Description
PQQ依存性PDHの酵素活性を、下記方法により測定した。
培地全体100mLに対して、ソルビトール 2g、酵母エキス 0.3g、肉エキス 0.3g、コーンスティープリカー 0.3g、ポリペプトン 1g、尿素 0.1g、KH2PO4 0.1g、MgSO4・7H2O 0.02g、CaCl2・2H2O 0.1gからなる培地(pH 7.0)100mLを調製し、500mL容の坂口フラスコに該培地80mLを移し、121℃、20分間オートクレーブ処理した。

Claims (4)
- グルコノバクター属に属する微生物による補欠分子族としてピロロキノリンキノンを含むポリオール脱水素酵素の製造方法において、炭素源としてソルビトールならびにグルコン酸およびグルコン酸塩の少なくとも一方を含む培地中で前記微生物を培養する段階を有することを特徴とする、補欠分子族としてピロロキノリンキノンを含むポリオール脱水素酵素の製造方法。
- 前記培地のpHが4〜7である、請求項1に記載の製造方法。
- 前記グルコン酸およびグルコン酸塩に対する前記ソルビトールの培地中質量比が1〜30である請求項1または2に記載の製造方法。
- 前記ソルビトール濃度が培地1Lに対して5〜30g/Lであり、前記グルコン酸およびグルコン酸塩の濃度が培地1Lに対して1〜30g/Lである、請求項3に記載の製造方法。
