Description
ジアゼニウムジオレート作用基を含む分岐ポリエチレンイミン(bPEI−NONOate)
1級アミン側鎖を有する反復単位と、エチレンイミンの反復単位との共重合体である3種の分岐ポリエチレンイミン(bPEI)(それぞれ、重量平均分子量600;10,000;25,000)を(米国Polysciences社、アミノ基の比率は、およそ約1級:2級:3級=25:50:25)、それぞれ0.5gずつ、それぞれの100mLビーカーに入れて10mLのメタノールと20mLのテトラハイドロピュとは、(THF)を加えて30分間撹拌した。
1)ジアゼニュムジオルレート作用基形成の確認
得られたそれぞれのbPEI−NONOateが、実際にジアゼニウムジオレート作用基を有するか否かということを、核磁気共鳴(NMR)を介した定量で決定した。図2は、前記反応生成物の1H−NMRスペクトル結果である。化学式3のbPEI分子で、3級アミンのα−水素(図2で、a位置)は、1H−NMRスペクトルで、δ=2.6ppmで示されるが、ジアゼニウムジオレート作用基が生成されれば、以前の3級アミンのα−水素は、1H−NMRスペクトルで、δ=3.1ppm(図2のb位置)で示される。図2に示したように、生成物のNMRスペクトルを分析した結果、bPEIの2級アミンのうち、分子量600であるbPEI(bPEI600)の場合は、15.3%(図2(a)参照)、分子量10,000であるbPEI(bPEI10K)の場合は、13.8%(図2(b)参照)、分子量25,000であるbPEI(bPEI25K)の場合は、16.0%(図2(c)参照)がジアゼニウムジオレートに転換されたということを確認した。以下の明細書記載で、図2(a)の1H−NMRスペクトルで確認した分子量600のbPEIジアゼニウムジオレートは、bPEI600−NONOate、図2(b)の1H−NMRスペクトルで確認した分子量10,000のbPEIジアゼニウムジオレートは、bPEI10K−NONOate、及び図2(c)の1H−NMRスペクトルで確認した分子量10,000のbPEIジアゼニウムジオレートは、bPEI25K−NONOateとそれぞれ略称する。
bPEIの2級アミンからジアゼニウムジオレート作用基が生成されれば、紫外線吸光分析で、一般的に250ないし260nm近傍で吸収ピークが示される。図3は、200ないし400nmで合成したbPEI−NONOateの紫外線スペクトルを測定したグラフである。図3で、bPEI600、bPEI10K及びbPEI25Kは、それぞれ反応前の分子量600;10,000及び25,000のbPEIであり、bPEI600−NO、bPEI10K−NO及びbPEI25K−NOは、それぞれそれらの当該ジアゼニウムジオレート化物を示す。図3に図示されたように、紫外線分光の結果、bPEI600−NO、bPEI10K−NO及びbPEI25K−NOの吸収ピークが、反応前のbPEIとは異なり、250ないし260nmで示されるということを確認した。
一酸化窒素放出分析は、一般的な検出方法であるGriess分析法(H.H.H.W.Schmidt and M.Kelm、inMethodsinNitricOxideResearch、ed.M.Feelish and J.S.Stamler、Wiley&Sons、New York、1996参照)を使用して行った。反応性の大きい一酸化窒素は、反応溶媒中の酸素や水気と反応して二酸化窒素を生成し、この二酸化窒素は、スルファニルアミド(sulfanilamide)と反応して活性化され、さらにこの活性化された物質は、ナフチレンジアミンと反応し、波長548nmの光を吸収するアゾ化合物を生成する。
ジアゼニウムジオレート作用基を含まない主鎖−側鎖複合体
(1)F127−bPEIの合成
分子量12,600のPluronicF127(BASF社登録商標)10.0gを、250mL丸底フラスコに入れ、これを塩化メチレン70mL中に溶解させた。次に、前記フラスコを窒素でパージした。後続として、1.29gのクロロホルム酸パラニトロフェニル(p−NPC)を塩化メチレン70mL中に溶解させ、この混合溶液を、滴下漏斗(dropping funnel)中に入れた。PluronicF127を窒素雰囲気下で撹拌しながら、ここに、p−NPCを前記フラスコに滴下させ、それらの間で12時間反応を進め、末端活性化されたPluronicF127を得た。この生成物の分離は、ジエチルエーテル沈澱法を使用した。次に、末端活性化されたPluronicF127にbPEIを反応させ、その末端にbPEIを連結させた。F127−bPEIの合成スキームは、以下の化学式4に図示してある。化学式4で、F127−bPEIは、bPEIが末端に結合されたPluronicF127を示す。
(1)PluronicF127とbPEIとの結合確認
前記3種の主鎖−側鎖複合体で、PluronicF127とbPEIとがカルバン酸連結部を介して連結されたか否かということは、1H−NMRを介した定量で決定した。前記3種の生成された主鎖−側鎖複合体の1H−NMRスペクトルは、図5に示した。一般的には、図5でaとcで表示したように、PluronicF127は、1H−NMRスペクトルがメチル水素(a)とメチレン水素(c)とに大別される。プロピレングリコールから誘導された反復単位のメチル水素aの1H−NMRシグナルは、δ=1.15ppmで示され、残りのメチレン水素cの1H−NMRシグナルは、δ=3.2ないし4.0ppmで示される。また、bPEIの炭素結合水素bの1H−NMRシグナルは、δ=2.6ppm近傍で示される。図5で(1)は、反応していないPluronicF127、(2)は、生成されたF127−bPEI0.8、(3)は、生成されたF127−bPEI1.0、そして(4)は、生成されたF127−bPEI1.3の1H−NMRスペクトルをそれぞれ示す。図5の結果から、PluronicF127の主鎖の末端ヒドロキシ基のうち、F127−bPEI0.8は、40.0%、F127−bPEI1.0は、50.0%、F127−bPEI1.3は、62.5%がbPEIに結合しているということを確認した。
PluronicF127とbPEIとからなる前記主鎖−側鎖複合体が、PluronicF127のように、温度感応性水和ゲルを形成することができるか、そしてそうであるならば、ゲル化特性にいかなる変化があるということを見るために、ゲル化試験を行った。さまざまな濃度(質量%)で、水に分散され主鎖−側鎖複合体を隅々見たとき、溶液が流れれば、溶液(ゾル)状態と判断し、流れなければ、ゲル状態と判断することにより、温度及び濃度によるゾル−ゲル存在いかんを記録した(inverted gelation test)。試験結果は、図6にグラフで示した。生成された主鎖−側鎖複合体は、いずれもPluronicF127よりさらに低い濃度でもゲルを形成することができた。安定性の側面では、F127−bPEI0.8、F127−bPEI1.0及びF127−bPEI1.3は、この順序で安定したゲルを形成した。従って、3種の複合体は、いずれも伝達体を製造する用途に適する前駆体として使用される。

F127−bPEI0.46gを100mLビーカーに入れ、5.3Mのメトキシ化ナトリウム0.23mL、メタノール2.07mL、テトラヒドロフラン2.3mLを入れた。このビーカーを、図1に図示した反応器中に入れ、20psiのアルゴンで2回パージした。その後、80psiの一酸化窒素を前記反応器に加え、前記反応器を3日間維持した。この反応の生成物をジエチルエーテルで沈澱させて分離した。この反応スキームは以下の化学式5に示した。
1)ジアゼニウムジオレート作用基形成の確認
F127−bPEI1.3−NONOateにジアゼニウムジオレート作用基が生成されたか否かということは、NMRを介した定量で決定した。1H−NMRスペクトル結果を図7に図示した。一般的に、F127−bPEIは、1H−NMRスペクトルで3個の位置でピークを有する。1H−NMRスペクトルで、PluronicF127のプロピレングリコールから誘導された反復単位のメチル水素のピーク(図7のa)は、δ=1.15ppmで示され、メチレン水素のピーク(図7のc)は、δ=3.2ないし4.0ppmで示される。また、1H−NMRで、bPEIのメチレン水素(図7のb)は、一般的に、δ=2.6ppm近傍で示されるが、bPEIのメチレン水素のうちジアゼニウムジオレート作用基について、α−位置のメチレン水素(図7のd)は、δ=2.9ppm近傍で示される。
F127−bPEI−NONOate伝達体について、温度感応性水和ゲル形成実験を行った結果、図6に示したF127−bPEI複合体の場合と類似して水和ゲルを形成するということを確認した(データ示さず)。前述の3種のF127−bPEI−NONOateは、いずれもF127−bPEIと共に、温度感応性水和ゲルを安定して形成した。
bPEI−NONOateでのように、Griess分析法で水和ゲルを形成したF127−bPEI−NONOate伝達体の一酸化窒素の放出を分析した。図8に、分析結果を図示した。bPEI600−NONOateで、全ての放出一酸化窒素の半減期は、1分以内(図8)であったが、F127−bPEI0.8−NONOate、F127−bPEI1.0−NONOate及びF127−bPEI1.3−NONOateの全ての放出一酸化窒素の半減期は、約1.6時間ないし2.4時間の範囲にあり、これは、F127−bPEI0.8−NONOate、F127−bPEI1.0−NONOate及びF127−bPEI1.3−NONOateが、bPEI600−NONOateより100倍以上の遅い速度で一酸化窒素を放出するということを意味する。
F127−bPEI0.8−NONOate水和ゲルについて、細胞毒性実験(日本DOJINDO社のCCK−8キットで分析)を行った。試験群では、血管内皮細胞株(cell lines)であるヒト臍帯静脈内皮細胞(HUVEC:humanumbilicalveinendothelialcells)、または血管平滑筋(SMC)細胞株である管状動脈平滑筋細胞(coronaryarterysmoothmusclecell)をF127−bPEI0.8−NONOate水和ゲル上で培養し、培養された細胞数を測定した。対照群として、同じ細胞を一般的な24ウェルプレートの上で培養した。試験結果は、図9に図示した。図9のデータは、平均値/標準偏差(n=3)でもって表示される。tテストを使用して統計分析を行った。記号**及び***は、それぞれp<0.001及びp<0.0001のレベルでの水準での統計的有意性を示す。
Claims (8)
- 一酸化窒素の調節放出用伝達体であって、
前記伝達体は、
有機高分子の主鎖と、
前記主鎖に共有結合で連結され、ジアゼニウムジオレート作用基を有する側鎖と、を含む重合体を含み、
前記主鎖の骨格は、エチレングリコール/プロピレングリコール共重合体、乳酸/エチレングリコール共重合体、乳酸−グリコール酸−エチレングリコール共重合体、乳酸−グリコール酸−プロピレングリコール−エチレングリコール共重合体、ε−カプロラクトン/エチレングリコール共重合体、3−ヒドロキシブチル酸/プロピレングリコール共重合体、フマル酸化プロピレン/エチレングリコール共重合体、有機ホスファゼン/エチレングリコール共重合体及びポリペプチド系ブロック共重合体からなる群から選択されるブロック共重合体を含み、
前記側鎖の骨格は、ポリエチレンイミン、ポリプロピルイミン、ポリリシン及びキトサンから選択されるものを含み、
前記ジアゼニウムジオレート作用基は、下記式で示され、
(式中、左側の窒素の2つの単結合は、前記側鎖の骨格に連結する。)
前記伝達体の前記重合体は、昇温するとき、ゾルからゲルに相転移を起こし、前記相転移の臨界温度は、25ないし35℃の範囲にあって、前記伝達体の前記重合体は、前記臨界温度以上の温度と生体内pH条件とで、化学架橋のない水和ゲルを形成する一酸化窒素調節放出用伝達体。 - 前記主鎖の骨格は、エチレングリコール/プロピレングリコール共重合体であることを特徴とする請求項1に記載の一酸化窒素調節放出用伝達体。
- 前記側鎖の骨格は、ポリエチレンイミン、ポリプロピルイミン、ポリペプチド及びキトサンからなる群から選択される重合体を含むことを特徴とする請求項1に記載の一酸化窒素調節放出用伝達体。
- 前記水和ゲル内に、血管内皮成長因子(VEGF)、消炎剤、ビタミンC、アセチルサリチル酸、高脂血症剤、血栓溶解剤、抗血栓剤、ヘパリン、抗癌剤及び抗増殖活性を有するオリゴヌクレオチドからなる群から選択された少なくとも1種の薬理効果物質をさらに含むことを特徴とする請求項に1記載の一酸化窒素調節放出用伝達体。
- 前記薬理効果物質は、前記伝達体に共有結合で連結されず、前記水和ゲル内に分散していることを特徴とする請求項4に記載の一酸化窒素調節放出用伝達体。
- 前記薬理効果物質は、前記ジアゼニウムジオレート作用基の酸素原子に共有結合で連結された結合体であることを特徴とする請求項4に記載の一酸化窒素調節放出用伝達体。
- 請求項1に記載の一酸化窒素調節放出用伝達体で表面が被覆された人工体内挿入物。
- 前記人工体内挿入物は、ステント、カテーテル、導管、皮下移植片、化学センサ、心臓拍動器、移植血管片、傷用ドレッシング、男性器挿入物、挿入用心拍発生装置(IPG)、挿入用心臓除細動器及び神経興奮器からなる群から選択されることを特徴とする請求項7に記載の人工体内挿入物。
