免疫原およびアジュバントをコードするPNSを保有するマイコバクテリウムベクター類を構築することが可能で、それらは、前記ベクターおよびPNSがコードした免疫原に対して強化宿主応答を惹起する点で有用である。これとは別に、アジュバントをコードするPNSを保有するマイコバクテリウムベクター類を構築することが可能で、それらは、少なくとも1個の免疫原をコードするPNSを保有する他のマイコバクテリウムベクター類と混合して投与することが可能で、パートナーの前記マイコバクテリウムベクターによってコードされた前記免疫原に対する宿主応答を高める。
さらに別のアプローチでは、免疫原およびサイトカインをコードする少なくとも1個のPNSを保有するマイコバクテリウムベクターの用途を包含し、それらを用いて、PNSがコードした免疫原マイコバクテリウムベクターに対する強化宿主応答を惹起する。これとは別に、サイトカインのみをコードするPNSを保有するマイコバクテリウムベクターを構築することもでき、免疫原をコードするPNSを保有する少なくとも1個の他のマイコバクテリウムベクターとそれらを混合し用いて、パートナーであるマイコバクテリウムベクターによって発現されたPNSがコードした免疫原類に対する宿主応答を高めることもできる。
下記の実施例は、本発明のさまざまな面を例示するためとみなすべきであり、本発明の実施に関して限定することを意図していない。当業者は、これらとは別の材料類、条件類、および操作に変化させることもでき、それら変形が、本明細書の教示として本発明の概括的範囲から逸脱することなく可能であることを認識するであろう。
下記の章で述べた各実験のため、制限エンドヌクレアーゼ類(本文で、“REs”);New England Biolabs Beverly,MA)、T4 DNAリガーゼ(New England Biolabs Beverly,MA)、およびTaqポリメラーゼ(Life Technologies,Gaithersburg,MD)を製造業者の指示に従い用いた;プラスミドDNAは、小型(Qiagen MiniprepRキット、Santa Clara、CA)または大型(Qiagen MaxiprepRキット、Santa Clara、CA)プラスミドDNA精製キット類を製造業者のプロトコール(Qiagen、Santa Clara、CA)に従い用いて、調製した;ヌクレアーゼを含まない分子生物学等級のmilli−Q水、Tris−HCl(pH7.5)、EDTA pH8.0、1M MgCl2,100%(v/v)エタノール、超純粋アガロース、およびアガロースゲル電気泳動緩衝液は、Life Technologies、Gaithersburg,MDから購入した。RE消化、PCRs、DNA連結反応およびアガロースゲル電気泳動は、周知の操作に従った(Sambrookほか、Molecular Cloning:A Laboratory Manual.1,2,3;1989);(Strausほか、Proc Natl Acad Sci USA、Mar;87(5):1889−93;1990)。下記の章で述べる各組換えプラスミドのDNA配列を検証するためのヌクレオチド配列決定は、従来の自動化DNA配列決定法によりApplied Bisystems自動化シークエンサー、モデル373Aを用いて完了した。
先行技術では、マイコバクテリウム株類に改変対立遺伝子類を導入する方法を教示しており、当業者は、このような方法を理解しかつ実行できるだろう(Parishほか、Microbiology 146:1969−1975;2000)。対立遺伝子交換プラスミドを調製するための新しい方法では、合成DNAを用いることを含む。この手法の利点はといえば、調製プラスミドが極めて明らかな調製の経緯を有しており政府規制に沿ったコンプライアンスを有していることであろう。一方これまでに用いられてきた方法は効果的ではあるが、ラボでの培養記録記載は満足のいくものではなく、したがって米連邦規則に適合しているとはいえないであろう。上記規制へのコンプライアンスは、もしヒトでの使用のために製品のライセンスを米国および欧州政府当局から取得しようとするならば、必須である。
選択したBCG株類は、Middlebrook7H9またはSaulton Synthetic Mediumのような液体培地中で好適には37℃において培養する。前記株類は、静止または攪拌培養物として維持できる。さらに、BCGの増殖速度は、オレイン酸添加(0.06%v/v;Research Diagnostics カタログ番号01257)およびTyloxapolのような界面活性剤類の添加(0.05%v/v;Research Diagnosticsカタログ番号70400)によって高めることができる。BCG培養物の純度は、リン酸緩衝生理食塩水(PBSと称する)中で系列希釈(例えば、原液〜10−8の10段階)したBCG培養物のアリコット100mclを、25−30mlのMiddlebrook 7H10のような固体培地を含む3.5インチのプレート上に均等に塗擦することによって、評価できる。さらに、前記培養物の純度は、チオグリケート培地(Science Lab、カタログ番号1891)およびダイズ−カシン培地(BD、カタログ番号211768)などの市販培地を用いて、評価できる。
エンドソームエスケープ可能なrBCG−PfoA株類の構築は、ureC遺伝子の隣接領域の対立遺伝子交換によって実施した。結果として、PfoA遺伝子セグメントはureC遺伝子に置き換わり、PfoAの安定な染色体発現が可能となった。具体的詳細は下記に述べる。
対立遺伝子交換プラスミドは、下記のDNAセグメント類から構成されている:大腸菌中でプラスミドが複製するためのoriE配列、大腸菌およびマイコバクテリウムの両者において選択するためのカナマイシン耐性遺伝子、およびHsp60プロモータによって発現される付加的抗生物質選択マーカー(ゼオシン耐性遺伝子)。第2のマーカーを用いて二重選択を行い、従って、このプロセスにおいてカナマイシン自然耐性を防止する。対立遺伝子交換プロセスにおいて陰性選択のため、ショ糖感受性遺伝子を用いた。最後に、標的BCG Danish1331株のためのureC遺伝子の1kb左および右隣接配列類を、PfoA遺伝子とともにその間に含めた。PfoA遺伝子は、Ag85Bプロモータの制御下に発現する。Ag85Bリーダーペプチド配列をPfoAオリジナル分泌シグナル配列の代わりにPfoA分泌のために用いた。最後に、これらの成分類全てを、Picoscript Inc(Houston,TX)により合成し組み立てた。生成したプラスミドはマイコバクテリウム自殺ベクターで、得られたプラスミドについてそのマップを図1に示した。生成したプラスミド構築体を、図5で示したものであると確認した。
対立遺伝子交換プロセスを図4に概略示し、本操作の主要段階を概略示している。それらの段階について、詳細に説明する。
実施例2のための材料類および方法類
マイコバクテリウムの培養
下記の実験のため、BCG株類を、10%OADC添加Middlebrook 7H9培地(BD biosciences)中37℃で培養した。Tyloxapol(0.05%v/v、Research Diagnostics,カタログ番号70400)を用いて、菌を分散させた。増殖を異なる株間で比較する実験において、接種後異なる時点で光学密度(600nm)を測定した。カナマイシンに対する感受性を試験するため、培養物を調製し増殖を上記のように測定したが、ただし、カナマイシンを最終濃度50μg/mlで添加した。固体培地を用いて菌を培養する時、Middlebrook 7H10寒天(BD biosciences)を用いた。適切である時には、カナマイシンを最終濃度50μg/mlで添加し、ショ糖を最終濃度3%で添加した。
実施例1に述べたショ糖プレートから生成したコロニーを最初に、ウレアーゼ活性欠損について、ウレアーゼ試験キット(BD Difico)を用いて製造業者の指示に従いスクリーニングした。簡単に述べると、1ループの菌を透明試験管中の製造業者供給試験緩衝液中に再懸濁させた。BCG Danish1331株をウレアーゼ陽性コントロールとして用いた。緩衝液単独は、陰性コントロールとして用いた。反応混合物を室温で30分間、インキュベーションし、結果を、製造業者の指示に従い判定した。
フォワードプライマー〔acggctaccgtctggacat〕(配列番号4)およびリバースプライマー〔cgatggcttcttcgatgc〕(配列番号5)によるPCRを行い、Pfo特異的挿入対立遺伝子のDNA配列とureC遺伝子に隣接するBCGゲノムDNA配列類を増幅した。PCRパラメータは下記のようであった:段階1:95℃で4分間を1サイクル;段階2:95℃で1分間、60℃で1分間、およびその後72℃で1分間を総計30サイクル;段階3:72℃で10分間を1サイクル;段階4:4℃で保存。生成したPCR生成物はアガロースゲル電気泳動で分析し、自動化ジデオキシヌクレオチド配列決定法により配列を決定し、ureC遺伝子の代わり全長PfoA遺伝子の存在(すなわち、ΔureC::PfoA)を確認した。
rBCG AFV102株のインサイチュにおける増殖を、感染マクロファージ中におけるマイコバクテリウムコロニー形成単位(CFU)を定量することによって、J774A.1マクロファージ様細胞中で試験した。マイコバクテリウム貪食の効果は、J774A.1細胞の感染3時間後、細胞内CFUを試験することによって、決定した。その後の長期細胞内生存は、先行文献にも説明されているとおり(Sunほか、2004)PBSで5回洗浄後、細胞を溶解させ細胞内菌を放出させ、CFUを計数することによって実行した。
AFV102によるPfoAの分泌を評価するため、本株を上記のように、対数相中期になるまで増殖させた。次に、培養上清と菌を採取した。AFV102菌ペレットを、96ウェルのV底プレート中0.1%BSA含有PBS(pH7.0)100μl中に再懸濁させた。PfoAタンパク質が培養上清に分泌されたかどうかを試験するため、液体培養物を回転して落とし、この上清を試験に用いた。発現PfoAをそのpH非依存性溶血活性について試験するため、サンプルを上記のように調製したが、ただし、反応緩衝液として異なるpH値のPBS緩衝液を用いた。1%洗浄ヒツジ赤血球100μlを各ウェルに添加した。反応物をゆっくりと混合し、37℃で1時間、攪拌しながらインキュベーションした。BCG Danish1331株の菌を、溶血陰性コントロールとして用いた。ヘモライシン活性が公知単位となっているα−ヘモライシン(Sigma)を、溶血陽性コントロールとして系列希釈して用いた。インキュベーション終了時点で、反応物を500gで15分間遠心しペレットとし、次に、V底プレートからの上清を平底96ウェルプレートの同じ位置に移し、光学密度を測定した(450nmの吸光度マイナス540nmにおける吸光度)。PfoA分子の溶血活性は、赤血球溶解後の色変化の光学密度を測定することによって、定量した。測定した色の強度は、赤血球溶解量に比例しており、それは、ヘモライシンの量に比例することになる。公知標準を用いることで、サンプルの値を標準曲線で読み取る。溶血単位は、ヒツジ赤血球の50%が溶解したサンプル希釈度として定義した。
組換え株のJ774A.1マクロファージ(ATCC番号A TIB−67)の細胞毒性を、“Cell Titer 96 Aqueous One Solution Cell Proliferation Assay”キット(Promega,カタログ番号G3580)を製造業者の指示に従い用いて、感染細胞から放出された乳酸デヒドロゲナーゼ(LDH)を測定することによって、求めた。簡単に述べると、細胞にAFV102菌を感染多重度10で感染させた。感染後異なる時点で、上清について細胞から放出されたLDH量を測定し、それを、BCG Danish 1331株の値と比較した。非感染正常細胞は、陰性コントロールとして用いた。生細胞の百分率は、陰性コントロール細胞(100%細胞生存)から放出されたLDH量に対する感染細胞からのそれに基づいて、計算した。
AFV102構築:
AFV102構築時において、ノックアウトプラスミドとの相同DNAセグメントの対立遺伝子交換によりその染色体上に全ノックアウトプラスミドを保有している選択メロジプロイド菌を、ショ糖含有Middlebrook 7H10プレート上で培養し、最終的な対立遺伝子交換を行い、ureC遺伝子をPfoA発現カセットにより置き換えた。ショ糖プレート上で産生されたコロニーのうち、コロニー1個Pfo−105−5(AFV102と再度命名)がウレアーゼ陰性で、このことは、ureC遺伝子がPfoA発現カセットにより置き換えられたことを示唆している。この菌コロニーをさらにPCRにより、ΔureC::ΩpfoA遺伝子型について遺伝子型分析に供した。生成したPCR反応物を、1.2%アガロースゲル上でゲル電気泳動により分析し、その結果を図6に示した。明らかであろうが、この菌をテンプレートとして用いたPCRにより、予測した大きさのPCR産物が産生され、それは、親BCG Danish1331株のそれよりも大きい。ΔureC::ΩpfoA遺伝子型についてDNAバンドについて予測した大きさは、2180bpsであり、親BCG Danish1331株についての大きさは、1967bpsである。コロニー105−5についてのPCR産物をさらにゲルで精製し、Johns Hopkins University(Baltimore,MD)の市販配列決定機器で配列を決定した。配列決定結果は、このコロニーが予測したΔureC::ΩpfoA遺伝子型を有していることを明らかに示した。さらに、AFV102株由来のカナマイシン遺伝子およびsacB遺伝子を増幅することを目的としたPCRは、いかなるPCR産物も産生できなかった(データを示さず)。これらの知見は、AFV102が最終的対立遺伝子交換段階を受けたこと、そして所望のΔureC::ΩpfoA遺伝子型を有していることを示唆している。
ウレアーゼ活性試験と遺伝子型決定結果に基づき、クローン105−5(AV102と再度命名)が、予測した表現型と所望のΔureC::ΩpfoA遺伝子型を示した。AFV102をさらに、親BCG Danish1331株と比較して7H9増殖培地で増殖する能力について試験した。結果を図7に示した。明らかであるように、AFV102構築体は、7H9増殖培地中で親株と非常に類似した増殖曲線を有している。さらに、カナマイシン存在下において、AFV102増殖は、親BCG Danish1331株と同程度に低下し、このことは、予測したとおり、それがカナマイシンに対して同様の感受性を有していることを示唆している。
PfoAタンパク質における1個のアミノ酸置換(コドン137のGlyをコードするggaからのGlnをコードするcagへの置換変異)の結果、哺乳類細胞に対する毒性が消失した。しかしながら、前記タンパク質は、小胞から菌エスケープを媒介する能力を保持したままである(Portnoy同上、1996)。AFV102から発現したタンパク質の毒性は、J7741A細胞にAFV102菌を感染させることによって評価した。感染後異なる時点で非感染正常細胞コントロールと比較すると、AFV102は、現在使用されているBCG Danish1331ワクチン株よりも優れた有意な細胞死を起こすことはなかった(図8)。
前記構築体がマクロファージ内部で生存できるかどうかを調べるため、対数相中期の培養物を用いて、J774A.1肺胞マクロファージに感染させた。細胞は、1:1の感染多重度(MOI)で感染させた。マイコバクテリウムの細胞内生存を、感染後さまざまな時点で菌をプレート計数し、モニタリングした。図9からわかるように、AFV102構築体は、親株のそれと同様の持続的表現型を示し、このことは、J774A.1細胞中においてこの構築体の細胞内生存に全く傷害がないことを示している。
AFV102構築体を含む菌によるPfoAタンパク質の分泌は、BCG Danish1331株と比較して溶血活性が増強されているか、菌培養上清を測定することによって、試験した。AFV102およびBCG Danish1331株両者の培養上清を、同一光学密度で採取し、それらの赤血球溶解能力を比較した。結果を図10に示した。これまでの報告と同じく、BCG培養上清は、増殖時における菌放出代謝物の結果として溶血活性が基準値レベルを示し、それが赤血球溶解を起こすことになるだろう(Grodeほか、Journal of Clinical Investigation,115:2472−2479;2005)。対比して、AFV102培養上清は、BCG Danish1331株と比較して有意に高いレベルの溶血活性を示し、それは、培養上清中PfoA分子の分泌と一致していた。さらに、PfoAのpH非依存性溶血活性をさらに試験し、pH5.5と7.0の両者で比較し、結果を図10に示した。この図から理解されるとおり、AFV102由来上清は、pH5.5と7.0の両者で同様の溶血能力を有しており、このことは、pfoAタンパク質によって分泌された溶血活性が予測どおりpH非依存性であることを示唆している。
マイコバクテリウム病原体の中心的パラダイムは食細胞成熟の停止である。ArmstrongおよびHart(1971)は、M.tuberculosis食細胞がフェリチン標識リソソームと混合しないことを確立し、そのことは、食細胞−リソソーム融合阻害と称されている。ワクチン株M.bovis(BCG)はまた、食細胞コンパートメント中に存在していることが見出され、このコンパートメントは、末端エンドサイトーシスオルガネラから隔離されている(Clemens and Horwitz,1995;Hasanほか、1997;Viaほか、1997)。ClemensおよびHowitz(1995)は、マイコバクテリウム食細胞が形質膜上のものと同等の密度でトランスフェリンレセプター(TfR)を持続的に染色することを見出した。トランスフェリンレセプターは、一般的に、エンドソームから迅速に除去され(半減期t1/2は分単位)、形質膜に再度運ばれる;しかし、マイコバクテリウム食細胞中において、このプロセスが停止し、食細胞がトランスフェリンレセプターを含有するだろう。発明者らがマイコバクテリアを含む食細胞を可視化できたのは、この現象による。食細胞をトランスフェリンレセプターに対する抗体で染色した。同時に、このマイコバクテリアを、食細胞内部にいったん入った後の菌のその後を可視的にモニタリング可能とする蛍光色素により染色した。結果は、rBCG−Pfo構築体が細胞感染後、エンドソームエスケープができるようになったことを明らかに示している。
菌および細胞:BCG Danish1331およびrBCG−ΔureC::ΩpfoAG137Q(AFV102)を、10%(v/v)OADCおよび0.05%(v/v)Tyloxapol(を増殖培地に)添加した7H9培地中でOD600が約0.8−1.0になるまで増殖させた。感染後、菌体をPBS中Alexa Flour568スクシンイミジルエステル(Molecular Probes、Eugene,OR)で製造業者の指示に従い、室温で1−1.5時間標識した。この色素は、菌表面上のタンパク質に局在する一級アミン類に対して非常に安定なアミド結合を形成する。簡単に述べると、菌培養物10mlをペレットとし、PBS中0.625μg/mlのAlexa Flour568 25mL中に再懸濁させ、室温で1−1.5時間インキュベーシンし、菌を標識した。標識した菌体を次にPBSで3回洗浄、7H9増殖培地中に再懸濁後、冷蔵庫で一晩保存した。J774A.1細胞は、文献既出のとおり(Sunほか、2004)、ヒトフィブロネクチン塗布カバースリップを有する6ウェル細胞培養プレート中DMEM培地中で培養した。細胞を3×106細胞/ウェルの密度で塗布し、5%CO2および湿度を有する37℃のインキュベーター中で2日間、培養した。感染中標識菌はペレット化し、DMEM+10%FBS培地中に再懸濁させ、各細胞について感染多重度(MOI)10でJ774A.1に直接添加した。20分、8時間および24時間後、細胞を室温(RT)でリン酸緩衝生理食塩水(PBS,pH7.2)により洗浄した。細胞を次に、RTでPBS(pH7.2)中2%パラホルムアルデヒドで20分間、固定した。固定細胞を次に、PBS(pH7.2)中0.1%Triton X−100でRTにおいて10分間、透析し、次に、PBS(pH7.2)で2回洗浄した。RTで少なくとも2時間ブロッキングを行うかまたは4℃でPBS(pH7.2)中3%ウシ血清アルブミン(BSA)、5%健常ヤギ血清(NGS)および0.5%アジ化ナトリウムにより一晩、ブロッキングを行った。ブロッキング緩衝液を除去後、ラット抗マウストラスフェリンレセプター−FITC(US Biological,Swampscott,MA)を、1%BSA,3%NGSおよび0.5%アジ化ナトリウム含有PBS(pH7.2)による1:50希釈で添加し、その後、RTで少なくとも1時間、インキュベーションした。細胞を次にPBSで2−3回洗浄し、ガラススライド上のヴェクトシールド(vectsheild)マウンティングメディアにより載せた。分析は、Retiga EXI Monoを付属させたNikon TE2000倒立顕微鏡、12ビット冷却IRフィルター付きデジタルカメラを用いて、倍率1500倍でイメージングを行った。
顕微鏡で調べると、BCGおよびrBCG−PfoA菌の両者ともに感染15分後に宿主細胞によって取り入れられたことがわかった。しかし、感染8時間後において、BCG菌と対比してrBCG−PfoA菌がエンドソーム外部で観察され、それらは、ほとんどが宿主食細胞内部に局在していることがわかった。これらの結果を図11A−Dに概略示したが、それらは、マクロファージの菌侵入(図11A)、エンドソーム内部におけるBCGの持続(図11B)、初期エンドソーム内部におけるAFV102菌(図11C)および、組換えPfoAの分泌によるエンドソームからのAFV102菌の細胞の細胞質へのエスケープ(図11D)を示している。菌の計数は、138個中100個(72%)のAFV102が感染後8時間でエンドソームをエスケープし、一方、100個中29個(26%)のBCGが食細胞をエスケープしたことを示していた。感染サンプル24時間後において菌を調べ、同様の結果が得られた。この知見は、PfoA発現が、食細胞からのAFV102放出を増加させることを示している。

ワクチン処方の戦略は、製造プロセス全てにおいて、最大の生存度と安定性を求めるための研究に基づいて、作成する。これには、マイコバクテリウム菌の培養のために一般的に用いられるさまざまな培地を利用した培養時において、最大菌生存度(生きているか死んでいるか)を調べることが含まれ、培養には、グリセロール、糖類、アミノ酸類、および界面活性剤類または塩類の添加を含む。培養後、遠心分離または接線フローろ過により培養細胞を採取した後、凍結または凍結乾燥プロセスにおいて細胞を保護できる安定化培地に再懸濁する。一般的に用いられる安定化剤には、グルタミン酸ナトリウム、またはアミノ酸またはアミノ酸誘導体類、グリセロール、糖類または一般的に使用される塩類が挙げられる。最終処方は、筋肉内、経皮注入、潅流または経口投与により運搬されるだけの十分な生存菌を提供し、それらは、維持のための十分な安定性と市販および使用のための適切な保存期間を有しているだろう。
一般的安全性試験
1群6匹のBALB/cマウスを、問題のrBCG株(類)および同族の親株類2×106CFUで腹腔内感染させる。感染後14日間、前記動物を全身健康と体重についてモニタリングする。BCGおよびrBCG株類を投与した動物は健康のままであり、観察期間中体重減少を起こすこともなく、あるいは病気の明らかな症状も示さない。
BALB/cマウス15匹を1群とし、これらに静注でそれぞれrBCGおよびBCG親株 2×106CFUを感染させる。感染第1日において、各群のマウス3匹を屠殺し、脾臓、肺および肝臓におけるCFUを分析し、各動物が同等の感染量であるようにする。感染後第4、8、12および16週において、各群のマウス3匹を屠殺し、脾臓、肝臓および肺におけるCFUを得て、親BCG株と比較してrBCG株類のインビボ増殖を評価する。
各群10匹としてSCID(重症複合免疫不全症)を有する免疫妥協マウスに、静注でそれぞれrBCGおよびBCG親株 2×106cfuを感染させる。感染後第1日において、各群のマウス3匹を屠殺し、脾臓、肝臓および肺におけるcfuを分析し、接種用量を確認する。各群7匹の残りの動物は、全身状態および体重をモニタリングする。これらのマウスの生存を追跡し、全観察期間において親株感染動物とrBCG感染マウスの生存を比較する。
rBCG株類の安全性をまた、ヒトにおける十分に確立された安全性を有している親BCGワクチンと比較しモルモットモデルで評価する。最初に、動物の全般健康状態に及ぼすワクチンの効果を検討するが、それには、体重も含める。モルモットは、組み換えおよび親株類を107(ワクチン用量の100倍)CFUだけ筋肉注射で免疫し、6週間、全般健康状態と体重をモニタリングする。この6週間という期間の前に死亡した動物については、死後検査も行う。感染後6週間の期間が終わった時点で、動物は全て屠殺し、総合病理検査を行う。この試験中確認すると体重減少および異常行動は全くなく、この6週時剖検において全ての臓器は正常のように見え、rBCG−PfoAワクチンについて、全く健康に対する悪影響が見られず、しかも、親株を接種した動物に比較してrBCG−PfoAワクチン接種動物は、正常速度で動物が体重を増やしている。
rBCG株類の毒性を評価するため、(各群12匹の)モルモットを、それぞれ、ヒト用rBCG株類、BCG親株または生理食塩水の単回投与量よりも4倍の1回投与量または4分の1の1回投与量で、筋肉内投与によりワクチン接種する。ワクチン後第3日において、動物6匹を屠殺し、これらの動物に及ぼすワクチンの急性効果を評価する。ワクチン後第28日において、残りの動物6匹を屠殺し、動物に及ぼす慢性効果を評価する。両方の時点で各動物の体重を調べ、全般病理状態と注射部位の様子を調べる。血液を採取し、血液検査を行い、内臓および注射部位の組織病理を調べる。
1群13匹としたC57Bl/6マウス(雌性、5−6週齢)を、皮下から106CFUのrBCG、親BCGまたは生理食塩水で免疫する。別のマウス群は、健常対照として使用する。免疫後8週時点で、マウスに対して、M.tb Erdman株(または、H37Rv Kan耐性株)を総計107CFU含む10mlの単細胞懸濁液から作製したエアゾールとして、このチャレンジ株によりチャレンジするが、この量は、文献に既出のとおり(Brodinほか、J Infect Dis.190(1):115−122;2004)各動物の肺におよそ100個の生きている菌を伝播する。チャレンジしていない動物とともに、実験動物の生存をモニタリングする。チャレンジ後、体重減少と全般健康状態についても動物をモニタリングする。チャレンジ後第1日において、各群3匹のマウスを屠殺し、肺におけるCFUを調べ、チャレンジ用量を確認し、動物1匹は、脾臓および肺の組織病理検査のために屠殺する。チャレンジ後第5週において、各群の動物9匹を屠殺し、動物の組織病理検査と顕微鏡検査を行う。マウス6匹の肺と脾臓組織を、CFU計測のために評価する(選択サプリメントを有するプレートを用いて、チャレンジ株からワクチン株を識別する)。もしH37Rv−kan耐性株でチャレンジするならば、KanまたはTCH(チオフェン−2−カルボン酸ヒドラジド)を用いてワクチン株からチャレンジ株を識別する。もしM.tb Erdman株をチャレンジに使用するならば、TCHを用いてチャレンジ株からワクチン株を識別する(BCGは感受性であるが、M.tbは本来耐性である)。
SPFモルモットを、rBCGまたは親BCG株103により筋肉注射で免疫する。免疫後9週で、動物背部の毛を剃り、リン酸緩衝生理食塩水100μl中PPD(タンパク質精製誘導体)10μgを筋肉注射する。24時間後、硬い硬変部の直径(DTH)を測定する。rBCG株は、親BCG株類で誘導したものに等しいかそれよりも大きいDTHを誘発する。
M.tbチャレンジに対してのrBCGワクチン類の有効性を調べるため、各群12匹のモルモット(若い成熟SPFハートレイ(Hartley)、250−300グラム、雄性)を、それぞれ、rBCG、親BCG株または生理食塩水で免疫する。ワクチン類および対照は、106cfuを筋肉注射で投与する。免疫後10週時点で、rBCG免疫動物、BCG免疫動物および偽免疫動物に対して、M.tbを総計107CFU含む10mlの単細胞懸濁液から作製したエアゾールでチャレンジする;この操作では、文献に既出のとおり(Brodinほか、2004)各動物の肺に約100個の生きている菌を伝播する。チャレンジ後、ワクチン接種していない未チャレンジの健常群とともに、動物の生存、体重減少および全般健康状態をモニタリングする。各群から6匹のマウスをチャレンジ後第10週で屠殺し、各群中残りの6匹は、チャレンジ後第70週で長期評価のため屠殺した。両方の時点において、動物の組織病理検査と顕微鏡検査を行う。肺および脾臓組織を、組織病理検査とCFU計測のために評価する(選択サプリメントを有するプレートを用いて、チャレンジ株からワクチン株を識別する)。もしH37Rv−kan耐性株でチャレンジするならば、KanまたはTCHを用いてワクチン株からチャレンジ株を識別する。もしM.tb Erdman株をチャレンジに使用するならば、TCHを用いてチャレンジ株からワクチン株を識別する(BCGは感受性であるが、M.tbは本来耐性である)。チャレンジ後偽免疫動物が最も早く死亡し、一方、rBCG免疫動物がBCG親株免疫動物よりも長く生存すれば、チャレンジ研究が成功したことになる。
さらに最近になって非ヒト霊長類を用いてM.tbに対するワクチン接種の評価が行われた。ヒトと非ヒト霊長類の進化上の関係とこれらの種における結核の臨床および病理的表れ方が似ていることにより、TBとワクチン効果についての実験的研究にとってこの非ヒト霊長類モデルが魅力的となっている。
安全性と毒性試験
規制ガイドラインにより規制されている前臨床安全性および毒性試験を、上記のような前臨床毒性および安全性試験として行う。これらを調べた後、ヒト安全性試験を行う。これらの研究は、健常クアンティフェロン(Quantiferon)陰性成人で当初行い、その後、小児および新生児に年齢を低下させて行う。
マウスおよび霊長類における免疫原性研究では、INFγのような細胞免疫評価のための標準的手法類、ELISPOTおよび/またはフローサイトメトリを短期および長期抗原またはペプチド刺激とともに用いるが、これらに限定されない。同様の方法論は、ヒト応答を評価するために利用する。テトラマー研究を、ヒトワクチン後のCD4およびCD8応答を評価するために用いる。
rBCGは、TBまたは関連トランスジーン類を発現するように遺伝子工学的に作製した他の疾患に対する単独ワクチンとしても、良好に作用する。本文でTBに対するワクチンとしてまたは他の疾患に対する防御のための抗原類を発現するワクチンとして述べたrBCGは、また、アジュバントまたはウイルスまたは細菌ベクター抗原類と混合した組み換えタンパク質類による追加免疫のための免疫系をプライムするためにも、非常によく作用する。動物前臨床研究およびヒト前臨床研究の両者において、BCGプライムとその後の組み換えタンパク質/アジュバントまたはベクターブーストを、投与法および用量の観点から最適化する。これらのプライムブースト戦略は、特に本発明の態様にあるBCGのプライム能力のゆえにヒトにおいて免疫を誘発するためおよびその後組み換えタンパク質またはベクターが免疫系のブースター応答を集束させ増強させるための最も強力な手段である。
C57BL/6マウスを用いて、潜伏感染を確立し;低用量感染によって無視可能なM.tb特異的免疫性が誘発された時点およびM.tb特異的免疫性がメモリT細胞で低下され圧倒される別の時点で、治療用ワクチンをマウスに投与する。その後ワクチン類の治療上の利点が、最終処置用ワクチン投与後2ヶ月および5ヶ月にマウスで、各マウスの肺および脾臓におけるcfuを計数することにより、評価する。このcfu計数値を、マウス群で標準的統計方法により分析し、その結果を用いて治療的ワクチン接種がマウスにおいて潜伏M.tb感染を有意に低下させるかどうかを分析する。同様の方法論を、必要に応じて、他の動物の応答評価のために利用する。
本発明のrBCGによる標的動物の経口ワクチン投与はまた、文献に既出の方法類を用いて行うことができる(Millerほか、Can Med Assoc J 121(1):45−54;1979)。経口投与した本発明rBCGの量は、対象の種により、ならびに治療しようとしている疾患または状態に応じて変わる。一般的に、使用した投与量は、生きている菌体約103乃至1011個であり、好適には生きている菌体約105乃至109個であろう。
先にも述べたとおり、BCGにおけるLloの発現は、エンドソームエスケープ促進において実質的に有効でないことが証明されており、それはおそらく、BCGが存在する修飾エンドソームというミクロな環境での中性pHでLloの活性が低いことによるのであろう(Hessほか、Proc Natl Acad Sci 95:5299;1998;Grodeほか、J.Clin.Invest.,J.Clin.Invest.,115(9):2472;2005)。にもかかわらず、Lloの発現は、エンドソームを十分に修飾し、SCIDマウスにおいてBCG−Llo+の毒性を低下させた(Grodeら、2005)。この観察は、宿主細胞の細胞質にBCGを再度標的化させることが、この昔からの生TBワクチンの免疫原性と安全性を両方ともに改善する潜在力を有していることを示唆している。本研究の目的は、従って、SCIDマウスでPfoAG137Qを発現するエンドソームエスケープ株AFV102の安全性を決定することである。
候補TBワクチン株AFV102のMtbチャレンジに対する能力を測定するため、8匹(若い大人のSPF Hartleyモルモット(250−300グラム)による群を表4に示したように免疫する。
アポトーシスはプログラムされた細胞死であり、その誘導と結果という観点からして壊死性細胞死とは大きく異なる。外来性抗原類を含む細胞のアポトーシスは、このような抗原類に対する細胞性免疫の公知の強力な刺激物である。抗原含有細胞のアポトーシスが細胞性免疫につながるプロセスは、クロスプライミングと称されることもある。1,2,3抗原特異的細胞媒介免疫増強を起こすアポトーシス誘導メカニズムにはいくつかある。カスパーゼ8媒介アポトーシスは、抗原特異的細胞性免疫防御につながる。4 エンドソームエスケープを行った組換えBCGによる細胞の細胞質におけるカスパーゼ8の産生は、組換えBCG発現外来性抗原という文脈において、BCGに対しておよび組換えBCGにより過剰発現した他の結核抗原類に対してならびに高レベルの抗原特異的細胞免疫につながるBCGそれ自体の抗原類に対して、プログラムされた細胞死を誘導する強力なもうひとつの方法であろう。TRAIL−R2(TRAILレセプター2)またはTNFR−SF−10B(腫瘍壊死因子−スーパーファミリメンバー10B)としても公知のデスレセプター−5(DR−5)はまた、カスパーゼ8媒介アポトーシスを媒介する。4 レオウイルス誘発アポトーシスは、TRAIL−DR5により媒介され、このウイルスがその後クリアランスされることになる。5 組み換えBCGによるDR−5発現は、rBCG発現抗原類に対する抗原特異的細胞性免疫の誘導に対して強力なアジュバント効果を付与する。抗原発現細胞はまた、抗原特異的細胞性免疫応答の誘導に強力な刺激物であるFas連結を介してアポトーシスを受けるように誘導することができる。6 FasまたはFas細胞質ドメイン/CD4エクトドメイン融合タンパク質を発現する組み換えBCGは、アポトーシスおよび抗原特異的細胞性免疫応答を誘発するであろう。
