Description
(1)L−ロイシンを、総必須アミノ酸中のモル組成比で35〜66%含有する、高齢者の骨格筋量減少を防止又は改善するための経口用アミノ酸含有組成物。
(2)L−ロイシンが、総必須アミノ酸中のモル組成比で35〜57%含有する、(1)項の経口用アミノ酸含有組成物。
(3)L−ロイシンを、総必須アミノ酸中のモル組成比で35〜50%含有する、(1)項記載の経口用アミノ酸含有組成物。
(4)L−ロイシンを、総必須アミノ酸中のモル組成比で35〜45%含有する、高齢者の骨格筋量減少を防止・改善するアミノ酸含有組成物。
(5)総必須アミノ酸中の各必須アミノ酸の組成が下記の数値範囲のモル組成比(%)である、高齢者の骨格筋量減少を防止又は改善するための経口用アミノ酸含有組成物:L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜66、L−リジン8.0〜16、L−メチオニン2.0〜10、L−フェニルアラニン2.5〜8.0、L−スレオニン7.0〜14、L−バリン5.0〜15、L−トリプトファン0.0〜2.0。
(6)総必須アミノ酸中の各必須アミノ酸の組成が下記の数値範囲のモル組成比(%)である、(5)項記載の経口用アミノ酸含有組成物:L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜57、L−リジン8.0〜16、L−メチオニン2.0〜10、L−フェニルアラニン2.5〜8.0、L−スレオニン7.0〜14、L−バリン5.0〜15、L−トリプトファン0.0〜2.0。
(7)総必須アミノ酸中の各必須アミノ酸の組成が下記の数値範囲のモル組成比(%)である、(5)項記載の経口用アミノ酸含有組成物:L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜50、L−リジン8.0〜16、L−メチオニン2.0〜10、L−フェニルアラニン2.5〜8.0、L−スレオニン7.0〜14、L−バリン5.0〜15、L−トリプトファン0.0〜2.0。
(8)総必須アミノ酸中の各必須アミノ酸の組成が下記の数値範囲のモル組成比(%)である、(5)項記載の経口用アミノ酸含有組成物:L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜45、L−リジン8.0〜16、L−メチオニン2.0〜10、L−フェニルアラニン2.5〜8.0、L−スレオニン7.0〜14、L−バリン5.0〜15、L−トリプトファン0.0〜2.0。
(9)アミノ酸の組成が下記の数値範囲のモル組成比(%)である高齢者の骨格筋量減少を防止又は改善するためのアミノ酸含有組成物:L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜45、L−リジン12〜16、L−メチオニン3.0〜10、L−フェニルアラニン3.5〜8.0、L−スレオニン11〜14、L−バリン8.5〜15、L−トリプトファン0.0〜1.0、L−アルギニン0.0〜10。
(10)アミノ酸を、(1)〜(3)、(5)〜(8)のいずれか1項記載の量で含有する、高齢者の骨格筋量減少を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
(11)アミノ酸を、(1)〜(3)、(5)〜(8)のいずれか1項記載の量で含有する、高齢者の骨格筋量減少を防止又は改善するとともに、インスリン感受性、糖尿病、糖代謝異常を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
(12)アミノ酸を、(1)〜(3)、(5)〜(8)のいずれか1項記載の量で含有する、高齢者の骨格筋量減少を防止又は改善するとともに、高脂血症、脂肪肝、脂質代謝異常を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
(13)アミノ酸を、(4)又は(9)項記載の範囲内の量で含有する、高齢者の骨格筋量減少を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
(14)アミノ酸を、(4)又は(9)項記載の量で含有する、高齢者の骨格筋量減少を防止又は改善するとともに、インスリン感受性、糖尿病、糖代謝異常を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
(15)アミノ酸を、(4)又は(9)項記載の量で含有する、高齢者の骨格筋量減少を防止又は改善するとともに、高脂血症、脂肪肝、脂質代謝異常を防止又は改善するための経口用医薬品、食品、栄養剤またはサプリメント。
表1に示すホエイタンパク質のアミノ酸組成からなる必須アミノ酸混合物を10名の高齢者(男性7名、女性3名、平均年齢66.7歳、平均体重81.7kg)に、また同じく表1に示すロイシンの配合を高めた高ロイシン必須アミノ酸を10名の高齢者(男性5名、女性5名、平均年齢66.5歳、平均体重74.5kg)に経口摂取させた。摂取させたアミノ酸の総量は、ともに6.726gとした。安定同位体標識L−フェニルアラニンを使用し、アミノ酸の摂取から3.5時間後までの筋タンパク質の合成比速度(Fractional Synthesis Rate)を求め、摂取前の合成比速度からの変化を算出した。また、動・静脈のフェニルアラニンの濃度差と血流速度を測定し、アミノ酸摂取から3.5時間のフェニルアラニンの下肢へのネットの取り込み量を、筋タンパク質の蓄積量の指標として求めた。
表2に示すアミノ酸混合物を8名のインスリン抵抗性を有する高齢者(男性4名、女性4名、平均年齢66.1歳、平均体重80.1kg)に16週間摂取させた。アミノ酸は1回11gを1日2回、食間に経口摂取させた。アミノ酸摂取開始前と、開始後4週ごとに、骨格筋量の指標として除脂肪体重(LBM: lean body mass)を二重エネルギーX線吸収測定法(DEXA: dual energy X-ray absorptiometry)により測定した。脚の筋力をアミノ酸摂取開始前と、開始後8週目、12週目、16週目に測定した。膝屈筋と膝伸筋それぞれの最大挙上重量(1RM: one repetition max)の和を脚の筋力とした。また、4週ごとに75gの経口ブドウ糖負荷試験(OGTT: oral glucose tolerance test)を行ない、インスリン感受性を評価するとともに、血液中脂質濃度を測定した。さらに、肝臓中脂質濃度を8週ごとに磁気共鳴画像法(MRI: magnetic resonance imaging)により測定した。
10週齢のSD系雄性ラットを用い、表3に組成を示すアミノ酸の3.5%水溶液を20ml/kgの用量で経口投与した(アミノ酸の投与量は0.7g/kg)。アミノ酸の組成は、ホエイたんぱく質のアミノ酸組成を基本として構成した。つまり基本のホエイたんぱく質のアミノ酸組成であるL−ロイシン含量が総必須アミノ酸中のモル比で25%であり非必須アミノ酸も含有する総合アミノ酸組成物(Leu/EAA 25% TAA)、L−ロイシン含量が総必須アミノ酸中のモル比で49%である必須アミノ酸のみの組成物(Leu/EAA 49% EAA)、L−ロイシン含量が総必須アミノ酸中のモル比で58%であり非必須アミノ酸も含有する総合アミノ酸組成物(Leu/EAA 58% TAA)、あるいはL−ロイシン含量が総必須アミノ酸中のモル比で66%であり非必須アミノ酸も含有する総合アミノ酸組成物(Leu/EAA 66% TAA)とした。投与後30分から50分までの20分間の筋タンパク質合成比速度を、安定同位体標識フェニルアラニンを使用した大量投与法により求めた。
10週齢のSD系雄性ラットを用い、表4に組成を示すアミノ酸の3.5%水溶液を20ml/kgの用量で経口投与した(アミノ酸の投与量は0.7g/kg)。アミノ酸の組成は、ホエイたんぱく質のアミノ酸組成を基本とし、L−ロイシンの投与量が各群で同じとなるように設定した。投与後30分から50分までの20分間の筋タンパク質合成比速度を、安定同位体標識フェニルアラニンを使用した大量投与法により求めた。
7週齢のSD系雄性ラットを用い、ホエイたんぱく質あるいは、表5に組成を示すアミノ酸となるようにホエイたんぱく質とアミノ酸を1:1で混合したもの、あるいは、表5のアミノ酸組成物の3.5%水溶液を20ml/kgの用量で経口投与した(投与量は0.7g/kg)。投与後30分から50分までの20分間の筋タンパク質合成比速度を、安定同位体標識フェニルアラニンを使用した大量投与法により求めた。
2型糖尿病モデルであるKK-Ayマウス雄性5週齢を、25%脂質含有高脂肪食で2週間飼育した。2週間後、14%脂質含有12%カゼイン食(Control)、あるいは表6に示すL−ロイシンが総必須アミノ酸中にモル比で53.5%となるように配合したアミノ酸混合物をControl食に8%添加した飼料(Leu/EAA 54%)で飼育を開始し、4週間後に1g/kg BWの経口ブドウ糖負荷試験(OGTT: oral glucose tolerance test)を行い、その後ヘモグロビンA1c(HbA1c)、肝臓中脂質含量の測定を行った。
Claims (6)
- アミノ酸の組成が下記の数値範囲のモル組成比(%)である高齢者の骨格筋量減少を防止又は改善するためのアミノ酸含有組成物:
L−ヒスチジン0.0〜3.5、L−イソロイシン5.0〜15、L−ロイシン35〜45、L−リジン12〜16、L−メチオニン3.0〜10、L−フェニルアラニン3.5〜8.0、L−スレオニン11〜14、L−バリン8.5〜15、L−トリプトファン0.0〜1.0、L−アルギニン0.0〜10。 - アミノ酸含有組成物を3〜11g含有する、請求項1記載のアミノ酸含有組成物。
- 1回あたり3〜11gのアミノ酸含有組成物を、1日あたり7〜22g摂取するように用いられる請求項1記載のアミノ酸含有組成物。
- 毎日続けて経口摂取される、請求項3記載のアミノ酸含有組成物。
- さらに、賦形剤、矯味剤又は色素を含有する、請求項1〜4のいずれか1項記載のアミノ酸含有組成物。
- 粉体混合物、液体混合物又はゼリーの形態である、請求項1〜5のいずれか1項記載のアミノ酸含有組成物。
