CYVaVの全長構造を、SHAPE構造プロービング、並びにウチワサボテン、イチジク、及びトウモロコシにおけるCYVaV近縁種との系統発生学的比較によって決定した(図9)。記録部位(図10を参照)及びISS様(I字型構造)3’CITE(図11を参照)を同定し、これと共に、インサートに対応するための領域は、例えば二重線で囲まれた領域によって示されており、インサートのための例示的な位置に関して更に詳細に議論する。
N. benthamianaの葉に、CYVaV、又はCVEV、又はCYVaV+CVEVをアグロ浸潤させた。CYVaVはCVEVのビリオンにカプシド化され、ビリオンは1週間後に単離されて、カプシド化されたRNAはPCR分析に供された(図5及び6を参照)。CYVaVの蓄積は、推定ヘルパーウイルスCVEVの存在下で有意に増大した。rRNAローディングコントロールは下側に示されている。p14サイレンシングサプレッサは、全ての葉に同時浸潤された。黄化症状は、CYVaV+CVEVを用いた葉においてわずかに重篤であった(図7、パネルB)。
蛍光in situハイブリダイゼーション(FISH)撮像により、CYVaVのプラス鎖が明確に検出され、これは完全にSE、CC、及びPPCに限定されていた(図8)。
N. benthamiana 16C植物に、(これらの植物においてサイレンシングされる)GFPを発現する構築物と、CYVaV p21若しくはp81を発現する構成物、又は既知のサイレンシングサプレッサp19(由来TBSV)若しくはp38(由来TCV)を発現する構築物とをアグロ浸潤させた(図13、パネルA)。p19及びp38だけがGFPのサイレンシングを抑制し、緑色蛍光を発現させる(図13、パネルB)。GFPオリゴヌクレオチドでプローブしたノーザンプロットは、GFP RNAがp21又はp81の存在下で依然としてサイレンシングされることを示した(図13、パネルC)。
CYVaVの感染性クローンが生成された。野生型RNA転写産物(CYVaV)、又はRdRpの合成を排除するため複製しない、「スリップしやすい」記憶部位に突然変異を含有する転写産物(CYVaV‐fsm)を、Arabidopsis原形質体に接種した。RNAを抽出し、ノーザンブロットを30時間後に実施した。なお、接種されたCYVaV‐fsmの転写産物は、30時間の時点において依然として原形質体中に存在していた(従来のウイルスでは4時間後には検出されなくなる)。
ノーザンブロットによって、N. benthamianaの浸潤済みの葉に蓄積されるCYVaVのレベルを決定した(図15、パネルA)。CYVaVが浸潤された植物は散発的に全身症状を示した(図15、パネルB;図16も参照)。これらの植物は、高いレベルのCYVaVを蓄積した。全身的に感染した植物の個々の葉におけるCYVaVのレベルを決定した(図15、パネルC)。葉4及び5には、CYVaVがアグロ浸潤された。最も若い葉にCYVaVが相当量蓄積されていることに注目されたい。
葉4及び5には、CYVaVがアグロ浸潤された。全身的に感染した植物の最初の兆候は、「カップ状の」葉(図16)であり、これは略常に葉9であった。次の数週間で、葉粒が植物の頂端分裂組織及び各節に出現した。全身的に感染した植物は、根粒も有しており、これは植物のノーザンブロットによって証明されるように、相当な量のCYVaVを含んでいた。
全身的に感染したN. benthamianaからの樹液をトマトの葉柄に注入した(図17)。4つの植物のうちの1つが極めて強い症状を示し、PCRによりCYVaVに関して陽性であった。図示されている植物は感染後53日目であり、同一年齢の植物と共に示されている。
標識された全長CYVaVは、ノースウェスタンブロットにおいて実証されるように、顕著なタンパク質に結合する(図18)。タンパク質は、SDSゲル電気泳動後に復元された。このタンパク質は、RRMモチーフを含有する既知の高度に保存されたRNA結合タンパク質であると考えられ、上記RRMモチーフは、RNAに対して伴細胞からキュウリの師部の師要素へのシャペロンとして機能することが知られている。電気泳動後にタンパク質が変性したままである場合には、結合は見られなかった。
タバコエッチウイルス(TEV)内部リボソーム侵入部位(IRES)の下流のナノルシフェラーゼの3つの別個のインサートの位置を同定した(図19)。上記3つの構築物の小麦胚芽抽出物における試験管内翻訳を評価した。ナノルシフェラーゼタンパク質(Nluc)の位置は、ゲルの底部付近である。ナノルシフェラーゼの発現を、原形質体で生体内で調査した(図19、パネルC)。(A)に示されている構築物の全長RNA転写物を、原形質体に形質転換し、18時間後に総タンパク質を抽出し、ナノルシフェラーゼ活性をルミノメータで測定した。

一実施形態では、CLas細菌によって引き起こされる柑橘類産業における病気(HLB)を治療するための、iRNA系ベクターが提供される。CYVaVの分離株をベクターとして利用して、上記細菌、及びこの細菌を樹木の中へと送達するキジラミを標的とする。上述のように、CYVaVは師部に限定され、この師部において、最良の植物ウイルスに匹敵する極めて高いレベルまで複製及び蓄積される。更に、サイズが比較的小さいため、遺伝子的な操作が極めて容易である。このように、CYVaVの構造及び生物学を考察することで、師部の通過のためのベクター及びモデルシステムとしてのこの新規の感染因子の開発が支援された。

Claims (43)
- 1つ以上の複製要素及び1つ以上の異種セグメントを含む、一本鎖プラス鎖リボ核酸(RNA)ベクターであって、
前記RNAベクターは、1つ以上の機能性コートタンパク質のオープンリーディングフレーム(ORF)、及び1つ以上の機能性移行タンパク質のORFを含まない、一本鎖プラス鎖リボ核酸(RNA)ベクター。 - 前記RNAベクターは、配列番号4及び/又は配列番号5の1つ以上の核酸配列を有する3’キャップ非依存的翻訳促進領域(3’ Cap Independent Translation Enhancer:3’CITE)を含む、請求項1に記載のRNAベクター。
- 前記3’CITEは、配列番号3の核酸配列を有する、請求項2に記載のRNAベクター。
- 前記1つ以上の複製要素は、配列番号10、配列番号11、配列番号12、配列番号13、及び/又は配列番号14の核酸配列を有する、1つ以上の保存ポリヌクレオチド配列を有する、請求項1~3のいずれか1項に記載のRNAベクター。
- 上記1つ以上の複製要素は更に、配列番号15及び/又は配列番号16の核酸配列を有する、1つ以上の保存ポリヌクレオチド配列を有する、請求項1~4のいずれか1項に記載のRNAベクター。
- カンキツ葉脈黄化ウイルス(配列番号1)、又はカンキツ葉脈黄化ウイルスの近縁のiRNAに由来する、請求項1~5のいずれか1項に記載のRNAベクター。
- 宿主植物内における、全身的な及び師部に限定された移行及び複製が可能である、請求項1~6のいずれか1項に記載のRNAベクター。
- 感染後少なくとも1か月にわたって、前記宿主植物内での複製、移行、及び/又は翻訳に関して機能的に安定している、請求項7に記載のRNAベクター。
- 前記1つ以上の異種セグメントは、レポータ分子、ペプチド、及びタンパク質からなる群から選択される少なくとも1つのポリペプチドをコードするポリヌクレオチドを含む、請求項1~8のいずれか1項に記載のRNAベクター。
- 前記ポリペプチドは殺虫剤、抗細菌剤、抗ウイルス剤、又は抗真菌剤である、請求項9に記載のRNAベクター。
- 前記抗細菌剤は酵素抗生物質である、請求項10に記載のRNAベクター。
- 前記抗細菌剤は、Candidatus Liberibacter種の細菌を標的とする、請求項10又は11に記載のRNAベクター。
- 前記Candidatus Liberibacter種はCandidatus Liberibacter asiaticus(CLas)である、請求項12に記載のRNAベクター。
- 前記1つ以上の異種セグメントは、ノンコーディングRNA小分子及び/又はRNA干渉分子を含む、請求項1~13のいずれか1項に記載のRNAベクター。
- 前記ノンコーディングRNA小分子及び/又は前記RNA干渉分子は、昆虫ベクター、ウイルス、又は真菌を標的とする、請求項14に記載のRNAベクター。
- 前記ノンコーディングRNA小分子及び/又は前記RNA干渉分子は、前記昆虫ベクター、前記ウイルス、又は前記真菌の核酸を標的とする、請求項15に記載のRNAベクター。
- 前記ウイルスは、カンキツベインエネーションウイルス(CVEV)及びカンキツトリステザウイルス(CTV)からなる群から選択される、請求項15又は16に記載のRNAベクター。
- 前記1つ以上の異種セグメントは、第2の1つ以上の異種セグメントを更に含む第1の異種セグメントであり、
前記1つ以上の複製要素は、前記第1の異種セグメントと前記第2の異種セグメントとの間にある、請求項1~17のいずれか1項に記載のRNAベクター。 - 前記1つ以上の異種セグメントは、表現型の特色を改変するタンパク質又はペプチドをコードするポリヌクレオチドを含む、請求項1~18のいずれか1項に記載のRNAベクター。
- 前記表現型の特色は、農薬耐性、除草剤耐性、昆虫耐性、カロース産生の減少、成長速度の上昇、及び萎縮症からなる群から選択される、請求項19に記載のRNAベクター。
- 請求項1~20のいずれか1項に記載のRNAベクターを含む、宿主植物であって、
前記宿主植物は、植物全体、植物器官、植物組織、又は植物細胞である、宿主植物。 - 前記宿主植物は、ミカン属、ブドウ属、イチジク属、及びオリーブ属からなる群から選択される属のものである、請求項21に記載の宿主植物。
- 前記宿主植物は、柑橘類の樹木又は柑橘類の樹木の移植片である、請求項22に記載の宿主植物。
- 請求項1~20のいずれか1項に記載のRNAベクターに感染した植物、植物器官、植物組織、又は植物細胞を含む、組成物。
- 前記植物は、ミカン属、ブドウ属、イチジク属、及びオリーブ属からなる群から選択される属のものである、請求項24に記載の組成物。
- 前記植物は、柑橘類の樹木又は柑橘類の樹木の移植片である、請求項25に記載の組成物。
- 1つ以上の異種セグメントを宿主植物に導入する方法であって、
前記方法は、前記宿主植物に、請求項1~20のいずれか1項に記載のRNAベクターを導入するステップを含む、方法。 - 前記導入するステップは、前記RNAベクターを含む植物の植物器官又は植物組織を、前記導入の前には前記RNAベクターを含まない別の植物の植物器官又は植物組織に移植するステップを含む、請求項27に記載の方法。
- 前記RNAベクターは、前記宿主植物に全身的に感染する、請求項27又は28に記載の方法。
- 植物、植物器官、植物組織、又は植物細胞中で、1つ以上の異種セグメントを生成するプロセスであって、
前記プロセスは、前記植物、前記植物組織、又は前記植物細胞に、請求項1~20のいずれか1項に記載のRNAベクターを導入するステップを含む、プロセス。 - 前記植物は、ミカン属、ブドウ属、イチジク属、及びオリーブ属からなる群から選択される属のものである、請求項30に記載のプロセス。
- 請求項1~20のいずれか1項に記載RNAベクターを含む、キット。
- 1つ以上の異種セグメントを、植物、植物器官、植物組織、又は植物細胞に導入するための、請求項1~20のいずれか1項に記載のRNAベクターの使用。
- RNAベクターを、導入の前には前記RNAベクターを含まない植物器官又は植物組織に導入するための、請求項21~23のいずれか1項に記載の宿主植物の使用、又は請求項24~26のいずれか1項に記載の組成物の使用。
- 前記導入は、前記RNAベクターを含む植物の植物器官又は植物組織を、前記RNAベクターを含まない別の植物の植物器官又は植物組織に移植するステップを含む、請求項34に記載の使用。
- 植物と共に使用するためのベクターを作製する方法であって、
前記方法は、1つ以上の異種セグメントをRNAに挿入するステップを含み、
前記RNAは:CYVaV;CYVaVの近縁種;CYVaVと少なくとも70%のRdRp同一性を有する他のRNAベクター;及び別のiRNAからなる群から選択される、方法。 - 請求項36に記載の方法によって生成されるベクター。

- 前記RNAは、挿入された1つ以上の異種セグメント、例えば酵素抗生物質によって改変される、請求項38又は39に記載の使用。
- 植物の病気又は状態の治療のための薬剤の製造におけるものであることを特徴とする、RNAの使用であって、
