Description
本発明は、宿主細胞における改良されたコドンの使用及び/又は希に存在するコドン(rarely occurring codon)をコードするtRNAの発現による、原核宿主細胞中で異種組換えタンパク質を生産するための有利な方法に関する。多数の細菌、特にグラム陽性細菌が、例えば異種組換えタンパク質、例えば分泌タンパク質を菌体内毒素を有しない環境下で生産するための宿主細胞として使用されてきた(参考文献8、9、19)。例えば、肉及び魚の発酵用に食品工業で使用されている細菌スタフィロコッカス・カルノサス(S.カルノサス)(Staphylococcus carnosus)は、この目的に適した細菌である。この細菌は、その上清中に毒性因子及びタンパク質分解活性を有さず、多量のタンパク質を分泌することができる(10)。更に、宿主細胞によりコードされて分泌されるタンパク質の量は少量であり、このことが生成タンパク質の精製をより容易にする。細菌酵素(7、4、17)が例えばS.カルノサス中で組換え的に生成されかつ発現させられてきている。例えばストレプトコッカス(Streptococcus)タンパク質G等の抗原をその表面上に発現する組換えS.カルノサス株は、生ワクチンとして特に有望である(5、12)。
多くの細菌系の1つの重大な不利益は、当該細菌系がヒト遺伝子におけるコドン優先度(codon preference)とは非常に異なる、希に存在するコドン(rare codon)を使用することである。大腸菌(E. coli)中の希に存在するコドンの存在は、組換え遺伝子の遅延した又は減少した発現を導いた(2、6)。それゆえ、本発明の課題は、先行技術の不利益を克服すること及び改善された性質を有する原核生物発現系を提供することである。

発明の詳細な説明
前述の課題は、本願の特許請求の範囲及び発明の詳細な説明の範囲内で解決される。
特許請求の範囲又は発明の詳細な説明における単数形又は複数形の使用は、どんな場合であっても本発明を限定することを意図するものではなく、本発明は代替の形態をも含んでいる。
本発明は、原核宿主細胞中で異種組換えタンパク質を生産する方法であって、宿主細胞遺伝子に対する宿主細胞によるコドンの使用を決定し、宿主細胞内で異種組換えタンパク質をコードする核酸配列中にある宿主細胞中にまれに存在するコドンを頻繁に存在するコドン(frequent codon)で置換し、組換えタンパク質をコードする核酸で宿主細胞を形質転換し、組換え核酸を発現させることを特徴とする方法に関する。本発明の方法を用いると、従来技術から既知の方法と比較して、有意に良好な異種タンパク質の発現速度を達成することができる。本発明の方法は、原核生物の表面に組換えタンパク質を発現させるのに特に有利である。なぜなら、原核細胞の細胞壁への共有結合性固着(covalent anchoring)は無傷のC末端に依存するところ、無傷のC末端が存在せず分泌性の切断タンパク質が本発明の方法では生産されないからである。

「異種」とは、原核宿主細胞中のタンパク質が天然タンパク質ではないこと、すなわち当該タンパク質が当該宿主細胞に特有のタンパク質として存在していることを意味する。「組換え」とは、分子生物学的方法により生産されることを意味する。異種組換えタンパク質は、当業者に既知の全てのタンパク質であってよく、例えば、インシュリン、hGH、tPA、サイトカイン、例えばインターロイキン(IL)、例えばIL−1、IL−2、IL−3、IL−4、IL−5、IL−6、IL−7、IL−8、IL−9、IL−10、IL−11、IL−12、IL−13、IL−14、IL−15、IL−16、IL−17、IL−18、インターフェロン(IFN)α、IFNβ、IFNγ、IFNΩ、IFNτ、腫瘍壊死因子(TNF)、TNFα、TNFβ、TRAIL;G−CSF、GM−CSF、M−CSF、MCP−1及びVEGFである。原核宿主細胞は、当業者に既知の全ての宿主細胞であってよく、特にグラム陽性及びグラム陰性の宿主細胞である。例えば、大腸菌(Escherichia coli)、バチルス・サブチリス(Bacillus subtilis)、ストレプトミセス(Streptomyces)、プロテウス・ミラビリス(Proteus mirabilis)、好ましくはスタフィロコッカス(Staphylococcus)、例えばスタフィロコッカス・カルノサス(Staphylococcus carnosus)、特に公的収集機関、例えば the Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH, Braunschweig, Germanyから入手可能なもの例えば、TM300株(DSM 4600、4601又は4602)である。
本発明の方法の範囲内において、最初に、宿主細胞の基本コーディング遺伝子配列中のできるだけ多くの天然タンパク質について、コドン、すなわちコーディングヌクレオチドトリプレットの使用を分析する。例示として、宿主生物スタフィロコッカス・カルノサスTM300株(10)について51遺伝子のコドン頻度を分析した。分析結果を下記表1に示す。

例えば、ブトウ球菌(Staphylococci)のコーディング配列は約30%という非常に低いG+C含量を有し、A+Tに富むコドンが好ましいことが見いだされた。それゆえ、本発明にしたがう2つの方法により、組換えタンパク質、例えばhGHの生産量が増加する。(i)コーディング遺伝子配列中の希に存在するコドンを頻繁に存在するコドンで交換すること(実施例1も参照のこと)及び/又は(ii)希に存在するtRNAを発現させること(実施例2も参照のこと)。
それゆえ本発明は、異種組換えタンパク質を生産する方法であって、S.カルノサスについては表1に示されるように、本発明にしたがうその他全ての宿主細胞についても同様に、希に存在するコドンの一部又は全部を頻繁に存在するコドンで置換する、例えば、表1に示される分析結果にしたがいGluコドンGAGをGAAで置換する、又は、LeuコドンCTC、CTG及びCTAをTTA、TTG又はCTTで置換することを特徴とする方法に関する。
それゆえ本発明は、異種組換えタンパク質を原核宿主細胞中で生産する方法であって、宿主細胞遺伝子に対する宿主細胞によるコドンの使用を決定し、希に存在するコドンに特異的なtRNAをコードする核酸を有する宿主細胞を組換えタンパク質をコードする核酸で形質転換し、組換え核酸を発現させることを特徴とする方法(実施例2も参照)に関する。当業者は、遺伝子又はcDNAを分析することにより(又はmRNAを逆転写することにより)、宿主細胞の希に存在するコドンを決定することができる。例えば表1に示される宿主細胞S.カルノサスTM300株(10)について決定を行った。本発明は、宿主細胞中の希に存在するtRNAの1つ〜全てに対し、tRNA又はこれをコードする核酸を導入することを含む。前記の宿主細胞は希に存在するコドンに特異的なtRNAを有しているので、本発明にしたがう異種組換えタンパク質は従来技術の既知の方法よりも大量に発現する。この驚くべき本発明の有利な特性は実施例2においても示される。大量の発現は、前記の方法単独又は異種タンパク質をコードする核酸中の、宿主細胞に対しては希に存在するコドンを頻繁に存在するコドンで置換することと組み合わせることにより起こるだろう。

それゆえ本発明は、宿主細胞中の異種組換えタンパク質をコードする核酸において、希に存在するコドンを頻繁に存在するコドンにより置換し、かつ、宿主細胞を希に存在するコドンをコードするtRNA及び組換えタンパク質をコードする核酸で形質転換することを特徴とする好ましい方法を含んでいる。
本発明は、宿主細胞中で使用されかつ15%未満の平均頻度を有するコドンを少なくとも15%の平均頻度を有するコドンで置換することを特徴とする好ましい方法、好ましくは宿主細胞中で使用されかつ7〜12%の平均頻度を有するコドンを12%をこえる平均頻度を有するコドンで置換することを特徴とする方法、特に好ましくは宿主細胞中で使用されかつ7〜10%の平均頻度を有するコドンを7%をこえる又は10%をこえる平均頻度を有するコドンで置換することを特徴とする方法を含んでいる。別の好ましい態様は、宿主細胞中に最も低い頻度で存在するコドンを当該宿主細胞中で最も頻繁に存在するコドンで置換することを特徴とする好ましい方法である。前記の最も低い頻度で存在するコドンが発現につき絶対的に必須である場合、当該コドンの置換は除外されるだろう。
更に本発明は、宿主細胞中で使用されかつ10%未満の平均頻度を有するコーディング核酸コドンを少なくとも10%の平均頻度を有するコドンで置換することを特徴とする好ましい方法を含んでいる。
更に本発明は、宿主細胞が、公的な収集機関、例えばthe Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH, Braunschweig, Germanyから入手可能な大腸菌、例えば大腸菌K12 JM107株(DSM3950)であることを特徴とする好ましい方法を含んでいる。
更に本発明は、宿主細胞がスタフィロコッカス属から選ばれる宿主細胞であることを特徴とする好ましい方法を含んでいる。
更に本発明は、宿主細胞がスタフィロコッカス・カルノサスであることを特徴とする方法(実施例1及び2参照)を含んでいる。
本発明の好ましい方法において、組換えタンパク質は抗体タンパク質である。抗体タンパク質とは、フラグメント、例えば、Fabフラグメント(抗原結合性フラグメント)、F(ab’)2フラグメント、Fvフラグメント(可変部分のフラグメント)又は単鎖Fv(scFv)、ミニボディ(minibody)、ジアボディ(diabody)、トリアボディ(triabody)及びテトラボディ(tetrabody)をも意味する。

実施例1:S.カルノサス特異的コドンを使用した合成hGH遺伝子の構築
タンパク質の例として、ヒト成長ホルモン(hGH)をS.カルノサスTM300株中で発現させた。
成熟hGHのコドン使用とS.カルノサスにおいて示されるコドン使用との比較は、成熟hGHのコドンのうちの51コドン(全コドン数の27%を表す)が、S.カルノサスでのコドン頻度が7%以下のコドンを構成することを示している(表2)。エンテロキナーゼ切断部位を成熟hGHのDNA配列の5’末端に導入して、S.カルノサス特異的コドンを用いてhGHを合成した。合成遺伝子中のコドンは、S.カルノサス遺伝子中における頻度とほぼ同じ頻度で使用した。S.カルノサスより10%未満の頻度で使用されるコドンについては、制限切断部位を導入するために必要なものを除いて使用しなかった(表2を参照)。

合成hGH DNA配列配列は、インビトロでハイブリダイズ及び結合したオリゴヌクレオチドをオーバーラップさせることにより合成した。得られたDNAフラグメントを大腸菌プラスミドpSL1190(3)へクローニングし、配列決定により検証し、hGH発現カセット中に挿入して、NsiIとPstI切断部位との間のhGH cDNAを置き換えた(図1)。新規な発現カセットをBglII−ClaIフラグメントとして単離し、ブトウ球菌(Staphylococci)発現ベクターpTX15のコンパチブルなBamHI及びNarI切断部位へクローニングし、プロトプラスト形質転換によりS.カルノサス TM300を形質転換した。得られた発現ベクターにおいて、lip−hGH融合遺伝子はpTX15のxylプロモーターの制御下で発現した。0.5%キシロースを栄養培地へ添加することによりxylプロモーターのリプレッサーであるXylRを不活性化した後、発現を得た。インビトロでのクローニング及び配列決定は標準的な方法(1)により行った。hGH遺伝子の発現は、正確なN末端を有する成熟hGHを融合タンパク質から遊離させることを許容するエンテロキナーゼ切断部位を含むスタフィロコッカス・ハイカス(Staphylococcus hyicus)由来のlip−シグナル及びプロペプチドとの融合物として行った(図1)。得られた発現ベクターpTX−LipP−hGH4は、成熟hGHのコドン使用以外はベクターpTX−LipP−hGH2と同一である。前記プラスミドのうちの1つを含むS.カルノサス株を、発現ベクターのxylプロモーターを活性化するために0.5%キシロースを補充した改変LB培地(1%ダイズ抽出物、0.5%酵母抽出物、0.5% NaCl)中で培養した。改良hGH DNAの使用は、S.カルノサスにおけるhGH産生効率に対し顕著な効果を有した。Lip−hGH収率は少なくとも2倍に改善し、切断Lip−hGHタンパク質の量は有意に減少した(図2、トレース2及び3)。切断Lip−hGHタンパク質は、細胞外タンパク質分解活性の結果物ではない。なぜなら、Lip−hGHはS.カルノサスの上清中で安定であるからである。S.カルノサス(pTX−LipP−hGH4)の上清中における切断タンパク質の存在の減少は、hGHが最適化DNA配列により早く翻訳されるため、細胞膜を通して分泌される前に細胞内プロテアーゼが当該タンパク質を切断する機会が少なくなったことを示している。

実施例2:hGH cDNA中に希に存在するコドンに特異的なt−RNAの同時発現
アルギニンコドンAGGは、S.カルノサス遺伝子中に極めて希に存在し、アルギニンコドンのわずか0.8%において使用される。対応するtRNA遺伝子はS.カルノサス中で同定されていない。それゆえ、本発明者等はAGGコドンに対する大腸菌tRNAをS.カルノサス中で発現させた。コドンAGG/AGA及びAGGに対するtRNAをコードする大腸菌遺伝子argU及びargWを、その天然のプロモーター及びターミネーターと共に、プラスミドpUBS520又はpSB101(2、16)へクローニングした。フラグメントをSalI−SpHIフラグメント(argU)及びXmaI−EcoRIフラグメント(argW)として単離し、シャトル(転移)ベクターpRB572(4)の対応の制限切断部位へクローニングした。得られたプラスミドpRBargU及びpRBargWで、hGH cDNAを発現するS.カルノサス(pTX−LipP−hGH2)を形質転換した。
得られたS.カルノサス株の上清におけるタンパク質パターンは、S.カルノサス(pTX−LipP−hGH4)と同等であり、Lip−hGHタンパク質の産生が増大しかつ切断タンパク質の産生は低下した。それゆえ、本発明したがう希に存在するtRNAの発現は、例えばS.カルノサスにおけるヒトタンパク質調製に対し有利な特性を有している。

【図1】 図1はプラスミドpTX−LipP−hGH2又はpTX−LipP−hGH4におけるLip−hGH融合物の発現を示す。遺伝子融合物は、スタフィロコッカス・ハイカス由来のシグナルペプチド(SP)及びプロペプチド(PP)並びに成熟タンパク質のhGH部分(黒色)をコードするDNA配列から構成され、xylプロモーター(灰色の三角として示される)により転写される。hGH cDNA又は合成DNA配列をNsiI切断部位とPstI切断部位との間に挿入した。図中の遺伝子の下部に示されるように、hGH DNA配列の5’末端を、エンテロキナーゼ切断部位「DDDDK」、続いて成熟hGH配列をコードするように修飾した。
【図2】 図2はS.カルノサス上清中におけるLip−hGH融合タンパク質の検出を示す。等量の上清を、15%アクリルアミドを含有するTris−グリシンゲル中、SDS−PAGEにより分離し、クーマシーブルーで染色した。トレース1〜5は、空の対照ベクターpTX16(15)、pTX15の誘導体(トレース1)、改良hGH遺伝子を有するプラスミドpTX−LipP−hGH4(トレース2)、hGH cDNAを有するpTX−LipP−hGH2(トレース3)、pTX−LipP−hGH2+pRBargU(トレース4)及びpTX−LipP−hGH2+pRBargW(トレース5)を含むS.カルノサス株の上清を含んでいる。標準タンパク質及びその分子量(kDa)は左側に示されている。上清を、トリクロロ酢酸を用いた沈殿により濃縮した。従来技術の標準的方法によりSDS−PAGEを行った。
【図3】 図3は本発明のhGHヌクレオチド配列及びアミノ酸配列(LipP−hGH4、配列番号3)を示す。関連する制限切断部位には一本下線を付した。シャイン−ダルガーノ配列には二本下線を付した。シグナルペプチダーゼ1及びエンテロキナーゼ切断部位には波線下線を付した。BglII部位とNdeI部位との間及びNsiI部位とPstI部位との間の領域のみを合成的に作成し、最適化した。残りの部分はスタフィロコッカス・ハイカス由来のシグナルペプチド及びプロペプチドのオリジナルの配列から構成される。

Claims (6)
- スタフィロコッカス・カルノサス中で配列番号3の配列を含むLipP-hGH融合遺伝子を発現させる方法であって、
ヒト成長ホルモン(hGH)遺伝子を、スタフィロコッカス・ハイカスリパーゼ(LipP)のリップシグナル及びプロペプチドのコード配列に融合させ、前記LiP-hGH融合遺伝子が正確なN末端を有する成熟hGHの放出を許容する、LipPとhGHの間のエンテロキナーゼ切断サイトを含んでおり、及び10%未満の平均頻度でスタフィロコッカス・カルノサス中で使用されるhGH遺伝子をコードするコドンが、少なくとも10%の平均頻度を有するコドンにより置き換えられ、及び/又は
前記スタフィロコッカス・カルノサスの宿主細胞が10%未満の平均頻度で用いられるコドンをコードするtRNA、複数のtRNA又はこれらをコードする核酸で形質転換される前記方法。 - hGHのG又はCに富むコドンを、A又はTを含むコドンで置換することを特徴とする、請求項1に記載の方法。
- スタフィロコッカス・カルノサスが、AGG-tRNA又はAGA-tRNAをコードする核酸で形質転換される、請求項1又は2に記載の方法。
- スタフィロコッカス・カルノサスが、コドンAGG/AGA及びAGGのためのtRNAをコードするE.coli遺伝子argU及びargWの核酸を用いて形質転換される、請求項1〜3のいずれか1項に記載の方法。
- LipP-hGH融合タンパクが、配列番号3の配列を含む核酸分子でコードされる、請求項1〜4のいずれか1項に記載の方法。
- 配列番号3の配列を含む、LipP-hGH融合タンパクをコードする核酸分子。
