・完全アゴニスト:アゴニストにより誘発された受容体刺激がその系の最大応答能に達した場合、それはその系の最大応答をもたらし、それはその系の完全アゴニストである。いくつかのアゴニストが同等の最大応答を惹起することもあり、その実験系においてそれらは総て完全アゴニストである。
・部分的アゴニスト:所与の組織において、特定の条件下で、同じ系で同じ受容体を介して作用する完全アゴニストほど大きな作用を惹起することができない分子(総ての受容体が占有されるはずの高濃度で適用した場合でも)。部分的アゴニストは一般に、完全アゴニストが共存する場合に、それらは該完全アゴニストの最大応答をそれら固有の最大応答まで引き下げることから、部分的アンタゴニストでもある。この完全アゴニストと部分的アゴニストという表記は系に依存し、ある系またはある測定での完全アゴニストは、別の系または別の測定では部分的アゴニストである場合がある。
・拮抗作用は、アゴニストおよびアンタゴニストの結合が互いに相容れない場合に競合的であり得る。これはアゴニストとアンタゴニストが同じ結合部位をめぐって競合し、重複する隣接部位と一緒になるためである可能性がある。第三の可能性は、異なる部位が関与するが、それらは受容体高分子にアゴニスト分子と、アンタゴニスト分子とが同時には結合できないような影響を及ぼすということである。
・非競合的拮抗作用はアゴニストと、アンタゴニストとが同時に受容体に結合できる場合に見られ、アンタゴニストの結合はアゴニストの作用を低下させるか、または妨げ、そのアゴニストの結合に影響を及ぼす場合と及ぼさない場合がある。

c)高ストリンジェンシー条件下で配列番号15〜21および50〜77の少なくとも一つの配列とハイブリダイズし得る少なくとも18ヌクレオチドの核酸。
原型チロシンキナーゼ受容体(c−Met受容体)を標的とする数種のマウスMabを、マウス可変ドメインと、ヒト定常ドメインとを有するキメラMabとして再構成した。それらの固有の活性を、リガンド(HGF)依存的なc−Met受容体リン酸化の阻害を測定する機能的アッセイに基づき分析した。マウス可変ドメイン配列(VH、VL)のPCR−クローニングの際、マウスVHまたはVL配列のいずれかを有する{NheI−BclI}制限断片を、IgG1免疫グロブリンのヒト軽鎖Cκまたはヒト重鎖[CH1−ヒンジ−CH2−CH3]のいずれかの定常ドメインの全コード配列を有するpCEP4ベクター(InVitrogen, US)に連結してキメラMabを構築した。クローニング工程は総て、Laboratory manual (Sambrook and Russel, 2001)に記載されているような慣例の分子生物学的技術に従って、または供給者の説明書に従って行った。各遺伝子構築物をBig Dyeターミネーターサイクルシーケンシングキット(Applied Biosystems, US)を用いたヌクレオチド配列決定により完全にバリデートし、3100 Genetic Analyzer(Applied Biosystems, US)を用いて分析した。対応するキメラMabの作製は6mMグルタミンを添加した血清不含培地Excell 293(SAFC Biosciences)で増殖させた、懸濁に適応したHEK293 EBNA細胞(InVitrogen, US)を用いて分析した。直鎖状25kDaポリエチレンイミン(PEI)(Polysciences)を用い、一時的トランスフェクションを行った。培養工程を、細胞生存率とMab生産に基づいて測定した。Aタンパク質樹脂(GE Healthcare, US)上での慣例的クロマトグラフィーアプローチを用い、Mabを精製した。
上記の所見に基づけば、マウスIgG1 MabをヒトIgG1 Mabへ再構成した際に見られた薬理学的特性の障害はヒトIgG1ドメインによるものであったとの仮説が立てられる。一方、文献では、c−Met受容体の活性化がその二量体化と関連していること、およびc−Met受容体の二量体化の阻害がc−Met受容体のリン酸化および下流のシグナル伝達を阻害し得ることが知られていた。

A549細胞を12マルチウェル(MW)プレートの完全増殖培地[F12K+10%FCS]に播種した。細胞を16時間飢餓状態にした後、HGF[100ng/ml]で刺激し、リガンド刺激の15分前に各供試Mabを終濃度30μg/mlで加えた。氷冷溶解バッファーを加え、15分後にHGFを加えてリン酸化反応を停止させた。細胞を機械的にこすり落とし(scaped)、細胞溶解液を4℃、13000rpmにて10分間の遠心分離により回収し、細胞溶解液は上清相に相当する。タンパク質含量はBCAキット(Pierce)を用いて定量し、使用まで−20℃で保存した。c−Metのリン酸化状態をELISAにより定量した。ヤギ抗c−Met Mab(R&D, ref AF276)を捕捉抗体として用い(一晩4℃でコーティング)、TBS−BSA5%バッファーを用いた飽和工程(室温で(RT)1時間)の後、25μgのタンパク質溶解液を、コーティングした96MWプレートの各ウェルに加えた。室温で90分インキュベートした後、プレートを4回洗浄し、検出抗体(位置1230、1234および1235のリン酸化されたTyr残基に対する抗ホスホ−c−Met Mab)を加えた。さらに1時間インキュベートし、4回洗浄した後に、HRP(Biosource)に結合された抗ウサギ抗体を室温で1時間加え、ルミノールを加えることにより発光の検出を行った。発光の読み取りはMithras LB920マルチモードプレートリーダー(Berthold)にて行った。
記号Δは欠失を意味する。
第一の実験セットでは、無関連のマウスIgGl、ヒトIgG1およびヒトIgG2は両BRETモデルにおいてHGFにより誘導されるBRETシグナルの効果は無かったことを対照とした(図3)。これらのMabをさらに対照として用いた。
組換えc−Metの種々のキメラ形態およびヒト化形態の結合能を調べるために直接的ELISAを設定した。要するに、R&D Systemsからの組換え二量体c−Metを96ウェルImmunlon IIプレートに1.25μg/mlでコーティングした。4℃で一晩インキュベートした後、ウェルを0.5%ゼラチン/PBS溶液で飽和させた。次に、プレートを37℃で1時間インキュベートした後、2倍希釈の供試抗体を加えた。プレートをさらに1時間インキュベートした後、ネズミ抗体を検出するためのヤギ抗マウスIgG HRPとキメラおよびヒト化抗体認識のためのヤギ抗ヒトκ軽鎖HRPを加えた。プレートを1時間インキュベートし、ペルオキシダーゼ基質TMB Uptimaを5分間加え、H2SO4 1Mで中和した。図6Aおよび6Bに示す結果は、供試した総ての形態でc−Met認識は匹敵するものであったことを示した。
ATCCからのNCI−H441細胞を通常、RPMI1640培地(Invitrogen Corporation, Scotland, UK)、10%FCS(Invitrogen Corporation)、1%L−グルタミン(Invitrogen Corporation)中で培養した。増殖アッセイのため、プレーティング前にコンフルエント増殖期となるように使用の3日前に細胞を分割した。NCI−H441細胞を96ウェル組織培養プレートの200μlの血清不含培地(RPMI1640培地および1%L−グルタミン)中に3.75×104細胞/ウェルの密度でプレーティングした。プレーティング24時間後に、供試抗体をNCI−H441に加え、37℃で30分間インキュベートし、その後、HGFを終濃度400ng/ml(5nM)でさらに142時間加えた。各抗体の試験用量範囲は10〜0.0097μg/ml(各ウェル中の終濃度)である。この実験では、ネズミIgG1 Mabをネズミアイソタイプ対照として加え、供試抗体は次のものであった:それぞれ[224G11]chim、[11E1]chimおよび[227H1]chimとして識別されるm224G11、m11E1、m227H1およびそれらのヒトIgG1キメラ形態。細胞単独−/+HGFをプレーティングしたウェルも含めた。次に、細胞を0.25μCiの[3H]チミジン(Amersham Biosciences AB, Uppsala, Sweden)で7時間30分刺激した。トリクロロ酢酸に不溶なDNAに組み込まれた[3H]チミジンの規模を液体シンチレーション計数により定量した。結果は、腫瘍細胞に単独で加えた場合の抗c−Met Mabで見られ得る潜在的な固有アゴニスト活性をより良く評価するために未変換のcpmデータとして表す。
NCI−H441は乳頭状肺腺癌に由来し、高レベルのc−Metを発現し、c−Met RTKの構成的リン酸化を示す。

PKSCDCHCPPCPに相当する[TH7]ヒンジ配列によりヒトIgG1以外の免疫グロブリンアイソタイプ骨格に誘導される薬理学的特性の調節を評価するために、本発明者らは上記の[TH7]配列をヒトIgG2およびIgG4骨格へ遺伝的に移入した。ヒンジ領域の改変は、{Nhe1I−Bcl1}制限断片を[TH7]改変を有する均等部分と交換することにより行った。なお、この{NheI−Bcl1}断片はグローバルな遺伝子合成(Genecust, LU)により合成される。クローニング工程は、Laboratory manual (Sambrook and Russel, 2001)に記載されているような慣例の分子生物学的技術に従って、または供給者の説明書に従って行った。各遺伝子構築物をBig Dyeターミネーターサイクルシーケンシングキット(Applied Biosystems, US)を用いたヌクレオチド配列決定により完全にバリデートし、3100 Genetic Analyzer(Applied Biosystems, US)を用いて分析した。得られた結果をTH7操作ヒトIgG2およびIgG4アイソタイプ(重鎖のみ、軽鎖は他の総てのIgG1系構築物に用いられているc224G11/ヒトCκと同じ)のそれぞれアミノ酸配列とヌクレオチド配列に関して配列番号78、79、80、および81として記載する。これらの新規な構築物を上記実施例2のようにキメラ224G11抗c−Met Mabに適用した。
TH7ヒンジをIgG2およびIgG4キメラ224G11 Mabにも導入し、c−Met受容体リン酸化アッセイで試験した。14Aおよび14Bに示されるように、c224G11[IgG2TH7]およびc224G11[IgG4TH7]は微弱ながらc224G11 Mabよりも有意に弱いアゴニスト作用のみを誘導し、224G11 Mabのネズミ形態(m224G11)に匹敵するアンタゴニスト作用を示した。この結果をc−Met活性化BRETモデルで確認したところ(図15)、c224G11[IgG2TH7]およびc224G11[IgG4TH7]もc224G11 Mabより弱いアゴニストを示した。
PC3前立腺癌細胞を、トリプシンを用いてディッシュから剥がし、血清不含F12k培地で3回洗浄し、同じ培地に再懸濁させた。細胞(100,000細胞/ウェル)を、1μg/mlのラミニン1をコーティングした96ウェルプレートにプレーティングした。以下の形態の供試抗CD151 Mabを同時に終濃度10μg/mlで加えた:ネズミIgG1 Mab m214B2、c214B2と呼ばれる非改変キメラIgG1抗体形態およびcTH7−214B2と呼ばれるTH7改変を伴うキメラIgG1抗体形態。
Claims (14)
- 特定の標的分子に対するモノクローナル抗体、またはその二価の機能的断片もしくは誘導体の拮抗活性を改良する方法であって、該抗体は該標的分子の1以上の生物活性を阻害することができ、少なくとも一つのアミノ酸の欠失、付加、または置換によるヒンジ領域のアミノ酸配列の改変からなるヒンジ領域の再構成工程を含んでなり、アミノ酸配列の改変によって得られた再構成されたヒンジ領域が、配列番号1、2、4、6、7、22〜29、33〜39、および42〜49からなる群から選択される配列を有するヒンジ領域である、方法。
- モノクローナル抗体が二価抗体である、請求項1に記載の方法。
- モノクローナル抗体がキメラ抗体である、請求項1または2に記載の方法。
- モノクローナル抗体がヒト化抗体である、請求項1または2に記載の方法。
- モノクローナル抗体がヒト抗体である、請求項1または2に記載の方法。
- モノクローナル抗体がIgG1である、請求項1〜5のいずれか一項に記載の方法。
- 標的分子が膜貫通受容体である、請求項1〜6のいずれか一項に記載の方法。
- 膜貫通受容体がチロシンキナーゼ受容体、テトラスパニン、およびGPCRからなる群から選択される、請求項7に記載の方法。
- 特定の標的分子に対するモノクローナル抗体、またはその二価機能的断片または誘導体をアンタゴニストに関してスクリーニングする方法であって、該抗体は該標的分子の1以上の生物活性を阻害することができ、
(a)該標的分子の該一以上の生物活性の、初期レベルの阻害を有する初期抗体を選択する工程、
(b)該初期抗体のヒンジ領域のアミノ酸配列を、請求項1〜8のいずれか一項に記載の方法により改変する工程、
(c)工程(b)の改変抗体を、その該標的分子の該一以上の生物活性を阻害する能力に関して評価する工程、および
(d)該標的分子の該一以上の生物活性の阻害レベルが、該阻害の初期レベルよりも高い工程(c)の抗体を陽性結果として選択する工程
を含んでなる、方法。 - 配列番号2、4、6、22〜26、28、29、33〜39、および42〜49からなる群から選択されるアミノ酸配列を含んでなる、モノクローナル抗体。
- ヒト抗体である、請求項10に記載のモノクローナル抗体。
- IgGl抗体である、請求項10または11に記載のモノクローナル抗体。
- 請求項10〜12のいずれか一項に記載のモノクローナル抗体をコードする単離核酸。
