配列番号7で表される塩基配列、又は配列番号7で表される塩基配列と90%以上の相同性を有する塩基配列(以下「請求項7に記載の塩基配列」とする。)、または配列番号8で表される塩基配列、又は配列番号8で表される塩基配列と90%以上の相同性を有する塩基配列(以下「請求項8に記載の塩基配列」とする。)、のいずれかを有する領域である。
E−N primer: 配列番号9で表されるE−G primer
M−N primer: 配列番号10で表されるM−G primer
を使用した。
E−N primer: 配列番号11で表されるE−A primer
M−N primer: 配列番号12で表されるM−C primer
を使用した。
PCR反応プログラム: 94℃5分間処理、続いて、94℃30秒間→56℃1分間→72℃1分間のサイクルを20サイクル行う。その後、72℃10分間処理、最後に4℃に冷却後保温する。
a)配列番号1に基づいた配列番号13、及び2に基づいた配列番号14で表されるテンサイ黒根病抵抗性品種選抜プライマーのペア
b)配列番号3に基づいた配列番号15、及び4に基づいた配列番号16で表されるテンサイ黒根病抵抗性品種選抜プライマーのペア
c)配列番号3に基づいた配列番号15、及び5に基づいた配列番号17で表されるテンサイ黒根病抵抗性品種選抜プライマーのペア
PCR反応プログラム: (第1増幅反応)94℃5分間処理し、次に94℃30秒間→65℃30秒間→72℃1分間のサイクルを13サイクル行う。このとき、アニーリング温度である65℃は、1サイクルごとに0.7℃ずつ低下するタッチダウンで行う。(第2増幅反応)第1増幅反応に引き続き、94℃30秒間→65℃30秒間→72℃1分間のサイクルを20サイクル行う。その後、72℃10分間処理、最後に4℃に冷却後保温する。

- 配列番号6で表される塩基配列からなるポリヌクレオチド。
- 配列番号7で表される塩基配列からなるポリヌクレオチド。
- 配列番号8で表される塩基配列からなるポリヌクレオチド。
- Beta属の細胞からDNAを抽出するDNA抽出工程と、
AFLP法に基づいて、
抽出したDNAを制限酵素で切断するDNA切断工程と、
前記DNA切断工程で得られるゲノムDNA断片を鋳型に、配列番号13で表される塩基配列からなるテンサイ黒根病抵抗性品種選抜プライマー及び配列番号14で表される塩基配列からなるテンサイ黒根病抵抗性品種選抜プライマーをペアとして核酸増幅を行う核酸増幅工程と、
前記核酸増幅工程で増幅された核酸中から請求項1に記載の塩基配列を含むポリヌクレオチドを検出する検出工程と、
によってテンサイ黒根病抵抗性品種の選抜を行うテンサイ黒根病抵抗性品種選抜方法。 - Beta属の細胞からDNAを抽出するDNA抽出工程と、
AFLP法に基づいて、
抽出したDNAを制限酵素で切断するDNA切断工程と、
前記DNA切断工程で得られるゲノムDNA断片を鋳型に、配列番号15で表される塩基配列からなるテンサイ黒根病抵抗性品種選抜プライマー及び配列番号16で表される塩基配列からなるテンサイ黒根病抵抗性品種選抜プライマーをペアとして核酸増幅を行う核酸増幅工程と、
前記核酸増幅工程で増幅された核酸中から請求項2に記載の塩基配列を含むポリヌクレオチドを検出する検出工程と、
によってテンサイ黒根病抵抗性品種の選抜を行うテンサイ黒根病抵抗性品種選抜方法。 - Beta属の細胞からDNAを抽出するDNA抽出工程と、
AFLP法に基づいて、
抽出したDNAを制限酵素で切断するDNA切断工程と、

前記核酸増幅工程で増幅された核酸中から請求項3に記載の塩基配列を含むポリヌクレオチドを検出する検出工程と、
によってテンサイ黒根病抵抗性品種の選抜を行うテンサイ黒根病抵抗性品種選抜方法。 - 前記Beta属は、テンサイである請求項4から6のいずれか一に記載のテンサイ黒根病抵抗性品種選抜方法。
