本出願は、2010年6月21日に出願した米国特許出願公開第12/820,101号の利益を主張する。この出願の主題は引用により本明細書に組み込まれる。
基材をコーティングするために使用可能である材料を含むバチルス ステアロサーモフィラス(Bacillus stearothermophilus)由来の細菌中性プロテアーゼの製造。
生物汚染の調製及びコートされた基材への塗布。実験手順の典型的な略図を、図2に示す。60gの凍結乾燥コオロギを、ミキサー(Oster, 600 watt)で10分間切り刻んで粉末にする。2gのコオロギ粉末と6mlのDI水とを激しく混合することにより、汚染溶液を調製する。均一な間隔のテンプレートを使用して、コーティング表面に汚染を塗布する。コオロギ汚染を40℃で5分間乾燥し、続いてコーティングパネルをガラス皿に載せ、室温で様々な時間、300rpmで振盪しつつ200mlのDI水で洗浄する。汚染除去の時間を記録する。確率的誤差を低減するために、最初及び最後に除去された領域の時間を含めない。8つの汚染スポットの平均洗浄時間を、汚染除去時間として平均する。
乾燥時間は汚染除去時間に影響を与える。実施例1のようなコーティング及び実施例2のような虫汚染で調製された、汚染されたコートされた基材パネルを、様々な時間、40℃で乾燥させる。汚染領域の洗浄時間は、乾燥時間に強力に依拠する。対照のプロテアーゼを含まないコーティングでは、5分間乾燥した後、3時間洗浄により除去されない、強固に付着した汚染を生じる(表1)。
増加した洗浄強度は、汚染除去時間を低減する。実施例1及び2のように調製されたパネルを、様々な洗浄強度に供する。バチルス ステアロサーモフィラス(B. stearothermophilus)TLP含有コーティングをした基材に関して、洗浄時間の減少を、洗浄強度を増加することにより達成する(表3)。
様々な酵素を含むコーティングを、実施例1のように調製する。それぞれのコーティングについて、コートされた基材にコオロギ汚染を塗布し、40℃で5分間乾燥し、300rpmの強度で水中で洗浄する標準プロトコルを使用して、平均洗浄時間の測定により能力を分析する。対照及び様々なプロテアーゼを含むコーティングを、粗さ、接触角、及び光沢に関しても評価する。結果を表4に示す。
プロテアーゼの機能について表面加熱の影響。実施例1のようにバチルス ステアロサーモフィラス(B. stearothermophilus)TLPに基づくコーティングでコートされたパネルを、100℃で10日間焼成し、その後、表面酵素活性の変化を測定する。プロテアーゼ含有コーティングのタンパク質分解表面活性を、Folin and Ciocalteau, J. Biol. Chem., 1927; 73: 627-50の方法に従って測定する。この主題は、引用により本明細書に組み込まれる。簡潔には、1mlの2%(w/v)カゼイン含有リン酸ナトリウム緩衝溶液(0.05M、pH7.5)を、200μlの酢酸ナトリウム、5mM酢酸カルシウム(10mM、pH7.5)と一緒に基質として使用する。基質溶液を3分間ウォーターバスで予備インキュベートし、37℃に達する。反応を、バチルス ステアロサーモフィラス(B. stearothermophilus)TLPに基づくコーティング(1.2×1.9cm2)でコートされた試験プレートを1枚添加することにより開始し、続いて200rpmで10分間振盪し、その時点で1mlの110mMトリクロロ酢酸(TCA)溶液を添加することにより、反応を停止する。遠心分離前に、混合物を37℃で30分間インキュベートする。400μlのTCA可溶性画分中のチロシン等量を、200μlの25%(v/v)フォリン‐チオカルト(Folin-Ciocalteau)試薬、及び1mlの0.5M炭酸ナトリウムを使用して、660nmで測定する。1ユニットの活性は、37℃で1分あたり1.0μmolのチロシンに相当する吸光度を生じるカゼインを加水分解する酵素量として規定する。この結果をユニット/cm2に変換する。図6は、バチルス ステアロサーモフィラス(B. stearothermophilus)TLP表面活性が、長期間の高温焼成後、約50%低下したことを示す(図6A)。同時に、汚染洗浄時間も増加する(図6B)。
サーモリシン、並びにバチルス セレウス(Bacillis cereus)、ラクトバチルス種(Lactobacillis sp.)、バチルス メガテリウム(Bacillis megaterium)、アリシクロバチルス アシドカルダリウス(Alicyclobacillis acidocaldarious)、バチルス カルドリティカス(Bacillis caldolyticus)、バチルス サーモプロテオリティカス(Bacillis thermoproteolyticus)、バチルス ステアロサーモフィラス(Bacillus stearothermophilus)、バチルス サブチリス(Bacillus subtilis)、バチルス アミロリケファシエンス(Bacillus amyloliquefaciens)、及びリステリア モノサイトゲネス(Lysteria monocytogenes)由来のサーモリシン様タンパク質の0.2%(A)、2.0%(B)、4.0%(C)、6.0%(D)、及び8.0%(E)の酵素ロード濃度で、実施例1のように基材パネル上に調製され及びコートされたコーティングにおいて、実施例2のように塗布された虫汚染で、酵素ロードを測定する。パネルを40℃で5分間焼成し、3時間300rpmで洗浄する。増加したプロテアーゼロードは、増加した洗浄能力と相関する(図7A‐Eは、バチルス ステアロサーモフィラス(B. stearothermophilus)TLPの結果を示す)。
虫汚染除去に関する様々なプロテアーゼ種の比較。コーティングを、想定されるシステインプロテアーゼとしてプロテアーゼN(バシロリシン(bacillolysin))、想定されるセリンプロテアーゼとしてプロテインSD AY10(バチルス リケニフォルミス(Bacillus licheniformis)由来のスブチリシン(subtilisin))、典型的な金属プロテアーゼとしてプロテアーゼA、並びにサーモリシン及び実施例7のサーモリシン様タンパク質を使用して実施例1のように調製し、及び実施例2のように虫汚染を伴って実施例1のように基材上をコートする。異なる酵素を含むコーティングを、40℃で5分間焼成後、3時間300rpmで洗浄して比較した。驚くべきことに、サーモリシンに基づくコーティングのみが、セリンプロテアーゼ、システインプロテアーゼ、又は他の金属プロテアーゼを含むコーティングによっては観察されない改善された自己洗浄機能を示す(表5及び図8)。
実施例1のように試験パネルを調製し、試験自動車のフロントバンパー上に載せる。路上試験プロトコルの略図を、図9に示す。実際の虫を、運転により路上から採取する。自動車を夏の夕方の間、約500マイル運転し、虫の体を採取する。平均運転速度は、65mphである。
pH6.4及びpH11の緩衝液を使用して、実施例1のように酵素含有コーティングを調製する。コートされたアルミニウムプレートを、実施例9のように虫汚染に供する。両pHレベルで調製された酵素含有コーティングは、対照よりも優れている(図13)。
Claims (16)
- 基材又はコーティング上の生物汚染の除去を促進するための方法であって、
有機架橋性ポリマー樹脂から形成される基材又はコーティングを準備し、
サーモリシン又はサーモリシン様プロテアーゼが、生物汚染成分を酵素的に分解できるように、前記サーモリシン又はサーモリシン様プロテアーゼと前記基材又は前記コーティングとを結合させること
を含み、前記サーモリシン様プロテアーゼが、バチルス ステアロサーモフィラス(Bacillus stearothermophilus)由来の細菌中性サーモリシン様プロテアーゼである、方法。 - 前記サーモリシン又はサーモリシン様プロテアーゼが、300000ユニット/gを超える比活性を有する、請求項1に記載の方法。
- 前記基材又は前記コーティングの表面活性が、0.0075ユニット/cm2以上である、請求項1又は2に記載の方法。
- 前記サーモリシン又はサーモリシン様プロテアーゼが、前記基材又は前記コーティングと共有結合する、請求項1又は2に記載の方法。
- 前記サーモリシン又はサーモリシン様プロテアーゼが、前記基材若しくは前記コーティングと非共有結合し、又は前記基材若しくは前記コーティング内に混合される、請求項1又は2に記載の方法。
- 前記基材又は前記コーティングが、有機架橋性ポリマー樹脂を含む、請求項1又は2に記載の方法。
- 前記有機架橋性ポリマー樹脂が、アセトアセテート、酸、アミン、カルボキシル、エポキシ、ヒロドキシル、イソシアネート、シラン、ビニル、又はそれらの組み合わせの官能基を含む、請求項6に記載の方法。
- 前記有機架橋性ポリマー樹脂が、アミノプラスト、メラミンホルムアルデヒド、ポリウレタン、ポリアクリレート、エポキシ樹脂、ポリカーボネート、アルキド、ビニル樹脂、ポリアミド、ポリオレフィン、フェノール樹脂、ポリエステル、ポリシロキサン、又はそれらの組み合わせを含む、請求項6に記載の方法。

生物汚染成分を分解できるサーモリシン又はサーモリシン様プロテアーゼであって、前記基材又は前記コーティングと結合されている、前記サーモリシン又はサーモリシン様プロテアーゼ
を含み、前記サーモリシン様プロテアーゼが、バチルス ステアロサーモフィラス(Bacillus stearothermophilus)由来の細菌中性サーモリシン様プロテアーゼである、組成物。 - 前記サーモリシン又はサーモリシン様プロテアーゼが、300000ユニット/gを超える比活性を有する、請求項10に記載の組成物。
- 前記基材又は前記コーティングが、アセトアセテート、酸、アミン、カルボキシル、エポキシ、ヒロドキシル、イソシアネート、シラン、ビニル、又はそれらの組み合わせの官能基を有する有機架橋性ポリマー樹脂を含む、請求項10又は11に記載の組成物。
