Description
幼児や小児における細気管支炎や肺炎のような重篤な兆候を含んでなる下部気道感染の3つの主要な原因は、大きい順に、呼吸器多核体ウイルス(RSV)、ヒトのメタニューモウィルス(hMPV)、及びパラインフルエンザウイルス(PIV)ファミリーのメンバーである。本開示は、パラミクソウイルスファミリーのウイルス病原体に対して防御し、免疫スケジュールの最適化を提供して日常の免疫付与に匹敵するワクチン接種スケジュールに従って乳児の防御を増強する組み合わせワクチンを記載する。
用語
本来、パラミクソウイルスのFタンパク質は、F0と命名された単一のポリペプチド前駆体として発現される。生体内で、F0は、小胞体にてオリゴマー化され、フリンプロテアーゼによってタンパク分解処理を受けて、2つのジスルフィド結合断片からなるオリゴマーを生成する。RSVのFのF0前駆体が2つのフリン認識部位で切断されてpep27と命名された介在ペプチドを放出するのに対して、ほかのパラミクソウイルス(PIV及びhMPVのタンパク質を含んでなる)のFタンパク質は、単一部位で切断される。フリン切断によって生じる2つの断片の小さい方はF2と呼ばれ、F0前駆体のN末端を起源とする。大きい方のC末端F1断片は、24アミノ酸の細胞質尾部に隣接する疎水性アミノ酸の配列を介して膜にてFタンパク質を固定する。略記、F0、F1及びF2は科学文献では一般にF0、F1及びF2と示されることが当業者によって認識されるであろう。
本開示の別の態様は、上述のようなPreF抗原をコードする組換え核酸に関する。さらに明確には、そのような核酸は、パラミクソウイルスのFタンパク質ポリペプチドのF2ドメイン及びF1ドメインを含んでなるFタンパク質ポリペプチド抗原を含んでなるポリペプチドをコードし、それは、(i)ヘテロ三量体化ドメインを含んでなるアミノ酸配列の付加、(ii)少なくとも1つのフリン切断部位の欠失、(iii)少なくとも1つの非フリン切断部位の欠失、及び(iv)Fタンパク質の細胞外ドメインの疎水性ドメインにおける親水性アミノ酸の少なくとも1つの置換又は付加から選択される少なくとも1つの修飾を含んでなる。任意で、そのようなポリヌクレオチドはグリコシル化部位で修飾を伴うPreF抗原をコードする。さらに、RSVのPreF抗原の場合、修飾は、pep27ドメインの1以上のアミノ酸の欠失も含んでなる。そのようなポリペプチドの性質及び構造の詳細は上記で詳細に開示している。当業者は、配列表で提供される、さもなければ本開示に(たとえば、参照による組み入れによって)含まれる例となる配列を含んでなる本明細書での教示に基づいた記載されるポリペプチド配列のいずれか及びすべてをコードするヌクレオチド配列を容易に決定することができる。
組換えタンパク質の発現及び精製について定評のある手順を用いて、本明細書で開示されるPreF抗原(PreF−G抗原及び適宜、G抗原も含んでなる)を製造する。当業者を導くのに十分な手順は以下の文献で見つけることができる:SambrookらのMolecular Cloning:A Laboratory Manual,Cold Spring Harbor Laboratory Press,Cold Spring Harbor,NY,200;及びAusubelらのShort Protocols in Molecular Biology、第4版、John Wiley & Sons,Inc.,999。追加の及び特定の詳細は以下に提供される。
本明細書で開示されるPreF抗原は、免疫原性組成物の製剤化、特に、幼児における重篤な下部呼吸器疾患を含んでなる呼吸器疾患の有意な原因である少なくとも2つの異なったパラミクソウイルスに由来する抗原との組み合わせを含んでなるものの製剤化で有用である。通常そのような免疫原性組成物は、少なくとも2つのパラミクソウイルス及び薬学上許容可能なキャリア又は賦形剤を含んでなる。たとえば、特定の実施形態では、免疫原性組成物は、hMPV抗原及び、たとえば、PIV−3抗原又はPIV−1抗原のようなPIV抗原を含んでなる。他の実施形態では、免疫原性組成物は、RSV抗原及び、hMPV抗原又はPIV抗原(たとえば、PIV−3抗原又はPIV−1抗原)のいずれかを含んでなる。他の実施形態では、免疫原性組成物は、3つの抗原を含んでなる。たとえば、特定の実施形態では、免疫原性組成物は、hMPV抗原、PIV抗原、及びRSV抗原(たとえば、hMPV抗原、PIV−3抗原、及びRSV抗原)を含んでなる。別の実施形態では、免疫原性組成物は、hMPV抗原と2つの異なったPIV抗原(たとえば、PIV−3抗原及びPIV−1抗原)を含んでなる。そのような組成物は任意でRSV抗原を含んでなることもできる。好ましくは、本明細書で開示されるように、抗原はパラミクソウイルスのPreF抗原である。
実施例1:例となるPreF抗原
呼吸器多核体ウイルス
例となるパラミクソウイルスPreF抗原は、あらゆる目的で本明細書に組み入れられるPCT/CA2008/002277で記載されたようにRSVのFタンパク質に基づいて作出した。Fの前融合立体構造に対して生成される免疫応答が、膜の融合に関与する結合、立体構造シフト及び/又はそのほかの事象を妨げる抗体を優先的に含んでなり、それによって防御的応答の有効性を高めるという予測に基づいて、前融合立体構造でタンパク質を安定化するためにRSVのFタンパク質にて修飾を行った。
hMPV及びPIV−3の融合タンパク質に相当する追加のPreF抗原を作出した。これらのPreFポリペプチドの配列はそれぞれ配列番号12及び14によって表される。RSVのPreF抗原で示したように、hMPV及びPIV−3のPreFポリペプチドは溶液中で三量体に自己形成する。

Claims (24)
- ヒトのメタニューモウイルス(hMPV)、パラインフルエンザウイルス3(PIV−3)及び呼吸器多核体ウイルス(RSV)からなる群から選択される異なるパラミクソウイルスの少なくとも2つのFタンパク質抗原を含んでなり、
前記少なくとも2つのパラミクソウイルスのFタンパク質抗原がそれぞれ、パラミクソウイルスのFタンパク質ポリペプチドのF2ドメインおよびF1ドメインを含んでなるFタンパク質ポリペプチドを含んでなり、
hMVPおよびPIV−3のFタンパク質ポリペプチドは、天然に存在するFタンパク質前駆体(F0)に存在するフリン切断部位を消失させる少なくとも1つのアミノ酸の付加、欠失又は置換を含んでなり、ヘテロコイルドコイル三量体化ドメインをさらに含んでなり、
RSVのFタンパク質ポリペプチドは、膜貫通ドメインを欠いており、かつ、天然に存在するFタンパク質前駆体(F0)に存在するフリン切断部位を消失させるpep27の欠失、少なくとも1つのアミノ酸の付加、欠失又は置換を含んでなり、ヘテロコイルドコイル三量体化ドメインをさらに含んでなる、
免疫原性組成物。 - 前記ヘテロ三量体化ドメインが、F1ドメインのC末端に位置する、請求項1に記載の免疫原性組成物。
- 前記Fタンパク質ポリペプチドの少なくとも1つが、前記F2ドメインと前記F1ドメインの間に未変化の融合ペプチドを含んでなる、請求項1または2に記載の免疫原性組成物。
- 前記Fタンパク質ポリペプチドの少なくとも1つが、
(a)少なくとも1つの酵素切断部位の欠失、
(b)Fタンパク質の細胞外ドメインの疎水性ドメインにおける親水性アミノ酸の少なくとも1つの置換又は付加、及び
(c)グリコシル化を変化させるアミノ酸置換
から選択される少なくとも1つの修飾を含んでなる、請求項1〜3のいずれか一項に記載の免疫原性組成物。 - 前記少なくとも2つのパラミクソウイルスのFタンパク質抗原が、ヒト呼吸器多核体ウイルス(RSV)のFタンパク質抗原と、ヒトのメタニューモウイルス(hMPV)のFタンパク質抗原及びパラインフルエンザウイルス3(PIV−3)のFタンパク質抗原のうち少なくとも1つとを含んでなる、請求項1〜4のいずれか一項に記載の免疫原性組成物。
- 前記少なくとも2つのパラミクソウイルスのFタンパク質抗原が、ヒトのメタニューモウイルス(hMPV)のFタンパク質抗原及びパラインフルエンザウイルス3(PIV−3)のFタンパク質抗原を含んでなる、請求項1〜5のいずれか一項に記載の免疫原性組成物。
- 前記Fタンパク質ポリペプチドの少なくとも1つが、天然に存在するFタンパク質前駆体(F0)に存在するフリン切断部位を消失させる少なくとも1つのアミノ酸の付加、欠失又は置換を含んでなる、請求項1〜6のいずれか一項に記載の免疫原性組成物。
- 前記Fタンパク質ポリペプチドの少なくとも1つが、グリコシル化を変化させる少なくとも1つの修飾をさらに含んでなる、請求項1〜7のいずれか一項に記載の免疫原性組成物。
- 前記少なくとも2つのパラミクソウイルスのFタンパク質抗原の1以上が、
(a)配列番号10を含んでなるか、又はそれからなるRSVのFタンパク質ポリペプチド、配列番号12を含んでなるか、又はそれからなるhMPVのFタンパク質ポリペプチド、及び配列番号14を含んでなるか、又はそれからなるPIV−3のFタンパク質ポリペプチド、
(b)配列番号9によってコードされるRSVのFタンパク質ポリペプチド、配列番号11によってコードされるhMPVのFタンパク質ポリペプチド、配列番号13によってコードされるPIV−3のFタンパク質ポリペプチド、及び配列番号9、11及び13の少なくとも1つにストリンジェントな条件下で実質的に全長にわたってハイブリダイズするポリヌクレオチド配列によってコードされるPreFタンパク質ポリペプチド;並びに
(c)配列番号10と少なくとも89%の配列同一性を持つPreFポリペプチド、配列番号12と少なくとも94%の配列同一性を持つPreFポリペプチド、及び配列番号14と少なくとも95%の配列同一性を持つPreFポリペプチド
からなる群から選択される、請求項1〜8のいずれか一項に記載の免疫原性組成物。 - 前記Fタンパク質抗原の少なくとも1つが、ポリペプチドの多量体を含んでなる、請求項1〜9のいずれか一項に記載の免疫原性組成物。
- 前記Fタンパク質抗原の少なくとも1つが、ポリペプチドの三量体を含んでなる、請求項1〜10のいずれか一項に記載の免疫原性組成物。
- キャリア又は賦形剤をさらに含んでなる、請求項1〜11のいずれか一項に記載の免疫原性組成物。
- アジュバントをさらに含んでなる、請求項1〜12のいずれか一項に記載の免疫原性組成物。
- 前記免疫原性組成物が、hMPV、PIV−3及びRSVのうち2以上による感染を軽減する又は予防する、請求項1〜13のいずれか一項に記載の免疫原性組成物。
- 前記免疫原性組成物が、hMPV、PIV−3及び/又はRSVによる感染に続く病的な症状又は疾患を軽減する又は予防する、請求項1〜14のいずれか一項に記載の免疫原性組成物。
- 請求項1〜11のいずれか1項に記載の組換えパラミクソウイルスFタンパク質抗原をコードするポリヌクレオチド配列を含んでなる、組換え核酸。
- 前記核酸が、
(a)配列番号9、配列番号11及び配列番号13を含んでなるか、又はそれらからなるポリヌクレオチド配列;
(b)(a)のポリヌクレオチド配列にストリンジェントな条件下で実質的に全長にわたってハイブリダイズするポリヌクレオチド配列;
(c)(a)のポリヌクレオチド配列と少なくとも90%の配列同一性を持つポリヌクレオチド配列
から選択されるポリヌクレオチド配列を含んでなる、請求項16に記載の組換え核酸。 - 請求項1〜11のいずれか一項に記載のFタンパク質抗原または請求項16又は17に記載の核酸を含んでなる、RSV感染の治療に用いるための医薬組成物。
- hMPV、PIV−3及びRSVのうち1以上による感染を予防的に治療する目的で投与される、請求項18に記載の医薬組成物。
- 対象においてhMPV、PIV−3及びRSVのうち1以上に対する免疫応答を誘発することに用いるための請求項1〜15に記載の免疫原性組成物。
- 前記免疫応答がTh1優位の免疫応答である、請求項20に記載の免疫原性組成物。
- 前記免疫応答が、hMPV、PIV−3及びRSVのうち1以上による感染を軽減する又は予防する防御的免疫応答を含んでなる、請求項20又は21に記載の免疫原性組成物。
- 医薬に使用するための請求項1〜11のいずれか一項に記載の免疫原性組成物。
- RSVに関連する疾患の予防又は治療に使用するための請求項1〜11のいずれか一項に記載の免疫原性組成物。
