Description
試薬は以下のものを用いた。
ビタミンB1:チアミンラウリル硫酸塩[ビタゲン(登録商標)、田辺製薬株式会社製]
マルチトール:結晶マルチトール[レシス(登録商標)、東和化成工業株式会社製]
ポリペプトン:新日本製薬株式会社製、ポリペプトンS
酵母エキス:新日本製薬株式会社製、粉末酵母エキスD−3
グルコース:試薬特級
培養試験には以下の菌を用いた。
枯草菌(Bacillus subtilis IFO 3009)
セレウス菌(Bacillus cereus IFO 3001)
黄色ブドウ球菌(Staphylococcus aureus IFO 12712)
(1)基礎培地として、ポリペプトン0.5%、 酵母エキス0.25%、グルコース0.1%を水に溶解して用いた。
(2)前培養培地として、基礎培地を水酸化ナトリウム水溶液でpH7.0に調整したもの5mlを試験管(Φ18×130mm)に入れ、オートクレーブで121℃、15分滅菌したものを培養に用いた。
(3)本培養培地として、基礎培地にビタミンB1及び/又はマルチトールを添加して塩酸水溶液でpH6.0に調整し10mlを試験管(Φ18×180mm)に入れ、前培養培地と同様に滅菌したものを培養に用いた。但し、ビタミンB1は加熱溶解した後、ポアサイズ0.2μmのメンブランフィルターでろ過した後に基礎培地に添加し、混合した。
各菌を保存してある市販の寒天培地(普通寒天培地、日水製薬株式会社製)から1白金耳の菌をそれぞれ前培養培地に植菌し、30℃で24時間振とう培養して前培養液とした。
本培養培地に上記供試菌株の前培養液100μlを摂取し、30℃で48時間振とう培養を行い、経時的に濁度を測定して濁度が低いほど菌の増殖抑制効果が高いと判断し、菌の増殖抑制の程度を評価した。濁度の測定は、本培養に用いた試験管をそのまま菌濃度測定用光度計(BACT−550、株式会社ジコー製)の所定の位置に装着し、660nmの波長を測定することにより実施した。
配合表1の組成に調製した本培養培地を48時間培養して増殖抑制試験を行った。

本発明の日持ち向上剤を食品に用いた場合の効果を確認する為、配合表の食品(ポテトサラダ)を調整し、セレウス菌を植菌して保存試験を実施した。
配合表4に示すように、下茹でしてつぶしたジャガイモに、マヨネーズ、食塩及びコショウを加え、均一になるように混合し、ポテトサラダを調製した(試料A)。また、試料Aと同様にポテトサラダを調製する過程で、グリシン、酢酸Na及びDL−リンゴ酸を添加したもの(試料B)、グリシン、酢酸Na、DL−リンゴ酸、ビタミンB1及びマルチトールを添加したもの(試料C)をそれぞれ調製した。
実施例2と同様にして、セレウス菌を前培養し、菌液を滅菌水で希釈した菌液2mlをポテトサラダ100gに、初発菌が10〜102(cfu/g)となるように植菌した。植菌後、よく混合してから小分けして温度30℃で保存し、各試料の一般生菌数の経時変化を測定した。
試料中の一般生菌数の測定は以下の方法で行った。まず、所定時間、所定温度で保存した試料10gと希釈液90gを、ストマッカー用の滅菌袋に入れてストマッカーSH−IIM(株式会社エルメックス製)で30秒程度、完全に乳化するまでホモジナイズし、乳化状物を得た。次に、予め希釈水4.5mlが分注された滅菌済み試験管に、得られた乳化状物0.5mlを加えて十分に攪拌し10倍希釈液を得た。以下、測定する試料中の菌の増殖具合により、さらに希釈液4.5mlに対し先の希釈液0.5mlを添加する同様の方法により10倍希釈を繰り返し、一般生菌数測定用の試料溶液とした。次いで、所定濃度に希釈した試料溶液1mlを、20mlの寒天培地を加えたプレート上表面に均一になるように加え、培地表面が乾燥した後、培地を上に向けて、30℃の恒温槽で培養を行い、24〜48時間後にコロニー数をカウントし、希釈倍率に基づき、試料1g中の一般生菌数を求めた。尚、試料の希釈に使用する希釈水は、塩化ナトリウム0.85%、ペプトン0.1%、蒸留水99.05%の組成を有し、塩化ナトリウムは試薬特級(和光純薬工業株式会社製)、ペプトンはポリペプトンS(新日本製薬株式会社製)、水は滅菌した蒸留水をそれぞれ使用した。また、一般生菌数の測定に用いる寒天培地には、標準寒天培地(栄研器材株式会社製)23.5gを1000mlの滅菌蒸留水に溶かしたものを使用した。
ポテトサラダの一般生菌数の経時変化測定結果を結果表17に示す。
配合表5に示すように下茹でしてつぶしたジャガイモに、マヨネーズ、食塩及びコショウを加え均一になるように混合してポテトサラダを調製し、これを官能検査の基準品(試料D)とした。
Claims (3)
- ビタミンB1及びマルチトールを有効成分として含有する日持ち向上剤。
- 有機酸、有機酸塩、アミノ酸、卵白リゾチームから成る群より選ばれる1種又は2種以上を有効成分として更に含有する、請求項1に記載の日持ち向上剤。
- 請求項1又は2に記載の日持ち向上剤を食品中に0.2〜2重量%含有させることを特徴とする食品の保存方法。
