Abstract
Description
広い応用の見通しがある。
メージングのための緑色蛍光プローブとしてプリン修飾されたAuCを使用している(非特許文献6)。この種類の文献は、AuCの蛍光特性を利用するものであり、AuC自体の医薬活性を必要とするものではない。
(1)HAuCl4をメタノール、水、エタノール、n−プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M〜0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M〜0.18Mである溶液Bを得る工程と、
(3)工程(1)における溶液Aと工程(2)における溶液Bとを、HAuCl4とリガンドYとの間のモル比1:(0.01〜100)(好ましくは1:(0.1〜10)、より好ましくは1:(1〜10))で混合し、それらを氷浴中で0.1時間〜48時間(好ましくは0.1時間〜24時間、より好ましくは0.5時間〜2時間)にわたり撹拌し、0.025M〜0.8MのNaBH4溶液(好ましくはNaBH4の水溶液、NaBH4のエタノール溶液、又はNaBH4のメタノール溶液)を添加し、次いで氷水浴中で0.1時間〜12時間(好ましくは0.1時間〜2時間、より好ましくは1時間〜2時間)にわたりNaBH4とリガンドYとの間のモル比1:(0.01〜100)(好ましくは1:(0.1〜8)、より好ましくは1:(1〜8))で撹拌し続ける工程と、
(4)工程(3)における反応溶液を8000回転/分〜17500回転/分で10分間〜100分間にわたり遠心分離することで、AuC沈降物を種々の平均粒度で得る工程、好ましくはMWCOが3K〜30Kの限外濾過チューブを使用して、工程(3)における反応溶液を8000回転/分〜17500回転/分で10分間〜100分間にわたり勾配により遠心分離することで、AuCを種々の平均粒度で得る工程と、
(5)工程(4)において得られた種々の平均粒度のAuC沈降物を水中に溶解させ、それを透析バッグに入れて、水中にて室温で1日間〜7日間にわたり透析する工程と、
(6)上記透析バッグ中のAuC溶液を12時間〜24時間にわたり凍結乾燥させることで、AuC含有物質を得る工程と、

この実施形態は、リガンド修飾されたAuCを製造する方法であって、以下の工程:
(1)HAuCl4をメタノール、水、エタノール、n−プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M〜0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M〜0.18Mである溶液Bを得る工程と、
リガンドYには、限定されるものではないが、L(D)−システイン及びその他のシステイン誘導体、例えばN−イソブチリル−L−システイン(L−NIBC)、N−イソブチリル−D−システイン(D−NIBC)、N−アセチル−L−システイン及びN−アセチル−D−システイン、限定されるものではないが、ジペプチド、トリペプチド、テトラペプチド及びその他のシステイン含有ペプチドを含むシステイン含有オリゴペプチド及びそれらの誘導体、例えばL−システイン−L−アルギニンジペプチド(CR)、L−アルギニン−L−システインジペプチド(RC)、L−システイン−L−ヒスチジン(CH)、グリシン−L−システイン−L−アルギニントリペプチド(GCR)、L−プロリン−L−システイン−L−アルギニントリペプチド(PCR)、L−グルタチオン(GSH)、グリシン−L−セリン−L−システイン−L−アルギニンテトラペプチド(GSCR)及びグリシン−L−システイン−L−セリン−L−アルギニンテトラペプチド(GCSR)並びにその他のチオール含有化合物、例えば1−[(2S)−2−メチル−3−チオール−1−オキソプロピル]−L−プロリン、チオグリコール酸、メルカプトエタノール、チオフェノール、D−3−トロロボール及びドデシルメルカプタンの1つ以上が含まれ、上記溶剤は、メタノール、酢酸エチル、水、エタノール、n−プロパノール、ペンタン、ギ酸、酢酸、ジエチルエーテル、アセトン、アニソール、1−プロパノール、2−プロパノール、1−ブタノール、2−ブタノール、ペンタノール、エタノール、酢酸ブチル、トリブチルメチルエーテル、酢酸イソプロピル、ジメチルスルホキシド、酢酸エチル、ギ酸エチル、酢酸イソブチル、酢酸メチル、2−メチル−1−プロパノール及び酢酸プロピルの1つ以上である;
(3)溶液Aと溶液Bとを、HAuCl4とリガンドYとの間のモル比が1:(0.01
〜100)であるように混合し、それらを氷浴中で0.1時間〜48時間にわたり撹拌し、0.025M〜0.8MのNaBH4の水溶液、エタノール溶液、又はメタノール溶液を添加し、氷水浴中で撹拌し続けて、0.1時間〜12時間にわたり反応させる工程と、
NaBH4とリガンドYとの間のモル比は1:(0.01〜100)である;
(4)MWCOが3K〜30Kの限外濾過チューブを使用して、上記反応溶液を8000回転/分〜17500回転/分で反応完了後10分間〜100分間にわたり勾配により遠心分離することで、リガンド修飾されたAuC沈降物を種々の平均粒度で得る工程(具体的な勾配遠心分離は、実施形態2の(4)に記載される通りである。種々のMWCOの限外濾過チューブのための濾過膜の開口が、その膜を通過し得るAuCのサイズを直接的に決定する)と、
この工程は省くことができる。言い換えれば、工程(3)の完了後に、工程(5)を直接開始することで、混合されたAuCが種々のサイズで得られる;
(5)工程(4)において得られた種々の平均粒度のAuC沈降物を水中に溶解させ、それを透析バッグに入れて、水中にて室温で1日間〜7日間にわたり透析する工程と、
(6)AuCを透析後12時間〜24時間にわたり凍結乾燥させることで、粉末状物質又は綿状物質、すなわちリガンド修飾されたAuCを得る工程と、
を含む、方法を開示している。
リガンドL−NIBCを例にとって、リガンドL−NIBCで修飾されたAuCの製造及び確認を詳説する。
(1)1.00gのHAuCl4を秤量し、それを100mLのメタノール中に溶解させることで、0.03Mの溶液Aを得る。
(2)0.57gのL−NIBCを秤量し、それを100mLの氷酢酸(酢酸)中に溶解させることで、0.03Mの溶液Bを得る。
(3)1mLの溶液Aを計量し、それをそれぞれ0.5mL、1mL、2mL、3mL、4mL、5mLの溶液B(すなわち、HAuCl4とL−NIBCとの間のモル比はそれぞれ1:0.5、1:1、1:2、1:3、1:4、1:5である)と混合し、氷浴中で撹拌しながら2時間にわたり反応させ、溶液が山吹色から無色に変わったら1mLの新たに調製した0.03M(11.3mgのNaBH4を秤量し、それを10mLのエタノール中に溶解させることにより調製される)のNaBH4水溶液を素早く添加し、溶液が暗褐色に変わった後30分間にわたりその反応を継続させ、そして10mLのアセトンを添加することで、その反応を止める。
(4)上記反応後に、その反応溶液を勾配遠心分離にかけることで、種々の粒度を有するL−NIBC修飾されたAuC粉末が得られる。具体的な方法:上記反応が完了した後に、その反応溶液を、MWCOが30Kの50mLの容量を有する限外濾過チューブに移し、10000回転/分で20分間にわたり遠心分離し、内側チューブ中の保持液を超純水中に溶解させることで、約2.6nmの粒度を有する粉末が得られる。次いで、外側チューブ中の混合された溶液を50mLの容量を有するMWCOが10Kの限外濾過チューブに移し、13000回転/分で30分間にわたり遠心分離する。その内側チューブ中の保持液を超純水中に溶解させることで、約1.8nmの粒度を有する粉末が得られる。次いで、外側チューブ中の混合された溶液を50mLの容量を有するMWCOが3Kの限外濾過チューブに移し、17500回転/分で40分間にわたり遠心分離する。その内側チューブ中の保持液を超純水中に溶解させることで、約1.1nmの粒度を有する粉末が得ら
れる。
(5)勾配遠心分離により得られた3つの異なる粒度の粉末を沈降させ、それぞれ溶剤を除去し、粗生成物をN2で風乾させ、それを5mLの超純水中で溶解させ、それを透析バッグ(MWCOは3KDaである)中に入れ、それを2Lの超純水中に入れ、1日おきに水を交換し、7日間にわたり透析させ、それを凍結乾燥させ、後で使用するために保持する。

試験粉末(実施形態2で製造されたL−NIBC修飾されたAuC試料及びL−NIBC修飾された金ナノ粒子試料)を、試料として超純水中に溶解させて2mg/Lとし、次いで試験試料をハンギングドロップ法によって製造した。具体的な方法:5μLの試料を超薄カーボンフィルム上に滴り落とし、水滴が消えるまで自然に蒸発させ、次いで該試料の形態を、JEM−2100F STEM/EDS電界放出型高分解能TEMにより観察した。
試験粉末を、濃度が10mg・L−1になるまで超純水中に溶解させ、室温でUV−vis吸収スペクトルにより測定した。スキャン範囲は、190nm〜1100nmであり、試料セルは、1cmの光路を有する標準的な石英キュベットであり、参照セルは、超純水で満たした。
約520nmに吸収ピークを有したことを示している。その吸収ピークの位置は、粒度と関連している。粒度が3.6nmである場合に、UV吸収ピークは516nmに現れ、粒度が6.0nmである場合に、UV吸収ピークは517nmに現れ、粒度が10.1nmである場合に、UV吸収ピークは520nmに現れ、そして粒度が18.2nmである場合に、その吸収ピークは523nmに現れる。上記4つの試料のいずれも、560nmより高いところに吸収ピークを一切有しない。
赤外スペクトルは、Bruker社により製造されたVERTEX80Vフーリエ変換赤外分光計において固体粉末の高真空の全反射モードで測定した。スキャン範囲は4000cm−1〜400cm−1であり、スキャン数は64である。実施形態2において製造されたL−NIBC修飾されたAuC試料を例にとると、該試験試料は、3つの異なる粒度を有するL−NIBC修飾されたAuC乾燥粉末であり、コントロール試料は、純粋なL−NIBC粉末であった。それらの結果を図3に示す。
消失と、560nmより高いところでの吸収ピークの出現を示す。この吸収ピークの位置は、リガンド及び粒度とともに僅かに変化するにすぎない。一方で、フーリエ変換赤外スペクトルはまた、リガンドのチオールの赤外吸収ピーク(図7〜図11のパネルBにおける点線の間)の消失を示すが、一方でその他の赤外の特徴的なピークは全て維持されるので、全てのリガンド分子はAuCの表面にうまくつながっており、かつ本発明では、表1中に列挙されるリガンドで修飾されたAuCを得ることに成功したことが示唆される。
この実施形態は、Aβの凝集動態のin vitro実験によりリガンド修飾されたAuCの機能を確認し、リガンド修飾された金ナノ粒子及びリガンド分子の独立した使用によるAβの凝集動態に対する効果を比較することで、その機能がリガンドではなくAuCに起因するものであることが分かった。それらの実験では、Aβ(1−40)の凝集及び線維化の動態を特性評価するためにThT蛍光標識法が使用された。
アミロイドポリペプチドAβ(1−40)の凍結乾燥粉末(Invitrogen Corp.社)を、ヘキサフルオロイソプロパノール(HFIP)中に溶解させることで、1g/LのAβ(1−40)溶液を得て、その溶液を、密封後に室温で2時間〜4時間にわたりインキュベートし、次いでHFIPを、高純度窒素(N2、99.9%)を用いて適切な流速でドラフトチャンバ内にて風乾(約1時間にわたり)させた。最後に、乾燥されたAβ(1−40)を200μLのDMSO中に溶解させ、密封後にその溶液を、後に使用するために1週間以内にわたり冷凍機中で−20℃に保った。使用前に、アミロイドポリペプチドのDMSO溶液を、Aβ(1−40)の濃度が20μMに達するまで、大量のリン酸緩衝溶液(PBS、10mM、pH=7.4)で希釈することで、Aβ(1−40)PBS溶液を得た。実験のための全てのAβ(1−40)PBS溶液は新たに調製された。
リガンド修飾されたAuC及び金ナノ粒子を、それぞれ20μMのAβ(1−40)PBSに添加することで、種々の濃度及び種々の粒度の、種々のリガンドで修飾されたAuCの試料を形成し、相応して種々のリガンドで修飾された金ナノ粒子の試料を形成した。それらの試料を、96ウェルプレート中で37℃においてThT蛍光標識法によって連続的にインキュベートし、蛍光強度をマイクロプレートリーダにより10分毎に1回モニタリングした。Aβ(1−40)凝集の動態過程を、ThTの蛍光強度の変化によって特性
評価した。
BC修飾された金ナノ粒子の添加がAβの凝集動態の成長段階及びプラットフォーム段階の両方を前倒すので(金ナノ粒子の濃度が20ppmである場合に、Aβの凝集動態の成長段階は第12時間目まで前倒され、そしてプラットフォーム段階は第16時間目まで前倒された)、L−NIBC修飾された金ナノ粒子はAβの凝集を加速させ得ることが示唆され(図5のパネルB及びパネルC)、金ナノ粒子の直径が6.0nm以下である場合には(図5のパネルD及びパネルE)、Aβの凝集の開始時間が遅延され得るので(L−NIBC修飾された金ナノ粒子の濃度が20ppmである場合に、Aβの凝集動態の成長段階は第54時間目まで遅延された)、金ナノ粒子はAβの凝集に対して阻害効果を有することが示唆されることを示している。しかしながら、図5は、上記濃度が非常に高い(20.0ppm)としても、L−NIBC修飾された金ナノ粒子の添加は完全に阻害する(すなわち、成長段階は現れず、蛍光曲線は完全に平坦にする)ことができなかったことを示している。他方で、L−NIBC修飾された金ナノ粒子の添加後に、ThTの蛍光発光ピークは515nmに位置している一方で、L−NIBC修飾された金ナノ粒子のプラズモン共鳴吸収ピークは520nm近くに位置するので、ここで観察されたThT蛍光強度の減少は、該金ナノ粒子のThT蛍光に対するプラズモン共鳴効果の部分的クエンチングであり、L−NIBC修飾された金ナノ粒子のAβ(1−40)凝集に対する阻害効果によるものではないはずである。
そしてより大きい金ナノ粒子は、Aβの凝集及び線維化の促進さえもするが、しかしながら、リガンド修飾されたAuCは、Aβの凝集及び線維化に対して優れた阻害効果を有し、かつその濃度が5ppm〜10ppmを上回る場合に、完全な阻害のために必要とされる最小濃度は、リガンド及びAuCの粒度に伴い僅かに変動するが、完全な阻害効果が達成され得る。同様に、これらのリガンド修飾されたAuCは、本発明において定義されるAuC含有物質に分類される。
細胞生存性を、この実施形態の実験における指標として使用した。CCK−8法の試験結果により、リガンド修飾されたAuC又は金ナノ粒子試料のAβ(1−40)の毒性に対する効果を反映し、リガンド修飾されたAuC又は金ナノ粒子が、アミロイドタンパク質のミスフォールディングの病因に対する神経保護効果を有するかどうかが示された。その実験で使用される細胞は、SH−SY5Y神経芽腫細胞系統であった。Aβ誘導されたAD細胞モデルは、文献(R. Liu, H. Barkhordarian, S. Emadi, C. B. Park, M. R. Sierks, Neurobiology of Disease 2005, 20, 74)での記載に従って樹立した。具体的な方法:
1)対数増殖期におけるSH−sy5y細胞(細胞は6世代まで継代培養した)を、完全培地(MEM+10%FBS+1%ペニシリン−ストレプトマイシン)で希釈して、5×104/mLの密度で細胞懸濁液を得た。その懸濁液を96ウェルプレートに1ウェル当たり200μLで接種し、5%CO2を有するインキュベーター中にて37℃で培養した。細胞がウェルに接着されたときに、試料を添加した。
実験1:
1)表1に列挙されるリガンドで修飾されたAuCをそれぞれ1.0g秤量し、水100mL中にストック溶液として溶解させ、後で使用するために4℃で貯蔵した。小容量のストック溶液を取り、使用前に水中で希釈した。
を凍結させ、切片にした。海馬内のAβアミロイド沈着の分布を、免疫組織化学法により調べた。

の試験システムは、円形プール及び自動型ビデオ及び分析システムからなっていた。プールの上方にあるカメラは、コンピュータに接続されていた(図13に示される)。水迷路は、直径120cm及び高さ60cmの円形プール、並びに直径9cmのプラットフォームからなっていた。液体の水準はプラットフォームより0.5cm高く、水温は22±0.5℃であった。白色色素を使用して、水を乳白色に染めた。該位置付け実験を使用することで、水迷路中のマウスの学習能力及び記憶能力を測定し、それを4日間にわたり継続した。図13に示されるように、水迷路を東(E)、西(W)、南(S)及び北(N)の4つの方向で十字形として4つの四分円に分けた。プラットフォームは、SW四分円の真ん中に位置しており、その位置は実験を通して固定した。トレーニング中に、マウスは、頭をプール壁に向けて内壁の近くで、異なる四分円において1/2ラジアンから優しく水中に入れた。マウスが隠れたプラットフォームに登るのに費やした時間(逃避潜時)を、カメラ追跡システムにより記録するか、又は記録時間が60秒間に達したら実験を止めた。マウスは、プラットフォーム上に登った後30秒間にわたりそのプラットフォーム上で
留まらせた。マウスが60秒間以内にプラットフォームを見つけられなかった場合に(この場合に逃避潜時は60秒として記録した)、実験者はそのマウスをプラットフォームに登るよう誘導し、30秒間にわたりプラットフォーム上で留まらせるものとする。実験後に、全てのマウスを取り出し、優しくぬぐって乾かした。それぞれのマウスは、4連日にわたり1日に4回、トレーニング期間の合間に15分〜20分の間隔を設けてトレーニングした。
鏡により計数した。各々の試料は、左心室及び右心室の両方を含む2つのスライスを並行試料として含んでいた。統計分析のために平均値を計算した。全てのデータをSPSSソフトウェア(SPSS 21)により処理し、それらのt検定又は1元配置分散分析を行った。P<0.05が、統計学的に有意差があることを意味した。

0%増加し(P<0.05)、大脳皮質において65±11.1%増加した(P<0.05)。モデルマウスがより高齢になるほど、記憶機能及び認知機能に対する影響はより大きくなるであろうことが示唆された。図17のパネルA、パネルB、及びパネルCは、それぞれ投与の第150日目における高用量群、低用量群、及びモデルコントロール群における海馬及び大脳皮質のAβ(1−40)の典型的な免疫組織化学的スライスの結果であった。図17のパネルDは、統計学的結果であった。それらの結果により、高用量群においては、Aβ(1−40)は、マウスの海馬及び大脳皮質の両方において明らかに減少し(海馬において59.0±11.1%(P<0.05)だけ減少し、かつ大脳皮質において36.4±4.5%(P<0.05)だけ減少した)、一方で、低用量での投与は、海馬においてAβ(1−40)斑の形成に対して大きな効果を有しなかったが(P>0.05)、大脳皮質においてはAβ(1−40)斑は大幅に減少した(26.9±2.1%(P<0.05)ことが示された。AuCは、第150日目にAβ(1−40)斑の形成に対して大幅な阻害効果を有することが示された。この効果はまた、用量依存的な関係性を示した。さらに、SPSSソフトウェアを使用して、150日目の水迷路実験におけるAβ(1−40)斑の数とプラットフォームを横切る回数との間の相関を分析した。その分析により、海馬及び大脳皮質中のAβ(1−40)斑の数が、プラットフォームを横切る回数と大きな負の相関を有することが明らかになった(海馬:R=−0.848、P<0.01、大脳皮質:R=−0.802、P<0.05)。さらに、この結果により、AuC投与により誘導された海馬及び大脳皮質中のAβ(1−40)斑の減少と、AuC投与により誘導されたマウスの記憶能力及び学習能力の改善との間の相関が確認された。
この実施形態は、in vitroでのα−synの凝集動態実験によりリガンド修飾されたAuCの機能を確認し、それを、リガンド分子を単独で使用した場合のそのα−synの凝集動態に対する効果と比較し、こうしてその機能がリガンドではなくAuCに起因するものであることが実証された。
ある。ThTをポリペプチド又はタンパク質の単量体と一緒にインキュベートする場合に、その蛍光はそれほど変化しない。ThTが線維構造を有するアミロイドポリペプチド又はタンパク質に出くわすと、そのアミロイドポリペプチド又はタンパク質と直ちに結合し、その蛍光強度は指数関数的に増加することとなる。この理由のため、ThTは、ペプチド又はタンパク質のアミロイドーシスをモニタリングするためのマーカーとして広く使用されている。この実施形態は、AuCの存在下でのα−synの線維化凝集の動態的過程をモニタリングするためにThT蛍光標識法を利用する。具体的な実験方法は、以下の通りであった。
、GSH、N−アセチル−L−システイン(L−NAC)、L−システイン(L−Cys)及びD−システイン(D−Cys)で修飾されたAuC(用量は全て10ppmであった)の、α−synの凝集及び線維化に対する阻害効果が示された。種々のリガンドで修飾されたAuCについても同様の現象が観察され、同じ結論に達し得た。これらのリガンド自体は、α−synの凝集及び線維化に影響を及ぼし得ないが、リガンド修飾されたAuCは、α−synの凝集及び線維化に対して優れた阻害効果を有した。その濃度が10ppmに達した場合には、完全な阻害効果を全てが達成し得た。完全な阻害のために必要とされる最小濃度は、種々のリガンドで僅かに変動する。同様にこれらのリガンド修飾されたAuCは、本発明において定義されるAuC含有物質に分類された。表1に列挙されるその他のAuCは、同様の効果を有していた。α−synの凝集及び線維化の完全な阻害のために必要とされるAuCの濃度だけは異なっていた。それは詳細に記載しないこととする。
実験1:

PC12細胞のMPP+(100mM)誘導されたアポトーシスのモデルを、フローサイトメトリー技術と組み合わせて使用して、この実験においてAuCのMPP+誘導された細胞の傷害及びアポトーシスに対する保護効果を観察した。具体的な方法:ブランクコントロール群は、MPP+及びAuCを添加しない群であり、MPP+モデル群は、MPP+のみを添加した群であり、AuCコントロール群は、AuCのみを添加した群であり、そして実験群は、MPP+及びAuCの両方を添加した群であった。実験群において、1.8nmの平均粒度を有するL−NIBC修飾されたAuCの溶液を、PC12細胞懸濁液(AuCの最終濃度は20ppmであった)に添加し、半時間後にMPP+を添加し、その混合物を24時間にわたりインキュベートし、Annexin V−FITC/PI細胞アポトーシス検出キット(Roche社から購入)及びFACSCaliburフロー
サイトメーターを使用して、細胞の増殖活性及びアポトーシスを検出し、そのデータを取得し、CellQuest Proにより分析した。
実験1:
実験動物:8週齢の体重25g〜30gの80匹のC57bl/6雄マウス;各々のケージ中の3匹のマウスを全て、室温22℃〜27℃の環境下で、12時間の概日リズムで、飲食を自由にして飼育し、7日間にわたり馴化させた。
0匹のマウスで無作為に分けた。MPTPモデル群及びAuC処置群において、20mg/kg(遊離塩基)MPTPを、2時間毎に1回で4回にわたり皮下注射した。ブランクコントロール群においては、20mg/kgの生理食塩水を、2時間毎に1回で4回にわたり皮下注射した。最後の注射より8時間後に、ブランクコントロール群及びMPTPモデル群において、10μLの生理食塩水を毎日静脈注射し、一方で、AuC正常コントロール群及びAuC処置群においては、表1中に列挙されるリガンド修飾されたAuCの10μLの生理食塩溶液(AuC濃度は10g/L)を、それぞれ毎日腹腔内注射した。上記注射は、7日間にわたり継続した。それらの動物を、清浄なパディングを有する給餌ボックス中に入れ、水と食料とを自由に与えた。

1回で7日間にわたりそれぞれ連続的に腹腔内注射した。ブランクコントロール群においては、30mg/kgの生理食塩水を、1日に1回で7日間にわたり連続的に皮下注射した。低用量AuCコントロール群及び低用量AuC処置群においては、1.8nmの平均粒度を有する1g/LのL−NIBC修飾されたAuCの100μLの生理食塩溶液を、それぞれ1日に1回で腹腔内注射し、一方で、高用量AuCコントロール群及び高用量AuC処置群においては、1.8nmの平均粒度を有する4g/LのL−NIBC修飾されたAuCの100μLの生理食塩溶液を、それぞれ1日に1回で7日間にわたり連続的に腹腔内注射した。それらの動物を、清浄なパディングを有する給餌ボックス中に入れ、水と食料とを自由に与えた。
(1)自発運動試験:動物をケージから自発運動検出装置に移した。それらの動物を新しい環境に5分間にわたり順応させた後に、それらの自発運動及び変化を5分間内で記録した。それらの動物の5分間内での運動距離及び移動速度を用いて、動物の運動量を測定した。

ウスをモリスの水槽中に入れた。水の深さは60cmであり、温度は22℃であった。それらの動物の10分間内での遊泳距離及び遊泳時間を記録して、それらの動物の運動量を測定した。
行動試験の後に、各々の群内の5匹のマウスを取り出し、黒質及び線条体における免疫組織化学的検出を行った。1mLの0.5%のペントバルビタールナトリウムにより腹部麻酔をした後に、開胸し、15mLの0.9%の生理食塩水を使用して、最初に大動脈からの血液をすすぎ、次いで4%のパラホルムアルデヒドを含む100mLの0.1mol/Lのリン酸緩衝溶液(PBS、pH7.2)を使用して、灌流(最初は速く、その後ゆっくり)及び固定を1時間にわたり行った。灌流固定の後に脳を取り出し、4%のパラホルムアルデヒド中に入れ、パラフィンで包埋し、マウスの脳マップを参照して黒質及び線条体を冠状断面で切片にした。脳スライスの厚さは、3μm/切片であった。得られた脳スライスを、免疫蛍光による超高感度の2段階免疫組織化学法及びその他の実験において使用した。免疫化学的染色工程は、以下の通りであった。得られた脳スライスを、30分間にわたり0.3%のH2O2メタノール溶液(30%H2O2 1mL+メタノール 80mL+PBS 19mL)中に入れ、30分間にわたり0.3%のTriton X−100 PBS中に入れ、マウス抗チロシンヒドロキシラーゼ(TH)モノクローナル抗体(1:200)又はIBa1(希釈比1:250)中に浸漬させ、48時間(4℃)にわたりインキュベートし、ビオチニル化ウサギ抗マウス二次抗体(1:500)中に浸漬させ、2時間(室温)にわたりインキュベートした。それらを蒸留水で素早くすすぎ、20分間〜30分間にわたる硫酸ニッケルアンモニウムで増感されたDAB青色反応法により発色させた。陽性の生成物が暗青色で、バックグラウンドが澄明となったら、それらの脳スライスを蒸留水で3回にわたりすすいで、発色を止めた。上記工程のそれぞれの後に、上記脳スライスは、0.01mol/LのPBSにより3回にわたり毎回10分間に
わたってすすぐべきである。そしてここで使用された一次抗体は、1%ウシ血清及び0.3%Triton X−100を含むPBSで希釈され、二次抗体及びABCの複合体は、PBSで希釈された。脳スライスの付着、脱水、及び透明な中性ガムでの封止を行った。
チロシンヒドロキシラーゼ(TH)は、ドーパミン(DA)の生合成経路における主要な酵素であり、THの免疫組織化学法は、黒質及び線条体中のDA作動性ニューロンにおいて変化を示す(D. Luo, J. Zhao, Y. Cheng, S. M. Lee, J. Rong, Molecular Neurobiology 2017, DOI: 10.1007/s12035-017-0486-6)。行動試験の後に、各々の群から5匹のマウスを取り出し、線条体のWB検出を行い、必要とされる脳組織を氷上に取り出し、RIPA溶解物中で破砕し、4℃及び12000gで30分間にわたり遠心分離して均質化し、タンパク質を抽出して試料を調製し、SDS−ポリアクリルアミドゲル電気泳動を55V〜60Vで4.5時間にわたり行い、そしてタンパク質を、セミドライ法により60mAの一定電流で約1.5時間にわたりメンブレンに転写させた。上記メンブレンを、5%のスキムミルクを用いて室温で1時間にわたりブロッキングし、TBST希釈されたウサギTH抗体(希釈比1:300)を添加し、4℃で一晩静置し、抗体を回収し、そのメンブレンをTBSTで3回にわたり毎回10分間洗浄し、TBST希釈されたIRDye
R 680RDヤギ抗ウサギ(希釈比1:3000)を添加し、そのメンブレンをTBSTで3回にわたり毎回10分間洗浄し、タンパク質シグナルを、2色赤外線レーザーイメージングシステムによりスキャンした。
1. SH−sy5y細胞系統を、AuC含有物質のバイオセーフティを細胞レベルで評価するために採用した。
具体的な方法:表1に列挙される種々のリガンド修飾されたAuC(実施形態2における1.8nmの平均直径を有するL−NIBC修飾されたAuCを例にとる)に関して、60匹の成体マウスを選び、各々の群内に15匹のマウスで、1つのコントロール群及び3つの実験群である4つの群に分けた。コントロール群においては、マウスには通常の給餌を行ったが、3つの実験群においては、マウスには、経口投与(強制栄養により)により、それぞれ1日につき0.1g/体重kg、0.3g/体重kg、及び1g/体重kgの用量で通常の食餌条件下で1週間にわたりAuCを供給した。AuCの供給が終わった後に、それらのマウスには30日間にわたり通常の給餌を行った。マウスの異常な応答を観察した。
実験1:
作業工程:80匹のマウスを、各々の群内に20匹のマウスで4つの群に無作為に分け、表1に列挙されるリガンド修飾されたAuCを経口投与(強制栄養により)により上記群においてそれぞれ100mg/kg、20mg/kg、5mg/kg、及び1mg/kgの用量で供給した。AuCを供給した後で、各々の群内の20匹のマウスを、各小群内に5匹のマウスを有する4つの小群に無作為に分けた。それらのマウスを、供給後のそれぞれ2時間、6時間、24時間、及び48時間の時点で屠殺した。心臓、肝臓、脾臓、肺、腎臓、及び脳の組織を別々に採取した。各組織を秤量し、2mLの水を添加して均質化し、次いで2mLの王水を添加し、ボルテックス混合し、振動装置上で72時間にわたり振動させた。2重量%の硝酸溶液を添加して10mLの最終容量にし、15000rpmで15分間にわたり遠心分離した。4mLの上清を吸引し、組織液中の金元素の含量を、原子吸光分光法により測定した。

含量は、100ppmの用量での検体を除き、検出限界を下回るまで減少した。上記の結果により、AuC含有物質が動物レベルでも良好なバイオセーフティを有し、血液−脳関門を通過することができ、体内での明らかな蓄積を有しないことが示され、こうしてAD又はPDを治療する医薬の製造での用途において良好な展望を有した。

- 前記AuCの金コア直径は、3nm未満、好ましくは0.5nm〜2.6nmである、請求項1に記載の物質。
- 前記リガンドYは、限定されるものではないが、L(D)−システイン及びその誘導体、システイン含有オリゴペプチド及びそれらの誘導体、並びにその他のチオール含有化合物の1つ以上を含む、請求項1又は2に記載の物質。
- 前記L(D)−システイン及びその誘導体は、好ましくはL(D)−システイン、N−イソブチリル−L(D)−システイン(L(D)−NIBC)、又はN−アセチル−L(D)−システイン(L(D)−NAC)等である、請求項3に記載の物質。
- 前記システイン含有オリゴペプチド及びそれらの誘導体は、好ましくはシステイン含有ジペプチド、システイン含有トリペプチド、又はシステイン含有テトラペプチドである、請求項3に記載の物質。
- 前記システイン含有ジペプチドは、好ましくはL−システイン−L−アルギニンジペプチド(CR)、L−アルギニン−L−システインジペプチド(RC)、L−ヒスチジン−L−システインジペプチド(HC)、又はL−システイン−L−ヒスチジンジペプチド(CH)等である、請求項5に記載の物質。
- 前記システイン含有トリペプチドは、好ましくはグリシン−L−システイン−L−アルギニントリペプチド(GCR)、L−プロリン−L−システイン−L−アルギニントリペプチド(PCR)、L−リジン−L−システイン−L−プロリントリペプチド(KCP)、又はL−グルタチオン(GSH)等である、請求項5に記載の物質。
- 前記システイン含有テトラペプチドは、好ましくはグリシン−L−セリン−L−システイン−L−アルギニンテトラペプチド(GSCR)、又はグリシン−L−システイン−L−セリン−L−アルギニンテトラペプチド(GCSR)等である、請求項5に記載の物質。
- 前記その他のチオール含有化合物は、好ましくは1−[(2S)−2−メチル−3−チオール−1−オキソプロピル]−L−プロリン、チオグリコール酸、メルカプトエタノール、チオフェノール、D−3−トロロボール、N−(2−メルカプトプロピオニル)−グリシン、又はドデシルメルカプタン等である、請求項3に記載の物質。
- 前記物質は、粉末又は綿状である、請求項1〜9のいずれか一項に記載の物質。
- 請求項1〜10のいずれか一項に記載のAuC含有物質の製造方法であって、
以下の工程:
(1)HAuCl4をメタノール、水、エタノール、n−プロパノール、及び酢酸エチルの1つに溶解させることで、HAuCl4の濃度が0.01M〜0.03Mである溶液Aを得る工程と、
(2)リガンドYを溶剤中に溶解させることで、リガンドYの濃度が0.01M〜0.18Mである溶液Bを得る工程と、
(3)工程(1)における溶液Aと工程(2)における溶液Bとを、HAuCl4とリガンドYとの間のモル比1:(0.01〜100)(好ましくは1:(0.1〜10)、よ
り好ましくは1:(1〜10))で混合し、その混合物を氷浴中で0.1時間〜48時間(好ましくは0.1時間〜24時間、より好ましくは0.5時間〜2時間)にわたり撹拌し、0.025M〜0.8MのNaBH4溶液(好ましくはNaBH4の水溶液、NaBH4のエタノール溶液、又はNaBH4のメタノール溶液)を添加し、次いでその反応系を、氷浴中で0.1時間〜12時間(好ましくは0.1時間〜2時間、より好ましくは1時間〜2時間)にわたりNaBH4とリガンドYとのモル比1:(0.01〜100)(好ましくは1:(0.1〜8)、より好ましくは1:(1〜8))で撹拌する工程と、
(4)工程(3)における反応溶液を8000回転/分〜17500回転/分で10分間〜100分間にわたり遠心分離することで、種々の平均粒度を有するAuC沈降物を得る工程、好ましくはMWCOが3K〜30Kのチューブ型限外濾過器を使用して、工程(3)における反応溶液を8000回転/分〜17500回転/分で10分間〜100分間にわたり勾配により遠心分離することで、種々の平均粒度を有するAuCを得る工程と、
(5)工程(4)において得られた種々の平均粒度を有するAuC沈降物を水中に溶解させ、それを透析バッグに入れて、水中にて室温で1日間〜7日間にわたり透析する工程と、
(6)前記透析バッグ中のAuC溶液を12時間〜24時間にわたり凍結乾燥させ、AuC含有物質を得る工程と、
を含む、方法。 - 工程(2)における溶剤は、メタノール、酢酸エチル、水、エタノール、n−プロパノール、ペンタン、ギ酸、酢酸、ジエチルエーテル、アセトン、アニソール、1−プロパノール、2−プロパノール、1−ブタノール、2−ブタノール、ペンタノール、エタノール、酢酸ブチル、トリブチルメチルエーテル、酢酸イソプロピル、ジメチルスルホキシド、酢酸エチル、ギ酸エチル、酢酸イソブチル、酢酸メチル、2−メチル−1−プロパノール、及び酢酸プロピルの1つ以上である、請求項11に記載の方法。
- 触媒製造又は分子触媒作用、キラル認識、分子検出、生物医学的検出、及びイメージングの分野における近赤外蛍光プローブへの請求項1〜10のいずれか一項に記載のAuC含有物質の使用。
- Aβの凝集及び線維化、及び/又はα−synの凝集及び線維化と関連する疾患のための医薬の製造における請求項1〜10のいずれか一項に記載のAuC含有物質の使用。
- ADの予防及び治療のための医薬の製造における請求項1〜10のいずれか一項に記載のAuC含有物質の使用。
- PDの予防及び治療のための医薬の製造における請求項1〜10のいずれか一項に記載のAuC含有物質の使用。
- Aβの凝集及び線維化と関連する疾患のための医薬の製造におけるAuCの使用。
- 前記Aβの凝集及び線維化と関連する疾患は、ADである、請求項17に記載の使用。
- 前記AuCは、L−グルタチオン(GSH)、N−アセチル−L(D)−システイン(L(D)−NAC)、N−イソブチリル−L(D)−システイン(L(D)−NIBC)、L−システイン−L−アルギニンジペプチド(CR)、L−アルギニン−L−システインジペプチド(RC)、1−[(2S)−2−メチル−3−チオール−1−オキソプロピル]−L−プロリン(Cap)、又はL(D)−システイン(L(D)−Cys)等で修飾されている、請求項17又は18に記載の使用。
- α−synの凝集及び線維化と関連する疾患のための医薬の製造における、AuCの使用。
- 前記Aβの凝集及び線維化と関連する疾患は、PDである、請求項20に記載の使用。
- 前記AuCは、L−グルタチオン(GSH)、N−アセチル−L(D)−システイン(L(D)−NAC)、N−イソブチリル−L(D)−システイン(L(D)−NIBC)、L−システイン−L−アルギニンジペプチド(CR)、L−アルギニン−L−システインジペプチド(RC)、1−[(2S)−2−メチル−3−チオール−1−オキソプロピル]−L−プロリン(Cap)、又はL(D)−システイン(L(D)−Cys)等で修飾されている、請求項20又は21に記載の使用。
