Description
本出願は、引用をもってその全文をここに援用することとする、2007年8月3日出願の米国仮出願NO.60/953,789号に基づく優先権を主張するものである。
本発明は部分的にナショナル・インスティテュート・オブ・ダイアビーティス・アンド・ダイジェスティブ・アンド・キドゥニー・ディジージズ(NIDDK)により付与される5R43DK069727の番号による政府支援を受けてなされた。合衆国政府はここに提供する主題について何らかの権利を有するであろう。
本開示は、生物学的に活性なペプチドを含有する錯体の組成物、このような組成物の調合物及び使用法を提供するものである。部分的には、本発明は、生物学的に活性なペプチド及びたんぱく質を送達すると共にそのinvivoでの半減期を延長するように構成された、患者への投与用のペプチド錯体に関する。
例えば、本発明は以下を提供する:
(項目1)
一般式:A-(Cm)x-ペプチドを有する組成物であって、但し式中:
(a)前記ペプチドは100個以下のアミノ酸を有する生物学的に活性なペプチドであり;
(b)Cmは:
(i)Gly部分(この場合、x は0−6の整数である);
(ii)Ala部分(この場合、x は0−6の整数である);
(iii)Arg 部分(この場合、x は0−6の整数である);
(iv)Lys部分(この場合、x は0−6の整数である);

本開示は、生物学的に活性なペプチド錯体の組成物、患者の治療のためのこのような組成物の調合物及び使用法を提供する。本組成物と、同組成物の作成法及び使用法は数多くの所望の結果及び特徴を達成するであろう。ある局面では、本発明は、生物学的に活性なペプチドを、患者への投与後にコントロールされた態様で送達する、放出する、及び/又はその半減期を延長するように構成されたペプチド類似体の錯体及びポリマ担体分子に関する。別の局面では、本発明は、当該ペプチドを安定化させることで患者への投与に適した調合物となるペプチド類似体の錯体及びポリマ担体分子に関する。本開示は、疎水性及び/又は金属結合性の基の結合性部分を共有結合させて持つペプチド(「ペプチド類似体」)を提供するものであり、該ペプチドは、疎水性ポリマ担体のコアを含有するポリマ担体に、又は、共有結合させた金属結合ドメイン及びキレートさせた金属を持つポリマ担体に、可逆的に結合することで、患者への投与に適した錯体を形成することができる。更に本開示は、疎水性のポリマ担体のコアと一緒に、又は前記コアなしで、ミセル又はリポソームを形成することのできる、アルキル基を持つペプチドも提供するものである。更なる実施態様を挙げると、本ペプチドを、選択に応じて、生物学的流体又は組織中のホストプロテアーゼで切断することのできる切断可能なリンカー部分により疎水性又は金属結合性の基に共有結合させてもよく、それにより、送達用錯体からの活性ペプチドのコントロールされた及び/又は持続的な放出が可能になる。
本発明は、疎水性基又は金属結合性基などの結合性部分を含むように修飾することで、この結合性部分を通じてペプチドをポリマ担体分子に結合可能にしたペプチドを提供するものである。修飾には、本発明のペプチド類似体を作製するための疎水性部分又は金属結合性部分を含めることができる。例えば、疎水性基は6乃至36個の炭素原子を有するアルキルであってよい。金属結合性基は、例えばHisタグ(His6など)であってよい。いくつかの実施態様では、結合性部分は、送達しようとするペプチドの直接、付着している。他の実施態様では、結合性部分は、Gly-Gly-Glyなどのアミノ酸配列や以下に詳述する通りの切断が置き易いアミノ酸配列を含め、切断可能な部分を介してペプチドに付着している。このように、本発明の目的のためには、「ペプチド類似体」とは、共有結合による結合性部分を持つペプチドや、共有結合による切断可能部分及び共有結合による結合性部分を持つペプチドを意味する。切断可能部分が体内で切断すると、当該ペプチドはポリマ担体から放出される。このように、本発明のペプチド組成物は一般式:結合性部分−選択的な切断可能部分−ペプチド、を有し、この場合、前記結合性部分及び選択的な切断可能部分は、例えば共有結合により、当該ペプチドに、このペプチドのいずれかの適した位置で結合することができる。切断可能部分は、結合性部分がペプチドの生物学的活性と干渉する場合にその用途がある。
本発明の目的のためには、「ペプチド」とは、5乃至100個の範囲のアミノ酸の、生物学的に活性なポリアミノ酸を意味する。過去において、51個を越えるアミノ酸のポリアミノ酸はたんぱく質と呼ばれ、51個以下のアミノ酸より小さいものはペプチドと呼ばれた。たんぱく質とペプチドの間の区別は単に大きさの問題である。たんぱく質などの長いポリアミノ酸の合成は、過去、ペプチド合成装置の何らかの技術的限界が原因で課題が多かった。この困難の結果、ペプチドとたんぱく質の区別が生まれた。しかし、ペプチド合成技術の進歩により、最高で100アミノ酸までの長いポリペプチドの合成が今や可能である。また、技術により、5乃至100個のアミノ酸のペプチドの自動ペプチド合成装置で可能になり、ペプチドの古い定義が広くなったために、このようなポリペプチドもペプチドの定義に含めることも、本発明の意図である。大半の大型のたんぱく質は未だに生物系において組換え法で作製されているが、ペプチドはペプチド合成装置を用いて作製されている。アルキル基、ヒスチジン、ニトリロ三酢酸、又はイミノ二酢酸のペプチドへの添加は、ペプチド合成中にペプチド合成装置を用いて容易に行うことができる。組換えたんぱく質の場合、これらのアルキル基、ニトリロ三酢酸、又はイミノ二酢酸を成熟たんぱく質中に導入することができようが、たんぱく質からたんぱく質への反応予測性には限界がある。本発明の目的のための「ペプチド」には、ポリリジン、ポリグルタミン酸などのアミノ酸のホモポリマは、それらは生物学的に活性ではないために含まれない。本発明の目的のための用語「生物学的に活性な」とは細胞内で第二メッセンジャ系を通じて伝令を送り、そうして生物学的応答を引き起こすことのできる細胞受容体に、当該ペプチドが結合することができることを意味する。これらが、本明細書で概略したように修飾された場合の本発明のペプチドであるペプチド/たんぱく質である。

本開示は、ペプチド錯体を形成するためのポリマ担体への化学的な結合を可能にする実施態様のペプチド及びポリマ担体分子に付着させることのできる疎水性又は金属結合性基の結合性部分を提供するものである。
ある局面では、修飾されたペプチドの結合性部分は、例えば6乃至36個の炭素ユニットのアルキル基など、疎水性基である。このような疎水性基は、担体分子中の疎水性基に非共有結合的に結合させることができる。ある実施態様では、前記結合性部分のアルキル基は式CH3(CH2)n-CO-を有する直鎖状アルキルカルボニル基(但しこの場合、nは4乃至34の整数である)、又は、6乃至36個の炭素ユニットを持つ対応するアミノ−アルキル基(但しこの場合、各アルキル基は当該ペプチドのN−又はC−末端か、あるいは、当該ペプチドのアミノ酸の側鎖に付着させることができる。別の実施態様では、結合性部分のアルキル基はそれぞれ6乃至36の炭素ユニットを当該ペプチドのN又はC末端又は当該ペプチドのアミノ酸の側鎖に付着させた分枝状アルキルカルボニル基又は分枝状アミノ−アルキル基である。更に疎水性基は6乃至36個の炭素ユニットの環状化合物であってもよい。

本発明のポリマ担体には、直鎖状又は分枝状構造のポリマ及びコポリマ、あるいはその結合体、ミセル、乳濁液、コロイド及び固体表面が含まれ、その上この場合のポリマは少なくとも二種のポリマを含む超分子構造に自己集合してもよい。コポリマには、主なポリマ要素の一方として、疎水性結合性基又は金属結合性基としての骨格コアポリマと、そしていくつかの実施態様では、PEG又はmPEGなどの保護側鎖とが含まれる。
患者の投与に向け、本ペプチド錯体組成物は薬学的に許容可能な形で提供される。ある実施態様では、当該バソプレッシン類似体ペプチド又はその塩は、例えば0.22μmのフィルタなどを通して滅菌ろ過され、実質的には無発熱源である製剤として、薬学的に許容可能な賦形剤に入れて提供される。好ましくは、調合しようとするバソプレッシン類似体ペプチドが、HPLC上で単一又は個別化したピ−クで泳動し、その解析時には均一かつ真正なアミノ酸組成及び配列を示し、他の点では、薬学的製品の質を調節する多様な国立組織が設定した標準に合致するとよい。6乃至36炭素ユニットのアルキル基を持つ類似体はミセルを形成することができ、また適した溶媒に入れて投与することができる。代替的には、6乃至36炭素ユニットのアルキル基を持つ類似体を、米国特許出願第11/613,183号に解説されたものなどの疎水性のコアを持つ担体に導入することもできる。代替的には、当該のアルキル含有類似体をミセル又はリポソ−ムに取り入れてもよい。2乃至6個のヒスチジン残基を持つ類似体は、米国特許番号第7,138,105号に解説されたものなどの金属含有担体に取り入れることができる。これらの担体は更に、血液からの急速な除去が後に続くであろう、それらの急速な活性化を妨げることにより、これらの類似体の血液循環中半減期を延長するであろう。
ここで開示する新規な組成物は、提供するペプチド類似体及びポリマ担体の組合せや、患者の基礎疾患又は生理条件、及び/又は、分子タ−ゲット及びその位置に応じて、患者の処置法での使用に選択することができる。本ペプチド類似体組成物は、非経口、肺内、及び鼻腔内を含むいずれの適した手段又は経路によっても、そして必要に応じて、局所注射でも、投与することができる。非経口投与経路には、筋肉内、静脈内、動脈内、腹腔内、又は皮下投与がある。
実施例1
二つのシステイン残基(Cys4及びCys9)間をジスルフィド結合させたCH3(CH2)10CO-Gly-Gly-Gly-Cys-Tyr-Phe-Gln-Asn-Cys-Pro-Lys-Gly-NH2(SEQ ID NO: 34)(リジンバソプレッシン類似体であるC12TerA とも言及する)の合成が、カリフォルニア州サンホセのAnaspec社で本発明者の明細に従って行われた。上記の式の公知の標準的アミノ酸は当業者に公知の三文字略語で表されている(化学式全体については図2を参照されたい)。このリジンバソプレッシン類似体においては、ラウリン酸をペプチドのN末端アミノ基に、ラウリン酸のカルボキシル基と、ペプチドのグリシン残基のアミノ基との間に形成されるアミド結合を介して付着させた。上記の式の公知の標準的アミノ酸は当業者に公知の三文字略語で表されている。合成の最後に決定された分子量はプロトン付加されたペプチド、即ち質量分光解析での分子イオン・ピ−クに相当する1409.5Daであることが判明した。これはプロトン付加したときの理論上の分子量1410.4 Da と合致する。このペプチドは水溶性の限られた白色粉末であり、 この場合は90%を超える純度で容易に得られた。このペプチドは水中でミセルを形成することができ(図10を参照されたい)、それによりペプチドがバソプレッシンへの急速な活性化(表1を参照されたい)及び最終的な失活から保護される。図11及び12に見られるように、血清中で2時間、インキュベ−トした後では、バソプレッシン類似体(C12TerA) はヒト臍帯内皮細胞培養株でカルシウムの内向き流入を誘導することができ、生物学的活性の存在が示された。アルキル基(C12)をペプチドに加えると、天然のバソプレッシン(リジンバソプレッシン又はアルギニンバソプレッシン)又はテルリプレッシンなどの他の長期作用性類似体よりもその生物学的活性が長くなる(図1及び表1を参照されたい)。ミセルの形成に加え、このバソプレッシン類似体は(図17及び18を参照されたい)、引用をもってここに援用することとする米国特許出願番号第11/613,183号に解説されたものなどの疎水性基を含有するポリマ薬物担体に装薬することができる。

イミノ二酢酸 (IDA) 又はニトリロ三酢酸 (NTA) をC末端に付加的に含むGLP-1 類似体 SEQ ID NO: 2、SEQ ID NO: 3、SEQID NO: 4、及びSEQ ID NO: 5 の合成が、カリフォルニア州サンホセのAnaspec社により、本発明者の明細及び手法に従って行われた(付着は図20及び21に示す通りであり、この場合の当該ペプチドはSEQID NO: 2での通り、GLPである)。合成は、イミノ二酢酸又はニトリロ三酢酸誘導体 (遮断されたイプシロン・アミノ基を持つN’,N’,ビス(カルボキシメチル)-リジン)のワン樹脂への付着で開始され、続いて残りのカルボキシル基が遮断された。この合成の結果、以下の配列:

ヒト臍帯動脈内皮細胞(HUAEC)のバソプレッシン類似体刺激後のカルシウム内向きフラックスを用いて、多様なバソプレッシン類似体の生物学的活性を検査した。用いた材料は:マサチュ−セッツ州ベッドフォ−ド、BDBioSciences社、75 cm2組織培養フラスコ、(カタログ番号137787)に0.2 μm の通気プラグ・シ−ル・キャップ(カタログ番号353136)を付けたもの;メリ−ランド州ウォ−カ−ズビル、Lonza社、ヒト臍帯動脈内皮細胞(HUAEC) (カタログ番号 CC-2520); EGM-2 完全補充培地(カタログ番号CC-3162);カリフォルニア州カ−ルスバッド、Invitrogen/Molecular Probes社、Fura-2 AM(カタログ番号 F-1221)(Fura-2AM は、レシオメトリック及び紫外光惹起性の高アフィニティの細胞内カルシウム指標子である); アセトキシメチル (AM) エステル型は細胞透過性であるため、非浸潤性の細胞内装薬にとって有用である。細胞内にいったん入ると、このエステルが切り離されて、Fura−2分子が細胞非透過性となるため、中に閉じ込められる。);本発明者の明細に従ってカリフォルニア州サンホセのAnaspec社により注文作製されたテルリプレッシン誘導体;ミズ−リ州セントルイス、Sigma社、ヒト血清プ−ル(カタログ番号H4522);ニュ−ジャ−ジ−州フェアロ−ン、Fisher Scientific社製、リン酸緩衝生理食塩水(PBS) (カタログ番号 BP399-500);カリフォルニア州カ−ルスバッド、Invitrogen/Gibco社製、トリプシン-EDTA溶液、0.25% (カタログ番号25200-072);カリフォルニア州カ−ルスバッド、Invitrogen/Gibco社製、熱不活化ウシ胎児血清(FBS) (カタログ番号10438-026);ニュ−ヨ−ク州コ−ニング、黒ポリスチレン96ウェル・平底検定プレ−ト(カタログ番号 3916);ワシントンDC、Bioscan社により流通、フィンランド、ツルク、Hidek社製、カメレオン・プレ−ト・マルチラベル検出リ−ダ−;ニュ−ジャ−ジ−州フェアロ−ン、FisherScientific 社製、ジメチルスルホキシド (DMSO)(カタログ番号 D-136-1)、である。
ラットの耳モデルを用い、本発明の金属結合性部分を持つバソプレッシン類似体 His6Terを、長時間のその血管収縮維持能について評価した。該類似体は、40PLPEG537-Zn-キレ−トを含有するポリマ担体に2%の装薬で調合された。該ペプチド類似体−ポリマ錯体を一組のラットの背中に皮下注射し、テルリプレッシンのみも別の組のラットの同様に注射した。耳を基線及び間隔を置きながら最高48時間まで調べた。ポリマ担体に入れたペプチド類似体を注射されたラットの耳は、公知の長期作用性類似体テルリプレッシンよりも長い薬理学的効果を有しており、それは、His6Ter類似体プラス・ポリマ担体で処理されたラットの耳は6時間目及び24時間の時点の両方で青白いままだった一方、未調合のテルリプレッシン処理ラットの耳は、6時間目では青白かったが、24時間目では肉眼でも明白なほど正常な血流のある基線色に戻っていたことが見出されたことでも実証された。
当業者であれば、ごく慣例的な実験を用いるのみで、ここに解説された本発明の具体的な実施態様の均等物を数多く、認識し、又は確認できることであろう。このような均等物は以下の請求項の包含するところと意図されている。
Claims (26)
- 一般式:A−(Cm)x−ペプチドを有するペプチド類似体であって、式中:
a.Cmはグリシン、アラニン、リジンおよびアルギニンのうちのいずれか1つであり;
b.xは2乃至6の整数であり:
c.Aは、−OCH(CH3)CO−リンカーによって−(Cm)x−ペプチドに付着された、6乃至36個の炭素ユニットを有するアルキル基を含む化学基又は部分であり;そして
d.前記A−(Cm)x−ペプチドは、血清の非存在下、細胞培養において前記A−(Cm)xを持たない対応するペプチドよりも低い生物学的活性を有し、ここで、前記ペプチドは、バソプレッシン、テルリプレッシン、グルカゴン様ペプチド(GLP)、レプチンフラグメント、胃抑制ポリペプチド(GIP)、上皮成長因子(EGF)受容体リガンド、EGF、トランスフォーミング成長因子アルファ(TGF−アルファ)、ガストリン/コレシストキニン受容体リガンド、ガストリン、コレシストキニン、アウリスタチン、ナイシン、インシュリン、インシュリン様成長因子、副甲状腺ホルモン(PTH)、心房性ナトリウム利尿因子、ソマトスタチン、ゴナドトロピン放出ホルモン、黄体ホルモン放出ホルモン、および血管作用性腸管ペプチド(VIP)から選択される、
ペプチド類似体。 - 請求項1に記載のペプチド類似体と、各々が8乃至36個の炭素の複数の疎水性基を持つポリマ担体とを含む組成物であって、前記ペプチド類似体の基Aは、疎水性相互作用によって前記ポリマ担体の複数の疎水性基に非共有結合され、前記ポリマ担体は、ポリアミノ酸、ポリエチレングリコールおよびポリカルボン酸からなる群より選択される、組成物。
- Aが直鎖状アルキルである、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- Aが分枝状アルキルである、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 前記ペプチドがGLP−1であり、A−(Cm)x−が前記ペプチド又は側鎖のC末端に付着される、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- A−(Cm)xが前記ペプチドに対してN末端において付着される、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 前記(Cm)x基が-Gly-Gly-Gly(SEQ ID NO: 58)である、請求項1に記載のペプチド
類似体または請求項2に記載の組成物。 - xが3乃至6の整数である、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 請求項1、3および4のいずれか1項に記載のペプチド類似体または請求項2〜4のいずれか1項に記載の組成物であって、前記ペプチドがバソプレッシンであり、ここで:
(a)バソプレッシンが一般式:Cys-Tyr-Phe-Gln-Asn-Cys-Pro-B-C-D(SEQ ID NO: 53)を含み、式中:
Cysがシスチン又はシステインであり;
Tyrがチロシンであり;
Pheがフェニルアラニンであり:
Glnがグルタミンであり;
Asnがアスパラギンであり;
Proがプロリンであり;
Bがリジン又はアルギニンであり;
Cがグリシン又はアラニンであり;そして
DがNH2であり;そして
(b)CmがGly部分である、
ペプチド類似体または組成物。 - Aが8乃至12個の炭素を有するアルキル基を含む化学基又は部分である、請求項9に記載のペプチド類似体または組成物。
- 前記ペプチド類似体が式:CH3(CH2)10CO-OCH(CH3)CO-Gly-Gly-Gly-Cys-Tyr-Phe-Gln-Asn-Cys-Pro-Lys-Gly-NH2(SEQIDNO:35)である、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 前記ペプチド類似体が式:CH3(CH2)6CO-OCH(CH3)CO-Gly-Gly-Gly-Cys-Tyr-Phe-Gln-Asn-Cys-Pro-Lys-Gly-NH2(SEQIDNO:37)である、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 前記ペプチド類似体が式:CH3(CH2)10CO-OCH(CH3)CO-Gly-Gly-Gly-Cys-Tyr-Phe-Gln-Asn-Cys-Pro-Arg-Gly-NH2(SEQIDNO:79)である、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 前記ペプチド類似体が式:CH3(CH2)6CO-OCH(CH3)CO-Gly-Gly-Gly-Cys-Tyr-Phe-Gln-Asn-Cys-Pro-Arg-Gly-NH2(SEQIDNO:80)である、請求項1に記載のペプチド類似体または請求項2に記載の組成物。
- 薬学的に許容可能な担体中に、請求項1および3〜14のいずれか1項に記載のペプチド類似体または請求項2〜14のいずれか1項に記載の組成物を含む、薬学的組成物。
- ペプチド類似体を含む組成物の作製方法であって、
樹脂中のペプチドと基A−(Cm)x−(反応性基)との間に共有結合を形成させて、請求項1に記載のペプチド類似体を含む組成物に対する樹脂に結合された前駆体を形成する工程であって、ここで、前記ペプチド類似体が、血清の非存在下、細胞培養において前記A−(Cm)xを持たない対応するペプチドよりも低い生物学的活性を有する、工程
を含む、方法。 - 請求項5に記載のペプチド類似体または組成物を含む、糖尿病の処置のための治療薬。
- 請求項1および3〜15のいずれか1項に記載のペプチド類似体または請求項2〜15のいずれか1項に記載の組成物を含む治療薬であって、注射又は注入による投与に適している、治療薬。
- 請求項9〜14のいずれか1項に記載のペプチド類似体または組成物を含む、血液量減少、内臓血管拡張、全身性血管拡張、低血圧、食道静脈瘤出血、肝腎障害症候群、又は敗血症の処置のための治療薬。
- 血液量減少又は低血圧の処置のための、請求項19に記載の治療薬。
- 穿刺術を原因とする低血圧の処置のための、請求項19に記載の治療薬。
- 食道静脈瘤出血の処置のための、請求項19に記載の治療薬。
- 肝腎障害症候群の処置のための、請求項19に記載の治療薬。
- 敗血症の処置のための、請求項19に記載の治療薬。
- 肝硬変により引き起こされる肝腎障害症候群の処置のための、請求項23に記載の治療薬。
- 前記肝硬変がアルコール又は肝炎により引き起こされる、請求項25に記載の治療薬。
