【発明の属する技術分野】
本発明は、光などに起因するシワを防止又は改善するのに有用な、皮膚外用剤に関する。

【従来の技術】
紫外線などに代表される光が皮膚に与える影響は非常に大きい。短波長紫外線を一時に皮膚に多量に照射すれば紅斑や浮腫を伴う重度の炎症を引き起こすことは既に知られていることである。又、炎症を起こすほど強くない量の短波長紫外線照射であっても、この様な照射を繰り返すことにより色素沈着などを起こすことも既に知られている。加えて近年では、短波長紫外線のみならず、長波長紫外線も炎症などの急性の症状は引き起こさないものの、繰り返しの照射により、DNAの開裂やコラーゲン線維の寸断等を引き起こし、シワ形成の原因になっていることが報告されている。

シワは人間の老化に伴って増えるものであり、このシワ形成が容姿に与える影響は大きく、シワの増加によって、自分の容貌の衰えを意識するのが世の常である。この容貌の衰えは男女にかかわらず、だれしも避けたいと願うものであり、シワ形成を抑制する手段が種々求められていたが、この様な手段として成功したのは僅かにレチノイドにより角化を亢進させる手段と珪酸ソーダ等を配合した化粧料を投与し、乾燥させて被膜を形成させる手段ぐらいしかない。これは、シワの原因は二、三知られているものの、シワ形成のメカニズムについてはまだ良く解明されていないのが現状であった為である。この様な状況を踏まえて本発明者等はシワの原因を求めて鋭意研究を重ねた結果、シワ形成の原因は、真皮コラーゲン線維束の乱れであることを見いだした。更にこの様な真皮コラーゲン線維束の乱れは、ウルソール酸、ウルソール酸の生理的に許容される塩、ウルソール酸の誘導体、バラ科ビワのエッセンス、バラ科モモのエッセンス、クロウメモドキ科ナツメのエッセンス、スイカズラ科セイヨウニワトコのエッセンス、キク科ヤグルマギクのエッセンス、シソ科タイムのエッセンス、シソ科ローズマリーのエッセンス、シソ科セージのエッセンス、シソ科シソのエッセンス、シソ科オドリコソウのエッセンス及びシソ科セイヨウハッカのエッセンスの投与によって再構築し、皮膚状態を改善しうることを見いだしている。しかしながら、再構築までに要する時間は長いため、更に再構築までの時間を減少させる技術が求められていた。

一方、上記の真皮コラーゲン線維束改善作用を有する成分と炎症抑制作用を有する成分とを組み合わせる技術思想は存在していないし、この組み合わせによって、より短時間真皮コラーゲン線維束の乱れを改善し、皮膚の状態を改善しうることも知られていない。又、更に、ウルソール酸、ウルソール酸の生理的に許容される塩、ウルソール酸の誘導体、バラ科ビワのエッセンス、バラ科モモのエッセンス、クロウメモドキ科ナツメのエッセンス、スイカズラ科セイヨウニワトコのエッセンス、キク科ヤグルマギクのエッセンス、シソ科タイムのエッセンス、シソ科ローズマリーのエッセンス、シソ科セージのエッセンス、シソ科シソのエッセンス、シソ科オドリコソウのエッセンス及びシソ科セイヨウハッカのエッセンスから選ばれる1種乃至は2種以上とバラ科ワレモコウのエッセンス、キク科カワラヨモギのエッセンス、キク科ゴボウのエッセンス、ヘンルーダ科イヌザンショのエッセンス、ボタン科ボタンのエッセンス、ユキノシタ科コジソウのエッセンス、ユリ科ハカタユリのエッセンス、ボタン科シャクヤクのエッセンス、キク科アルニカのエッセンス、キク科ローマカミツレのエッセンスから選ばれる1種乃至は2種以上とを組み合わせることも知られていない。

【発明が解決しようとする課題】
本発明はこの様な状況を踏まえてなされたものであり、真皮コラーゲン線維束を速やかに再構築させ、皮膚状態を改善する皮膚外用剤を提供することを課題とする。

【課題の解決手段】
本発明者等は、かかる実状に鑑みて、真皮コラーゲン線維束を速やかに再構築させ、皮膚状態を改善する皮膚外用剤を求めて鋭意研究を重ねた結果、ウルソール酸、ウルソール酸の生理的に許容される塩、ウルソール酸の誘導体、バラ科ビワのエッセンス、バラ科モモのエッセンス、クロウメモドキ科ナツメのエッセンス、スイカズラ科セイヨウニワトコのエッセンス、キク科ヤグルマギクのエッセンス、シソ科タイムのエッセンス、シソ科ローズマリーのエッセンス、シソ科セージのエッセンス、シソ科シソのエッセンス、シソ科オドリコソウのエッセンス及びシソ科セイヨウハッカのエッセンス等の真皮コラーゲン線維束改善作用を有する物質とバラ科ワレモコウのエッセンス、キク科カワラヨモギのエッセンス、キク科ゴボウのエッセンス、ヘンルーダ科イヌザンショのエッセンス、ボタン科ボタンのエッセンス、ユキノシタ科コジソウのエッセンス、ユリ科ハカタユリのエッセンス、ボタン科シャクヤクのエッセンス、キク科アルニカのエッセンス、キク科ローマカミツレのエッセンス等の炎症抑制作用を有する物質とを含有する皮膚外用剤にその様な作用があることを見いだし、発明を完成させるに至った。以下、本発明について、実施の形態を中心に更に詳細に説明する。

【発明の実施の形態】
(1)本発明の必須成分である真皮コラーゲン線維束改善作用を有する成分
本発明で用いることの出来る真皮コラーゲン線維束改善作用を有する成分としては、真皮コラーゲン線維束改善作用を有するものであれば特段の限定無く用いることが出来、この様な真皮コラーゲン線維束改善作用は、例えば、マウスを用いた紫外線照射シワ形成モデルでスクリーニングし、評価・選択することが出来る。以下に、このモデルによる評価例を示す。本発明で言う真皮コラーゲン線維束改善作用を有する成分とは、下記のスクリーニングに於いて真皮コラーゲン線維束改善値が1.1以上のものを意味する。

<試験例1>
光老化モデルでの皮膚の状態の変化の検討
ヘアレスマウス(Skh:HR−1、雌性、8週齢)に紫外線B(東芝SEランプ、60mJ/cm)を連日照射し、照射開始後2、5、10週間に皮膚及び皮膚表面形態レプリカを採取した。採取皮膚はNaOH法によりコラーゲン線維束構造を走査電子顕微鏡により観察した。図1に倍率50倍での皮膚表面形態(A:非照射コントロール、B:照射2週間、C:照射5週間、D:照射10週間)、図2に倍率50倍での真皮表面形態(E:非照射コントロール、F:照射2週間、G:照射5週間、H:照射10週間)、図3に倍率500倍での真皮コラーゲンの線維束の構造(I:非照射コントロール、J:照射2週間、K:照射5週間、L:照射10週間)、図4に倍率2500倍での真皮コラーゲンの線維束の構造(M:非照射コントロール、N:照射2週間、O:照射5週間、P:照射10週間)を示す。これらの図より、しわが形成される際、それに対応するように真皮の表面にも溝が形成されており、皮膚表面の形態の変化は真皮の表面の形態の変化対応していること、更に真皮表面の変化は真皮に於けるコラーゲン線維束の構造の変化、即ち、線維束が明確でなくなる等の線維束の秩序の低下を反映していることが判る。ここで、図4の顕微鏡像を次の判定基準でスコアーを付した。即ち、スコアー0:観察領域全域で線維束構造が認められない、スコアー1:過半領域で線維束構造の崩壊又は異常構造への変移が認められる、スコアー2:一部に線維束構造の崩壊又は変性が認められるが、全体的にはほぼ正常な構造が認められる、スコアー3:全面に亘り正常な線維束構造が認められ、崩壊・変性はほぼ認められないの基準である。この結果を図5に示す。又、皮膚表面の構造について、レプリカへの入射角20度でのキセノンランプによる光照射を行い出来たシワの陰影を画像解析により定量し、シワの生成量とした。この測定結果を図6に示す。このシワ量とスコアー値の平均との相関係数を算出したところ、0.91であり、シワの形成と真皮コラーゲン線維束の乱れ(秩序)の間に強い関係があり、真皮コラーゲン線維束の乱れがシワ形成のメカニズムであることがわかる。

<試験例2>
上記の試験例1の動物モデル例を用い、光照射により生じたシワ等の皮膚の悪化状態の変化が上記真皮コラーゲン線維束改善剤によりどの様に変化するかを調べた。上記の光照射ヘアレスマウス(Skh:HR−1、雌性、8週齢)を用い、投与群には上記真皮コラーゲン線維束改善作用を有する成分の0.1%エタノール溶液を、対照群はエタノールのみをそれぞれ0.05mlづつ8週間連日投与した。この動物の皮膚を上記と同様に処理し、コラーゲン線維束構造を観察した。これよりコラーゲン線維束スコアー値を算出し、対照群のスコアー値で除し、真皮コラーゲン線維束の改善値を算出した。コラーゲン線維束の改善値を表1に示す。尚、植物のエッセンスは表1に示す植物の部位の50%エタノール水溶液抽出物の溶媒除去物を、ウルソール酸の誘導体としてはメチルエステルとベンジルエステルを用いた。結果を表1に示す。何れのサンプルも真皮コラーゲン線維束を改善していることが判る。

即ち、本発明で用いることの出来る真皮コラーゲン線維束構造の改善作用を有する物質として、ウルソール酸、ウルソール酸の生理的に許容される塩、ウルソール酸の誘導体、バラ科ビワのエッセンス、バラ科モモのエッセンス、クロウメモドキ科ナツメのエッセンス、スイカズラ科セイヨウニワトコのエッセンス、キク科ヤグルマギクのエッセンス、シソ科タイムのエッセンス、シソ科ローズマリーのエッセンス、シソ科セージのエッセンス、シソ科シソのエッセンス、シソ科オドリコソウのエッセンス及びシソ科セイヨウハッカのエッセンスが好適に例示できることが判る。ここで、ウルソール酸の生理的に許容される塩としては、例えば、ナトリウムやカリウム等のアルカリ金属、カルシウムやマグネシム等のアルカリ土類金属、トリエチルアミンやトリエタノールアミン等の有機アミン塩、アンモニウム塩、塩基性アミノ酸塩が好適に例示できる。又、ウルソール酸の誘導体としては、メチルエステル、ブチルエステル、ラウリルエステル、ステアリルエステル、ベンジルエステル、フェネチルエステル等の炭素数1〜20のエステル等が好適に例示できる。この中では特に炭素数1〜4のアルキルエステルとベンジルエステルが好ましい。又、本発明で言う、植物のエッセンスとは、植物体の全部又は一部そのもの、植物体の全部又は一部を粉砕、細切、乾燥した加工物、植物体の全部又は一部、或いは加工物を溶媒で抽出した抽出物、抽出物の溶媒除去物、抽出物乃至は抽出物の溶媒除去物を分画精製した分画物等の総称を意味する。この際、抽出はエタノール、メタノール、1,3−ブタンジオール等のアルコール、水、アセトン、メチルエチルケトン等のケトン類、ジエチルエーテル、テトラヒドロフラン等のエーテル類、アセトニトリル等のニトリル類、塩化メチレンやクロロホルム等のハロゲン化炭化水素類、酢酸エチルや蟻酸メチル等のエステル類、ノルマルヘキサン、石油エーテル等の炭化水素類等から選ばれる1種乃至は2種以上に、室温であれば数日、沸点付近の温度であれば数時間浸漬すればよい。これらの内好ましいエッセンスはアルコール水溶液抽出物の溶媒除去物である。これをダイアイオンHP−20に吸着させて水乃至は20%エタノール水溶液で溶出させたフラクションが特に好ましい。本発明ではこれらの真皮コラーゲン線維束改善作用を有する成分を単独で用いることもできるし、2種以上を組み合わせて用いることもできる。本発明の皮膚外用剤における真皮コラーゲン線維束改善作用を有する成分の好ましい含有量は、0.01〜10重量%であり、より好ましくは0.05〜5重量%であり、更に好ましくは0.1〜3重量%である。

(2)本発明の必須成分である炎症抑制作用を有する成分
本発明で用いることの出来る炎症抑制作用を有する成分としては、炎症抑制作用を有していれば特段の限定無く用いることが出来、炎症抑制作用としては、例えば、ラットカラゲニン浮腫モデル等の既存の炎症モデルを用いて、評価選択することが出来る。本発明者等が確認した好ましい炎症抑制作用を有する成分としては、バラ科ワレモコウのエッセンス、キク科カワラヨモギのエッセンス、キク科ゴボウのエッセンス、ヘンルーダ科イヌザンショのエッセンス、ボタン科ボタンのエッセンス、ユキノシタ科コジソウのエッセンス、ユリ科ハカタユリのエッセンス、ボタン科シャクヤクのエッセンス、キク科アルニカのエッセンス、キク科ローマカミツレのエッセンスが特に好ましい。又、エッセンスとしては、エタノール等のアルコール水溶液の抽出物の溶媒除去物が特に好ましい。これをダイアイオンHP−20に吸着させて水乃至は20%エタノール水溶液で溶出させたフラクションが特に好ましい。本発明の皮膚外用剤にはこれら炎症抑制作用を有する成分をただ一種で含有させても良いし、二種以上を組み合わせて含有させても良い。本発明の皮膚外用剤における炎症抑制作用を有する成分の好ましい含有量は、0.01〜10重量%であり、より好ましくは0.05〜5重量%であり、更に好ましくは0.1〜3重量%である。これらの炎症抑制作用を有する成分の炎症抑制作用を1%含有親水軟膏を検体として用い、ラットカラゲニン浮腫モデルで調べた結果を表2に示す。尚、エッセンスは何れも50%エタノール水溶液抽出物の溶媒除去物を用いた。何れも炎症抑制作用を有していることが判る。

上記、炎症抑制作用を有する成分について、上記試験例2の方法で真皮コラーゲン線維束改善作用を調べた結果、何れも真皮コラーゲン線維束改善作用を認めなかった。

(3)本発明の皮膚外用剤
本発明の皮膚外用剤は、上記真皮コラーゲン線維束改善作用を有する成分から選ばれる1種乃至は2種以上と上記炎症抑制作用を有する成分から選ばれる1種乃至は2種以上とを含有することを特徴とする。本発明の皮膚外用剤では、これら必須成分以外に皮膚外用剤で通常使用されている任意成分を含有させることが出来る。かかる任意成分としては、例えば、ワセリンやマイクロクリスタリンワックス等のような炭化水素類、ホホバ油やゲイロウ等のエステル類、牛脂、オリーブ油等のトリグリセライド類、セタノール、オレイルアルコール等の高級アルコール類、ステアリン酸、オレイン酸等の脂肪酸、グリセリンや1,3−ブタンジオール等の多価アルコール類、非イオン界面活性剤、アニオン界面活性剤、カチオン界面活性剤、両性界面活性剤、エタノール、カーボポール等の増粘剤、防腐剤、紫外線吸収剤、抗酸化剤、色素、粉体類等が例示できる。本発明の皮膚外用剤の適用可能な製剤は、例えば、クリーム、乳液、化粧水、パック等の基礎化粧料、アンダーメークアップ、ファンデーションなどのメークアップ化粧料、抗炎症皮膚外用医薬等の医薬何れもが例示でき、これらの内では基礎化粧料が特に好ましい。本発明の皮膚外用剤は上記の如く、シワの原因である真皮コラーゲン線維束を改善する作用に優れるため、シワを予防・改善するために用いるのが好ましく、シワの中でも光照射によるシワを予防・改善する光老化防止用に適用するのが特に好ましい。

以下に実施例を挙げて、本発明について更に詳細に説明を加えるが、本発明がこれら実施例にのみ限定を受けないことは言うまでもない。

実施例1および参考例1〜13
下記に示す処方に従ってローション剤(化粧料)を作製した。即ち、処方成分を良く攪拌し可溶化させて、ローション剤(化粧料)を得た。表3の植物エッセンスは何れも50%エタノール水溶液抽出物の溶媒除去物を用いた。併せて、真皮コラーゲン線維束改善値を示す。これより真皮コラーゲン線維束改善作用を有する成分と炎症抑制作用を有する成分の併用効果によって優れた真皮コラーゲン線維束改善作用を示していることが判る。

実施例2および3、参考例14〜20
下記に示す処方に従ってローション剤(化粧料)を作製した。即ち、処方成分を良く攪拌し可溶化させて、ローション剤(化粧料)を得た。表4の植物エッセンスは何れも50%エタノール水溶液抽出物の溶媒除去物を用いた。併せて、真皮コラーゲン線維束改善値を示す。これより真皮コラーゲン線維束改善作用を有する成分と炎症抑制作用を有する成分の併用効果によって優れた真皮コラーゲン線維束改善作用を示していることが判る。

下記に示す処方に従って、クリームを作製した。即ち、イを混練りし、80℃に加熱しておいたロを加え希釈し、80℃に加熱した。これに予め80℃に加熱しておいたハを攪拌しながら徐々に加え乳化し、攪拌冷却しクリームを得た。このものの真皮コラーゲン線維束改善値は2.4であった。

キク科ゴボウ1Kgに50%エタノール10lを加え、還流条件で3時間加熱攪拌し、不溶物を濾過で取り除き、溶媒を減圧溜去しエッセンス(50%エタノール抽出物の溶媒除去物)を得た。これを水に分散させダイアイオンHP−20にチャージし、水、20%エタノール、50%エタノール、エタノールの順でそれぞれ1lづつ流し、それぞれのフラクションを集め、減圧溜去し、それぞれゴボウエッセンス1、ゴボウエッセンス2、ゴボウエッセンス3、ゴボウエッセンス4を得た。これらを用いて表5に示す処方に従って化粧水を作成した。即ち、処方成分を加熱攪拌し可溶化し、冷却し化粧水を得た。又、上記の方法で真皮コラーゲン線維束改善値を求めた。この結果も表5に示す。この表より、ゴボウのエッセンスとしては、ダイアイオンカラムクロマトグラフで非極性部を取り除いた、水フラクションと20%エタノール水溶液フラクションが好ましいことが判る。

参考例26のクリームを用いて、使用テストにて本発明の皮膚外用剤の作用を調べた。即ち、任意に選択したシワに悩むパネラー1群10名に本発明の皮膚外用剤である参考例26のクリーム、参考例26のクリーム中のローズマリーのエッセンスを水に置換した比較例1、ボタンのエッセンスを水に置換した比較例2、ローズマリーのエッセンスとボタンのエッセンスを共に水に置換した対照例1を3ヶ月間使用して貰いシワの改善をアンケートによって答えて貰った。結果を出現例数として表6に示す。これより、本発明の皮膚外用剤はシワ改善作用に優れること及び抗炎症成分との併用により真皮コラーゲン線維束改善作用を有する物質の真皮コラーゲン線維束改善作用が高められることが判る。
