結合している前記核酸を、前記第1pHより高い第2pHで放出させるステップと、

前記固体基板上に固定化される官能基が、下記化学式(3)または下記化学式(4)で表されることを特徴とする核酸の分離方法を提供する。:
本実施例では、下記化学式(7)の官能基が固定化された固体基板を製造し、第1pHでDNAを結合させた後、第2pHで、前記DNA−基板複合体から核酸を回収した。
実施例2:化学式(7)の基が固定化された基板を利用した核酸の回収
本実施例では実施例1と同じ方法で製作された化学式(7)の各官能基が固定化された1枚の基板を用いた。本実施例では、pH3でDNAを結合させ、洗浄した後、pH3で蛍光強度を測定した。その後、基板を再度洗浄し、pH7.9で蛍光強度を測定した。
Claims (14)
- 核酸を含む試料と、アミノ基およびカルボキシル基を含み、第1pHで正電荷を帯びる二官能性物質とを、前記第1pHで接触させて、前記核酸を前記二官能性物質に結合させるステップと、
結合している前記核酸を、前記第1pHより高い第2pHで放出させるステップと、
を含み、
前記二官能性物質は、下記化学式(2)で表されることを特徴とする、核酸の分離方法:
- 前記核酸を放出させるステップは、核酸−二官能性物質の複合体を混合物から分離して、前記第2pHである溶液中で前記核酸を溶出するステップを含む請求項1または請求項2に記載の方法。
- 前記第1pHは2〜3.5であり、前記第2pHは5〜10である請求項1〜3のいずれか1項に記載の方法。
- 前記化学式(2)中の、−OHと−NH(CH2)mY’との割合が、モル比で1:1〜3:1である請求項1〜4のいずれか1項に記載の方法。
- 核酸を含む試料と、アミノ基およびカルボキシル基を含み、第1pHで正電荷を帯びる官能基が固定化されている固体基板とを、前記第1pHで接触させ、前記核酸を前記固体基板に結合させるステップと、
結合している前記核酸を、前記第1pHより高い第2pHで放出させるステップと、を含み、
前記固体基板上に固定化される官能基が、下記化学式(4)で表されることを特徴とする核酸の分離方法:
- 前記固体基板を構成する材料は、シリコン、ガラスおよびプラスチック物質からなる群から選択される請求項6または7に記載の方法。
- 前記核酸を放出させるステップは、核酸−官能基が固定化された固体基板の複合体を混合物から分離して、前記第2pHである溶液中で前記核酸を溶出するステップを含む請求項6〜8のいずれか1項に記載の方法。
- 前記第1pHは2〜3.5であり、前記第2pHは5〜10である請求項6〜9のいずれか1項に記載の方法。
- 前記化学式(4)中、−OHと−NH(CH2)mY’との割合が、モル比で1:1〜3:1である請求項6〜10のいずれか1項に記載の方法。
- 前記固体基板を構成する材料は、シリコン、ガラスおよびプラスチック物質からなる群から選択されることを特徴とする請求項12に記載の固体基板。
- 前記化学式(4)中の、−OHと−NH(CH2)pY’との割合が、モル比で1:1〜3:1の割合である請求項12または請求項13に記載の固体基板。
