本明細書に記載される化合物を薬学的に許容される活性または不活性物質と混合して医薬組成物を調製し得る。医薬組成物製剤化のための組成物および方法は、特に限定されないが、投与経路、疾患の程度、または投与する用量を含めた多数の基準によって決まる。
以下の実施例には、DNM2を標的とするリード化合物を特定するのに用いたスクリーニング工程を記載する。スクリーニングした3,000を超えるオリゴヌクレオチドのうち、強力で耐容性のあるオリゴヌクレオチドが多数特定され、化合物951799,949935,950023,950089,951372,950060,および950132が上位のリード化合物として浮上した。特に化合物951799は、効力および耐容性の点で最も優れた特性の組合せを示した。
本願に添付する配列表では、各配列が必要に応じて「RNA」または「DNA」として特定されるが、実際には、これらの配列が化学修飾の任意の組合せで修飾されていてもよい。当業者には、修飾オリゴヌクレオチドを記載するこのような「RNA」または「DNA」という名称は、特定の場合には任意のものであることが容易に理解されよう。例えば、2’−OH糖部分とチミン核酸塩基とを含むヌクレオシドを含むオリゴヌクレオチドは、RNA糖を有するDNAまたはDNA核酸塩基を有するRNAと記載され得る。
1つまたは複数のダイナミン2(DNM2)核酸に相補的な修飾オリゴヌクレオチドを設計し、ダイナミン2 mRNAの発現に対するそのin vitroの効果を試験した。培養条件がほぼ同じである一連の実験で修飾オリゴヌクレオチドを試験した。

上の実施例に挙げた選択されたオリゴヌクレオチドをA431細胞で、自由取込みにより様々な用量で試験した。細胞を1ウェル当たり細胞10,000個の密度で培養し、下の表に明記するように、8nM、40nM、200nM、または1,000nMの修飾オリゴヌクレオチドで処置した。約48時間後、全RNAを単離し、プライマープローブセットRTS36027(実施例1に記載したもの)を用いてRT−PCRにより解析し、GADPHで正規化した。下の表に示すように、DNM2に相補的な修飾オリゴヌクレオチドによって、DNM2 mRNA発現が用量依存性に阻害された。
上の実施例に挙げた選択されたオリゴヌクレオチドをA431細胞で、自由取込みにより様々な用量で試験した。細胞を1ウェル当たり細胞10,000個の密度で培養し、下の表に明記するように、7.8nM、31.25nM、125nM、500nM、または2,000nMの修飾オリゴヌクレオチドで処置した。約48時間後、全RNAを単離し、プライマープローブセットRTS36027(実施例1に記載したもの)を用いてRT−PCRにより解析し、GADPHで正規化した。下の表に示すように、DNM2に相補的な修飾オリゴヌクレオチドによって、DNM2発現が用量依存性に阻害された。

上の実施例に挙げた選択されたオリゴヌクレオチドをSkMC細胞(ヒト骨格筋、Lonza CC−2561)で、様々な用量で試験した。細胞を1ウェル当たり細胞20,000個の密度で培養し、下の表に明記するように、7.8nM、31.25nM、125nM、500nM、または2,000nMの修飾オリゴヌクレオチドをエレクトロポレーションによりトランスフェクトした。約16時間後、全RNAを単離し、プライマープローブセットRTS36027(実施例1に記載したもの)を用いてRT−PCRにより解析し、ribogreenで正規化した。下の表に示すように、DNM2に相補的な修飾オリゴヌクレオチドによって、DNM2発現が用量依存性に阻害された。
CD1(登録商標)マウス(Charles River社、マサチューセッツ州)を上の実施例から選択した修飾オリゴヌクレオチドで処置し、様々な血漿化学マーカーのレベルの変化を評価した。6週齢の雄CD1マウスのグループに週1回、下の表に挙げる修飾オリゴヌクレオチドを50mg/kgで6週間皮下注射した(用量50mg/(kg・週))。各グループにはマウス4匹を含めた。1つの雄CD1マウス対照群には、PBSを週1回、6週間皮下投与した。最後の投与から48時間後、マウスを屠殺し、のちの解析に臓器および血漿を回収した。

- 長さが連結ヌクレオシド14〜30個であり、
i)配列番号2879の少なくとも14個の連続する核酸塩基を含む核酸塩基配列;
ii)配列番号1の核酸塩基87,359〜90,915;90,968〜97,263;83,573〜87,287;3,404〜44,737;97,378〜104,979;81,061〜81,199;または116,048〜116,903の等しい長さの部分に100%相補的な少なくとも14個の連続する核酸塩基の部分を含む核酸塩基配列、
iii)ヒトDNM2プレmRNAもしくはヒトDNM2 mRNAのイントロン12、イントロン13、イントロン11、イントロン1、イントロン14、エクソン10、もしくは3’−UTRに相補的な核酸塩基配列、または
iv)配列番号7〜3134のいずれか1つの核酸塩基配列の少なくとも12個、少なくとも13個、少なくとも14個、もしくは少なくとも15個の連続する核酸塩基を含む核酸塩基配列

- 薬学的に許容される塩の形態の請求項1〜13のいずれか1項に記載の化合物よりなり、好ましくは、前記薬学的に許容される塩が、ナトリウム塩またはカリウム塩である、化合物。
- 特定の立体化学的配置を有する少なくとも1つの特定のホスホロチオアートヌクレオシド間結合を含む修飾オリゴヌクレオチドが富化されている、請求項1〜14のいずれか1項に記載の化合物のキラル富化集団。
- 請求項1〜14のいずれか1項に記載の化合物、または請求項15に記載の化合物の集団と、少なくとも1つの薬学的に許容される希釈剤もしくは担体とを含む、医薬組成物。
- 治療法に使用する、請求項1〜14のいずれか1項に記載の化合物、請求項15に記載の化合物の集団、または請求項16に記載の医薬組成物。
- 中心核ミオパチー、デュシェンヌ型筋ジストロフィー、またはシャルコー・マリー・トゥース病の治療、予防、または改善に使用し、好ましくは、前記疾患が、X連鎖筋細管ミオパチー、常染色体劣性中心核ミオパチー、または常染色体優性中心核ミオパチーであり、より好ましくは、前記疾患が、MTM1、BIN1、およびDNM2のなかから選択される少なくとも1つの遺伝子の変異に関連するものである、請求項1〜14のいずれか1項に記載の化合物、請求項15に記載の集団、または請求項16に記載の医薬組成物。
- 細胞内でのDNM2発現を阻害するin vitroの方法であって、前記細胞と、DNM2核酸転写物のエクソン10、イントロン、または3’−UTRに100%相補的な修飾オリゴヌクレオチドを含む一本鎖化合物とを接触させ、それにより前記細胞内でのDNM2発現を阻害することを含む、方法。
