なお、本明細書では、上記発明のほか、下記式(1)
で表される基を示す。R 2 〜R 5 は、同一又は異なって、水素原子又は炭素数1〜6の直鎖状アルキル基を示す)
で表されるビアダマンタン誘導体についても説明する。前記式(1′)で表されるビアダマンタン誘導体は、前記式(1)で表されるビアダマンタン誘導体に包含される化合物である。
(式中、R2〜R5は、同一又は異なって、水素原子又は炭素数1〜6の直鎖状アルキル基を示す。X、X′はそれぞれ独立してハロゲン原子を示す。式(7)、(8)中、ベンゼン環におけるOH基の隣接位は単数又は複数のハロゲン原子及び/又はアミノ基で置換されていてもよい)
で表されるルートにより合成される。
前記反応工程式の第一段目及び第三段目におけるハロゲン化反応は、慣用の方法によって行われる。X(又はX′)が臭素原子の場合、臭素化剤(臭素源)としては、例えば臭素、t−ブチルブロミドなどの臭化アルキルなどが用いられる。また、他のハロゲン原子(フッ素、塩素、ヨウ素)の場合も慣用のハロゲン化剤にてハロゲン化を行うことができる。ハロゲン化剤の使用量は、通常、原料化合物に対して当量以上である。
前記反応工程式の第二段目におけるカルボキシル化反応において、アダマンタン骨格の橋頭位にカルボカチオンを形成可能な酸としては、例えば、硫酸、硝酸、塩酸、固体酸、ヘテロポリ酸、発煙硫酸などの強酸を使用できる。これらの酸は単独で又は2種以上を組み合わせて使用できる。酸の使用量は、式(4)の化合物に対して当量以上が好ましく、大過剰量用いてもよい。
前記反応工程式の第四段目におけるフェノール化反応において、X′におけるハロゲン原子としては、塩素原子、臭素原子、ヨウ素原子などが挙げられる。式(6)で表されるハロゲン化ビアダマンタンカルボン酸誘導体と式(7)で表されるフェノール類との反応は、反応に不活性な溶媒の存在下又は溶媒非存在下で行われる。前記溶媒として、例えば、ヘキサン、シクロヘキサン、トルエンなどの炭化水素;塩化メチレン、1,2−ジクロロエタン、クロロホルム、四塩化炭素、クロロベンゼンなどのハロゲン化炭化水素;ジエチルエーテル、ジメトキシエタン、テトラヒドロフラン、ジオキサンなどの鎖状又は環状エーテル;アセトニトリル、ベンゾニトリルなどのニトリル;酢酸エチルなどのエステル;酢酸などのカルボン酸;N,N−ジメチルホルムアミドなどのアミド;アセトン、メチルエチルケトンなどのケトン;ニトロメタン、ニトロベンゼンなどのニトロ化合物;これらの混合物などが挙げられる。
窒素雰囲気下、3,3′,5,5′−テトラメチル−1,1′−ビアダマンタン20g(61.2mmol)と四塩化炭素360g、臭素244g(1.53mol)の混合溶液を30℃で70時間撹拌した。反応溶液を300gの氷水に加え、亜硫酸ナトリウムで未反応臭素を還元した。還元後、溶液にクロロホルム120gを加えて分液し、有機層を水で洗浄した。有機層を40℃で濃縮した。濃縮後、シリカゲルカラムクロマトグラフィーにより単離したところ、3−ブロモ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンが29%の収率で得られた。
窒素雰囲気下、3−ブロモ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタン20g(49mmol)と濃硫酸302gの混合溶液を室温で2時間撹拌して溶解させた。これに、ギ酸22.7g(490mmol)を反応混合液が急激に泡立たないように3時間かけてゆっくりと滴下した。滴下終了後、2時間撹拌熟成した。次いで反応混合液を氷水1000gにゆっくりと注いで反応中間体を加水分解し、析出した沈殿物を濾別し、水で洗浄した。得られた結晶をアセトンで加熱溶解し、水を加え、析出した沈殿物を濾別した。これを乾燥することにより3−カルボキシ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンが70%の収率で得られた。
得られた3−カルボキシ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンをトリメチルシリルジアゾメタンでメチルエステル化してGC/MS分析を行った。
GC/MS−spectrometry : 384 m/z [EI法]
窒素雰囲気下、3−カルボキシ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタン10g(27mmol)と臭素96gの混合溶液を60℃で5時間撹拌した。次いで反応混合液にクロロホルム20gを加え、氷水200gにゆっくりと注ぎ、亜硫酸水素ナトリウムで過剰の臭素を還元した。沈殿物を濾別し、水で洗浄した。得られた結晶をテトラヒドロフランで加熱溶解した後、濃縮し、メタノールを添加した。析出した沈殿物を濾別した。これを乾燥することにより3−カルボキシ−3′−ブロモ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンが81%の収率で得られた。
得られた3−カルボキシ−3′−ブロモ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンをトリメチルシリルジアゾメタンでメチルエステル化してGC/MS分析を行った。
GC/MS−spectrometry : 463 m/z [EI法]
窒素雰囲気下、3−カルボキシ−3′−ブロモ−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタン6.0g(13mmol)とフェノール11.3gの混合溶液を160℃で4時間撹拌した。次いで反応混合液を80℃に冷却し、水10gを加え析出した沈殿物を80℃で濾別した。得られた結晶をテトラヒドロフランで加熱溶解した後、濃縮し、5℃に冷却して一時間撹拌した。析出した沈殿物を濾別した。これを乾燥することにより3−カルボキシ−3′−(4−ヒドロキシフェニル)−5,5′,7,7′−テトラメチル−1,1′−ビアダマンタンが68%の収率で得られた。
