I.ミトコンドリア外膜アンカリングペプチド、リンカー、ユビキチンおよび所望のタンパク質を含む融合タンパク質の調製
実施例1. p53を含む融合タンパク質の調製
実施例1.1. p53遺伝子の増幅
組換えタンパク質においてヒトp53を発現させるために、ヒト上皮細胞から全RNAを抽出し、そこからcDNAを合成した。具体的には、ヒト皮膚線維芽細胞を、5%二酸化炭素および37℃の条件下に10%血清培地中で培養した(1×106細胞)。その後、培養液を除去し、細胞にPBSバッファー液を加えて2回洗い、RNA抽出試薬(Trizol試薬、Thermo Fisher Scientific)0.5mlを直接加えた。RNA抽出試薬を加えた混合物を周囲温度で10分間置き、その後、0.1mlのクロロホルムを加え、15秒間攪拌した後、約12000×gで10分間遠心分離した。次いで、分離された上清を取り、同体積のイソプロピルアルコールを加えて、再び12000×gで10分間遠心分離した。その後、液体を除去し、75%エタノールで1回洗い、RNAを周囲温度で乾燥した。
実施例1.2.1. プラスミドpET15b−UB−p53の作製
ユビキチンが融合した形態のp53タンパク質を調製するために、下記の発現ベクターを作製した。ユビキチン遺伝子を得るために、NdeUBプライマーおよびT2UBプライマーを作製した。各プライマーの配列を表2に示す。
ミトコンドリア外膜に結合するTOM70およびユビキチンが融合した形態のp53タンパク質を調製するために、TOM70およびユビキチンが融合した形態のp53を発現することのできる発現ベクターを作製した。TOM70およびユビキチン遺伝子を得るために、NdeTOM70プライマー、TOM70−ASプライマー、TOM70UB−SプライマーおよびT2UB−ASプライマーを作製した。各プライマーの配列を表3に示す。
ミトコンドリア外膜に結合するTOM70、リンカー(GGGGSGGGGSGGGGS(配列番号70))およびユビキチンが融合した形態のp53タンパク質を調製するために、TOM70、リンカーおよびユビキチンが融合した形態のp53タンパク質を発現することのできる発現ベクターを作製した。TOM70に連結したリンカー遺伝子を得るために、TOM70(G)3−ASプライマー、(G)3UB−Sプライマー、Xp53(noT)プライマーを作製した。各プライマーの配列を表4に示す。
ミトコンドリア外膜に結合するTOM70、およびリンカー(GGGGSGGGGSGGGGS(配列番号70))が融合した形態のp53タンパク質を調製するために、TOM70およびリンカーが融合した形態のp53を発現することのできる発現ベクターを作製した。TOM70およびリンカーが融合したp53遺伝子を得るために、プライマー(B(G)3p53)を作製した。プライマーの配列を表5に示す。
ユビキチン、およびミトコンドリア外膜に結合するTOM70が融合した形態のp53タンパク質を調製するために、ユビキチン、p53およびTOMがこの順序で融合した形態のp53を発現することのできる発現ベクターを作製した。TOM70およびユビキチンが融合したp53遺伝子を得るために、Xp53(noT)プライマー、XTOM7プライマー、およびLTOM7プライマーを作製した。各プライマーの配列を表6に示す。
p53を発現することのできる動物細胞用発現ベクターを作製した。p53遺伝子を得るために、Rp53プライマーを作製した。プライマーの配列を表7に示す。
実施例1.3.1. 大腸菌から誘導された組換えTOM70−(GGGGS)3−p53タンパク質の単離および精製
組換えTOM70−(GGGGS)3−p53タンパク質を発現する大腸菌BL21(DE3)の産生株をLB液体培地に播種し、37℃の条件下に培養した。OD600の吸光度が0.4に達したら、0.5mMのIPTGを加え、さらに4時間の震盪培養を行って、TOM70−(GGGGS)3−p53タンパク質を発現させた。
TOM70−(GGGGS)3−UB−p53組換えタンパク質を発現する大腸菌を用いて、TOM70−(GGGGS)3−UB−p53タンパク質を、実施例1.3.1と同じ方法で単離し精製した。結果として、TOM70−(GGGGS)3−UB−p53タンパク質を溶出させた(図16)。この場合、図16のレーンMはタンパク質分子量マーカーを示し、レーン1はニッケルアフィニティークロマトグラフィーのローディングサンプルを示す。レーン2はニッケルアフィニティー樹脂に結合しなかったものを示す。レーン3は、8M尿素/50mMリン酸Na/500mM NaCl/50mMイミダゾール溶液による溶出の結果を示す。レーン4〜7は、8M尿素/50mMリン酸Na/500mM NaCl/100mMイミダゾール溶液による溶出の結果を示す。レーン8〜11は、8M尿素/50mMリン酸Na/500mM NaCl/250mMイミダゾール溶液による溶出の結果を示す。
ユビキチンが融合した成熟型のUB−p53タンパク質を発現するBL21(DE3)の産生株をLB液体培地に播種し、37℃の震盪インキュベーター内で培養した。OD600の吸光度が0.4の達したら、0.5mMのIPTGを加え、さらに4時間の震盪培養を行って、ユビキチンが融合した成熟型のUB−p53タンパク質タンパク質を発現させた。
ユビキチンが融合した成熟型のUB−p53−TOM7タンパク質を発現する大腸菌BL21(DE3)の産生株をLB液体培地に接種し、37℃の条件下に培養した。OD600の吸光度が0.4の達したら、0.5mMのIPTGを加え、さらに4時間の震盪培養を行って、ユビキチンが融合した成熟形態のUB−p53−TOM7タンパク質を発現させた。
実施例2.1. グランザイムB遺伝子の増幅
組換えタンパク質においてヒトグランザイムBを発現させるために、ヒトナチュラルキラー細胞から全RNAを抽出し、そこからcDNAを合成した。具体的には、ヒトナチュラルキラー細胞を、5%二酸化炭素および37℃の条件下に10%血清培地中で培養した(1×106細胞)。その後、RNAを、実施例1.1.と同じ方法で取得し、それをグランザイムB遺伝子のポリメラーゼ連鎖反応の鋳型として用いた。
実施例2.2.1. プラスミドpET11c−TOM70−(GGGGS)3−UB−グランザイムBの作製
ミトコンドリア外膜に結合するTOM70、リンカー(GGGGSGGGGSGGGGS)およびユビキチンが融合した形態のグランザイムBタンパク質を調製するために、TOM70、リンカーおよびユビキチンが融合した形態のグランザイムBタンパク質を発現することのできる発現ベクターを作製した。
ユビキチン、およびミトコンドリア外膜に結合するTOM7が融合した形態のグランザイムBタンパク質を調製するために、ユビキチン、グランザイムBおよびTOMがこの順序で融合した形態のグランザイムBタンパク質を発現することのできる発現ベクターを作製した。
TOM70−(GGGGS)3−UB−グランザイムBタンパク質を、実施例1.3.1.と同じ方法で単離および精製した。結果として、TOM70−(GGGGS)3−UB−グランザイムBタンパク質を溶出させた(図27)。この場合、図27のレーンMはタンパク質分子量マーカーを示し、レーン1はニッケルアフィニティークロマトグラフィーのローディングサンプルを示す。レーン2はニッケルアフィニティー樹脂に結合しなかったものを示す。レーン3および4は、8M尿素/50mMリン酸Na/500mM NaCl/50mMイミダゾール溶液による溶出の結果を示す。レーン5〜7は、8M尿素/50mMリン酸Na/500mM NaCl/100mMイミダゾール溶液による溶出の結果を示す。レーン8〜9は、8M尿素/50mMリン酸Na/500mM NaCl/250mMイミダゾール溶液による溶出の結果を示す。
実施例3.1. RKIP遺伝子の増幅
組換えタンパク質においてヒトRKIP(Rafキナーゼ阻害タンパク質)遺伝子を発現させるために、ヒト上皮細胞から全RNAを抽出し、そこからcDNAを合成した。ヒト上皮線維芽細胞を、5%二酸化炭素および37℃の条件下に10%血清培地中で培養した(1×106細胞)。その後、RNAを、実施例1.1.と同じ方法で取得し、それをRKIP遺伝子のポリメラーゼ連鎖反応の鋳型として用いた。
実施例3.2.1. プラスミドpET11c−TOM70−(GGGGS)3−UB−RKIPの作製
ミトコンドリア外膜に結合するTOM70、リンカー(GGGGSGGGGSGGGGS)およびユビキチンが融合した形態のRKIPタンパク質を調製するために、TOM70、リンカーおよびユビキチンが融合した形態のRKIPを発現することのできる発現ベクターを作製した。
組換えTOM70−(GGGGS)3−UB−RKIPを発現する大腸菌BL21(DE3)の産生株をLB液体培地に播種し、37℃の条件下に培養した。OD600の吸光度が0.3に達したら、それを冷蔵庫に入れて培養液の温度を下げ、インキュベーターの温度を18℃に変更し、その後、0.5mMのIPTGを加え、さらに1日間の震盪培養を行って、TOM70−(GGGGS)3−UB−RKIPタンパク質を発現させた。
実施例4.1. PTEN遺伝子の増幅
組換えタンパク質においてヒトPTEN(ホスファターゼテンシンホモログ)を発現させるために、ヒト上皮細胞から全RNAを抽出し、そこからcDNAを合成した。線維芽細胞(ヒト皮膚線維芽細胞)を、5%二酸化炭素および37℃の条件下に10%血清培地中で培養した(1×106細胞)。その後、RNAを、実施例1.1.と同じ方法で取得し、それをPTEN遺伝子のポリメラーゼ連鎖反応の鋳型として用いた。
実施例4.2.1. プラスミドpET11c−TOM70−(GGGGS)3−UB−PTENの作製
ミトコンドリア外膜に結合するTOM70、リンカー(GGGGSGGGGSGGGGS)およびユビキチンが融合した形態のPTENタンパク質を調製するために、TOM70、リンカーおよびユビキチンが融合した形態のPTEN遺伝子を発現することのできる発現ベクターを作製した。
組換えTOM70−(GGGGS)3−UB−PTENタンパク質を、実施例1.3.1.と同じ方法で単離および精製した。結果として、TOM70−(GGGGS)3−UB−PTENタンパク質を溶出させた(図37)。この場合、図37のレーンMはタンパク質分子量マーカーを示し、レーン1はニッケルアフィニティークロマトグラフィーのローディングサンプルを示す。レーン2はニッケルアフィニティー樹脂に結合しなかったものを示す。レーン3は、8M尿素/50mMリン酸Na/500mM NaCl/10mMイミダゾール溶液による溶出の結果を示す。レーン4は、8M尿素/50mMリン酸Na/500mM NaCl/50mMイミダゾール溶液による溶出の結果を示す。レーン5〜8は、8M尿素/50mMリン酸Na/500mM NaCl/100mMイミダゾール溶液による溶出の結果を示す。レーン9〜10は、8M尿素/50mMリン酸Na/500mM NaCl/250mMイミダゾール溶液による溶出の結果を示す。レーン111は、8M尿素/50mMリン酸Na/500mM NaCl/500mMイミダゾール溶液による溶出の結果を示す。
実施例5.1. 大腸菌から誘導された組換えUB−GFP−TOM7タンパク質の単離および精製
ユビキチンが融合した成熟型のUB−GFP−TOM7タンパク質を発現する大腸菌BL21(DE3)の産生株をLB液体培地に播種し、37℃の条件下に培養した。OD600の吸光度が0.3に達したら、それを冷蔵庫に入れて培養液の温度を下げ、インキュベーターの温度を18℃に変更し、その後、0.5mMのIPTGを加え、さらに1日間の震盪培養を行って、ユビキチンが融合した成熟型のGFP−TOM7タンパク質を発現させた。
組換えタンパク質TOM70−(GGGGS)3−UB−GFPを発現する大腸菌BL21(DE3)の産生株をLB液体培地に播種し、37℃の条件下に培養した。OD600の吸光度が0.3に達したら、それを冷蔵庫に入れて培養液の温度を下げ、インキュベーターの温度を18℃に変更し、その後、0.5mMのIPTGを加え、さらに1日間の震盪培養を行って、組換えタンパク質TOM70−(GGGGS)3−UB−GFPを発現させた。
実施例6. scFvHER2を含む融合タンパク質の調製
実施例6.1. scFvHER2遺伝子の合成
組換えタンパク質においてヒトscFvHER2を発現させるために、Bionics Co., Ltd.から遺伝子合成依頼によって入手したscFvHER2遺伝子をpUC57−scFvHER2と称し、scFvHERの塩基配列は配列番号37の塩基配列と同じであった。
実施例6.2.1. pET15b−UB−scFvHER2−TOM7
ユビキチン、およびミトコンドリア外膜に結合するTOM7が融合した形態のscFvHER2タンパク質を調製するために、ユビキチンおよびTOM7が融合した形態のscFvHER2遺伝子を発現することのできる発現ベクターを作製した。
ミトコンドリア外膜に結合するTOM7が融合した形態のscFvHER2タンパク質を調製するために、TOM7が融合した形態のscFvHER2を発現することのできる動物細胞用の発現ベクターを作製した。TOM7およびscFvHER2遺伝子を得るために、RscFvHER2プライマーおよびXTOM7(noT)プライマーを合成した。各プライマーの配列を表11に記載する。
UB−scFvHER2−TOM7タンパク質を、実施例1.3.1.と同じ方法で単離および精製した。結果として、UB−scFvHER2−TOM7タンパク質を溶出させた(図47)。この場合、図47のレーンMはタンパク質分子量マーカーを示し、レーン1はニッケルアフィニティークロマトグラフィーのローディングサンプルを示す。レーン2はニッケルアフィニティー樹脂に結合しなかったものを示す。レーン3は、8M尿素/50mMリン酸Na/500mM NaCl/10mMイミダゾール溶液による溶出の結果を示す。レーン4〜5は、8M尿素/50mMリン酸Na/500mM NaCl/50mMイミダゾール溶液による溶出の結果を示す。レーン6〜8は、8M尿素/50mMリン酸Na/500mM NaCl/100mMイミダゾール溶液による溶出の結果を示す。レーン9〜10は、8M尿素/50mMリン酸Na/500mM NaCl/250mMイミダゾール溶液による溶出の結果を示す。レーン11は、8M尿素/50mMリン酸Na/500mM NaCl/500mMイミダゾール溶液による溶出の結果を示す。
実施例7.1. scFvMEL遺伝子の合成
組換えタンパク質においてメラノーマに対する抗体フラグメントとしてのヒトscFvMELを発現させるために、Bionics Co., Ltd.から遺伝子合成依頼によって入手したscFvMEL遺伝子をpUC57−scFvMELと称し、scFvMELの塩基配列は配列番号42の塩基配列と同じであった。
実施例7.2.1. pET15b−UB−scFvMEL−TOM7の作製
ユビキチン、およびミトコンドリア外膜に結合するTOM7が融合した形態のscFvMELタンパク質を調製するために、ユビキチンおよびTOM7が融合した形態のscFvMELを発現することのできる発現ベクターを作製した。

組換えタンパク質においてヒトscFvPD−L1を発現させるために、Bionics Co., Ltd.から遺伝子合成依頼によって入手したscFvPD−L1遺伝子をpUC57−scFvPD−L1と称し、その塩基配列は配列番号46の塩基配列と同じであった。
実施例8.2.1. pCMV−scFvPD−L1−TOM7−myc/Hisの作製
ミトコンドリア外膜に結合するTOM7が融合した形態のscFvPD−L1タンパク質を調製するために、ユビキチンおよびTOM7が融合した形態のscFvPD−L1を発現することのできる動物細胞用の発現ベクターを作製した。
実施例9. 改変ミトコンドリアの作製
ミトコンドリア外膜結合部位と融合された蛍光タンパク質がミトコンドリア外膜に結合するかを確認するために、以下の実験を行った。まず、臍帯由来の間葉系幹細胞(UC−MSC)から遠心分離法によってミトコンドリアを単離した。次いで、それをMitoTracker CMXRos Redで染色した。それを、前記の大腸菌から精製された組換えタンパク質TOM70−(GGGGS)3−UB−GFPと混合し、周囲温度で約30分間インキュベートした。
臍帯由来間葉系幹細胞から遠心分離法によって単離されたミトコンドリアを、精製組換えタンパク質TOM70−(GGGGS)3−UB−p53またはUB−p53−TOM7と混合し、4℃の反応条件下に1時間、1:1の比で結合させた。対照群として、タンパク質と混合しないミトコンドリアを用いた。ミトコンドリアとp53の間の結合親和性を、ウエスタンブロット試験法によって確認した(図56)。
実施例11. 外来ミトコンドリアの単離および細胞内導入
臍帯由来間葉系幹細胞(UC−MSC)から遠心分離法によってミトコンドリアを単離した。単離ミトコンドリアをMitotracker CMX Rosで染色し、単離ミトコンドリアの濃度および総量をBCA定量法によって確認し、0μg、1μg、5μg、10μg、50μg、100μgのミトコンドリアを胃癌細胞株SNU−484細胞に遠心法によって導入した。試験の結果、細胞へのミトコンドリアの導入の程度は、ミトコンドリア量に濃度依存的であったことが蛍光顕微鏡によって確認された(図57)。
正常細胞由来のミトコンドリアが、癌細胞増殖およびROS産生に対してどのように作用するかを調べるために、以下の実験を行った。まず、肝細胞(WRL−68)、線維芽細胞、および臍帯由来間葉系幹細胞(UC−MSC)を、ミトコンドリアドナー細胞として選択した。細胞からミトコンドリアを遠心分画法によってそれぞれ単離した。ミトコンドリアレシピエント細胞として用いた癌細胞は、皮膚表皮癌細胞のA431細胞株であった。この場合、皮膚表皮癌細胞にミトコンドリアを、濃度にしたがい遠心力を利用して送達した(韓国特許出願番号10−2017−0151526)。
正常細胞由来のミトコンドリアが癌細胞に導入されたとき、癌細胞の特徴である薬剤耐性、抗酸化遺伝子の発現、癌転移に対してどのように作用するかを、以下の方法で調べた。まず、正常肝細胞(WRL−68)をミトコンドリアドナー細胞とし、細胞からミトコンドリアを遠心分画法によって単離し、ミトコンドリアを使用した。ミトコンドリアレシピエント細胞として用いる癌細胞として、肝臓癌細胞株のHepG2細胞を使用した。ミトコンドリアを肝臓癌細胞に、濃度にしたがい遠心力を利用して送達し、抗癌剤ドキソルビシンに対する薬剤耐性の観察の結果として、ミトコンドリアを受け取った癌細胞株は、より高い薬剤感受性を示したことを確認した(図60)。
正常細胞から単離されたミトコンドリアを濃度にしたがい肝臓癌細胞株のHepG2細胞に導入したとき、抗酸化タンパク質である酵素カタラーゼおよびSOD−2(スーパーオキシドジスムターゼー2遺伝子の癌細胞における発現が増加したことが確認された(図61)。
転移に関して、EMT(上皮間葉転換)に関与する遺伝子の1つであるα−平滑筋アクチン(α−SMA)遺伝子の発現があったかを確認した。この場合、ミトコンドリアを受け取った肝臓癌細胞の場合、ミトコンドリアを受け取らなかった肝臓癌細胞と比較して、α−SMAタンパク質の発現が、ミトコンドリア濃度に依存して顕著に低下したことがわかった。一方、細胞接着タンパク質の1つであるE−カドヘリンタンパク質は、ミトコンドリア濃度に依存して増加した(図62)。癌細胞に導入された正常ミトコンドリアによって、癌転移に関与することが知られるタンパク質の変化がもたらされ、したがって癌細胞の転移にも作用することが確認された。
ミトコンドリアを臍帯由来間葉系幹細胞から遠心分離法によって単離し、Mitotracker CMX Rosで染色し、精製組換えタンパク質TOM70−(GGGGS)3−UB−p53またはUB−p53−TOM7と混合し、1:1の比で、4℃の反応条件下に1時間インキュベートし、その後、遠心分離して未反応タンパク質を除去し、PBSバッファー液で2回洗った後、p53タンパク質が結合した形態のミトコンドリアを胃癌細胞株SNU−484細胞に遠心法によって導入した(図63)。この場合、対照群は、ミトコンドリアを用いない群、およびミトコンドリアを単独で用いる群と設定した。1日間の培養後、細胞に導入された外来ミトコンドリアに結合されたp53タンパク質を、免疫細胞化学(ICC)により蛍光顕微鏡で観察した。
実施例17.1. 胃癌細胞株を用いての、細胞に導入されたp53結合外来ミトコンドリアのアポトーシス能力の確認
臍帯由来間葉系幹細胞から遠心分離法によって単離されたミトコンドリアを、大腸菌から精製された組換えタンパク質TOM70−(GGGGS)3−UB−p53またはUB−p53−TOM7と混合し、4℃の反応条件下に1時間、1:1の比で結合させた。対照群として、TOM70を含まないUB−p53タンパク質、およびユビキチンを含まないTOM70−(GGGGS)3−p53を用いた。未結合タンパク質を遠心分離およびPBSによる洗浄プロセスによって除去し、タンパク質が結合したミトコンドリアを、p53遺伝子の変異によってp53能力を欠く胃癌細胞株SNU−484に遠心によって導入した(図66)。1日間の培養後、4%パラホルムアルデヒドによる固定を1時間行い、次いで、浸透化溶液(0.1% Triton-X-100を含む0.1%クエン酸ナトリウムバッファー、pH7.4)を用いて細胞の浸透化を誘導し、TUNEL溶液(in situ細胞死検出キット、TMR RED, Roche)と37℃で1時間反応させた。
レシピエント細胞において送達されたTOM70−(GGGGS)3−UB−p53タンパク質の生物学的活性が、実施例5.2.において得られたミトコンドリアに結合した形態のTOM70−(GGGGS)3−UB−p53タンパク質のレシピエント細胞への送達後に維持されるかを確認するために、レポーター遺伝子を用いる細胞ベースの分析を行った。p53タンパク質は転写因子であるから、p53転写因子が結合することのできる塩基配列RRRCWWGYYY(ここで、RはGまたはAを表し、WはAまたはTを表し、YはCまたはTを表す)が6回繰り返された遺伝子を、下記の配列で合成した。P53−プロモーター−Sの塩基配列は、以下のとおり(5’−GGG CAT GCT CGG GCA TGC CCG GGC ATG CTC GGG CAT GCC CGG GCA TGC TCG GGC ATG CCC−3’)(配列番号91)であり、P53−プロモーター−ASの塩基配列は、以下のとおり(5’−GGG CAT GCC CGA GCA TGC CCG GGC ATG CCC GAG CAT GCC CGG GCA TGC CCG AGC ATG CCC−3’)(配列番号92)である。
臍帯由来間葉系幹細胞から遠心分離法によって単離されたミトコンドリアを、精製組換えタンパク質TOM70−(GGGGS)3−UB−RKIPと混合し、4℃の反応条件下に1時間、1:1の比で結合させた。タンパク質が結合したミトコンドリアを、RKIPタンパク質の減少によって転移能が高まっていることが知られている乳癌細胞株MDA−MB−231に遠心によって導入した。
実施例19. 癌細胞を標的とする一本鎖可変フラグメント(ScFv)抗体の細胞内発現の確認、および細胞におけるミトコンドリアとの結合の確認
pCMV−ScFv−HER2−TOM7またはpCMV−ScFv−MEL−TOM7またはpCMV−ScFv−PD−L1−TOM7を動物細胞内で発現させるために、リポフェクタミンLTXおよびPLUSまたはリポフェクタミン2000を用いてDNAをCHO細胞にトランスフェクトした。対照群として、GFP−TOM7 DNAを用いた。それらが細胞内で発現され、同じ細胞内でミトコンドリアに結合することを確認するために、トランスフェクトした細胞からサイトゾルおよびミトコンドリアを遠心分離法によって単離し、BCAアッセイを用いて同じタンパク質量に調節してPAGE電気泳動を行い、結果をウエスタンブロットによって観察した。モノクローナルc−myc抗体を一次抗体として使用し、抗マウスIgG HRPを二次抗体として使用した。
pCMV−ScFv−HER2−TOM7またはpCMV−ScFv−PD−L1−TOM7をトランスフェクトしたCHO細胞からミトコンドリアを単離した。対照群として、形質転換していないCHO細胞のミトコンドリアを単離して使用した。各細胞から単離されたミトコンドリアを、Mitotracker CMX Rosで染色した。同量のミトコンドリアで胃癌細胞株SNU−484を処理し、翌日、ミトコンドリアの細胞導入の程度を蛍光顕微鏡を用いて比較し確認した。対照群と比較して、ScFv−HER2−TOM7またはScFv−PD−L1−TOM7が結合したミトコンドリアは、対照群から得たミトコンドリアよりも多く癌細胞に導入されたことが確認された(図72)。したがって、標的タンパク質が結合したミトコンドリアは、単独のミトコンドリアよりも容易に癌細胞に導入されることがわかった。
実施例21. 異種移植モデル(SNU−484)の作製、および試験材料の投与
実施例21.1. 癌細胞の準備
実験日に、胃癌細胞株のSNU−484細胞株を、マウス当たり5×106細胞とするように準備した。細胞の培地を除去し、PBSを加えて細胞を洗った。トリプシン−EDTA液で細胞を解離させ、細胞を50mLチューブに入れてPBSバッファー液で2回洗った後、20mLのPBSを加え、細胞の数および生存性を調べた。測定された細胞数に基づいてマウス当たりの細胞数を5×106細胞に調節し、細胞を群に分けることによって準備した。マウス当たりの移植体積は、同じ100μLとなるよう調節した。対照群として、100μLの癌細胞単独の群を用意した。
前記のように臍帯血間葉系幹細胞から単離されたミトコンドリアを、タンパク質濃度に基づきマウス当たり50μgの量で移植用に準備した。単独のミトコンドリアを投与する群の場合は、ミトコンドリアを、癌細胞を混合した100μLのPBSとよく混合することによって準備した。改変ミトコンドリア群の場合は、準備したその量のミトコンドリアを癌細胞と混合する前に、エッペンドルフチューブ内でTOM70−(GGGGS)3−UB−p53タンパク質と1:1の濃度比で混合し、周囲温度で1時間置いた。反応時間の経過後、20000×gで10分間の遠心分離によって上清を除去し、タンパク質が結合したミトコンドリア(MT+TOM70−(GGGGS)3−UB−p53)のペレットを得た。それをPBSバッファー液で2回洗った後、p53タンパク質が結合したミトコンドリア(MT+TOM70−(GGGGS)3−UB−p53)を、癌細胞を混合した100μLのPBSとよく混合することによって準備した。
群によって準備される移植サンプル用に、マトリゲル(BD)をPBSと同じ量で加え、細胞と軽く混ぜて、マウス当たり200μLの試験材料を調製した。この場合、操作はすべて氷上で行った。モデル作製のために、Balb/cヌードマウス(雌、7週齢)をRAONBIOから購入し、癌細胞移植のためにイソフルラン吸入により麻酔し、右背部域(動物に基づく)をアルコール綿で消毒した。その後、注射液を入れた1ml注射器を用いて試験動物の右背部域に200μLを皮下投与した。投与後、動物の体重および腫瘍サイズを週2回測定し、3週間まで観察を続けて結果を分析した(図73)。
腫瘍の長軸長および短軸長を計測し下記式に当てはめることにより腫瘍体積を計算した。

抗癌剤候補の投与によるマウスの生理学的および形態学的変化を観察するために、癌細胞および試験材料の投与時から、体積および腫瘍サイズの変化を週に2回測定した(図74)。
上記のように得たp53が結合したミトコンドリアを、皮膚癌細胞であるA431細胞に遠心法によって送達し、A431細胞の増殖を観察した。この場合、対照群として生理食塩液を使用し、対照試験群として、p53タンパク質を融合していない等量のミトコンドリアを使用した。アポトーシスを誘導するタンパク質であるp53タンパク質が結合したミトコンドリアは、対照群、および単独のミトコンドリアを用いた群と比較して、A431細胞の増殖を有意に抑制できることが確認された(図76)。
実施例23. 単離されたミトコンドリアの機能の確認:ATP量
臍帯由来間葉系幹細胞(UC−MSC)から細胞内ミトコンドリアを単離するために、シリンジを用いてホモジナイズして細胞を破壊し、次いで連続的遠心分離を行ってミトコンドリアを得た。単離されたミトコンドリアの機能を確認するために、単離されたミトコンドリアのミトコンドリアタンパク質濃度をBCAアッセイによって定量して、5μgのミトコンドリアを準備した。CellTiter-Glo発光キット(Promega, Madison, WI)を用いて、ミトコンドリア中のATP量を確認した。
単離されたミトコンドリアの膜電位を確認するために、JC−1色素(分子プローブ、カタログ番号1743159)を使用した。用意したミトコンドリアを50μlのPBS中に混合し、96ウェルプレートに準備した。対照群としてのミトコンドリアを含まないPBS(50μl)群、およびCCCP(R&D systems, CAS 555-60-2)処理群を準備した。ミトコンドリアのイオノフォアであるCCCPは、ミトコンドリア膜電位の脱分極によってミトコンドリアの機能を阻害する。CCCP群を50μMで、単離されたミトコンドリアと室温で10分間反応させた。
前記のように準備した5μgのミトコンドリアが傷害されているかを確認するために、単離されたミトコンドリア中のミトコンドリア活性酸素種を分析することのできるMitoSOX red指示薬(Invitrogen, カタログ番号M36008)の色素を使用した。準備したミトコンドリアを50μlのPBSと混合し、96ウェルプレートに準備し、対照群としてのミトコンドリアを含まない50μlのPBSと比較した。MitoSOX red色素を10μMの濃度となるように50μlのPBSと混合し、96ウェルプレートに入れ(終濃度5μM)、37℃のCO2インキュベーター内で20分間反応させた。反応の終了後、ミトコンドリア中のROS量をマイクロプレートリーダーを用いて測定した(励起510/蛍光580)。結果を図80に示す。
実施例26. 細胞外部でのミトコンドリア外膜タンパク質に結合した所望のタンパク質の解離の確認
ミトコンドリアと結合した活性タンパク質を細胞に導入したとき、所望のタンパク質を遊離形態で得るために、ユビキチンタンパク質がミトコンドリア外膜タンパク質と所望のタンパク質の間に挿入された形態の融合タンパク質(TOM70−UB−p53またはTOM−UB−GFP)を大腸菌から調製した。ユビキチン配列がユビキチン切断酵素UBP1によって切断されるかを確認するために、組換え融合タンパク質TOM70−UB−p53をUBP1酵素と37℃で1時間反応させた。
上記実施例において得た融合タンパク質(TOM70−(GGGGS)3−UB−p53またはTOM70−(GGGGS)3−UB−GFP)がミトコンドリアと結合した状態で細胞に入ったとき、細胞中に存在するユビキチン切断酵素によって活性タンパク質が解離されるかを観察した。まず、臍帯血間葉系細胞から得たミトコンドリアと融合タンパク質TOM70−(GGGGS)3−UB−GFPをマイクロチューブ内で1時間反応させて結合させ、その後、未結合融合タンパク質を遠心分離により除去し、PBSバッファー液で2回洗った。この場合、ユビキチンが除かれている融合タンパク質(TOM70−(GGGGS)3−p53)を対照群として使用した。

- 所望のタンパク質が、p53、グランザイムB、Bax、Bak、PDCD5、E2F、AP−1(Jun/Fos)、EGR−1、網膜芽細胞腫(RB)、ホスファターゼテンシンホモログ(PTEN)、E−カドヘリン、ニューロフィブロミン−2(NF−2)、ポリ[ADP−リボース]合成酵素1(PARP−1)、BRCA−1、BRCA−2、大腸腺腫性ポリポーシス(APC)、腫瘍壊死因子受容体関連因子(TRAF)、RAFキナーゼ阻害タンパク質(RKIP)、p16、KLF−10、LKB1、LHX6、C−RASSF、DKK−3PD1、Oct3/4、Sox2、Klf4、およびc−Mycからなる群から選択される任意の1つである、請求項36に記載の融合タンパク質。
- ミトコンドリア外膜アンカリングペプチドが、TOM20、TOM70またはOM45であるとき、ミトコンドリア外膜アンカリングペプチドおよび所望のタンパク質がN末端からC末端へと連結されている、請求項36に記載の融合タンパク質。
- ミトコンドリア外膜アンカリングペプチドが、TOM5、TOM6、TOM7、TOM22、Fis1、Bcl−2、Bcl−xおよびVAMP1Bからなる群から選択される任意の1つであるとき、所望のタンパク質およびミトコンドリア外膜アンカリングペプチドがN末端からC末端へと連結されている、請求項36に記載の融合タンパク質。
- 融合タンパク質が、ユビキチンもしくはそのフラグメントを、ミトコンドリア外膜アンカリングペプチドと所望のタンパク質の間にさらに含む、請求項36に記載の融合タンパク質。
- 融合タンパク質が、真核細胞中のタンパク質分解酵素によって認識されるアミノ酸配列を、ミトコンドリア外膜アンカリングペプチドと所望のタンパク質の間にさらに含む、請求項36に記載の融合タンパク質。
- 請求項36〜43のいずれかに記載される融合タンパク質をコードするポリヌクレオチド。
- 原核細胞、またはユビキチン分解酵素もしくはタンパク質分解酵素を持たない真核細胞に、請求項44に記載されるポリヌクレオチドを導入するステップ;および
融合タンパク質を取得するステップ
を含む、請求項36〜43のいずれかに記載される融合タンパク質を製造する方法。 - 細胞膜に存在するリガンドまたは受容体に結合する能力を有する標的標的化タンパク質、およびミトコンドリア外膜アンカリングペプチドを含む、融合タンパク質。
- ミトコンドリア外膜アンカリングペプチドが、TOM5、TOM6、TOM7、TOM22、Fis1、Bcl−2、Bcl−xおよびVAMP1Bからなる群から選択される任意の1つである、請求項46に記載の融合タンパク質。
- 細胞膜に存在するリガンドまたは受容体に結合する能力を有する標的標的化タンパク質、およびミトコンドリア外膜アンカリングペプチドが、N末端からC末端へと連結されている、請求項46に記載の融合タンパク質。
- 細胞膜に存在するリガンドまたは受容体に結合する能力を有する標的標的化タンパク質が、抗体またはそのフラグメントである、請求項48に記載の融合タンパク質。
- 抗体のフラグメントが、Fab、Fab’、scFvおよびF(ab)2からなる群から選択される任意の1つである、請求項49に記載の融合タンパク質。
- 請求項46〜50のいずれかに記載される融合タンパク質をコードするポリヌクレオチド。
- 単離されたミトコンドリアを、請求項36〜43のいずれかに記載される融合タンパク質、および/または請求項46〜50のいずれかに記載される融合タンパク質と混合するステップを含む、改変ミトコンドリアの作製方法。
- 請求項36〜41のいずれかに記載される融合タンパク質および/または請求項46〜50のいずれかに記載される融合タンパク質をコードするポリヌクレオチドを真核細胞に導入することにより形質転換した細胞から改変ミトコンドリアを調製する方法。
