[0016] 本発明はいずれかの動物、好ましくは哺乳動物、より好ましくは愛玩動物に関する。用語”愛玩動物”は、ヒトと密接な関係で生きているいずれかの動物を表わし、いずれかの品種のイヌ類およびネコ類が含まれるが、これらに限定されない。ただし本明細書においてこの用語は、その食事をヒトが管理でき、かつ本明細書に開示する配合物の給餌が有益となりうる、いかなる動物も含むことができるものとする。これらの動物には、たとえば家畜化された農場動物(たとえばウシ、ウマ、ブタなど)、および家畜化されていない動物であってたとえば動物園などに捕獲状態にあるものを含めることができる。好ましい動物にはイヌ類、たとえばイヌが含まれ、これには成長期のイヌ、たとえば子イヌが含まれる。
[0019] 本明細書中で用いる用語”子イヌ”は、一般に誕生と約12カ月の間の歳である未成熟のイヌ類を表わす。
[0029] 本明細書において、本発明方法には、表14〜27に記載する1以上の生物学的状態、経路または疾病状態および遺伝子について、動物の遺伝子発現における有益な改変、場合に応じて、特に、望ましくない生物学的状態もしくは経路もしくは疾病状態に関連する1以上の遺伝子の発現におけるダウンレギュレーション、および/または目的とする生物学的状態もしくは経路に関連する、または疾病状態に対してプラスもしくは予防効果をもつ可能性のある、1以上の遺伝子の発現におけるアップレギュレーションを生じさせるための方法であって、有効量の本発明組成物を動物に、直接にその動物に、または動物が子宮内にいる間は雌親に、投与することを含む方法も含まれるものとする。実際に、動物にゲノムレベルで影響を及ぼすことによる後記の実施例7および8に述べるように、直接にその動物に、または動物が子宮内にいる間は雌親に、本発明組成物を投与することは、表14〜27に挙げる1以上の遺伝子の発現に影響を及ぼすことによって、動物に健康上有益な、さらには予防的な効果をもつことができる。たとえば、有効量の本発明組成物を動物に投与すると、関節炎に関連する軟骨および関節の損傷に関連するインターロイキン1−ベータ、フィブロネクチン、ラクトランスフェリンなどの遺伝子(表14を参照)のダウンレギュレーションを引き起こすことにより、動物において骨および関節の健康状態を増進することができる。
イヌ類の遺伝子効果:受胎から1歳まで
[0035] 成長期動物の発達を増進するために配合した組成物を与えることの遺伝子型効果を解明するために、表1に示した組成物を与えた雌親からの1歳の子イヌにおいて遺伝子発現の変化を同定する実験を行なう。
[0037] 雌親に組成物Hまたは組成物Pを、受胎の前に少なくとも10日間与える。雌親を触診により妊娠が確認されるまでグループ施設で飼育し、次いで出産施設へ移動させる。雌親からの子イヌを雌親に与えたのと同じ飼料で1歳まで(離乳前および離乳後の期間を含む)飼育する。次いで血液試料を子イヌから採取し、mRNAを常法により単離する。Affimetrix Canine−2遺伝子チップを用いて常法によりマイクロアレイアッセイを行なう。
[0047] 離乳から1歳まで組成物Pを与えた子イヌにおける遺伝子発現に対する影響を、組成物Hを与えた子イヌに対比して判定するために実験を行なう。
Claims (22)
- 5〜70%のタンパク質、
0.5〜1.6%のメチオニン、
50〜200ppmのマンガン、
0.1〜0.5%のDHA、
0.1〜0.7%のEPA、
1200〜7500ppmのコリン、
1000〜2000ppmのタウリン、
2.5〜6%のリノール酸、
1〜3%の総n−3脂肪酸、
50〜1200IU/kgのビタミンE、
50〜500ppmのビタミンC、
50〜500ppmのカルニチン、および
2.5〜7gのリジン/1000kcal
を含む、イヌ類における遺伝子発現を調節するためのペットフード組成物。 - 0〜90重量%の炭水化物;
20重量%〜60重量%のタンパク質;
2重量%〜50重量%の脂肪;
0.1重量%〜20重量%の総食物繊維;ならびに
0〜15重量%のビタミン、無機質、およびアミノ酸
を含む、請求項1の組成物。 - 0.1%〜0.4%のDHAを含む、請求項1または2の組成物。
- 100ppm〜500ppmのカルニチンを含む、請求項1〜3のいずれか1項の組成物。
- 0.1%〜0.6%のEPAを含む、請求項1〜4のいずれか1項の組成物。
- 50ppm〜150ppmのマンガンを含む、請求項1〜5のいずれか1項の組成物。
- 0.8%〜1.6%のメチオニンを含む、請求項1〜6のいずれか1項の組成物。
- イヌ類における遺伝子発現を調節する方法であって、イヌ類に請求項1〜7のいずれか1項に記載の組成物を投与することを含む方法。
- イヌ類が子イヌである、請求項8の方法。
- イヌ類が、妊娠中に請求項1〜7のいずれか1項に記載の組成物を与えた雌親から産まれたものである、請求項8または9の方法。
- 子イヌが子宮内にいる、請求項10の方法。
- 雌親に、妊娠前に請求項1〜7のいずれか1項に記載の組成物を与える、請求項10の方法。
- 雌親に、大部分の妊娠期間中に請求項1〜7いずれか1項に記載の組成物を与える、請求項10または12の方法。

- 子イヌに、請求項1〜7のいずれか1項に記載の組成物からなる飼料組成物を与える、請求項16の方法。
- イヌ類に有効量の組成物を投与する、請求項8〜17のいずれか1項の方法。
- イヌ類に、有効な期間、組成物を投与する、請求項8〜18のいずれか1項の方法。
- イヌ類に、少なくとも分娩後1年間または離乳後1年間、組成物を投与する、請求項19の方法。
- 遺伝子が表2〜15に挙げたものから選択される、請求項8〜20のいずれか1項の方法。
- 動物において、表2〜15に記載したいずれか1以上の生物学的状態、経路または疾病状態について、表2〜15に示したいずれか1以上の遺伝子の発現における有益な改変を生じさせる方法であって、有効量の請求項1〜7のいずれか1項に記載の組成物を動物に、直接にその動物に、または動物が子宮内にいる間はその動物の雌親に、投与することを含む方法。
