本発明の抗体をペプチジルおよび非ペプチジルの双方のカップリングパートナーにカップリングする適切な技術が、当業者に既知である。適切な標識の例としては、放射性標識等の検出可能な標識、西洋ワサビペルオキシダーゼ等の酵素標識またはビオチン等の化学部分を含む。或いは、標識は機能標識、例えばリシンまたはプロドラッグを活性ドラッグに抗体結合部位で変換することができるプロドラッグであってもよい。

本発明の前記態様のある実施形態において、本発明は、抗体断片または抗体の抗原結合部位を提供する。結合断片の例は、ヘテロ四量体抗体VL,VH,CLおよびCH1領域からなるか、実質的にそれからなる一価断片のFab、Fab’およびF(ab’)2断片を含む。ある実施形態において、VL領域は配列番号:1のアミノ酸配列を有していて、VH領域は配列番号:4のアミノ酸配列を有する。ある実施形態において、CLおよびCH1領域は、免疫グロブリンのサブクラスIgG、およびアイソタイプIgG4のCL、並びにCH1領域のアミノ酸配列に基づくものであるか、図5の配列番号:5に示すようなCLおよびCH1領域である。ある実施形態において、本発明のこの実施形態のFab断片は、ブドウ膜炎およびAMD(加齢性黄斑変性症)を挙げる乾癬、皮膚炎および眼疾患(ただし、これらに制限されない)の治療または予防に使用することができる。
TLR2に特異的な結合を有し、TLR2機能に拮抗するヒト化モノクローナル抗体を提供するのに加えて、本発明は、さらに配列番号:1に規定するようなVL(軽鎖可変)領域のアミノ酸配列および配列番号:4に規定するようなVH(重鎖可変)領域のアミノ酸配列に基づく結合領域の一組を含む抗体以外の結合メンバーも包含する。特に、本発明は、本発明の抗体のヒト化抗体のVLまたはVH領域のいずれかに基づく単一結合領域にまでも及ぶ。
従来技術における当業者であれば、相補領域の配列(配列番号:7,8,9,10,11および12)を理解し、並びに、超可変および可変領域の配列は、TLR2特異的に結合する性能を欠失することなく修飾することができるであろう。例えば、本発明のヒト化抗体由来の結合メンバーのCDR領域は、ここに規定した配列番号:7,8,9,10,11および12のアミノ酸配列と相同性があるか、非常に高い相同性を有する(例えば95%以上の同一な配列)。ここに規定する「非常に相同性がある」の用語は、1から5アミノ酸の置換が、CDRの配列にされることを意味する。ある実施形態において、個々のCDR,超可変領域または可変領域およびそれらの非修飾対応物に存在する相同性は、少なくとも80%、好ましくは90%、より好ましくは少なくとも95%、および最も好ましくは98%を超える。かかる修飾配列は、本発明の範囲内に分類され、例えば修飾したか、相同する欠乏メンバーは、TLR2に特異的に結合する能力を保持し、その機能活性に拮抗する。
また、様々な態様において、本発明は、本発明のヒト化抗体、抗体断片および結合メンバーをコード化するポリヌクレオチド、特に単離されたポリヌクレオチドも包含する。
本発明のさらに他の態様は、配列番号:1のアミノ酸配列の重鎖可変領域および配列番号:4の軽鎖の可変アミノ酸配列を有する抗体を発現するハイブリッド細胞株(ハイブリドーマ)を提供する。
本発明のヒト化抗体もしくはその断片またはその結合メンバーは、Toll様受容体2に選択的に結合し、その機能に拮抗する。受容体のリガンド結合部位付近のそのリガンドによる結合によって、モノクローナル抗体は、受容体の活性をブロックすることができ、これは、リガンド結合部位にリガンドを接近させることを排除する立体障害を結果として生じる。
通常、本発明の抗体、抗体断片または結合メンバーによって拮抗されるTLR2は、哺乳類TLR2(またはその機能的変異体)、例えばヒトTLR2またはネズミTLR2である。ある実施形態にいて、拮抗したTLR2は、図7の配列番号:15に規定するようなアミノ酸配列を有するヒトの形状のTLR2であり、これは、Genbankアクセス番号AAC 34133 (URL www.ncbi.nlm.nih.gov)に規定するような784アミノ酸の全長ヒトToll様受容体配列を含む。
本発明の抗体および抗体断片は、完全なヒトのものである。即ち、抗体を含むアミノ酸残基は、例えばマウスと対象的なヒト由来のものである。したがって、本発明の抗体は、ネズミ抗体またはキメラ抗体、例えばマウス/ヒト抗体より低い免疫性がある。
本発明のヒト化抗体、断片および結合メンバーは、免疫抑制(免疫反応、最も特に、炎症性免疫反応)、特にToll様受容体2を介在した活性およびシグナリングを抑制することによって誘発することができる。Toll様受容体2のこの抑制は、疾患状態の治療または予防に有用性を有するとして、Toll様受容体2の活性化およびシグナリングが、疾患の発症または進行の原因となることが、発明者に同定された。

発明者は、ここに提供するヒト化抗TLR2抗体が、技術分野に既知の抗TLR2ネズミモノクローナル抗体より、治療的に好ましいことを確認した。
「抗体」は、天然または部分的もしくは完全に合成して産生した免疫グロブリンである。この用語は、結合領域を有するあらゆるポリペプチド、タンパク質またはペプチドの範囲を包含し、即ち本発明のヒト化抗体の結合領域に相合性がある。
本発明は、さらに本発明の抗体またはその結合断片に基づくペプチド模倣薬をも包含する。ここに使用する「ペプチド模倣薬」または「ペプチド模倣剤」の用語は、ポリペプチドでないが、それらの構造的態様を模倣し、本発明の拮抗するToll様受容体2の機能活性を生じる方法で、Toll様受容体に特異的に結合する分子を示す。それゆえに、本発明は、Toll様受容体2拮抗剤として働く本発明の抗体に基づくペプチド模倣薬をも包含する。
本発明の抗体および結合メンバーは、完全にまたは部分的に化学合成によって生成することができる。例えば、本発明の抗体および結合メンバーは、当業者に周知の技術、例えば標準の液体ペプチド合成または固相ペプチド合成法によって調製することができる。或いは、抗体および結合メンバーは、溶液中に液相ペプチド合成技術または具体的には固相、液相および溶液化学の組み合わせを用いて調製することができる。
本発明の完全なヒト化抗体、それに由来する結合メンバーまたはそれをコード化するポリペプチドに関するものである場合、「単離された」の用語は、前記抗体、結合メンバーまたは核酸(ポリヌクレオチド)が単離されたおよび/または精製された形状にあることを示し、即ち、それらが自然環境で別々にされ、単離されまたは精製され、実質的に精製されたか、異種形状であるかまたは核酸の場合、必要とされる機能を有するポリペプチドをコード化する配列以外の原点となる核酸もしくは遺伝子を含まないか、実質的にそれを含まない。したがって、このように単離した抗体、結合メンバーおよび単離した核酸は、前記調製が実験室内(in vitro)または生体内(in vivo)で実施したDNA技術によって調製された場合、それらが自然に関連する物質、例えばそれらの自然環境またはそれらが調製される環境で(例えば細胞培養)みられる他のポリペプチドまたは核酸を含まないか、実質的にそれを含まない。
本発明のモノクローナル抗体または結合メンバーは、単独で投与することができるが、好ましくは、意図する経路による投与に依存して選択される、適切な薬学的に許容可能な賦形剤、希釈剤またはキャリアを一般的に含む医薬組成物として投与される。適切な薬学的なキャリアの例としては:水、グリセロール、エタノール等を含む。
他に定められない限り、ここに使用される用語の全ての技術および科学用語は、本発明の分野の当業者に一般的に理解できる意味を有する。
本発明のTLR2拮抗抗体は、通常、完全なヒト化抗体である。即ち、抗体は、完全にヒト起源のものであるため、非ヒト類、例えばマウス由来の領域またはアミノ酸の組み合わせを含まない。
例えば相補性決定領域(CDR)を同定するため、Toll様受容体2に特異的な結合を有するネズミまたはキメラ抗体の分析工程を備える。その後、抗体の可変領域のタンパク質モデルは、Swiss PDBを用いた鋳型として既存の抗体構造を用いて生成する。抗体の結合特性に重要である可能性のある本来の抗体の可変領域部位内の重要な「抑制」アミノ酸を同定するため、その後、これらの構造モデルを分析する。フレームワーク残基の数と共にCDR領域内に含まれる残基(カバットおよびコチア(Chothia)の定義の双方を用いて規定した)は、通常、重要である。タンパク質モデルからの構造情報は、抗体の確認および既存の抗体構造および配列のデータベースと構造的に同一の結合に重要な残基を同定し、比較するために使用される。その後、これらアミノ酸は、1以上の最終的なヒト化配列の変異体を包含する候補とすることができる。本発明の抗体の調製において、本発明の完全なヒト化抗体に基づくネズミ抗TLR2抗体(ハイブリドーマクローンT2.5由来のマウスToll様受容体2(TLR2)抗体HyCult Biotechnology b.v., Cell Sciences, Canton,米国:カタログ番号1054)のVHおよびVK配列の双方は、典型的なフレームワーク残基、特に抗体が非常に一般的な配列構成を重要な位置、例えばカバット残基45−49(LEWIG)および92−94(CAR)で有するVHにおいて含むことを観測した。
驚くべきことに、本発明者は、完全にToll様受容体2機能を拮抗するため、T2.5 TLR2拮抗ネズミモノクローナル抗体がCD32への結合に必要であることを同定した。したがって、T2.5抗体の使用と関連のあるこの機能制限が、そのToll様受容体2の生物活性の中和効果を拮抗するためのOPN−305モノクローナル抗体の使用とも関連することを確認するため、実験を実施した。
THP−1細胞は、ヒト末消血単球細胞である。Toll様受容体リガンドに応じて、転写制御因子NF−κBおよび他の転写制御因子は、THP1細胞内で活性化されている。THP−1青色細胞株を、いくつかの転写制御因子、例えばNF−κBおよびAP−1による誘導性プロモーターの調節下、分泌型胎盤アルカリホスファターゼ(SEAP)遺伝子をコード化することが報告されているプラズミドを安定的に形質転換した。THP−1 CD14青色細胞株の変異体は、高い感度で細胞表面タンパク質を過剰発現する。結果として生じるTHP−1 CD14青色細胞は、選択的なマーカーであるゼオシンおよびブラストサイジンに抵抗性である。TLR刺激に応じて、THP−1青色細胞は、転写制御因子、その後、QUANTI−Blueレポーター系を用いて検出されるSEAP分泌を活性化する。QUANTI−Blueは、分泌型胎盤アルカリホスファターゼ(SEAP)活性を、細胞培養物の上清中で測定するために開発された比色分析の酵素分析である。SEAPの存在下におけるQUANTI−Blue培地は、紫青色への変化を肉眼で簡便に定性的に検出するか、625−655nmでの吸光度(OD)を計測することによって定量化することができる。
実施例2の結果は、本発明のOPN−305の完全なヒト化抗体によるToll様受容体2の拮抗剤が、CD32(CD32aまたはCD32bのいずれか)への抗体結合に依存しないことを示唆する。
THP−1 CD14青色細胞(Invivogen,サンディエゴ、米国)を、CD32aまたはCD32bをブロックする抗体の量で、前もってインキュベートした。
NF−κBに依存するSEAP生成は、細胞上清で測定された。図10Aは、OPN−301(T2.5ネズミ抗TLR2抗体)にさらされた細胞の結果を示す。図10Bは、OPN−305の完全なヒト化TLR2拮抗抗体にさらされた細胞に関する結果を示す。図10Aにおいて、カラムAは、Pam3CSK4のみで刺激された細胞に関する。カラムBは、OPN−301抗体にさらされた細胞に関する。カラムCは、0.4μg/mLの抗CD32a/bまたはコントロールヤギIgG抗体を加えられたOPN301抗体に関する。カラムDは、2μg/mL抗CD32a/b抗体またはコントロールヤギIgG抗体を加えられたOPN301抗体に関する。カラムEは、OPN301抗体と10μg/mL抗CD32a/b抗体またはコントロールヤギIgG抗体に関する。
本発明のOPN−305モノクローナル抗体が、Toll様受容体2、特にヒトToll様受容体2に特異的な結合を示すことを確認するため、実験を設計した。
TLR1/TLR2へテロ二量体の形成を可能とするため、Toll様受容体1およびToll様受容体2と安定的に形質転換したHEK 293(ヒト胎児由来腎臓293)細胞は、ビオチン化形状のネズミ抗TLR2抗体OPN−301(T2.5)1.0μg/mL単独で、または、完全なヒト化抗TLR2抗体OPN−305またはOPN−305 IgG4アイソタイプコントロール抗体1.0μgまたは10μg/mLの存在下のいずれかで30分間、氷の上でインキュベートした。
実施例4において、OPN−305がヒトToll様受容体2に特異的結合を示すことを測定した後、この実験は、OPN−305モノクローナル抗体が、Toll様受容体2種内交差反応であり、OPN−305が哺乳類Toll様受容体2の他の形状、例えばネズミToll様受容体2に結合することを可能とすることを示すかを評価した。
J774マウスマクロファージを、1×106/mLで、5μg/mLのOPN−305(OPN-305-21として示す)、OPN−301(T2.5)または関連のあるアイソタイプコントロール抗体(OPN−301へのネズミIgG1およびOPN−305へのヒトIgG4)の存在下、6時間、37℃で培養した。細胞を、TLR2拮抗剤HKLM(Invivogen,サンディエゴ、米国,カタログ番号tlr−kit2)にさらした。HKLMは、凍結乾燥され、熱殺菌されたグラム陽性通性細胞内細菌であるリステリア菌である。その後、上清を取り除き、ネズミTNF−αレベルを、RnD systems (R&D Systems)からの特定のELISAデュオセットによって測定した。
結果は図12のグラフに示す。これは、OPN−301の阻害と類似する方法において、OPN−305がネズミTLR2反応を抑制することを示す。したがって、それが表面またはネズミ細胞で発現するToll様受容体2に結合するため、OPN−305は、交差反応性がある。
さらなる実験は、OPN−305のサル細胞で発現されているToll様受容体の特異的結合を測定するため、実施された。これは、OPN−305抗体が、ヒトおよびネズミの形状のみより、種々の哺乳類形状のToll様受容体2に交差反応性であるかを同定した。
カニクイザルからの全血を、1.0μg/mLのOPN−305またはIgG4アイソタイプコントロール抗体で、30分間、室温でインキュベートして、その後、赤血球を溶解するため、PharmLyse(BD Biosciences)とインキュベートして、それからFITC標識された抗ヒトIgG4と検出した。
カニクイザルからの全血は、1.0μg/mLのビオチン化OPN−301単独で、または、OPN−305もしくはIgG4アイソタイプコントロール抗体(OPN305)1.0μgもしくは10μg/mLの存在下のいずれかで30分間、室温でインキュベートされた。細胞は、その後、洗浄され、PECy7に結合されたストレプトアビジンとさらに30分間インキュベートされた。
OPN−305抗体の分析は、免疫反応、例えばHAMA反応に結果として生じることがあり得る、抗体が投与される対象によって産生された抗体を免疫性エピトープの存在を見出すため、実施された。前記のように、T細胞エピトープの存在は、抗体に対する中和抗体反応の産生が、治療目的のために患者に投与するのにもはや適していない結果を生じるため、非常に望ましくない。したがって、T細胞エピトープが明らかに存在していないことを確認するため、OPN−305抗体をスクリーンした。
<ドナーPBMCの調製および選択>
末梢血単核球細胞(PBMC)をNational Blood Transfusion Service (Addenbrooke's Hospital,ケンブリッジ、英国)から健康な群のドナー軟膜(24時間以内に献血した)から単離した。PBMCは、軟膜からLYMPHOPREP(商標)(Axis-Shield,ダンディ、英国)密度遠心によって単離され、CD8+ROSETTESEP(商標)(StemCell Technologies Inc., ロンドン、英国)を用いて、CD8+T細胞は減少された。ドナーは、組織型判定キット(Biotest, Landsteinerstrasse,デンマーク)に基づくBiotest SSP−PCRを用いてHLA−DRハプロタイプを同定し、並びに、コントロール抗原であるスカシ貝ヘモシアニン(KLH)(Pierce,ロックフォード、米国)へのT細胞の反応を測定することによって特徴付けられた。PBMCは、その後凍結され、必要とされるまで液体窒素に保管された。
国際公開第2007/099341号に教示するように、T細胞エピトープの存在を決定するため、抗体を分析の対象とした。個々のドナーからのPBMCを解凍し、計数して生存率を評価した。細胞は、室温AIMV培養培地中(インビトロゲン、ペイズリー、英国)で回復され、AIMVで4.6×106PBMC/mLまで再懸濁された。計1mL増殖細胞ストックを24ウェルプレートに添加し、それぞれのドナーに対してバルク培養を確立した。サンプルあたり最終濃度50μg/mLが得られるように計1mLのそれぞれ希釈したテストサンプルをPBMCに添加した。それぞれのドナーに対して、陽性対象(100μg/mLのKLHでインキュベートした細胞)および陰性対象(培養培地のみでインキュベートした細胞)も含有した。最初の4ドナーに関して、付加的なコントロールは、T細胞反応の調節をテストサンプルでテストするために含まれ、テストサンプルおよびKLHの双方ともPBMCに添加した。KLH単独でのこれらサンプルの比較は、増殖へのテストサンプルの効果を評価するために使用することができる。培養物は、計8日間、37℃、5%CO2でインキュベートされた。5,6,7および8日目、それぞれのウェル内の細胞を徐々に再懸濁し、丸底96ウェルプレートのそれぞれのウェルに3×100μLアリコートで移した。培養物は、100μLのAIMV培養培地中1μCi[3H]−チミジン(Perkin Elmer(商標), Waltham,マサチューセツ州、米国)を適用し、さらにTomTec Mach III細胞ハーベスターを用いてフィルターマット(Perkin Elmer(商標), Waltham,マサチューセツ州、米国)上に収穫の18時間前インキュベートした。各々のウェルにつき1分あたりのカウント(cpm)を、MELTILEX(商標)(Perkin Elmer(商標), Waltham,マサチューセツ州、米国)シンチレーションカウントによってパララックス(paralux)のマイクロプレートβカウンターの低いバックグラウンドカウントモードで測定した。テストサンプルの潜在的毒性を評価するため、細胞生存率カウント(VICELL(商標)カウンターおよびトリパンブルー染色による除外を用いた)は、テスト培養物のサンプル、培地およびKLHコントロールに最初の10ドナー
に対して7日間インキュベートした後、実施された。
増殖分析に関して、2以上の刺激指数(SI)の実験的閾値(SI≧2)を、以前確立し、それによって前記増殖反応を誘発するサンプルは、陽性と考えられる(境界線上のSI>1.95をハイライトしたものを含む)。広範囲に亘る分析開発および以前の研究は、大きい数の偽陽性反応を検出することなく、これが最高感度を可能とする最小限のシグナルとノイズの閾値であることを示す。増殖データセット(n=3)について、陽性反応を統計学および実験による閾値によって規定した:
抗TLR2抗体OPN−305(VK5/VH4)を、均一になるまでプロテインA親和性クロマトグラフィで精製し、次いでサイズ排除クロマトグラフィによって精製した。
EpiScreen(商標)で、経時変化によるキメラ抗体に対するT細胞分析を観測した陽性CD4+T細胞増殖反応は、15−40%の期待範囲にあった。重大なことは、期待通りに分析が機能したことを示すコントロール抗原KLHに対して、頻繁、且つ強力なT細胞反応を観測した。結果は、完全なヒト化抗体OPN−305(VK5A/H4)が、いずれのドナーにおいて、あらゆる陽性反応を示すことができなかったことを示し、この抗体は、したがって、臨床における免疫性のリスクが低いことが考察された。
抗原および抗体の複合体の結合および親和性を同時に測定する免疫学的バイオセンサー、例えばBIACORE(商標)表面プラズモン共鳴(SPR)装置は、結合反応速度の解析を可能とする。化合物の分離速度およびその後の最適化は、生物薬剤抗体の開発に特に重要である。BIACOREは、表面結合分子「リガンド」および溶液「電解質」中の、その結合パートナーの間の相互作用を同時に観測するため、表面プラズモン共鳴(SPR)を使用する。SPRは、総内部反射率の条件下、光が層で反射したとき、金属層の表面で生じる電子電荷密度波現象である。発生する表面プラズモンは、反射光から金属層の反対側の媒体の屈折率におけるあらゆる変化に感受性を有する。表面で生じるタンパク質間相互作用は、媒体の屈折率に影響し、したがって、検出可能である。リガンド修飾したセンサー表面への分子結合は、検出した結合した分解質の量における小さな変化を可能とする結合した塊に比例する反応を生じる(低いピコグラムレベルまで)。技術は、10−5から10−12Mの範囲に亘る親和定数(KD)、103から107M−1s−1の間の親和性速度定数(ka)および10−1から10−6s−1の間の分離速度定数(kd)を測定するために使用することができる(1−3)。
<装置の準備>
あらゆるサンプルを実施する前、システムチェック(Biacore Preventative Maintenance Kit 2)を実施した。全てのシステム(試薬パンプ、屈折計、注射、ノイズ、混合およびバッファーセレクター)の試験に合格したことで、装置が製造者が設定した基準に従って作動することが示された。システムチェック後、脱着/消毒(Biacore Preventative Maintenance Kit 2)プログラムを、装置を清浄にするために実施した。
<システム調製>
CM5チップの挿入に応じて、システムは、BIAnormalizing solution(Biacore Preventative Maintenance Kit 2)で満たされ、正常化された。特に記載のない限り、全サンプルについて、25℃で試験した(サンプルラックを25℃でインキュベートした)。チップは、HBS−EP(ランニングバッファーとして使用)とともにシステムに投入した。チップ表面は、事前に30秒の50mM NaOHで2回処理し、安定した基準線が全てのフローセル(Fcs)内で得られるようにした。
T2.5ネズミ抗Toll様受容体2抗体(ハイブリドーマクローンT2.5 に由来するHyCult Biotechnology b.v., Cell Sciences, Canton,米国:カタログ番号1054)および本明細書に記載したOPN−305抗Toll様受容体2抗体の2つの抗体について分析を行った。双方のモノクローナル抗体は、補給品として保存され、全固定を実施するため、1μg.mL−1まで希釈された。受容体rhTLR−2およびrmTLR2/Fcを、乾燥粉末からHBS−EPを用いて最終濃度1000nMまで再構成し、4℃で保存した。キャリアタンパク質は、この溶液に添加しなかった。
前記試験の結果に基づいて、最適な条件であることが示された条件に従って、この試験についての直接的な分析手段を選択した。速度実験に関して、固定したリガンドの量は、チップ表面での物質移動効果を防ぐために制限する必要がある。
結果は図1に示す。会合速度(Kon)および分離速度(Koff)は、二価結合モデルを用いて算出された(BIAcore Evaluation Software version 3.2)。平衡解離定数(Ko)は、kon/konの比として算出された。
この実験は、TLR2に結合したとき、OPN−305モノクローナル抗体の生物活性を阻害するために必要とされる、最小の占有レベルを同定するために設計した。THP−1受容体細胞株を用いて結合に対する機能を観測した。特に、OPN−305の生物活性は、Pam3CSK4を誘発したTHP1−CD14細胞で評価した。
THP1−CD14細胞を、ウェルあたり1×106細胞の濃度で12ウェルディッシュに播種した。Pam3Csk4(200ng/mL)の添加前、OPN−305は、ウェルに様々な濃度(0.000488μg/mLから2μg/mLの範囲)で添加した。細胞は、一晩、38℃でインキュベートした。NF−κBによって促進されるSEAP活性は、160μL Quantiblue(Invivogen)試薬と熱不活性条件の培地を90分間、37℃でインキュベートし、650nmの吸光度により測定された。
結果は、図21A,BおよびCに示す。これらの結果は、Pam3Csk4で処理した後、NF−κB活性の阻害が用量依存的な方法で、観測されたことを示す(図21AおよびB)。OPN−305は、ほぼ完全にNF−κB活性を濃度2μg/mLで阻害する(図21C)。THP1−CD14細胞におけるOPN−305の特異的結合の割合は、前記一般式を用いて算出された。
<材料および方法>
THP1 CD14青色細胞を、ウェルあたり100,000細胞で96ウェルプレートに播種した。細胞は、0,1,10および100ng/mLのOPN−305で、一時的に様々な投与量のPam3Csk4で一晩、37℃で刺激する前に前もってインキュベートした。上清40μLを取り除き、65℃で10分間、熱不活性化した。NF−κBによって促進されるSEAP活性は、160μLQuantiblue(Invivogen)試薬と熱不活性条件の培地を90分間、37℃でインキュベートした後、650nmの吸光度より測定された。細胞に基づく分析における受容体は酵素と同様に反応することを仮定し、V(NF−κB活性)に対するS[Pam3CSK4]並びに二重逆数のラインウィーバー・バークプロット(1/v対1/s)をプロットした。
結果は図22に示し、図22Aは、NF−κBに依存するSEAP活性に対する[Pam3CSK4]を示す。一方、図22Bは、1/Vに対する1/Sのラインウィーバー・バークプロットを示す。
この実験は、OPN−305によるTLR2の阻害が、他のTLRの相当するそれらリガンドに影響するかを考慮した。
THP1 CD14青色細胞を、ウェルあたり25,000細胞で384ウェルプレートに播種した。様々な投与量の異なるToll様受容体リガンド拮抗剤で刺激する前、細胞は、120分間、0,1,10および100ng/mLのOPN−305またはヒトIgG4アイソタイプコントロール抗体で前もってインキュベートした。一晩、HEK青色検出用培地を用いて検出した後、NF−κBによって促進されるSEAP活性は、調整済みの培地中で直接測定した。
結果は、図23A,BおよびCに示す。OPN−305にさらされていないコントロール細胞と比較した場合、フラジェリン(図23C)およびLPS(図23B)への反応は、不変であった。予期したように、Pam3CSKおよびFSL−1の介在によるTLR2活性反応は、OPN−305によってブロックされた(図23A)。OPN−305にさらされていないコントロール細胞と比較した場合、フラジェリンおよびLPSへの反応は、不変であった。予期したように、Pam3CSKおよびFSL−1の反応は、OPN−305によってブロックされた。これは、OPN−305が、あらゆる予期しないTLR4またはTLR5のリガンドへの反応性の増減をもたらさないことを示唆する。
<材料および方法>
雌BALB/cマウスの群(n=4)は、100μgPam3Csk4処理30分前、OPN−305モノクローナル抗体で10mg/kg,2mg/kg、および0.4mg/kg、30分間処理した。全ての治療は、腹腔内投与によって実施された。4時間後、マウスを致死麻酔で屠殺し、血液を抽出した。血清を取り出し、サイトカイン濃度を、ELISAによって測定した。血清は、KC,IL−12p40については1/10、IL−6 ELISAについては1/5希釈して使用した。
結果を図24A,BおよびCに示す。OPN305は、Pam3Csk4によって誘発した敗血症を阻害することを示す。低い投与量(0.4mg/kg)の抗TLR2 OPN305は、Pam3Csk4によって誘発されたサイトカインをマウスで著しく阻害することができる。
<材料および方法>
NR8333細胞は、ATCCから購入され、ATCCの指示に従ってFK12培地で培養した。細胞は、ラット特異的抗CD32(マウスIgG1)で10分間、室温でブロックした。細胞は、OPN305(またはアイソタイプ)で15分間処理し、洗浄し、RPE抗ヒトIgG4で検出し、採取前2%ホルモル生理食塩水で固定した。NR8383細胞は、ポリクローナルラビット抗ラットTLR2抗体でも染色し、Alexa−Fluor 488抗ラビットIgGで検出した。
結果は、TLR2がラット肺胞マクロファージ(NR8383)で発現されていて、OPN305を用いて検出されたことを図26に示す。(A)無染色細胞、(B)陽性対象;ポリクローナルラビット抗ラットTLR2一次抗体、二次抗対は抗ラビットAlexa−Fluor 488;(C)ポリクローナルヒトIgG4で処理した細胞、二次抗体は、ヒト抗IgG4 PEであった;(D)OPN305染色した細胞、二次抗体抗ヒトIgG4 PE。
<材料および方法>
フィコールを用いて、ブタPBMCを血液から精製した。細胞数を計数し、チューブあたり1×106細胞で配置した。ブタ用の特定のCD32ブロッキング剤の不存在下において、細胞は、PBS中50% FCS/BSAマトリックスのヒト抗CD32を用いて、10分間、ブロックした。細胞は、OPN305(またはアイソタイプコントロール)で15分間、室温でインキュベートした。1H11(マウス抗ブタTLR2、Javier Dominguezからの寄贈)は、陽性対象として使用された。洗浄後、細胞はRPE結合した抗ヒトIgG4で、15分間、室温でインキュベートした。最終洗浄工程後、細胞を固定し、採取し、分析した。
結果を図27に示す。TLR2は、ブタPBMCで発現があり、OPN305で染色された。PBMCは、フィコールを用いた分離によって精製した。A−Cは、Pig 666からのPBMC、D−Fは、Pig 488からのものを示す。A,Dは無線色;B,Eは、コントロールポリクローナルヒトIgG4抗対で標識し、その後、PE標識した抗ヒトIgG4で二次染色した;C,Fは、OPN305、その後PE標識した抗ヒトIgG4で染色した。
Claims (18)
- Toll様受容体2に特異的に結合してToll様受容体2の機能に拮抗する抗体またはその抗原結合部位であって、
前記の拮抗は、抗体または抗原結合部位のCD32(Fcγ受容体II)への結合とは独立に介在され、
前記抗体は、配列番号:1のアミノ酸配列の軽鎖の可変領域又は配列番号:1のアミノ酸配列と95%以上の同一性を有するアミノ酸配列、及び
配列番号:4のアミノ酸配列の重鎖の可変領域又は配列番号:4のアミノ酸配列と95%以上の同一性を有するアミノ酸配列を含み、
前記軽鎖の可変領域が、配列番号:7、配列番号:8及び配列番号:9のそれぞれで表される、CDR1、CDR2及びCDR3を含み、
前記重鎖の可変領域が、配列番号:10、配列番号:11及び配列番号:12のそれぞれで表される、CDR1、CDR2及びCDR3を含む、
抗体またはその抗原結合部位。 - 軽鎖の可変領域がκ定常領域に接続される、請求項1に記載の抗体またはその抗原結合部位。
- 重鎖の可変領域は、免疫グロブリンGのサブクラスのアイソタイプ4(IgG4)の抗体由来の少なくとも1つの定常領域に結合する、請求項1または2に記載の抗体またはその抗原結合部位。
