Description
本願は、2008年1月24日付申請の米国仮出願特許第61/023,310の利益を主張し、その内容を、参照により本明細書中に組み込んでいる。
本明細書では、アセトアルデヒド濃度を減少させる、高アセトアルデヒド濃度の症状を予防する又は寛解させる、及び例えば、環境供給源由来の、又はアルコールの摂取によるインビボでのアルデヒド生成由来のアルデヒドへの曝露による、アルデヒドとの接触に起因する疾患又は障害の危険を低減させるのに有用な、ジンセノサイド組成物及び方法を提供する。
アルデヒドは、人及び動物において毒性作用を有する化学反応物質である。アルデヒドは、アルデヒド脱水素酵素(ALDH)によって、低反応性のカルボン酸に触媒され、そのようなものとして又は複合体として、身体から排出される(Lindahlの文献、Crit. Rev. Biochem. Mol. Biol. 1992, 27, 283-335)。例えば、サイトゾルのALDH1A1並びにミトコンドリアのALDH1B1及びALDH2を含むALDHの多形が存在し、広範囲のアルデヒドを触媒する(Yoshidaらの文献、Eur. J. Biochem. 1998, 251, 549-557;Vasiliouらの文献、Pharmacogenetics 1998, 9, 421-434)。
一態様において、例えば、ヒトなどの対象への投与に適したジンセノサイド組成物を提供する。例えば、該ジンセノサイド組成物は、1つ以上のプロトパナキサジオール系ジンセノサイド、例えば、ジンセノサイドRb1、Rb2、Rb3、Rc、Rd、Rg3、Rh2、Rs1、Rs2、R1、R4、F2などを含むことができる。
一般に、本明細書中で使用される命名法、及び本明細書中に記載される医薬品化学、生化学、及び薬理学における検査法は、当該分野で周知であり、かつ通常使用されるものである。別途定義しない限り、一般に、本明細書中で使用される全ての技術的及び科学的用語は、本開示が属する分野の当業者に通常理解されるものと同じ意味を有する。本明細書中で使用される用語について複数の定義が存在する場合、特に指定しない限り、本節のものが優先する。
一態様において、本明細書中に、1つ以上のプロトパナキサジオール系ジンセノサイド、例えば、ジンセノサイドRb1、Rb2、Rb3、Rc、Rd、Rg3、Rh2、Rs1、Rs2、R1、R4、F2などを含む、対象への投与に適した組成物を提供する。いくつかの実施態様において、提供される組成物は、基本的にプロトパナキサジオール系ジンセノサイドからなる。特定の実施態様において、提供される組成物は、基本的にジンセノサイドRb1、Rb2、Rc及び/又はRdからなる。
(8.1. アルコール脱水素酵素(ADH)活性)
この実施例は、全てではないがいくつかのジンセノサイドが、アルコール脱水素酵素(ADH)の触媒活性を低下させることを示している。特に、プロトパナキサジオール系ジンセノサイドは、ADH活性を低下させることが見出されたのに対して、プロトパナキサトリオール系のジンセノサイドは、ADH活性に対して有意な効果を有さなかった。
この実施例は、全てではないがいくつかのジンセノサイドが、ALDHの触媒活性を増大させることを示している。特に、プロトパナキサジオール系ジンセノサイドは、ALDH活性を増大させることが見出されたのに対して、プロトパナキサトリオール系のジンセノサイドは、ALDH活性に対して有意な効果を有さなかった。
この実施例では、種々のジンセノサイド栄養補助食品組成物の製造を説明する。
下記の実施例は、ヒト対象において、ジンセノサイドが、エチルアルコール不耐性に関連する症状を予防、又は軽減できることを示す。
この実施例では、エチルアルコールを飲むことによる、血液中アセトアルデヒド濃度の最大値が、ジンセノサイドの非存在下でエチルアルコールを飲んだ後の対象における最大濃度に対して、ジンセノサイドを投与後の対象においては、減少できることを示す。
処理群(ジンセノサイド及びエチルアルコール)及び対照群(偽薬及びエチルアルコール)に分類する。ジンセノサイド又は偽薬、及びエチルアルコールの投与の前後で、心拍数及び血液試料を各対象から取得する。液体組成物の形態のプロトパナキサジオールジンセノサイドを、0.8mgのジンセノサイド/体重kgの用量で、処理群に経口投与し、かつ偽薬を対照群に投与する。ジンセノサイド又は偽薬の投与の30分後に、エチルアルコール(0.5g/kg)を各対象に投与する。定期的に心拍数を監視し、血液試料を最大6時間の経時変化で取得する。また、対象は、頭痛、眩暈などの自己報告を求められる場合があり、紅潮及び/又は他の症状について評価され得る。およそ一週間後に、同じ対象について該実験を繰り返すが、処置を取り替える。つまり、前回に対照群だった対象は、処理群になり、前回に処理群だった対照も同様である。
上記に説明したように、特定の個人はエチルアルコールの影響に敏感であり、そのような個体においては、他のより大きな一般集団の人々では、通常はそのような症状を引き起こすことがない少量での摂取時に、顔面潮紅及び心拍数の上昇などの症状が引き起こされるであろう。この研究では、少量のアルコールに対してそのような症状を示す病歴を有する個体において、本明細書中に提供されるジンセノサイド組成物を投与することによって、顔面潮紅の重症度及び心拍数の上昇が、軽減され得ることを示す。紅潮の病歴を有する個体を、下記の研究の参加のための対象として選択する。
i. 座っている間の安静時のHR(心拍数)が、少なくとも30拍/分に上昇(30分間の飲酒中、15分ごとの自身の心拍数を監視する。)。;
ii. 顔面潮紅スコアが≧3+。;
iii. アセトアルデヒド曝露と関連した実験1以上の症状、例えば、頭痛、軽い眩暈、悪心、嘔吐など。
i. 任意のアルコールを飲む直前に、座りながら彼又は彼女の安静時の脈拍を測る;
ii. 家族又は友人に「赤み」の判断を依頼し、かつ顔面潮紅を自己評価する。飲酒前ベースラインの顔写真を撮る(本研究を通して全ての写真の照明条件は同じでなければならない。)。
iii. 上記の工程A1、b部と同一の白ワインを同量飲み、アルコール摂取の時間を記録する。
i. 任意のアルコールを飲む直前に、座りながら彼又は彼女の安静時の脈拍を測る;
ii. 家族又は友人に「赤み」の判断を依頼し、かつ顔面潮紅を自己評価する。飲酒前ベースラインの顔写真を撮る(本研究を通して全ての写真の照明条件は同じでなければならない。)。
iii. 上記の工程A1、b部と同一の白ワインを同量飲み、アルコール摂取の時間を記録する。
Claims (22)
- 非ジンセノサイド画分及びジンセノサイド画分からなる、対象への投与に適した、ジンセノサイド組成物であって、該ジンセノサイド画分が、Rb1、Rb2、Rc及びRdからなり、該ジンセノサイド組成物が医薬組成物である、前記ジンセノサイド組成物。
- 前記非ジンセノサイド画分が、医薬として許容し得る担体、賦形剤、希釈剤、又は溶媒を含む、請求項1記載のジンセノサイド組成物。
- 前記ジンセノサイド組成物が、対象への経口投与に適している、請求項1記載のジンセノサイド組成物。
- 非ジンセノサイド画分及びジンセノサイド画分からなる、対象への投与に適した、ジンセノサイド組成物であって、該ジンセノサイド画分が、Rb1、Rb2、Rc及びRdからなり、該ジンセノサイド組成物が、栄養補助食品組成物である、前記ジンセノサイド組成物。
- 前記非ジンセノサイド画分が、グルタミン又はシリマリンを更に含む、請求項1記載のジンセノサイド組成物。
- 対象におけるアセトアルデヒド濃度の減少速度を増加させるための、請求項1記載のジンセノサイド組成物。
- 対象における高アセトアルデヒド濃度の症状を予防する、又は寛解させるための、請求項1記載のジンセノサイド組成物。
- エチルアルコールの摂取に起因する疾患又は障害に対する患者の危険を低減させるための、請求項1記載のジンセノサイド組成物であって、アセトアルデヒドが、該対象によるエチルアルコール消費の生成物であり、かつアセトアルデヒドの異化反応を増加させることで、該対象における、エチルアルコールの摂取に起因する疾患又は障害の危険を低減させる、前記ジンセノサイド組成物。
- 前記対象がヒトである、請求項6〜8のいずれか一項記載のジンセノサイド組成物。
- 前記対象が、変化したエチルアルコール代謝を有する、請求項6〜8のいずれか一項記載のジンセノサイド組成物。
- 前記対象が、アルデヒド脱水素酵素サブタイプ2(ALDH2)活性が低下している、請求項10記載のジンセノサイド組成物。
- 前記アセトアルデヒドが、前記対象におけるエチルアルコール異化反応の生成物である、請求項6又は7記載のジンセノサイド組成物。
- 前記対象が、たばこの煙中又は該対象の外部環境中のアセトアルデヒドに曝露される、請求項6又は7記載のジンセノサイド組成物。
- 高アセトアルデヒド濃度の症状が、紅潮、高心拍数、動悸、低血圧、悪心、眩暈、頭痛、嘔吐、下痢、胃の不調、運動失調、又は意識の混乱を含む、請求項7記載のジンセノサイド組成物。
- 前記疾患又は障害が、宿酔、アルコール性胃炎又はアルコール性肝臓障害を含む、請求項8記載のジンセノサイド組成物。
- 前記疾患又は障害が、上気道消化管癌、消化管癌又は乳癌を含む、請求項8記載のジンセノサイド組成物。
- 前記上気道消化管癌が、食道癌、中咽頭癌、下咽頭癌、喉頭癌、頭部癌、又は頸部癌を含む、請求項16記載のジンセノサイド組成物。
- 前記消化管癌が、胃癌又は結腸癌を含む、請求項16記載のジンセノサイド組成物。
- 前記アセトアルデヒド濃度が、前記対象の血中、唾液中及び/又は組織中におけるものである、請求項6又は7記載のジンセノサイド組成物。
- 1日あたり、前記対象の体重1キログラムあたり、0.1μg〜100mgのジンセノサイドが投与されるように調製されている、請求項6〜8のいずれか一項記載のジンセノサイド組成物。
- 1日あたり400ミリグラム〜2グラムのグルタミンと組み合わせて投与されるように調製されている、請求項20記載のジンセノサイド組成物。
- 1日あたり、20ミリグラム〜800ミリグラムのシリマリンと組み合わせて投与されるように調製されている、請求項20記載のジンセノサイド組成物。
