Description
本出願は、米国特許仮出願第60/424,045号(2002年11月5日出願)に対する優先権を主張する。
感染、癌および他の障害の処置において有用な多くの異なる化合物の開発にも拘わらず、より低用量で有効であり得、より選択的かつ有効な、より少ない副作用を有するか、または既知の化合物に対する耐性が発生した疾患もしくは障害を処置し得る、新たな化合物を開発する必要が未だにある。
ラクトン構造を有する式Ia、IbおよびIcの天然化合物および合成化合物、ならびにその化合物を使用する方法および作製する方法、ならびにその化合物の投与のための組成物を開発した。その化合物は、抗菌剤、抗真菌剤、および抗炎症剤として有用であり、増殖性障害(例えば、黒色腫、白血病、乳癌、肺癌、卵巣癌、結腸癌、食道癌、肝臓癌、およびリンパ性癌)を処置するために有用である。その化合物はまた、炎症性疾患(例えば、アテローム硬化症、肺線維症、全身性エリテマトーデス、膵炎、類肉腫、糸球体炎、および器官移植拒絶の処置または予防のために有効である。さらに、それらは、細菌感染および真菌感染の処置または予防(消化性潰瘍、胃炎、消化不良および胃癌の処置を含む)、歯肉炎および歯周炎の処置または予防に有効である。

そのラクトンおよび機能的誘導体は、経腸投与、非経口投与、または局所的投与(例えば、Encyclopedia of Controlled Drug Delivery,Edith Mathiowitz,Ed.,John Wiley&Sons,Inc.,New York,1999を参照のこと)のための標準的な技術を使用して処方され得る。有効な投薬量は、当業者に公知のインビトロアッセイ(例えば、例において記載されるアッセイ)に基づいて決定され得る。
式Ia、Ib、およびIcのラクトンの合成は、以下の工程を包含する:a)そのラクトン構造を有する中間体もしくは前駆物質を形成する工程、およびb)その中間体と1以上の適切な化学薬剤とを、式Ia、Ib、およびIcのラクトンへと反応させる工程。
(A.処置される障害)
そのラクトンは、広い範囲の障害を予防または処置するために有用な、抗感染剤、抗増殖剤、および抗炎症剤として有用である。特に、そのラクトンは、例えば、癌、歯肉炎、歯周炎、Helicobacter pyloriにより引き起こされる疾患、細菌により引き起こされる疾患、真菌により引き起こされる疾患、および炎症性疾患を含む障害を処置するために有用である。
有効量は、処置されるべき疾患または障害、投与様式およびその処方物に基づいて決定される。有効投薬量は、実施例中に記載されるもののようなインビトロアッセイを使用して決定された有効投薬量に基づいて決定され得る。
を、口腔経路、直腸経路、非経口経路によって、または皮膚および粘膜上に局所的塗布としての局所経路によって投与する工程を包含するが、その好ましい経路は、口腔経路である。有用な一日用量は、その投与方法、処置される状態、および特定の化合物に依存して、1〜5mg/kgである。
そのラクトン組成物は、任意の適切な投与様式において温血動物に投与され得る。その投与様式は、局所投与または全身投与であり得る。その投与様式は、処置もしくは予防されるべき障害により変動し得る。

(結果および考察)
ラットを、手術で感染させて、感染を生じさせ、次いで、連続してSecurolideで処置した。とりわけ、Escherichia coli、Klebsiela pneumoniae、Pseudomonas aeroginosa、Staphylococo aureusに対するこれらのラクトンの高い活性、およびその低分子量は、有利である。その試験は、Securolideが、Pseudomonas(これは、排除するのが困難な微生物のうちの1つである)に対して高い活性を有することを明らかに示す。
材料および方法:
雌性マウス(体重18〜22g)を使用し、106CFU/マウス(CFU:コロニー形成単位)の割合の量のCandida Albicans 44858を尾静脈に投与した。そのマウスを5匹のマウスの5つのバッチに分け、それらを、以下の様式で処置した:
感染後1時間
群1:そのマウスを、25mg/kgで経口により生成物で処置した
群2:そのマウスを、25mg/kgの用量で、腹腔内により生成物で処置した
群3:そのマウスを、25mg/kgのケトコナゾールで経口により処置した
群4:そのマウスを、25mg/kg用量のケトコナゾールで腹腔内により処置した
群5:そのマウスに、いかなる抗真菌処置も与えなかった。
生成物の活性は、2つの投与法で使用された用量において優れていた。その同じ処置はまた、皮膚真菌(例えば、白癬菌)の致死量未満モデルでの「局所的処置」において有効であった。
Candida albicans細胞を、J.Antimicrobial Chemotherapy,38,579−587に示されるように調製し、0.1M リン酸溶液で3回洗浄し、その直後に使用して、最小阻害濃度(MIC)を決定した。そのMICを、Comite Nationalの実験室臨床標準の標準的方法に従って、マイクロタイタープレートの改変によって決定した。
48時間でMICを読み取った後、そのプレートを振盪し、10μlのウェルアリコートをウェルから取り出し、それを、デキストロース糖を含む矩形のディスク上に置いた。そのプレートを、35℃で48時間インキュベートし、その最小殺真菌濃度および、コロニー形成単位が全く見られない抗真菌剤の濃度を決定した。
薬物の抗増殖効果および細胞傷害性効果の定量を、テトラゾリウム塩(MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド))を用いて行い得る。この比色アッセイにおいて、その黄色のテトラゾリウム塩であるMTTは、生きた細胞のみにある、ミトコンドリア酵素である、スクシネート−デヒドロゲナーゼにより切断されて紫色のホルマザンになる。ホルマザンは、細胞膜に対して非浸透性であるので、生存細胞内に蓄積する。生成されたホルマザンの量は、薬物処置後の生細胞の量に比例する。このアッセイを使用して、多くの癌細胞株に対する本発明者らの化合物の細胞傷害性効果または抗新生物効果を検出する。

細胞増殖阻害の評価を、Skehanら,J.Nat.Cancer Inst.82:1107(1990)の方法に従って決定した。細胞を、400〜1200細胞/ウェルの間で96ウェルプレートにプレートし、薬物を添加して、細胞の結合を可能にする前に、37℃で15〜18時間インキュベートした。試験した化合物を、100% DMSOに溶解し、10mM HEPESを含有するRPMI−1640中でさらに希釈した。各細胞株を、化合物の10通りの濃度で処理した(5log範囲)。72時間のインキュベーション後、100mlの氷冷50% TCAを各ウェルに添加し、4℃で1時間インキュベートした。次いで、プレートを、水道水で5回洗浄して、TCA、低分子量代謝産物および血清タンパク質を除去した。スルホロダミンB(SRB)(0.4%、50mL)を各ウェルに添加した。室温で5分間インキュベーションした後、プレートを、0.1% 酢酸で5回すすぎ、風乾した。結合した色素を、旋回振盪機上で、10mM Tris塩基(pH10.5)で5分間溶解させた。光学密度を、570nmで測定した。

一般に、メトトレキサート、ドキソルビシン、およびパクリタキセル(LC50=0.1μM)は、0.001〜0.5μM未満の濃度で全ての癌の型の増殖活性を阻害した。唯一の例外は、メトトレキサートであり、これは、CaSKiに対する活性を欠いていた。

6mgの評価されるべき化合物を、0.6mlの100% ジメチルスルホキシド(DMSO)中に溶解し、次いで、これを滅菌ブルセラブロスで6mlにし、その溶解度を記録した。DMSOの最終濃度は、総容積の10%であった。連続2倍希釈液(3mL Securolide+3mL ブルセラブロス)を、次いで、滅菌ブルセラブロスで作製した。この系列内の各ブロス希釈液の2mLアリコートを、別個の滅菌ペトリ皿に入れ、そのペトリ皿に、18mlの7% ウマ血液を補充した溶解して冷却した(約50℃)ブルセラ寒天を添加した。これは、寒天中、最終的に、Securolideの1:10希釈液、および1%というDMSOの最終濃度を生じた。例えば、寒天中の薬物の最高濃度が100μg/mLである場合。寒天プレートを、接種する一日前に調製し、冷蔵庫に一晩入れておいた。
Helicobacter pylori培養物を、トリプティケースソイ−5% ヒツジ血液寒天プレート上で維持し、48時間毎に移した。Helicobacter mustelae培養物を、同じ寒天上で維持し、前に移した増殖物の程度に依存して、48〜60時間毎に移した。プレートを、パラジウム触媒を有する水活性化(10ml)CampyPak Plus(BBL Microbiol.Systems)袋とともに、GasPakジャーの中で、37℃でインキュベートした。
インキュベーション後、全ての試験プレートを、Securolide非含有増殖コントロールプレートに対して比較した。そのMICは、コントロールプレートと比較して、増殖を阻害する濃度である。より高濃度で見ることができる増殖の薄いフィルムは、考慮に入れず、真のMICと考えなかった。コントロール生物をまた、各プレートに接種し、接種物として使用するために1000倍希釈した。そのコントロール生物は、Campylobacter jejuni、およびE.coli[ATCC 35218、Lote 202602,Exp 05/2000(19−258)]、Pseudomonas aeroginosa[ATCC 27853,Lote 202992,Exp 08/2000(19−060)]、E.Cloacae、Providencia stuartiiおよびP.rettgeriのスクリーニング培養物を含む。プレートおよび/または接種物の移動は、2時間より長く微小嫌気環境の外にあるべきではない。Helicobacter培養物を含む全ての操作を、その培養物が糸状菌で汚染される機会を減少するために層流フード中で行った。
Securolide含有うがい薬を使用した歯肉炎に対する経口用組成物の評価(10週間にわたる30匹のビーグル犬の研究において行った)によって、歯肉炎に対する優れた有効性が明らかに示される。使用される手順は、硬いまたは軟らかい歯の沈着物の完全な除去を包含する。その後、そのイヌを、軟らかい食餌を与えて6週間飼育して、歯肉炎を発生させた。次いで、歯列を、試験液で1週間に5日、1日2回、口の片方に対して約15秒間処置した。その動物を試験し、歯肉の炎症の程度を、0〜3の尺度に従ってスコア付けした:
0=炎症なし、
1=軽度、局所的に水腫、および歯肉縁の赤み、穏やかに指で圧を加えても、出血はもたらされない、
2=中程度の浮腫、歯肉縁の赤み、および穏やかに指で圧を加えると、出血あり、
3=重度の浮腫ならびに歯肉縁および付着歯肉の潰瘍化、ならびに、穏やかに指で圧を加えなくても、出血あり。プラシーボのうがい薬および0.5% メトロニダゾールうがい薬を、それぞれ、陰性コントロールおよび陽性コントロールとして使用した。
プラシーボと比較すると、Securolideうがい薬は、歯肉炎の発生を有意に減少させた。その得られた生成物は、歯肉炎を調節し、歯周炎を処置するにあたって有効である。さらに、この生成物は、1日あたり1〜3回の口腔への適用という規則正しいレジメンで使用される場合、口腔衛生を改善する単純な手段を提供する。
この研究を、毒物学分析法についてのガイドラインを定めるUSC 79/831の基準に従って行った。
(方法)
19名の健常ボランティアを、研究に参加させた。その参加者の年齢は、21〜40歳の間であり、平均30.5±7.2歳であった。その体重は、150〜190ポンドの間であり、中央値は、163.2±14.3ポンドであった。その伸長は、1.8〜2.1メートルであり、平均1.9±0.1メートルであった。その被験体を、ヒトでの調査について確立されたその国内基準および国際基準に従って、無作為に選択した。
