実施例1
方法
ヒトの記憶B細胞を末梢血または扁桃腺から、最初にCD19 MACS(登録商標)ビーズ(Miltenyi Biotech)を使用するB細胞の正の選択によって精製する。次に記憶B細胞を、IgGを対象とする表面染色および細胞選別によって選択する。次にIgG+ B細胞を、CD40Lを発現するマウス線維芽L細胞とともに、マウスまたはヒトのIL-21の存在下で36〜48時間にわたって培養する。次に細胞を、Retronectin(登録商標)(Takara, Shiga, Japan)でコーティングされた組織培養プレートに移し、そこでヒトBCL6-IRES-GFPをコードするレトロウイルスで37℃で16時間かけて形質導入を行う。次に形質導入細胞をCD40L-L細胞上で、ヒトIL-2およびヒトIL-4の存在下で培養する。約3〜4週間後にGFP+細胞(すなわちBCL6+細胞)が培養100%に達し、その後はBCL6+細胞をIL-2およびIL-4とともに、またはヒトまたはマウスのIL-21とともに培養する。発明者らはフローサイトメトリーで、GFP、CD19、CD38、CD20、MHCクラスII、CD27(BD Biosciences)、および他のマーカーの発現のモニタリングを、標識抗体を使用して行う。発明者らは、細胞数を数えて成長をモニタリングし、およびIgの産生のモニタリングを、培養上清中におけるIgの酵素ELISA検出で行う(Dako, Glostrup, Denmark)。遺伝子の発現は、逆転写酵素ポリメラーゼ連鎖反応(RT-PCR, Invitrogen, Breda、Netherlands)によってモニタリングする。
記憶B細胞へのBCL6の導入は、培養物中における、正常B細胞を上回る、寿命のかなりの延長につながる(数か月対約3週間)。このような細胞は、CD19、表面Ig、MHCクラスIIを保持し、ならびに中レベルのCD38およびCD20を発現することから、記憶細胞の表現型を有することが示唆される(データは示していない)。IL-21の存在下における、CD40L-L細胞上におけるこれらの細胞の培養は、有意な成長上の利点(図1)、および形質芽細胞様の細胞表面表現型の獲得(0038hi CD20+、図2)につながる。重要な点は、IL-21とともに培養した細胞は、IL-2およびIL-4とともに培養した細胞と比較して300%多いIgGを分泌することである。以上のデータを総合すると、IL-21培養物は不死化B細胞集団中で、成長および抗体産生の亢進を示す形質芽細胞の発生を促進することがわかる。
本発明の1つの態様の非制限的モデルを図4に示す。
材料および方法
ヒトB細胞の維持および単離
標準的な手順で、CD19陽性のヒトB細胞を、血液バンクに由来するバフィーコート(他の供給源には、新鮮なヘパリンまたはACD血液、またはリンパ器官、例えば扁桃腺または脾臓などがある)から単離した。簡単に説明すると、全末梢血単核球(PBMC)をフィコール密度勾配分離(Amersham, Buckinghamshire, UK)で分離した。CD19標識ビーズを使用して、MACS細胞選別法(Miltenyi, Auburn, CA, USA)でB細胞を正に選択した。次に細胞を、CD19、CD27、IgG、IgM、CD3(Becton Dickinson (BD), Franklin Lakes, NJ, USA)、およびフィコエリトリン(PE)標識破傷風トキソイド(A. Radbruch, Berlin, Germanyから供与)、または他の任意の標識抗原に対するモノクローナル抗体(mAb)を適切に組み合わせて染色した。次に細胞をFACSAria(BD)で選別した。選別された細胞を洗浄し、ならびに8%ウシ胎仔血清(FCS)およびペニシリン/ストレプトマイシンを含むイスコフ改変D最小基本培地(IMDM)中の照射CD40L発現L細胞(5x104細胞/ml;DR. J. Banchereau, Schering Plough France, Dardilly Franceから供与)上で培養した(1.5〜2x105細胞/ml)。特に明記した部分を除いて、このようなCD40L発現L細胞は培養物中に常に存在する。
精製済みのB細胞を、caSTAT5b遺伝子で形質導入されるように刺激した。以下の2つの刺激プロトコルを使用した:(1)精製済みのB細胞をインターロイキン(IL)2(20 U/ml, Chiron, Emeryville, CA, USA)とともに3日間にわたって培養後に、IL-2およびIL-4(10 ng/ml, R&D, Minneapolis, MN, USA)とともに24時間にわたって培養するか、または(2)精製済みのB細胞を、組換え型のマウスIL-21(50 ng/ml, R&D)とともに36時間にわたって培養した。次に細胞を、組換え型のヒトフィブロネクチン断片CH-296(Hanenberg H., Nat., Med. 1996;RetroNectin, Takara, Japan)、およびヒト血清アルブミンで処理したプレート(Corning Life Sciences, Corning, NY, USA)上に、L細胞の非存在下でサイトカインIL-2/4またはIL-21とともに添加した。最後に細胞の形質導入を、エストロゲン受容体(ER、H. Kurata, DNAX Institute, Palo Alto, CA, USAから供与)と融合させたcaSTAT5b遺伝子(Ariyoshi K., JBC, 2000に記載、およびT. Kitamura, IMSUT, Tokyo, Japanから入手)を使用して行った。caSTAT5b-ER融合産物の活性は、ホルモンであるヒドロキシタモキシフェン(4HT, Sigma-Aldrich, St. Louis, MO, USA)によって制御可能である。形質導入は、文献(Heemskerk M.H., JEM, 1997;Heemskerk M.H., Cell Immunol. 1999;Scheeren F.A., Nat Immunol, 2005)に記載された手順で、レトロウイルスを使用して実施した。形質導入効率は、神経成長因子受容体の切断型のシグナル伝達不能変異体(ΔNGFR、C. Bonini, St. Raphael Hospital, Milan、Italyから供与)を抗体で染色して決定した。したがって、caSTAT5b遺伝子を含むB細胞の成長は4HTの存在に依存し、およびこのような細胞は、NGFR(Chromaprobe, Maryland Heights, MO, USA)に対する抗体染色によって検出可能である。
発明者らは、モノクローナル抗体を産生し、かつ100% caSTAT5b(=NGFR)陽性のB細胞株を開発した。これは、B細胞を記憶B細胞から抗体産生表現型に分化させ、およびcaSTA5b-ER-IRES-NGFRコンストラクトを使用する形質導入を行うことで達成された。caSTAT5bの作用のために、分化したB細胞は細胞死に対して非感受性となる。B細胞の分化は、サイトカイン混合物(IL-2、IL-4、IL-21、またはこれらのサイトカインとCD40Lの組み合わせ)を使用して、単離後の最初の2〜3週間で誘導される(図5)。4HTの添加によってcaSTAT5bが活性化される時点は、培養物の全体的な表現型に影響する。これはcaSTAT5bによって細胞の表現型の変化が阻害される、例えばさらなる分化が阻害されるためである。したがって、より長い期間にわたって4HTが除去されると、より多くのB細胞が抗体産生細胞に、または、このような培養条件で選択的に成長する細胞タイプに分化する(細胞タイプの例:ナイーブ細胞、濾胞細胞、記憶細胞、抗体産生細胞、血漿芽細胞、形質細胞、周辺帯細胞、類洞周囲細胞、または移行B細胞(これらのB細胞サブセットの多くはマウスでのみ確認されている)。培地中に4HTが存在すると、caSTA5b-ER-IRES-NGFR陽性のB細胞は、長期間の生存が可能となる(表2)。
caSTAT5b-ER-IRES-NGFRが形質導入されたヒトB細胞培養物の概略。PBMCを血液バンク由来のバフィーコートのフィコール勾配分離後に得た後に、CD19 MACSおよびCD27による選別、またはFACSAriaによる細胞選別を行った。次に、精製済みのB細胞をL細胞の存在下で、記載のサイトカインとともに培養した後に、caSTAT5b-ER-IRES-NGFR遺伝子コンストラクトを含むレトロウイルスによる形質導入を行った。
caSTA5b-ER-IRES-NGFR陽性B細胞の成長は一般に約4週間を要し、この後にクローン培養物が、限界希釈(LD)培養、またはフローサイトメトリー(FACSAria)による1細胞選別を行うことで得られる。このような培養物は、2500〜5000個のL細胞、正常濃度のIL-2およびIL-4、ならびに96ウェルあたりの細胞数が1個、5個、または10個(LDが100% NGFR+細胞を使用して実施時)、ならびに96ウェルあたりの細胞数が10個、100個、および1000個(NGFR+細胞がFACSAriaを使用して96ウェルに選別時)のいずれかからなる。
抗体産生が陰性か、または低いポリクローナル、オリゴクローナル、またはモノクローナルのcaSTAT5b-ER-IRES-NGFR陽性B細胞培養物を十分に洗浄後に、培養物から(1)4HT、IL-2、およびIL-4を除去後にIL-21とともに培養し、4〜10日後に上清をIgMおよびIgGの産生に関して検討するか、または(2)4HTを10日間かけて除去しながら、IL-2およびIL-4とともに培養し、ならびに10日目に、IL-2およびIL-4をIL-21と交換する。次に、さまざまな時点において、上清をIgMおよびIgGの産生に関して検討した。
B細胞の分化および増殖;「IL-2およびIL-4プロトコル」対「IL-21プロトコル」
IL-21で処理されたB細胞培養物は、IL-2およびIL-4の場合と比較して、最初の2〜3週間以内に増殖反応の亢進が認められた(図6a)。しかしながら、IL-2およびIL-4を使用時の培養物とは異なり、IL-21による連続的な刺激は、活性STAT5bの存在下であっても、増殖および細胞死の低下につながった(図6b)。IL-21は最終的に、IL-2およびIL-4と交換しなければならないことが示唆された。この仮説を詳細に検討するために、CD19+CD27+記憶B細胞を対象に、IL-21をIL-2およびIL-4と、36時間後、または5日後、10日後、15日後、および20日後に交換する時系列実験を行った。図7に示すように、大半の培養物は、IL-21が20日間にわたって添加された培養物であっても、IL-21の除去後に維持可能であった。
興味深いことに、IL-2およびIL-4を使用した培養物とは対照的に、IL-21で増幅された培養物は、抗体を比較的長期間にわたって産生可能であった(IgGおよびIgMはELISA(Dako, Glostrup, Denmark)で測定)(それぞれ図8aおよび図8b)。重要な点は、ドナーB18およびB19のポリクローナルの記憶B細胞培養物のうち、1細胞クローンがLD培養によって得られたことである(表3)。
CD19+CD27+NGFR+選別B細胞から単離されたクローンの頻度。細胞は96ウェル中に、1ウェルあたりの細胞数が1000個、100個、10個、または1個のいずれかとなるように選別した。ウェルは、5000個のCD40L発現L細胞、IL-2およびIL-4を含む。培地の1/4〜1/2を週に2回、新鮮なサイトカインおよび2500個のL細胞と交換した。
次に発明者らは、破傷風トキソイド(TT)に特異的な抗体を産生するB細胞を単離可能か否かを検討した。簡単に説明すると、以下のプロトコルを実施した:
パート1
(1)CD27+TT+ B細胞を選別し(回収率はドナー依存性であり、10000〜1000個の細胞)、
(2)IL-21とともに36時間にわたって培養し、
(3)caSTA5b-ER-IRES-NGFRにより形質導入を行い、ならびに
(4)時間を変えて(36時間〜3週間)IL-21とともに培養後、IL-21をIL-2、IL-4、および4HTと交換し、

(5)培養物が100% NGFR+の場合は、限界希釈(LD)でクローニングした。
100% NGFR+ TT特異的B細胞の限界希釈培養。単離クローンの総数、および単離条件(1細胞/ウェル、5細胞/ウェル、または10細胞/ウェルのいずれか)を示す。各条件(1細胞/ウェル、5細胞/ウェル、または10細胞/ウェル)につき1枚の96ウェルプレートを使用した。ウェルは、2500個のCD40L発現L細胞、IL-2およびIL-4を含む。培地の1/4〜1/2を週2回、新鮮なサイトカイン、および2500個のL細胞と交換した。
発明者らは、IL-21を刺激として使用して、IgMおよびIgGを産生するポリクローナルおよびモノクローナルのB細胞培養物を作製することもできた。それにもかかわらず、抗体産生は安定ではなかった。しかしながら驚くべきことに、これらのIL-21処理培養物は再刺激されると、IgG抗体およびIgM抗体を産生可能となった(それぞれ図12aおよび図12b)。これは、4HTの除去、およびIL-21による同時刺激によって達成された。このプロトコルを採用したところ、全抗体産生は、IgMについては2〜1000倍、およびIgGについては2〜25倍上昇した。再刺激されたモノクローナル培養物の上清の一部は、この時点で、IgGおよびIgM破傷風ELISAで陽性と判定された(図13)。
材料および方法
以下に関する方法の詳細は、実施例3に記載されている:
・ヒトB細胞の維持および単離;
・B細胞におけるマウスの構成的に活性なSTAT5bの形質導入および調節;
・抗体を分泌する100% caSTAT5b陽性B細胞株の作製;
・1細胞由来のクローン性B細胞培養物の作製;ならびに
・caSTA5b-ER陽性B細胞培養物による抗体産生の再刺激。
精製済みの初代CD19+CD27+ B細胞(B細胞)をIL-21、および照射済みCD40L発現L細胞(L細胞)で36時間にわたって刺激した後に、BCL6-IRES-NGFRまたはcaSTAT5b-ER-IRES-NGFRによる形質導入を行った。形質導入後に、BCL6形質導入細胞はIL-21とともに培養し、ならびにcaSTAT5b-ER形質導入細胞はIL-2およびIL-4とともに培養した。形質導入細胞は、2〜3日以内にNGFR陽性となり、および続けてFACSAriaで選別を行った。NGFR選別細胞を次に、100〜5000細胞/96ウェルの細胞密度で培養した(ミニバルク培養(mini bulk culture; MBC))。BCL6/IL-21によるMBCは、caSTAT5b-ER培養物と比較して強く増殖した。これらの培養物を抗体産生に関して検討し、24ウェルに拡張して凍結した(生細胞、細胞ペレット、および上清)。caSTAT5b-ER培養物を拡張し、2枚の平行96ウェルに分割した。1つのウェルでは、IL-2、IL-4、および4HTとともに培養し、ならびに別のウェルでは、IL-21とともに培養した(4HT非添加)。
強い増殖反応がEBVの存在と関連するか否かを検討するために、EBV RT-PCRを実施した。溶解したペレットからRNeasyミニキット(Qiagen)を使用して全RNAを単離した。RNAの逆転写を、5種類のファーストストランド緩衝液、500 μM dNTP、25 μg/l オリゴ(dT)、および200 U superscript II RT(Life Technologies)を含む20 μlの容量で行った。cDNA溶液の一部(1 μl)を、20 mM Tris-HCl、50 mM KCl、1.5 mM MgCl2、5 mM dNTP、2.5 U Taq DNAポリメラーゼ(Life Technologies)、および30 pmolの各プライマーを含む50 μlの溶液中でPCR増幅した。以下のPCR条件を用いた:94℃で7分間の変性段階と、これに続く94℃で30秒間、62℃で30秒間(HPRT1)、52℃(LMP-1)および58℃(EBNA1/2)、および72℃で30秒間を30サイクル、ならびに最終的な72℃で7分間の伸長。RT-PCRには以下のオリゴヌクレオチドを使用した。
発明者らの系におけるEBVの果たす役割を詳細に調べるために、発明者らは、天然のEBVを含むか、または含まない培養物から得られた非形質導入細胞、BCL6-IRES-NGFR形質導入細胞、およびcaSTAT5b-ER-IRES-NGFR形質導入細胞の増殖および抗体産生を決定するための実験を準備した。
IL-21およびL細胞を添加したBCL6 MBC(培養物Ce2-B9および培養物Ce2-G9)
IL-21、L細胞、およびEBVを添加したBCL6 MBC(培養物Be3-F3および培養物Be2-G9)
IL-21およびL細胞を添加したEBV MBC(培養物B28および培養物B29)
IL-2およびIL-4を添加した培養物も含めた。抗体レベルおよび細胞数を1週間間隔で決定した(表7)。培養物のEBV状態を図14に示す。
BCL6培養物のEBV感染のPCR判定
BCL6の形質導入と組み合わせて、2人のドナーを対象に発明者らが検討した、強いB細胞の増殖および分化をIL-21が誘導することが判明した(表5)。発明者らは実際に、発明者らがB細胞を1000 c/wおよび100 c/wの密度で培養した培養条件でB細胞の強い成長を見出した(表7)。しかしながら、1人のドナーでは、ほぼ全ての培養物がEBVに感染していることが、光学顕微鏡による表現型、培養物の上清の色、および多数の細胞の増殖を元に疑われた。同ドナー(B29)は実際にEBV感染者であることが判明した(図14)。EBV感染細胞の活発な成長から、BCL6とIL-21の組み合わせが、EBV感染細胞に成長上の利点をもたらすことがわかる。なぜなら特に、インビボにおけるEBV感染B細胞の頻度は比較的低いと考えられているからである。ドナーB29とは対照的に、ドナーB30はEBV陰性であったが、弱くて比較的小さいLMP1のバンドが試料Ce2-F2中に認められた(図14)。
細胞の成長に対してEBV、BCL6、IL-2、IL-4、およびIL-21が果たす役割を調べるために、増殖の動態を比較した(表5の実験設定を参照)。試料を、LMP-1 EBV PCR反応を元に選択した(図14)。EBV状態は、EBV感染細胞がL細胞の非存在下で成長する能力によって確認した。IL-2およびIL-4とともに培養したBCL6試料は、低い増殖能力を示し、維持不可能であった(表7)。IL-21を含む培地で培養した全ての試料には強い増殖が認められた。IL-2およびIL-4は、細胞が、BCL6との組み合わせではなく、EBVに感染している場合にのみ強い増殖を誘導可能であった。
EBVを含むか含まない場合の、または、IL-2およびIL-4、もしくはIL-21を含むか含まない場合の、BCL6形質導入B細胞および非形質導入B細胞の増殖;全ての試料がL細胞を含む。
IgG抗体産生を、長期の培養物を対象に、表5に記載された手順で判定した。記載の抗体産生レベルは、2人のドナーにおける6〜10の異なる測定結果の抗体産生の平均である(図15)。BCL6/EBV/IL-21培養物がBCL6/IL-21培養物より有意に多いIgGを産生することが明らかである。したがってEBVは、BCL6形質導入B細胞の抗体産生を有意に促進する。したがって、IL-21とEBVの組み合わせは、BCL6の非存在下ならびに存在下で高レベルの抗体産生につながる。
EBV感染細胞がBCRの発現を喪失することが報告されている。したがって、IL-21含有培養物中のB細胞(表5に記載)を、NGFR、CD19、カッパ、およびラムダに関して染色を行った。
BCL6の形質導入と平行して、caSTAT5b-ERを使用した形質導入を行った。興味深いことに、全てがEBVに感染していたBCL6培養物とは対照的に、同じドナー(B29)のcaSTAT5b-ER培養物は、EBV感染細胞の天然の分布(パーセンテージ)を維持しているようであった。複数の培養物に、EBV感染の明瞭な特徴が認められ、ならびにLMP1のPCRによる確認の結果、実際に陽性であることが判明した。caSTAT5b-ER培養物がEBV陽性である兆候の1つは、MBCが、4HTの非存在下でIL-21処理した際に生存する能力であった。このような4HT除去B細胞は、活性型のcaSTAT5bを欠き、および通常2〜3週間以内に死滅する。
EBVがcaSTAT5b-ER系において果たす役割を調べるために、表6に記載された手順で実験を行った。caSTAT5b-ER形質導入B細胞の反応の図式的概観を図8に示す。4HTの存在による、活性なcaSTAT5b-ERは、どのサイトカインが存在しても、またはB細胞がEBVに感染していてもB細胞の分化を阻害する。したがって、EBVに感染していて、IL-2およびIL-4、またはIL-21によって(4HTは非添加)維持されたcaSTAT5b-ER培養物は抗体を産生しない。4HTの除去は、分化と、分化に続く抗体産生につながる。好ましくは、4HTの除去中および/または除去後にIL-21が添加される。しかしながら、このような細胞は最終的には死滅する。なぜなら、caSTAT5bが不活性であるために、抗体が一定の期間しか産生されないからである。したがって、もしEBVが存在しない場合は、最終的にはIL-21が、実施例3で既に言及した、例えばIL-2やIL-4などの少なくとも他の1つの成長刺激因子と交換される。
方法
BCL6-IRES-YFP陽性細胞の形質導入を、STAT3/ER-IRES-GFPを使用して標準的な手順で行い、ならびに実施例1の方法セクションに記載された手順によってL細胞上でIL-2およびIL-4とともに増殖させた。STAT3の発現は、タモキシフェン(4HT)の有無によって調節された。YFP陽性細胞およびGFP陽性細胞を選別し、および等しい数を培養した。BLIMP1遺伝子の発現をRT-PCRでモニタリングし、ならびに抗体産生を、4HTの添加時および非添加時の培養物を対象に判定した。
細胞への4HTの添加は、細胞数の増加につながり、Blimp-1の発現を高めた。さらにIgG産生の促進が、図19a〜cに示すように測定された。
CD19陽性B細胞の形質導入を、対照となるYFP-IRES-YFP(cYFP);BCL6-IRES-YFP(BCL6-YFP)またはBcl-xL-GFP(Bcl-xL-GFP)を使用して行った。次に細胞を、CD40L上でIL-4を加えて維持し、ならびにYFPおよびGFPの単一陽性細胞および二重陽性細胞のパーセントを、非選別バルク培養物を対象としたFACSで経時的に決定した。
図20〜23から、BCL6およびBcl-xLを含む二重形質導入B細胞が、より高い累積的増殖率を示し、ならびに実際に、バルクの記憶B細胞培養物中で成長を示す優性な細胞でもあることがわかる。発明者らは、このような二重形質導入細胞が、単一形質導入細胞と比較して2倍速く分裂することも示す。IL-4またはIL-21と培養時に、BCL6培養物とBCL6/Bcl-xL培養物間で等しい抗体産生レベル(IgGおよびIgM)は、ここには示していない。
チロシンリン酸化型のSTAT5が陽性のホジキン細胞株L591を、L細胞(CD40刺激)およびサイトカインに依存せずに培養した。L591細胞の形質導入を、E47-IRES-GFPを含むレンチウイルス、またはGFPのみを有する対照ウイルスで行った(方法の詳細は実施例1に記載)。形質導入細胞を選別し、および細胞の成長を経時的に追跡した。
L591細胞がE47の発現に伴って速やかに分裂を停止することを図24に示す。これはE47の、その下流の標的(例えば、Socs1、Socs3、Id2、Eto2、およびXbp1)を介する作用が、B細胞の、抗体産生性のB細胞の表現型への分化を誘導することを示唆する。分化がE47によって誘導されたことは、STAT5の作用が消失したこと、またはE47が直接的または間接的に、機能性のSTAT5の量および作用に影響したことも意味する可能性がある。したがって、ホジキン細胞株L591に関して得られたデータを元に発明者らは、4HT、ならびにIL-2およびIL-4、またはIL-21が添加されたL細胞上で維持されるSTAT5b/ER陽性B細胞培養物が、E47が活性な場合に、抗体産生細胞に分化するように誘導され得ると主張したい。
Claims (27)
- 抗体産生B細胞にBCL6をコードする核酸配列を提供すること、ならびに
該抗体産生B細胞をIL-21および/若しくはIL-10の存在下で培養することにより、または該抗体産生B細胞にSTAT3をコードする核酸配列を提供することにより、該抗体産生B細胞内におけるBlimp-1の発現産物の量を高めること、
を含む、該抗体産生B細胞の複製寿命を延長する方法。 - BCL6およびBlimp-1が該B細胞において共発現する、請求項1記載の方法。
- 該B細胞に追加の不死化剤を提供することを含む、請求項1または2記載の方法。
- 不死化剤が形質転換剤を含む、請求項3記載の方法、
- 不死化剤がエプスタイン・バーウイルスを含む、請求項3または4記載の方法。
- 該B細胞をIL-21の存在下で培養した後、BCL6をコードする核酸配列を該B細胞に提供する、請求項3〜5のいずれか一項記載の方法。
- 該B細胞をIL-21の存在下で培養し、かつ該B細胞にエプスタイン・バーウイルスを提供する、請求項1〜6のいずれか一項記載の方法。
- 該B細胞にBcl-xLをコードする核酸配列を提供することをさらに含む、請求項1〜7のいずれか一項記載の方法。
- ナイーブB細胞または記憶B細胞を提供すること;
該B細胞をIL-21および/若しくはIL-10の存在下で培養することにより、または該B細胞にSTAT3をコードする核酸配列を提供することにより、該B細胞におけるBlimp-1の発現レベルを高めること;および
