他の実施形態においては、本発明は、本明細書に記載の単子葉植物科の植物に由来するプラスチドACCaseまたはその一部を包含する。
本明細書で用いられる「耐性」または「除草剤耐性」は、通常は非耐性(例えば、野生型)の植物またはその一部においては成長阻害を引き起こす量の除草剤の存在下で成長することができる植物またはその一部を示す。通常は非耐性植物の成長を阻害する除草剤のレベルは公知であり、当業者であれば容易に決定することができる。例としては、適用のために製造業者により推奨される量が挙げられる。最大速度は通常は非耐性植物の成長を阻害するであろう量の除草剤の例である。
本発明は、イネ科の除草剤耐性単子葉植物を提供する。イネ科を2つの主要なクレード:タケ亜科、エールハルタ亜科、およびイチゴツナギ(Pooideae)亜科を含むクレード(BEPクレード)と、キビ(Panicoideae)亜科、ダンチク(Arundinoideae)亜科、ヒゲシバ(Chloridoideae)亜科、ラッパグサ(Centothecoideae)亜科、ミクライロイデア(Micrairoideae)、アリスチドイデア(Aristidoideae)、およびダントニオイデア(Danthonioideae)を含むクレード(PACCMADクレード)に分けることができる。タケ亜科は、オリザ族を含む。本発明は、BEPクレードの植物、特に、タケ亜科およびエールハルタ亜科の植物に関する。本発明の植物は、典型的には、以下に記載の本発明のアセチル-コエンザイムAカルボキシラーゼ酵素を発現する結果として、アセチル-コエンザイムAカルボキシラーゼ活性を阻害する少なくとも1種の除草剤に対して耐性である。BETクレードは、タケ亜科、エールハルタ亜科、およびコムギ類(Triticodae)を含み、他のイチゴツナギ亜科群を含まない。BET作物は、BETサブクレードのメンバーである食物または飼料のために成長させた植物、例えば、オオムギ、トウモロコシなどである。
本発明は、対応する野生型植物中に認められるアセチル-コエンザイムAカルボキシラーゼ酵素のアミノ酸配列とは異なるアミノ酸配列を有するアセチル-コエンザイムAカルボキシラーゼ酵素を発現する植物を提供する。理解を容易にするために、本明細書で用いられるアミノ酸番号方式は、アロペクルス・ミオスロイデス[Huds.](ブラックグラスとも呼ばれる)に由来するアセチル-コエンザイムAカルボキシラーゼに用いられる番号方式であろう。アロペクルス・ミオスロイデスのアセチル-コエンザイムAカルボキシラーゼをコードするmRNA配列は、GenBankアクセッション番号AJ310767で入手可能であり、そのタンパク質配列はGenBankアクセッション番号CAC84161で入手可能であり、それらは特にいずれも参照により本明細書に組入れられるものとする。言及されるアミノ酸の番号の後に(Am)を付けて、アミノ酸が対応するアロペクルス・ミオスロイデス配列中のアミノ酸を示す。図18は、アロペクルス・ミオスロイデスのアセチル-コエンザイムAカルボキシラーゼのアミノ酸配列を提供する(GenBankアクセッション番号CAC84161)。本発明のアセチル-コエンザイムAカルボキシラーゼ中で変化してもよいアミノ酸を太字の下線付きで示し、図19は、アロペクルス・ミオスロイデスのアセチル-コエンザイムAカルボキシラーゼと整列させた野生型オリザ・サティバのアセチル-コエンザイムAカルボキシラーゼのアミノ酸配列を表し、いくつかの重要な残基を示している。

T0(「Tゼロ」)再生個体である前記イネ植物と、
それと平行して、そのような植物の対照集団を未処理の基準植物として用いる;
(2)テプラロキシジム(AI)および1%作物油濃縮物(COC)を含む組成物を200 L/haの噴霧容量で前記集団に適用して、50 g/haのテプラロキシジム(AI)に等しいAI適用率を提供する;
(3)伝統的な植物損傷評価体系(例えば、好ましくは、典型的な、少なくとも5段階の損傷評価尺度に従う、除草剤による日焼け、葉の形態変化、しおれ、黄変、および他の形態学的特徴の視覚的証拠の評価)に基づいて、それぞれの試験植物および基準植物に関する植物毒性スコアを決定する;
(4)収集されたデータを分析して、試験集団中の少なくとも75%の植物が平均植物毒性、すなわち、基準植物と比較して10%未満の損傷の増加を示すかどうかを決定する;ならびに
(5)そのように決定された陽性の結果を、二重突然変異ACCaseが許容可能なAITを提供することを証明するものとして同定する。
本発明は、通常は野生型植物の成長を阻害する濃度の除草剤に対して耐性である植物、例えば、イネ植物を提供する。前記植物は、典型的には、アセチル-コエンザイムAカルボキシラーゼ活性を阻害する除草剤に対して抵抗性である。アセチル-コエンザイムAカルボキシラーゼ活性を阻害する任意の除草剤を、本発明の植物と共に用いることができる。好適な例としては、限定されるものではないが、シクロヘキサンジオン除草剤、アリールオキシフェノキシプロピオナート除草剤、およびフェニルピラゾール除草剤が挙げられる。雑草を制御し、および/または除草剤耐性植物を成長させるいくつかの方法においては、少なくとも1種の除草剤を、セトキシジム、シクロキシジム、テプラロキシジム、ハロキシホップ、ハロキシホップ-Pまたはこれらの除草剤のいずれかの誘導体からなる群より選択する。表1は、アセチル-コエンザイムAカルボキシラーゼ活性を阻害し、本発明の除草剤耐性植物と共に用いることができるシクロヘキサンジオン除草剤(DIM、シクロヘキセンオキシムシクロヘキサンジオンオキシム除草剤;およびCHDとも呼ばれる)の一覧を提供する。当業者であれば、このクラスに他の除草剤が存在し、本発明の除草剤耐性植物と共に用いることができることを認識するであろう。また、アセチル-コエンザイムAカルボキシラーゼ活性を阻害し、本発明の除草剤耐性植物と共に用いることができるアリールオキシフェノキシプロピオナート除草剤(アリールオキシフェノキシプロパノエート;アリールオキシフェノキシアルカノエート;オキシフェノキシ;APP;AOPP;APA;APPA;FOPとも呼ばれる;これらのものは接尾辞「-oic」を用いて書かれることもあることに留意されたい)の一覧も表1に含まれる。当業者であれば、このクラスに他の除草剤が存在し、本発明の除草剤耐性植物と共に用いることができることを認識するであろう。
本発明の除草剤耐性植物を、それらが耐性である除草剤と共に用いることができる。当業者には公知の任意の技術を用いて、本発明の植物に除草剤を適用することができる。植物の栽培プロセスにおける任意の時点で除草剤を適用することができる。例えば、除草剤を、植える前に、植える時に、発芽前に、発芽後に、またはその組合せで適用することができる。
除草剤耐性作物は、雑草管理のためのさらなる選択肢を農業従事者に与える。現在、いくつかの作物系において、遺伝的に改変された(GMO)溶液が利用可能である。BASF社製の現在のCLEARFIELD溶液などの除草剤抵抗性を付与する変化した酵素、活性または構造を選択するために、さらなる突然変異技術が用いられてきた。米国では、CLEARFIELD米は被害地域において赤米を管理するための第一のツールである(USDA-ARS, 2006);しかしながら、1ヘクタールあたり最大170個のF1ハイブリッドで異系交配が報告されているため(Shivrainら、2007)、赤米とCLEARFIELD米との遺伝子流動は、AHAS耐性にとってかなり危険である。多くの他の側面、非CLEARFIELD米の変化を含む管理指針は、CLEARFIELD米の市場浸透を制限し得る。様々な作用様式(MOA)のイネ科雑草防除剤(graminicide)に対する耐性を有する栽培米の作製は、これらの危険性を低下させ、雑草管理のためのより多くのツールを提供するであろう。
in vitro組織培養突然変異誘発アッセイを開発して、アセチル-コエンザイムAカルボキシラーゼ阻害除草剤、例えば、テプラロキシジム、シクロキシジム、およびセトキシジムに対して耐性である植物組織(例えば、イネ組織)を単離および特性評価した。このアッセイは、in vitro組織培養物中に認められるソマクローナル変異を利用するものである。ソマクローナル変異から誘導される自然突然変異を、化学的突然変異誘発および高濃度の除草剤上での段階的様式でのその後の選択により増強することができる。
一度、組織培養条件を決定したら、シクロキシジム、テプラロキシジム、セトキシジム(図1)またはハロキシホップ(示さない)を用いる殺菌曲線における組織生存の分析により、さらなる選択条件を確立した。組織中の除草剤の蓄積、ならびに細胞および培養培地中でのその持続性および安定性を注意深く考慮した。これらの実験を通じて、突然変異した物質の初期選択に関する致死未満量を確立した。
耐性組織を再生させ、ACCase遺伝子配列突然変異について分子的に、および/または選択剤の存在下でのACCase活性の変化について生化学的に特性評価した。
クローン植物から葉組織を採集し、移植のために分離し、個別に分析した。製造業者により指示されるようにWizard(登録商標)96 Magnetic DNA Plant Systemキット(Promega、米国特許第6,027,945号および第6,368,800号)を用いて、ゲノムDNAを抽出した。1個のフォワードプライマーおよび1個のリバースプライマーを用いて、単離されたDNAをPCR増幅した。

選択された突然変異体およびエスケープを小さいポットに移した。野生型栽培品種および赤米の3種の次亜種を、種子から発芽させて、対照として供給した。
培地を使用のために選択し、殺菌曲線を上記で特定されたように生じさせた。選択のために、様々な技術を用いた。段階的選択を適用するか、または即時致死量の除草剤を適用した。いずれの場合でも、全てのカルスをそれぞれの新しい周回の選択のために移した。選択は、各サイクルにつき、3〜5週間で4〜5サイクルの培養であった。カルスをナイロン膜上に置いて、移動を容易にした(200ミクロンの孔径のシート、Biodesign, Saco, Maine)。膜を切って100 x 20 mmのペトリ皿に合わせ、使用前にオートクレーブした後、プレート毎に25〜35個のカルス(カルス1個あたりの平均重量は22 mgである)を用いた。さらに、1セットのカルスを液体培養培地中での選択にかけて、毎週二次培養した後、半固体培地上でさらに選択した。
方法:
特定されたように培地交換を行うことを除いて(詳細については付属の培地表を参照されたい)、本質的にはHieiおよびKomari(2008)に記載のように、インディカ1およびニッポンバレイネカルスの形質転換を実行した。形質転換における使用の前に、カルスをR001M培地上で4〜8週間誘導した。用いたアグロバクテリウムは、選択のためにRLM185(L. Mankin、未刊行:選択のためにDsRedおよび突然変異AHASを含む)、I1781(Am)Lを含有するACC遺伝子、I1781(Am)LおよびW2027Cを含有するACC遺伝子、I1781(Am)LおよびI2041(Am)Nを含有するACC遺伝子、またはI17181(Am)Aもしくは突然変異AHAS遺伝子をも含む野生型を含有するACC遺伝子で形質転換されたLBA4404(pSB1)(Ishidaら、1996)であった。固体培地上で1〜3日間増殖させたアグロバクテリウムをM-LS-002培地中に懸濁し、OD600を約0.1に調整した。カルスをアグロバクテリウム溶液中に約30分間含浸させた。液体を除去した後、半固形イネcc培地上での共培養のためにカルスを濾紙に移した。共培養は暗室中、24℃で3日間であった。イネカルスを含有する濾紙を、回収のためにチメンチンを含有するR001M培地に1〜2週間直接移し、暗室中、30℃で培養した。チメンチンを含み、100μMイマゼタピル、10μMシクロキシジムまたは2.5μMテプラロキシジムを補給した新鮮なR001M培地上にカルスを再分割した。3〜4週間後、カルスを新鮮な選択培地に移した。さらに3〜4週間後、増殖しているカルスを新鮮な培地に移し、増殖させた後、Taqman分析を行った。Taqman分析は、Nosターミネーターに関するものであり、選択されたカルスのトランスジェニック性質の分子的確認を提供するために行った。10μMシクロキシジムまたはハロキシホップ、2.5μMテプラロキシジムまたは100μMイマゼタピルを含有する培地上でカルスを二次培養することにより、トランスジェニックカルスの増殖を様々な選択剤を用いて測定した。最初の二次培養後、次いで、約1ヶ月の増殖後、カルスのサイズを走査画像から測定した。
I1781(Am)LおよびW2027(Am)Cを含有するACC遺伝子、ならびにI1781(AM)LおよびI2041(Am)Nを含有するACC遺伝子を用いるインディカ1イネ形質転換実験から、トランスジェニックカルスを取得した。テプラロキシジム選択後にI1781(Am)LおよびW2047(Am)Cを含有するACC遺伝子から1個のカルスが得られ、I1781(Am)LおよびI2041(Am)Nを含有するACC遺伝子から3個のカルスが得られた。シクロキシジム選択を用いる場合、I1781(Am)LおよびI2041(Am)Nを含有するACC遺伝子から1個のカルスが得られた。Nos Taqmanにより、これらのカルスは全部トランスジェニックであることが示された。突然変異AHAS選択マーカーのためのイマゼタピル(Pursuit-P)などの様々な選択剤の下で、カルスを増殖についてスクリーニングした。

- 前記ACCase-阻害性除草剤が、アロキシジム、ブトロキシジム、クレトジム、クロプロキシジム、シクロキシジム、セトキシジム、テプラロキシジム、トラルコキシジム、クロラジホップ、クロジナホップ、クロホップ、ジクロホップ、フェノキサプロプ、フェノキサプロプ-P、フェンチアプロプ、フルアジホップ、フルアジホップ-P、ハロキシホップ、ハロキシホップ-P、イソキサピリホップ、プロパキザホップ、キザロホップ、キザロホップ-P、キザロホップ-P-エチル、キザロホップ-P-テフリル、トリホップ、ピノキサデン、又はこれらの農学上許容し得る塩若しくはエステルの少なくとも1つを含む、請求項13に記載の種子。
- 耕地における雑草を制御する方法であって、
a. 請求項1〜6のいずれか1項に記載の植物を耕地で生育させること、及び
b. 耕地の植物及び雑草にアセチル-コエンザイムAカルボキシラーゼ-阻害性除草剤を対応する野生型植物の成長を阻害する量で適用すること、
を含み、それにより雑草を制御する方法。 - イネ植物を生育する方法であって、
a. 請求項1〜6のいずれか1項に記載の植物又は請求項9〜11のいずれか1項に記載のイネ種子を得ること、及び
b. 1つ以上のアセチル-コエンザイムAカルボキシラーゼ-阻害性除草剤を対応する野生型植物の成長を阻害する量で植物及び前記種子から成長する植物に適用すること
を含む、方法。 - 前記アセチル-コエンザイムAカルボキシラーゼ-阻害性除草剤が、アロキシジム、ブトロキシジム、クレトジム、クロプロキシジム、シクロキシジム、セトキシジム、テプラロキシジム、トラルコキシジム、クロラジホップ、クロジナホップ、クロホップ、ジクロホップ、フェノキサプロプ、フェノキサプロプ-P、フェンチアプロプ、フルアジホップ、フルアジホップ-P、ハロキシホップ、ハロキシホップ-P、イソキサピリホップ、プロパキザホップ、キザロホップ、キザロホップ-P、キザロホップ-P-エチル、キザロホップ-P-テフリル、トリホップ、ピノキサデン、又はこれらの農学上許容し得る塩若しくはエステルの少なくとも1つを含む、請求項15又は16に記載の方法。
- 請求項1〜6のいずれか1項に記載の植物、又は請求項9〜11のいずれか1項に記載の種子から調製された食品。
- 請求項1〜6のいずれか1項に記載の植物、又は請求項9〜11のいずれか1項に記載の種子から調製された消費者製品。
- 請求項1〜6のいずれか1項に記載の植物、又は請求項9〜11のいずれか1項に記載の種子から調製された工業製品。
- 請求項1〜6のいずれか1項に記載の植物、又は請求項9〜11のいずれか1項に記載の種子から調製された動物用製品。
- 配列番号2のアミノ酸配列を有し、かつ該配列番号2におけるアロペクルス・ミオスロイデス(Alopecurus myosuroides(Am))のプラスチドアセチル-コエンザイムAカルボキシラーゼ(ACCase)の位置1,781に対応するアミノ酸位置でのイソロイシンからロイシンへの置換を有するイネプラスチドACCaseをコードするACCase核酸分子であって、イネプラスチドACCaseが100 g ai/haのシクロキシジムに対する耐性をイネ植物に付与するACCase核酸分子。
- 前記プラスチドACCaseが、アロペクルス・ミオスロイデス(Am)のプラスチドACCaseの位置に対応する位置1,785、1,786、1,811、1,864、2,027、2,039、2,041、2,049、2,074、2,075、2,080、2,088、2,095、2,096又は2,098でのアミノ酸置換、位置2,075(Am)でのバリンの重複、又は位置2,080(Am)でのアミノ酸の欠失をさらに含む、請求項22に記載の核酸分子。
- 前記核酸分子が、
i. 位置1,785(Am)でのグリシン置換;
ii. 位置1,786(Am)でのプロリン置換;
iii. 位置1,811(Am)でのアスパラギン置換;
iv. 位置2,039(Am)でのグリシン置換;
v. 位置2,049(Am)でのイソロイシン、ロイシン、バリン、又はフェニルアラニン置換;
vi. 位置2,074(Am)でのロイシン置換;
vii. 位置2,075(Am)でのロイシン、イソロイシン、又はメチオニン置換;
viii. 位置2,075(Am)でのバリンの重複;
ix. 位置2,078(Am)でのトレオニン、グリシン又はリジン置換;
x. 位置2,080(Am)でのグルタミン酸置換;
xi. 位置2,088(Am)でのアルギニン、トリプトファン、フェニルアラニン、グリシン、ヒスチジン、リジン、ロイシン、セリン、トレオニン、又はバリン置換;

a. 植物又は種子から生体材料を得ること、
b. 前記生体材料中のACCase遺伝子のPCR又はハイブリダイゼーション試験を実施し、生体材料が請求項1〜6及び9〜11のいずれか1項に記載されるACCase核酸を含むかどうか決定すること、及び
c. ステップ(b)の結果に基づいて、ステップ(a)の植物が請求項1〜6及び9〜11のいずれか1項に記載される核酸を含むことを確認すること
を含む、方法。
