材料および方法:
a) DNA 調製/精製キット:
いくつかの 市販の DNA 調製および精製キットを提供されたマニュアルにしたがって用いた(表3参照)。
TOPO(登録商標) クローニングは提供されたマニュアルにしたがって行った ( pCR(登録商標)4Blunt-TOPO(登録商標)へのクローニングおよびpCR(登録商標)-Blunt II-TOPO(登録商標)へのクローニング)。常に 4 μlのPCR産物をクローニングに用いた。2および4 μlの各クローニング反応を One Shot(登録商標) 化学的コンピテント大腸菌 TOP10F' 細胞(Invitrogen Corp.、Carlsbad、CA、USA)に上記プロトコールにしたがって形質転換した。
ライゲーション反応およびプラスミドの大腸菌への形質転換はSEM (simple and efficient method)-プロトコールによって行った[16]。化学的コンピテント 大腸菌 TOP10F' 細胞を形質転換に用いた。
コンピテント・ピキア・パストリス 細胞の調製: 所望のピキア・パストリス宿主株のシングルコロニーを用いて、300 ml バッフル付き広首三角フラスコ中の50 ml YPD (2% グルコース)に接種した。30℃、一晩、60% 湿度、130 rpm (Pilot Shake(登録商標) RC-2 TE) でのインキュベーションの後、特定の体積のこの前培養物を用いて2 l バッフル付き広首三角フラスコ中の200 mlの YPD (2% グルコース)に接種し、595 nmでの吸光度(OD595)を約 0.1とした。培養物を前培養物と同じ条件下で培養して吸光度1.0〜 1.5とした。細胞を4℃、4000 rpmで10 分間ペレットとし、200 ml 氷冷滅菌水に再懸濁した。この手順を3回繰り返し、細胞をそれぞれ100 ml 水、10 ml 1 M ソルビトールおよび0.5 ml 1 M ソルビトールに再懸濁した。
所望のピキア株のシングルコロニーを100 μl マイクロチューブ中の100 μl ddH2Oに再懸濁し、95℃で5〜 10 分間加熱した。13,200 rpmで1 分間の遠心分離の後、10 μlの 上清をPCR 反応の鋳型として用いた。5 μlのこの第一PCRラウンドを第二PCRラウンドの鋳型として用いた。5 μlの第二PCRラウンドを対照ゲルのために用いた。PCR 反応には10 pmolの 各プライマー (AOX1_col および GFPrev)、200 μMの 各 dNTPおよび2.5 ユニットの Hot Star Taq(登録商標) DNA ポリメラーゼ (QIAGEN)またはTaq DNA ポリメラーゼ (Promega)が最終体積50 μlにて提供されたマニュアルにしたがったバッファー条件下で含めた。配列決定のために、第二PCR産物をQIAquick(登録商標) PCR Purification Kitを用いて精製した。
所望のピキア・パストリス株を一晩、回転容器上の滅菌 12 ml PP-チューブ中の5 ml YPDで30℃で最終 OD595が5-10となるまで培養した。1.5 mlの 培養液を使用して、Easy-DNA(商標) Kitを用いて提供されたプロトコールにしたがってDNAを単離した。
溶液中のタンパク質濃度の測定は生化学で長く用いられてきた。その主な用途の一つは、酸素消費速度についてこの例で行われているように総タンパク質量に対して多数の生化学的方法を規準化することである。もっとも一般的に用いられているタンパク質濃度の測定方法は Bradford法、Lowry 法および BCA(商標) 法である。これらの方法は感受性、ダイナミックレンジおよび特定の試薬に対する適合性の面で明確な限界を有する。これら3つのアッセイのなかでは、Bradford法および Lowry法がBCA(商標)と比べてより信頼でき、再現性が高い。一方 Lowry法およびBradford法は界面活性剤 および/または 還元剤が存在する場合に深刻な限界を有し、その場合、ブランクの値が高くなる。したがってBCA(商標) アッセイが化学的溶解の後に選択すべき方法である。タンパク質濃度をY-Per(登録商標)による化学的細胞溶解後にBCA(商標)-アッセイを用いて、BSAを標準として指示マニュアルにしたがって測定したので(Pierce Biotechnology Inc.)、主な工程のみを以下に簡単に記載する。200 μlの 培養液を4000 rpmで 4℃で5 分間遠心分離した。上清を捨てた後、ペレットをピペッティング操作により100 μl Y-Per(登録商標)に再懸濁した。懸濁液をThermomixer中で 室温で600 rpmで20 分間1.5 ml マイクロチューブ中でインキュベートした。細胞細片を13,200 rpmで 室温で10 分間ペレットとした後、上清を新しいマイクロチューブに移し、4℃でBCA(商標) アッセイまたはSDS-PAGEのために保存した。25 μl サンプルをマイクロプレートウェル中で 200 μl BCA(商標) ワーキング試薬 (試薬 A: 試薬 B = 50:1)と混合し、徹底的に30 秒撹拌し、プレートシーラー (Promega)で密封した。37℃での30 分間のインキュベーションおよび室温への冷却後、562 nmでの吸収をSpectramax Plus 384 プレートリーダーを用いて測定した。必要な場合、サンプルをddH2Oで希釈した後、BCA アッセイに用いた。
SDS-PAGE用のサンプルを上記のセクションで記載したように試薬としてY-Per(登録商標)を用いて化学的細胞溶解により調製した。10 μlの 溶解液を10 μl 2x SSB (sigma サンプルバッファー)と混合し、95℃で5-10分間インキュベートし、15 μlのこの混合物をタンパク質ゲルにロードした。電気泳動は180 Vで約 1時間行い、タンパク質バンドはクーマシー(商標) ブルー染色を用いて検出した。
グルコース濃度は除タンパクを行わずにグルコース- UV ヘキソキナーゼ法を用いて測定した(DIPRO med Handels GmbH、Weigelsdorf、Austria、Prod. no. D590522)。50 μlのピキア培養液をPCR マイクロプレートに移し、4000 rpmで5 分間遠心分離した。10 μlの上清をUV-Star マイクロプレート中の190 μlのヘキソキナーゼ 試薬に添加し、室温で15 分間インキュベートした。インキュベーション後、340 nmでの吸光度をSpectramax Plus 384 プレートリーダーを用いて測定した。
ピキア・パストリス株をBMD(1%)中で最終 OD595が1.5となるまで培養し、10倍ずつ希釈して最終希釈率 105とした。寒天プレートに48ピンレプリケータを用いて移した。プレートをコロニーが現れるまで30℃でインキュベートした(通常MD プレート上で2日間)。
すべての配列アラインメント はINRAホームページ (Institut National de la Recherche Agronomique、Paris、France) (prodes.toulouse.inra.fr/multalin/multalin.html)のMultAlin [17]またはEuropean Bioinformatics Institute (EBI、www.ebi.ac.ch/clustalw)のClustalW [18]を用いて行った。MultAlinによる配列比較のために常にDNA 配列類似性マトリックスを比較のために用いた。
転写因子分析をGenomatix Software GmbH サーバー のGenomatixSuite 1.6.1 におけるMatInspector Release professional 6.1 Jan. 2003 [15] を用いて行った。pPICZ B 由来のPAOX1 配列を用いてMatrix Family Library Version 3.1.1 April 2003 group ALL fungi.lib (www.genomatix.de)を用いて転写因子結合部位を検索した。

GFP-ZeoをAOX1プロモーター変異体により駆動される遺伝子発現のためのレポーターとして用いた。ATG 開始コドンの周囲の配列を酵母において高度に発現される遺伝子についてのKozak コンセンサス配列の最少必要条件を満たすよう構築した。GFP-Zeo 遺伝子のまえのプロモーター領域を変更するため、EcoRI 制限部位を重複伸長 PCR により挿入した(表 8)。
AOX1 プロモーターの左腕をフォワードプライマーとしてP(AOX1)forwおよびリバースプライマーとしてAOX n rev (n=1...9)を用いて合成した。右腕はフォワードプライマーとして10 pmolの AOX n forw (n=1...9) およびリバースプライマーとしてP(AOX1)revを用いて合成した。すべての腕は鋳型として12 ngのベクター pAOX および10 pgの 各 プライマーを用いて合成した。反応には適当なバッファー条件中10 pmolの 各 プライマー、200 μMの 各 dNTP および0.6 U Pwo DNA ポリメラーゼを含め最終体積を50 μlとした。PCRをサーマルサイクラーで 30 サイクル (95℃、1 分; 55℃、1 分; 68℃、1 分 30秒)行い、最初の変性工程を5 分95℃とし、最終伸長工程 を10分68℃とした。すべての腕はアガロースゲルで精製した後、重複伸長 PCRの鋳型として用いた。
ピキア・パストリス形質転換は以前に記載のようにして行った。PAOX1 (またはPAOX1Δ)-GFP-Zeo-AOX1 TTの組込みの選抜はアリコット中の形質転換された再生したピキア細胞をMSM-Zeo 寒天プレートに播くことにより行った。
ピキア・パストリス中でのGFP-Zeoの発現をSpectramax Gemini XS プレートリーダー中で、395 nmでの励起および507 nmでの発光によるGFPの蛍光検出により分析した。上記のようにディープ・ウェル・プレートで培養した50 μlの ピキア・パストリス培養液をFIA マイクロタイタープレート中でddH2Oにより1+3に希釈した。サンプル量が限られていたため 一度の測定のみを行った。得られたすべての平均 ± 標準偏差を少なくとも 3種類の培養液(ウェル)から算出した。
遺伝子のコード領域のPCR 突然変異誘発はよく開発され確立されているが、プロモーター領域の突然変異誘発に関しては全く知られていない。知識がないため、いくつかの突然変異誘発条件を行った: 突然変異範囲における偏りを最少にするために、Taq DNA ポリメラーゼとMutazyme(登録商標) DNA ポリメラーゼ (Stratagene Inc.)との2種類のポリメラーゼを用いた。プロモーター配列の進化のための突然変異頻度に関する知識が完全にないという事実により、いくつかの突然変異頻度 (理論的には 1〜 ~14/kb)を試験した。
I)レポーター系の特徴決定
今日まで、多数のGFP 変異体が分子生物学において用いられている。数個の点突然変異においてのみしか変わらないのに、それらの特徴は大幅に異なる。フォールディング特性の改善の他に、それらの蛍光スペクトルおよび量子収率も異なり、それゆえ強度もかなり異なる。緑色蛍光タンパク質はその励起極大に応じて2つの主な群に分けられる: 励起極大が395 nmでありより小さい極大が470 nmである野生型 GFP 変異体、および、励起極大 が480-490 nmである赤方にシフトした GFP 変異体。そのアミノ酸配列にしたがって、サイクル-3-GFP は野生型 GFP 変異体の群に属し、励起極大が395 nmである。
微生物細胞(例えば、酵母、細菌)のスモールスケール培養は通常振盪フラスコ培養で行われる。振盪フラスコ中の大きい微生物ライブラリーの接種および培養は労力と時間がかかり、コストが高くなる。近年、深ウェル・マイクロタイタープレートを用いるマイクロスケール培養系が替わりとして開発された。例えば、96 または384 株/培養の並行操作および必要材料が少ないことのために、マイクロタイター系は労力、時間そしてコストの点で振盪フラスコよりも優れている。いくつかの理由のため、マイクロタイター系の主な欠点である、サンプル体積が少なく、通気効率が低いことは、さほど問題ではない: (1) 分析系における技術的進歩により多数の化合物の検出限界が低くなった結果、非常に小さいサンプル体積しか必要ではない; (2)深ウェル・マイクロタイタープレートでの増殖方法および装置も改善された。いくつかの研究において、マイクロタイタープレートでの通気速度およびそれゆえ培養条件は振盪フラスコと同様であることが示された。出芽酵母(S. cerevisiae)におけるサイクル-3-GFPをレポーター タンパク質として用いるGAL1 プロモーターのリアルタイム研究が振盪フラスコ研究と一致することが示された。
以前に記載のように、いくつかの転写因子の結合部位についてのコンセンサス配列は知られている。AOX1 プロモーター配列の配列分析により、いくつかの推定転写因子結合部位が明らかとなり、特に興味深いいくつかのヒットがあった。熱ショック因子およびストレス応答要素モチーフのなかで、一般的にグルコース制御に関与していることが知られているいくつかの転写因子の結合部位がみられた。もっとも興味深い結合部位を表11および図2 に要約した。

いくつかの配列がメタノール誘導性遺伝子の制御に関与していることが文献に記載されている。ピキア・パストリス AOX2 プロモーターの欠失分析に基づいて3つの調節領域が記載され、2つは負に作用する領域(URS1およびURS2、上流抑制配列)であり、1つは正に作用する領域 (UAS、上流活性化配列)である [3]。 ハンセヌラ・ポリモルファ(H. polymorpha) MOX プロモーターについて、2つの上流活性化配列(UAS1およびUAS2)および1つの抑制因子結合部位 (URS1)が同様に記載されている [8]。
転写因子分析および複数配列アラインメントに基づき、 9つのプロモーター領域が先に記載のようにして重複伸長 PCRの欠失のために選択された。 AOX1 プロモーター欠失コンストラクトをpAOX ベクターにクローニングしてレポーター 遺伝子 GFP-Zeoの5'側の「野生型 AOX1」プロモーターを置換した。 プラスミドを直鎖化し、ピキア・パストリス ゲノムに組み込んだ。
このたび、転写因子結合部位のコア配列内の特異的突然変異の結果、そのエフェクター能力が有意に変化することが示された。おそらくいくつかの活性化因子および抑制因子タンパク質がAOX1 プロモーターに作用する結果、その非常に強力な制御が導かれる(グルコース下ではほとんど活性はなく、メタノール中では非常に活性は高い)。それゆえAOX1 プロモーターのランダム突然変異誘発の結果、抑制因子結合部位活性が破壊または低下したいくつかのプロモーター変異体が生じるはずである。様々な突然変異率の一連のPCR 反応を行った。その結果得られるプロモーター変異体をAOX1 遺伝子がGFP-Zeo株によって置換されているピキア・パストリス GFP-Zeo MutS A9 株に形質転換した。野生型 AOX1 プロモーターの突然変異誘発したプロモーター変異体による置換が特定の率で起こるはずである。グルコースが培地に存在する場合により高い発現速度を示すプロモーター 変異体のスクリーニングをMD-Zeo 寒天プレートで行った。
実施例1の結果に基づき、第二世代の欠失変異体を作製した。第一の群と異なり、これら新しい欠失コンストラクトにおいては、推定転写因子結合部位の短い特定の配列ストレッチのみ(5-15 bp)を欠失させた(表15)。
a) 突然変異誘発:
すべての欠失はWang et al.[34]にしたがって2段階部位特異的突然変異誘発-プロトコールを用いて導入した。第一段階において2つの別の反応 (1つはフォワードプライマーのため、1つはリバースプライマーのため)を評価した(100 ng pAOX 鋳型、15 pmol プライマー、200 μMの 各 dATP、dTTP、dCTPおよびdGTP、2.5 U PfuUltra(商標) ポリメラーゼ、適当なバッファー条件中総体積50 μl)。25 μlのこれら2つのPCR 反応を合わせ、第二のPCR 反応工程を行った。
上記のように構築したプラスミドを調製し、実施例1に記載のようにピキア・パストリスに形質転換した。
発現レベルに対する強力な効果が、すべての突然変異が有意な正または負のプロモーター活性に対する効果を有していた実施例1のより大きい欠失について既に記載したように、実施例2の短い突然変異によって観察された。特定の転写因子結合部位の短い欠失はプロモーター活性に強力な効果を有し、個々の制御部位 (例えば、転写因子結合部位)の調節特性に関するより正確な情報を与える。Gcr1は、その結合部位がΔ6 欠失に含まれるため特に興味深い。pAOXΔ6 欠失 突然変異体のプロモーター領域およびピキア・パストリス クローンのゲノム DNAのコロニー PCR産物の配列決定により、プロモーター領域におけるさらなる欠失が明らかとなった(配列番号1のヌクレオチド737〜 738 (-217〜 -216)の欠失)。このプロモーター変異体はプロモーター活性の上昇を導く結果、抑制解除条件下においてより高い発現速度が達成されるという事実により、このさらなる突然変異を本発明によるプロモーターに導入するとかかる条件下でタンパク質発現を上昇させることが出来る。
ほとんどの場合において多コピー株の作製の結果、GFP-Zeo 超発現株が生じた。多くの場合、これらの株は主にメタノール誘導条件下でd6*F10 株より高い発現レベルを有する。多コピー株の作製はいくつかのコンストラクト、特にΔ6* コンストラクト、二重欠失 d2d6、コンストラクト、例えばΔ1、Δ2 およびΔ6* 欠失ならびに例えば、Gcr1、Rap1、abaA、Hap2345_1、また例えば、QA-1F、Adr1、Hsf_2_Mat1MC およびHsf_2_Hap2345_1 (図7参照)により達成可能である。これらの株において誘導条件下でd6*F10 株と比較してより高い発現速度がみられる。一方、d6*F10 株は既知の抑制解除条件下まで、作製したいずれかのその他の株よりも多くのGFP-Zeoを産生することが出来た。Δ6* コンストラクトのピキア・パストリスへの形質転換の繰り返しの結果、多数の多コピー株が生じ、これらは特に抑制解除条件下でd6*F10 株と匹敵する活性であった(図8)。
すべてのGFP-Zeoの結果のその他の基本的によく発現する工業的に関連するタンパク質(例えば、酵素)への適用可能性を試験するために、いくつかのプロモーター変異体をかかるレポーター酵素(例えば、PaHNL5αおよびHRP)の前にクローニングした。
プロモーター変異体をベクターpPICZαB-HRP-WT [35]およびpGAPZ A-PaHNL5αにクローニングした。pPICZαB-HRP-WTにおけるプロモーター交換のために、NdeI 制限部位を部位特異的突然変異誘発によりプロモーターの5’末端に挿入した(鋳型として100 ng ベクター、プライマー Nde1PICZforおよびNde1PICZrev - 表18参照)。その結果得られたベクターをpPICZαB-NdeI-HRP-WTと称した。
ピキア・パストリス形質転換のために、すべてのHRP ベクターをNdeIで直鎖化し、すべてのPaHNL5α プラスミドをBglIIで直鎖化した。形質転換は実施例1に記載のようにして行った。ピキア・パストリス株の培養も実施例1に記載のようにして行ったが、わずかにそれを改変した。最初の BMD(1%)の量を350 μlに増やし、60 時間後100 μlの 培養液をとって遠心分離した(4000 rpm、4℃ 、10分間)。メタノール誘導は実施例1とまったく同様にして行った。
15 μlの上清をPS マイクロタイタープレート中の50mM NaOAc バッファー pH 4.5中の150 μl 1mM ABTS/2,9mM H2O2 に添加した。吸収は5 分間405nmでSpectramax Plus384 プレートリーダー (Molecular Devices、Sunnyvale、CA、USA)で測定した。
50 μlまたは10 μlの 上清をUV-Star マイクロタイタープレート中の100または140 μl 0.05M リン酸クエン酸バッファー pH 5.0に添加した。反応を50 μlの0.06 M マンデロニトリル溶液 (0.003 M リン酸クエン酸バッファー pH 3.5中)の添加により開始させ、 5 分間280 nmでSpectramax Plus384 プレートリーダー (Molecular Devices、Sunnyvale、CA、USA)で測定した。
代替のレポータータンパク質である PaHNL5αおよび HRP を用いた結果は、明らかにGFP-Zeoを用いて検出されたプロモーター活性の転換能力を示す(表17)。
GFPを有するプロモーター変異体を試験するために、実施例1および2に記載のプロモーター変異体をサイクル-3-GFP 遺伝子の前にクローニングしした。
ベクター pAOXにおけるサイクル-3-GFP中の内部 BamHIおよびXhoI 制限部位をプライマー Bam-del-fおよびXho-del-f (表19)および鋳型として100 ng ベクターを用いる部位特異的突然変異誘発によって欠失させた。GFP 断片をその結果得られたプラスミド (10 ng)からプライマーGFP-Zeo forw (配列番号 4、表 7、10 pmol)およびwtGFP-XhoI-r (表19、10 pmol)およびPhusion(商標) ポリメラーゼを用いる適当な条件下でのPCRにより増幅した。その結果得られたPCR産物をEcoRI/XhoI 制限切断およびT4 DNA リガーゼを用いるライゲーションの実施によってベクター pPICZ Bにクローニングした。その結果得られたプラスミドをpPICZ-GFPと称した。
ピキア・パストリス形質転換のためにすべてのプラスミドをBglIIによって直鎖化した。形質転換を実施例1に記載のように行った。形質転換および2時間の再生相の後、細胞を、100 μg/ml ゼオシンを含有するYPD-Zeo 寒天プレートに播いた。
この場合も、GFPをレポーター系として用いる結果は、 GFP-Zeoを用いて検出されるプロモーター活性の転換能力を示す(表20)。
実施例1および2に記載のように、ゼオシンを選抜マーカーとして用いる多コピー株の発生は非常に一般的である。多コピー株の頻度は、選抜プレート上のゼオシン濃度をそれぞれ500 および1000 μg/mlに上昇させることによって劇的に上昇させることが出来た。
ほとんどすべての転写因子結合部位を有さない系におけるAOX1 プロモーターの小部分を試験するために、AOX1 プロモーターを、位置 -176 および-194においてTATAボックスの前の数塩基対を切断して、基本プロモーター要素AOX176 および AOX194 (表21)を得た。基本プロモーター断片の前のプロモーター要素のクローニングならびに基本プロモーターのクローニングを可能とするために、 BspTI およびEcoRI 制限部位をそれぞれ5'および3’末端に挿入した。
基本 AOX1要素をpAOX (10 ng)から、プライマーAOX1basalrv (表21、10 pmol)およびAOXbasalfwn (10 pmol、AOX194)およびAOX176fw (10 pmol、AOX176)をそれぞれ用いて増幅した。PCR はPhusion(商標) ポリメラーゼ (0.6 u)を用いて適当な条件で総体積 50 μlにて行った。
ピキア・パストリス 形質転換のために、すべてのプラスミドをBamHIにより直鎖化した。形質転換を実施例1に記載のように行った。形質転換および2時間の再生相の後、細胞を100 μg/ml ゼオシンを含有するYPD-Zeo 寒天プレートに播いた。
この実験は、実施例1および2にて同定した小さい要素の付加が、AOX1 プロモーターまたは出芽酵母(S. cerevisiae) LEU2 プロモーター由来の基本プロモーター要素のプロモーター強度の上昇に利用できることを示す。
形質転換後にみられる多コピー株の発生および頻度は実施例4に記載のものとまったく同様であった。プラスミド内の直鎖化の部位が異なることは、多コピー株の作製に影響を与えなかった。

Claims (32)
- メタノール誘導条件下における高発現のための、配列番号1のヌクレオチド170〜235 (-784〜-719)が欠失した、野生型ピキア・パストリス AOX1 プロモーター(配列番号1)の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーター。
- 請求項1の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーターおよびタンパク質、ペプチドまたは機能的核酸をコードする少なくとも1種類の核酸を含む核酸分子であって、該プロモーターおよび該核酸が互いに作動可能に連結して1コピーまたは多コピー発現カセットを形成している核酸分子。
- 請求項1の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーターまたは請求項2の核酸分子を含むベクター。
- 請求項1の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーター、請求項2の核酸分子の少なくとも1つの断片であって請求項1の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーターおよびタンパク質、ペプチドもしくは機能的核酸をコードする少なくとも1種類の核酸を含む断片、または請求項3の少なくとも1つのベクターを含む細胞。
- 真核生物細胞である、請求項4の細胞。
- 酵母細胞である、請求項5の細胞。
- メチロトローフ酵母細胞である、請求項6の細胞。
- 該メチロトローフ酵母細胞が、カンジダ、ハンゼヌラ、ピキアおよびトルプロシスからなる群から選択される、請求項7の細胞。
- ピキア・パストリス細胞である、請求項4〜8のいずれかの細胞。
- 以下を含む選択されたタンパク質の発現のためのキット:
i) 請求項3のベクター、および
ii)請求項1のプロモーターの制御下で該タンパク質を発現することが出来る細胞。 - 該細胞が、酵母細胞である、請求項10のキット。
- 該細胞が、メチロトローフ酵母細胞である、請求項11のキット。
- 該メチロトローフ酵母細胞が、カンジダ、ハンゼヌラ、ピキアおよびトルプロシスからなる群から選択される、請求項12のキット。
- 細胞がピキア・パストリス細胞である、請求項10〜13のいずれかのキット。
- 以下の工程を含む、細胞において組換えタンパク質、ペプチドまたは機能的核酸を発現させる方法:
-請求項1のAOX1 プロモーターおよび、タンパク質、ペプチドまたは機能的核酸をコードする核酸を含み、該プロモーターが該核酸に作動可能に連結している、請求項2の核酸分子または請求項3のベクターを提供する工程、
-該細胞を該ベクターまたは該核酸分子で形質転換する工程、
-形質転換細胞を培地で培養する工程、および
-形質転換細胞において発現した該タンパク質、ペプチドまたは機能的核酸を単離する工程。 - 該タンパク質、ペプチドまたは機能的核酸の発現を誘導する工程をさらに含む、請求項15の方法。
- 該細胞が、酵母細胞である、請求項15または16の方法。
- 該細胞が、メチロトローフ酵母細胞である、請求項17の方法。
- 該メチロトローフ酵母細胞が、カンジダ、ハンゼヌラ、ピキアおよびトルプロシスからなる群から選択される、請求項18の方法。
- 細胞がピキア・パストリス細胞である、請求項15〜19のいずれかの方法。
- 請求項2の核酸分子、請求項3のベクターまたは請求項4〜9のいずれかの細胞の、タンパク質、ペプチドまたは機能的核酸の発現のための使用。
- 以下の工程を含む、超発現クローンを単離する方法:
a) 請求項1の突然変異体ピキア・パストリス・アルコールオキシダーゼ 1 (AOX1) プロモーターおよびタンパク質、ペプチドまたは機能的核酸をコードする少なくとも1種類の核酸およびマーカー耐性遺伝子を含む核酸分子であって、該プロモーターおよび該核酸が互いに作動可能に連結して1コピーまたは多コピー発現カセットを形成している核酸分子、または該核酸分子を含むベクターを、細胞に導入する工程、
b)工程a)の細胞を、誘導条件下での超発現クローンの選択的増殖のための、選抜マーカー、非抑制性炭素源およびメタノールを含む培地に移す工程、
c)工程b)からの細胞を該培地上でインキュベートする工程、
d)工程c)から得た細胞のコロニーを単離する工程、および、
e)該細胞の発現速度を測定することにより、超発現クローンを検出する工程。 - 選抜マーカーが、抗生物質である、請求項22の方法。
- 選抜マーカーが、ゼオシンまたはジェネテシンである、請求項23の方法。
- 選抜マーカーがゼオシンであり、マーカー耐性遺伝子がsh ble 遺伝子である、請求項22〜24のいずれかの方法。
- 細胞が、酵母細胞である、請求項22〜25のいずれかの方法。
- 細胞が、メチロトローフ酵母細胞である、請求項26の方法。
- 該メチロトローフ酵母細胞が、カンジダ、ハンゼヌラ、ピキアおよびトルプロシスからなる群から選択される、請求項27の方法。
- 細胞がピキア・パストリス細胞である、請求項22〜28のいずれかの方法。
- 非抑制性炭素源が、アラニン、マンニトール、ソルビトール、トレハロース、ラクトースおよびそれらの組合せからなる群から選択される、請求項22〜29のいずれかの方法。
- 核酸分子またはベクターが、形質転換により、またはプロトプラスト融合により、または粒子銃により、細胞に導入される、請求項22〜30のいずれかの方法。
- 形質転換が、エレクトロポレーションまたは化学的形質転換である、請求項31の方法。
