Description
本発明において、「不ケン化物」とは、微生物由来のものをいう。従って本発明で不ケン化物という用語は、後から添加したものを含まない微生物由来のものを指している。
すなわち、アラキドン酸生産能を有するモルティエレラ属のモルティエレラ亜属に属する微生物から得られるアラキドン酸含有油脂であって、不ケン化物含量が少なく、その中でも特に食経験のないシクロプロパン環を有するステロールを極力、含有しないアラキドン酸含有食用油脂からなる食品、特に乳幼児用調製乳の配合に適したものを提供することができる。
上記のようにして得られたアラキドン酸含有脂質は、その大部分がトリグリセリド(約70重量%以上)及びリン脂質(約30重量%以下)であり、この他にデスモステロール等の不ケン化物が含まれている。そして該不ケン化物の中には構造が解明されていない物質や食経験が知られていない物質、例えば食経験が知られていないシクロプロパン環を有するステロールが存在しており、具体的には24,25−メチレンコレスト−5−エン−3β−オールが存在している。
有機溶媒を蒸留などの方法により除去した後、さらに水蒸気蒸留で処理する。すなわち、水蒸気蒸留で微量の臭いの成分や低沸点の不ケン化物まで除去することができる。またクロマト法を用いた残存する微量の有機溶媒も併せて除去することができる。アラキドン酸を含有し不ケン化物を実質的に含有しない食用油脂組成物が得られる。なおカラムクロマト法と水蒸気蒸留、超臨界流体での分別蒸留の他に、公知の精製手段を併用することができる。
実施例1と同様に抽出し、カラムにはかけず、水蒸気蒸溜で脱臭し、これにトコフェロール0.04%を添加し、精製油脂を得た。
実施例1と比較例1で得られた精製油脂について、不ケン化物含量を下記の方法に従って測定した。結果を表1に示す。本発明において、不ケン化物含量とは、日本油化学協会の基準油脂分析試験法の不ケン化物の定量に記載された規定の方法に基づき、油脂をケン化したのち、定量に使用する溶剤にて抽出される物質より混入脂肪酸量を削除し試料に対する百分率として表したものをいう。但し、精製後に添加した例えばトコフェロールのような不ケン化物量は差し引くものとする。上記規定の方法の概略は以下の通りである(油化学、13、489(1996)参照)。

実施例1と比較例1で得られた精製油脂について、下記の方法に従って脂肪酸メチルを調製し、アラキドン酸含量をガスクロマトグラフィーで測定した。結果を表1に示す。

注入口温度 240℃
実施例1と比較例1で得られた精製油脂について、下記の方法に従って24,25−メチレンコレスト−5−エン−3β−オール含量を測定した。結果を表1に示す。
まず、ステロール組成分析法を説明する。原料油脂を30〜80mg、栓付き試験管内に秤量し、メタノール4ml及び33%水酸化カリウム水溶液1mlを添加し栓をする。これで80℃で緩く攪拌しながら1時間反応させた後、放冷し、脂溶成分をヘキサンで抽出する。得られたヘキサン溶液をフェノールフタレイン指示薬が水層に着色しなくなるまで水洗し、減圧濃縮により分析サンプルを得る。分析サンプルを少量のヘキサンに溶解し、以下に示す条件のガスクロマトグラフィーに供する。市販のコレステロールを内部標準に用い、FID検出面積/検出重量比が全てのステロールで同じとの前提に基づいて、原料油脂に対する重量比を求めた値を24,25−メチレンコレスト−5−エン−3β−オール含量とする。

注入口及び検出器温度 300℃
キャリアガス及びゲージ圧力 ヘリウム 1.2kg/cm2

スプリット比:20
カラム処理を行うことによって、アラキドン酸含量に影響を及ぼすことなく、不ケン化物含量及び24,25−メチレンコレスト−5−エン−3β−オール含量の少ない精製品を得ることができた。
Claims (18)
- 不ケン化物含量が0.8重量%以下で、かつ、アラキドン酸含量が20重量%以上である微生物由来のアラキドン酸含有油脂からなる食品配合剤。
- 不ケン化物含量が0.6重量%以下である請求項1の食品配合剤。
- さらに24,25−メチレンコレスト−5−エン−3β−オールが0.3重量%以下である請求項1又は2の食品配合剤。
- 24,25−メチレンコレスト−5−エン−3β−オール含量が0.15重量%以下である請求項3の食品配合剤。
- 前記微生物がアラキドン酸生産能を有するモルティエレラ属のモルティエレラ亜属に属する微生物である請求項1ないし4のいずれかの食品配合剤。
- 前記モルティエレラ亜属に属する微生物が、モルティエレラ属アルピナ種に属する微生物である請求項5の食品配合剤。
- 未熟児用調製乳、幼児用調製乳、幼児用食品、又は妊産婦用食品の配合剤である請求項1ないし6のいずれかの食品配合剤。
- アラキドン酸生産能を有する微生物の培養物から得られるアラキドン酸含有油脂原料を、カラムクロマト法及び/又は蒸留により、不ケン化物含量が0.8重量%以下で、かつ、アラキドン酸含量が20重量%以上である精製アラキドン酸含有油脂に精製することを特徴とする微生物由来のアラキドン酸含有食用油脂の製造方法。
- 前記精製アラキドン酸含有油脂の不ケン化物含量が0.6重量%以下である請求項8の微生物由来のアラキドン酸含有食用油脂の製造方法。
- さらに前記精製アラキドン酸含有油脂の24,25−メチレンコレスト−5−エン−3β−オール含量が0.3重量%以下である請求項8又は9の微生物由来のアラキドン酸含有食用油脂の製造方法。
- 24,25−メチレンコレスト−5−エン−3β−オール含量が0.15重量%以下である請求項10の微生物由来のアラキドン酸含有食用油脂の製造方法。
- 前記微生物が、アラキドン酸生産能を有するモルティエレラ属のモルティエレラ亜属に属する微生物である請求項8ないし11のいずれかの微生物由来のアラキドン酸含有食用油脂の製造方法。
- 前記モルティエレラ亜属に属する微生物が、モルティエレラ属アルピナ種に属する微生物である請求項12の微生物由来のアラキドン酸含有食用油脂の製造方法。
- 請求項8ないし13のいずれかの方法で製造したアラキドン酸含有食用油脂を配合することを特徴とする、食品の製造方法。
- 請求項8ないし13のいずれかの方法で製造したアラキドン酸含有食用油脂を配合することを特徴とする、未熟児用調製乳、幼児用調製乳、幼児用食品、又は妊産婦用食品の製造方法。
- 請求項8ないし13のいずれかの方法により製造されるアラキドン酸含有食用油脂。
- 請求項8ないし13のいずれかの方法により製造されるアラキドン酸含有食用油脂を配合してなる食品。
- 請求項8ないし13のいずれかの方法により製造されるアラキドン酸含有食用油脂を配合してなる、未熟児用調製乳、幼児用調製乳、幼児用食品、又は妊産婦用食品。
