Description
本発明は、バイオリアクターにおいて使用する細胞培養培地を処理するための方法であって、該細胞培養培地を紫外線C(UVC)光に曝露し、該細胞培養培地を滅菌フィルターに通し、該細胞培養培地をバイオリアクター内に導入することを含んでなる方法を提供する。本発明はまた、バイオリアクターにおいて使用する細胞培養培地を処理する方法であって、該細胞培養培地をUVC光に曝露し、該細胞培養培地をデプスフィルターに通し、該細胞培養培地を滅菌フィルターに通し、該細胞培養培地をバイオリアクター内に導入することを含んでなる方法を提供する。
本発明の方法は、バイオリアクターにおいて使用する細胞培養培地を処理するために用いられうる。浸透圧重量モル濃度、25℃における伝導度および254nmにおける吸光度に関して3種類の細胞培養培地を分析した(表Iを参照)。
UVC光による幾つかのウイルスの不活性化を判定するための研究を行った。表IIは、選択したモデルウイルス、すなわち、ゼノトロピックマウス白血病ウイルス(x−MuLV)、マウス微小ウイルス(MMV)、ブタパロウイルス(PRV)およびレオウイルス3(Reo3)を示す。
デプスフィルターを使用することにより細胞培養培地から包膜ウイルス粒子および他の望ましくない細胞物質を除去することに関する研究を行った。表VIIは3つの包膜ウイルスおよび非包膜ウイルスのウイルス不活性化を示す。これらの研究から決定されたLRVは、該デプスフィルターが細胞培養培地から包膜ウイルス粒子を効率的に除去しうることを示している。
Claims (14)
- バイオリアクターにおいて使用する細胞培養培地を処理する方法であって、
(a)該細胞培養培地を紫外線C(UVC)光に曝露し、
(a2)細胞培養培地をデプスフィルターに通し、
(b)該細胞培養培地を滅菌フィルターに通し、
(c)該細胞培養培地をバイオリアクター内に導入する
ことを含み、細胞培養培地が120〜320J/m 2 のエネルギー密度のUVC光に曝露される、方法。 - UVC光が254nmの波長を有する、請求項1記載の方法。
- 細胞培養培地を毎時1〜12リットルの流量でUVC光に曝露する、請求項1記載の方法。
- 細胞培養培地を毎時6リットルの流量でUVC光に曝露する、請求項3記載の方法。
- 対数減少値が4.85以上である、請求項1記載の方法。
- 対数減少値が6〜7である、請求項5記載の方法。
- 細胞培養培地を238J/m2のエネルギー密度のUVC光に曝露する、請求項1記載の方法。
- 滅菌フィルターが、200nmの最大サイズを有する細孔を有する、請求項1記載の方法。
- 細胞培養培地をUVC光に曝露する工程が、該細胞培養培地内のいずれかの非包膜ウイルスの核酸を損傷させるのに十分なものである、請求項1記載の方法。
- 処理された細胞培養培地を、哺乳類細胞または昆虫細胞の成長を支持するために使用する、請求項1記載の方法。
- 哺乳類細胞が抗体を産生しうる、請求項10記載の方法。
- 細胞培養培地を酸性pHでデプスフィルターに通す、請求項1記載の方法。
- 細胞培養培地をpH5.0でデプスフィルターに通す、請求項12記載の方法。
- 細胞培養培地をデプスフィルターに通す工程が、いずれかの包膜ウイルスを該細胞培養培地から除去するのに十分なものである、請求項1記載の方法。
