Description
Mikrobiologi 1996年、ISBN 91−7205−037−3)。齲蝕の開始には、歯垢内のストレプトコッカス・ミュータンスが比較的高比率である必要があるという多くの証拠がある。これらの細菌は、歯の表面に十分に付着し、他の細菌型よりも糖類から多量の酸を産生し、酸性環境において他の細菌よりも良好に生存でき、ショ糖から細胞外多糖類を産生できる。プラークにおけるストレプトコッカス・ミュータンスの比率が高い(2〜10%の範囲)場合、患者は、齲蝕に対する危険性が高い。その比率が低い(0.1%未満)場合、患者の危険性が低い。それらは、他の細菌よりも耐酸性であるので、プラーク内の酸性条件は、ストレプトコッカス・ミュータンスの生存および増殖に好ましい。

zeae)の株は、その表面に有害なストレプトコッカスを中和する抗体を産生するために遺伝子工学により修飾されているが(ハンマーストローム L.(Hammarstrom L.)、2002年7月発行のNature Biotechnology)、遺伝子修飾生物を用いるこのアプローチは、安全性の承認が不明であるという状況に直面している。
。ストレプトコッカス・ミュータンスを阻害するために、いわゆる「競争的排除」の考え方を用いて他の研究が実施されている。例えば、ラクトバチルス・ロイテリ株ATCC 55730は、ストレプトコッカス・ミュータンスを阻害することが示されている(ニカワ H.(Nikawa H.)ら、2002年7月11日、広島大学によりニュース報道)。日本でLSlと呼ばれ、市販されているラクトバチルス・サリバリウス(Lactobacillus salivarius)(LSl)(フレンテ(Frente)社、日本国)の株を含有する錠剤製品は、ストレプトコッカス・ミュータンスを阻害することが主張されている。

チルスGG ATCC 53103のストレプトコッカス・ミュータンス阻害作用もまた公知であり、他の幾つかの乳酸菌は、坑齲蝕症であることが主張されているが、口腔内のストレプトコッカス・ミュータンス数の減少、それによる齲蝕症を減少させる能力においてラクトバチルス株の間で相当な差異が存在し、このような株が選択できることは以前には知られていなかった。
Culture Collection(ATCC)(ロックビレ、メリーランド州)で公に入手でき、最後の2つの株は、2003年7月29日に寄託されている。
株の選択方法
本発明により用いられるラクトバチルス株の選択は、以下の3つのステップ様式で実行することができる。
阻害効果を測定するために使用する株の例は、ストレプトコッカス・ミュータンス、ATCC25175(The American Type Culture Collectionマナサス、ヴァージニア州、米国から入手できる)である。この単離体を、0.5%酵母抽出液(ディフコ(Difco)、デトロイト、米国)(TSBY)で補足されたトリプチカーゼダイズブロス(ディフコ)中で増殖する。細胞を、1000xgでの遠心分離により指数増殖期中に採取し、PBSで2回洗浄し、同じ緩衝液中に再懸濁する。細菌凝集物を分散させるために細胞懸濁液を低強度音波装置に供する。
遠心分離により指数増殖期中に採取し、リン酸緩衝生理食塩水(PBS;pH6.8)で2回洗浄し、同じ緩衝液中に再懸濁する。

試験されるラクトバチルス株を採取する。該菌は、MRSブロス(ディフコ)中、37℃で16時間増殖させる。プレートを、CO2+N2雰囲気下の嫌気性ジャー(GasPak System、BBL、ベクトン・ディッキンソンマイクロバイオロジーシステムズ(Becton Dickinson Microbiology Systems)、Cockeysville、メリーランド州、米国)中でインキュベートする。
Doc2000装置(バイオラッド・ラボラトリーズ(BioRad Laboratories)、Herkules、カリフォルニア州、米国)内でウェルの全面にわたり結合を測定する。全ての測定は、三重反復試験で行われる。

選択株を含有する製品の製造
ストレプトコッカス・ミュータンスの阻害およびムチンの結合に関して、上記の方法を用いた前述の実施例2の節における一般的に良好な増殖特性および好ましい結果に基づいて、本実施例において、ラクトバチルス・ロイテリFJ1「プロデンチス(Prodentis)」(ATCC PTA−5289)が該株をチューインガムに添加させるために選択される。ラクトバチルス・ロイテリ株を、産業界においてラクトバチルスを増殖させる標準法を用いて増殖させ、凍結乾燥する。
であるチューインガム製品の他の成分を、該製品の有効性に影響を及ぼすことなく使用できることは理解されている。すなわち、
1 融解。Softisan154(サソール・ジーエムビーエイチ(SASOL GMBH)、Bad Homburg、ドイツ国)を容器に溶解し、70℃に加熱して結晶構造の完全破壊を確実にする。次いで52〜55℃(その固化点のすぐ上)に冷却する。
2 顆粒化。ラクトバチルス・ロイテリの凍結乾燥粉末を、Diosna高せん断ミキサー/造粒機、または等価の機器に移す。融解したSoftisan154を、ラクトバチルス・ロイテリ粉末に凡そ1分間でゆっくりと加える。さらに集結させる時間は必要としない。添加時にチョッパを使用する。
3 湿性篩い分け。顆粒化直後、トルネードミルを用いて顆粒を1−mmの篩い分けネットに通過させる。篩い分けられた顆粒を、PVCコーティングアルミニウムフォイル製のアルポーチに包装し、乾燥ポーチと共にポーチを形成するためにヒートシーラにより密封し、混合まで冷凍保存する。顆粒化バッチを2つの錠剤用バッチに分ける。
4 混合。ミキサー中の全成分を均一混合物へと混合する。
5 圧縮。最終混合物を、回転式錠剤プレスのホッパに移し、Killianコンプレッサ中、全量が765mgの錠剤に圧縮する。
6 バルク包装。モレキュラーシーブの乾燥ポーチと共にアルバッグ中に該チューインガムを包装する。アルポーチをプラスチック製バケツに入れ、最終包装の前に少なくとも1週間冷所で保存する。
