本実施例は、多様なマウス異種移植片モデルの、2つの異なる薬物対抗体比(DAR)における、本開示のAADCである、抗CD71 TF02.13−2011−vc−MMAEの有効性を示す。

本実施例は、本開示の活性化可能抗CD71コンジュゲート抗体が、抗体依存性細胞媒介性細胞傷害作用(ADCC)及び補体依存性細胞傷害作用(CDC)など、エフェクター機能ベースの機構を媒介する能力を裏付けた。
表面プラズモン共鳴(Biacore)により、これらの、抗CD71 TF02.13−2011−vc−MMAE、抗CD71 Ab21.12(IgG1/κ)、AB095−MMAE−E2及びAB095(IgG1/κ)への結合について評価するのに、6×Hisタグを介して、組換えヒトFcγRII及びFcγRIIIを捕捉した。製造元のプロトコールに従うアミンカップリングにより、マウス抗6×His抗体を、CM5チップ上に、10000RUの密度まで、直接固定化させた。次いで、ヒトFcγRを、フローセル2、3及び4において捕捉して、250〜500RUの、FcγRの捕捉レベルを達成した。フローセル1を、基準表面として使用した。HBS−EP+緩衝液(GE、Healthcare)を、ランニング緩衝液として使用した。抗CD71 TF02.13−2011−vc−MMAE E2、抗CD71 Ab21.12(IgG1/κ)、AB095−MMAE−E2、AB095(IgG1/κ)及びトラスツズマブ(IgG1/κ)を、全てのフローセル上に、50μL/分の流速、FcγRIIについて、46.9〜12000nM、FcγRIIIについて、7.8〜4000nMの範囲の濃度(両方とも、2倍の系列希釈液)で、1〜2分間にわたり(FcγRIIb及びFcγRIIa H131については1分間にわたり、FcγRIIIa F158及びFcγRIIIa V158については2分間にわたり)注入した後、1〜5分間の(FcγRIIb、FcγRIIa H131については1分間にわたり、FcγRIIIa F158及びFcγRIIIa V158については5分間にわたる)解離時間を施した。チップ表面は、4つ全てのフローセル上、100μL/分の流速において、2秒間にわたる、100mMのHClの注入により再生した。全ての種類の実験誤差(計器、表面及び作業者)に対処するように、3つの異なるCM5チップの使用による、3つの実験を、各試料について(各々、2回繰り返して)行った。これらの3つの実験の結果を平均した。
抗CD71 TF02.13−2011−vc−MMAE E2は、ヒトIgG1/κアイソタイプFcを含有する。抗CD71 TF02.13−2011−vc−MMAE E2が、FcγRI、FcγIIa、FcγIIb、FcγIIIに結合することが予測されたことは、他の野生型IgG1/κ抗体と符合した。抗CD71 TF02.13−2011−vc−MMAE E2の、FcγRへの結合は、表面プラズモン共鳴、FcγRI競合結合ELISA(結果は示されていない)及びFcγRIII競合結合ELISA(結果は示されていない)により解析し、抗CD71 TF02.13−2011−vc−MMAE E2の、補体への結合は、ヒト及びカニクイザル補体の機能的活性化アッセイ(結果は示されていない)により解析した。これらのアッセイは、市販のヒト抗体及び社内において作出したヒト抗体をベンチマークとした。トラスツズマブの素材である、臨床グレードのヒト抗HER2 IgG1/κを、FcγR結合アッセイのための陽性対照として使用した。Fc受容体及び補体は、野生型IgG1抗体に結合することが公知である(Ravetch;Roopenian;Burtonら、Nature.1980年;288:338〜344頁;Hughes−Jonesら、Mol Immunol.、1979年;16:697〜701頁;及びHezarehら、J Virol.、2001年;75:12161〜12168頁)。抗CD71 TF02.13−2011−vc−MMAEについての検討は、それが、FcγRI、FcγIIa H131、FcγRIII及びFcRnに結合するが、陽性対照であるトラスツズマブと比較した場合、FcγRIIIへの結合のわずかな低減(SPRによる)が観察されることを明らかにした。抗CD71 TF02.13−2011−vc−MMAE E2の、FcγRIIbへの結合は、Biacore計器の検出下限である、1.0×105のKDを下回ったため、微弱過ぎて、決定できなかった(表10A及び10Bを参照されたい)。
本実施例は、抗CD71−TF02.13−2011−vcMMAE E2は、ヒト又はカニクイザルの末梢血単核細胞(PBMC)に結合しないが、IgG1抗体は、末梢血リンパ球上のFcγ受容体を架橋し、これにより、サイトカインの誘導を含む、様々な機能を媒介することが可能であることを示した。
本実施例は、SW−48細胞(ヒト結腸直腸がん)を使用する、マウス腫瘍異種移植片モデルにおける、本開示の活性化抗CD71コンジュゲート抗体の有効性を示す。
本実施例は、雌SCIDマウス又は雌NOGマウスにおける、HuPrime患者由来びまん性大細胞型B細胞リンパ腫(DLBCL)を使用する、5匹のマウスによる、患者由来異種移植片(PDX)腫瘍モデルにおける、本開示の活性化抗CD71コンジュゲート抗体の有効性を示す。
本実施例は、ヌードマウスにおいて、患者由来ヒト乳がん(CTG−0437、CTG−0869、CTG−1059)、非小細胞肺がん(NSCLC)(CTG−1082、CTG−0860、CTG−0160、CTG−1012)及び頭頸部小細胞癌(HNSCC)(CTG−1140、CTG−1082)の腫瘍移植片を使用する、9匹のマウスによる、患者由来異種移植片(PDX)腫瘍モデルにおける、本開示の活性化抗CD71コンジュゲート抗体の有効性を示す。
本実施例は、SCIDマウスにおいて、HuPrime患者由来ヒト膵臓がん(PA6237)を使用する、患者由来異種移植片(PDX)マウスモデルにおける、本開示の活性化抗CD71コンジュゲート抗体の有効性を示す。
本実施例は、3週間ごと(Q3W)又は2週間ごと(Q2W)のIVボーラス注射の後の、カニクイザル(群1つ当たり性別当たりのサル1〜2匹)における、6〜18mg/kgの範囲の投与レベルにおける、本開示の活性化可能抗CD71コンジュゲート抗体(AADC)である、抗CD71−TF02.13−2011−vcMMAE E2の、0.6〜6mg/kgの範囲の投与レベルにおける、抗CD71コンジュゲート抗体(ADC)である、Ab21.12−vcMMAE E2に対する相対毒性を示す。終了時における剖検は、29日目に(最終回投与の7日後に)、被験動物に対して実施した。研究時の評価は、臨床徴候、体重、飼料摂取、臨床病理学、解剖病理学、TK及び免疫原性を含んだ。パイロット毒性研究の概要を、表13に示す。
本実施例は、IVボーラス注射を介する、1日目及び22日目の、0、2、6又は12mg/kgの投与レベルにおける、本開示の活性化可能抗CD71コンジュゲート抗体(AADC)である、抗CD71−TF02.13−2011−vcMMAE E2についての、カニクイザルにおける、GLP反復投与毒性研究を示す。投与群を、表14に示している。
本実施例は、DARを、約3へと強化するのに使用される方法に加えて、vc−MMAEを、活性化可能抗CD71抗体である、抗CD71 TF02.13−2011へとコンジュゲートさせて、各抗CD71 TF02.13−2011分子につき、2つずつのMMAE分子による薬物ロードを含有する、抗CD71 TF02.13−2011−vc−MMAEを創出するのに使用される方法について概括する。
E2強化型抗CD71 TF02.13−2011−vc−MMAE:抗CD71 TF02.13−2011の溶液(13.95mg/mL)、39gの溶液、544mg、3.5ナノモル)に、0.5MのEDTA(0.39mL)及びTCEP(0.77mL、WFI中に10mM、7.7ナノモル、2.2当量)を、2〜8℃において、一晩にわたり添加した。次いで、還元された抗体を、DMSO(2.29mL)により希釈し、DMSO(17.9ナノモルのDMSO中に、9.49mMのvcMMAEが1.89mL、5.1当量)中のvcMMAEにより、室温において、1時間にわたり処理した。コンジュゲート抗体を、N−アセチルシステイン(29ナノモルのWFI中に、100mMが0.29mL、8.2当量)により処理して、過剰なvcMMAEをクエンチングした。クエンチングした溶液を、HIC(疎水性相互作用クロマトグラフィー)により、>E4のDAR分子種(高DAR分子種、上記の調製物中に20.9%)について解析した。
コンジュゲーション反応物を、東ソー株式会社から市販されているHIC樹脂(例としては、Toyopearlアルキル)などのHIC樹脂の一部と共に、室温において、2時間にわたり懸濁させ、高DAR分子種を、標的レベル(一般に<2%)へと低減した。処理された溶液を濾過し、樹脂を、PBSE(125mMのリン酸カリウム、150mMのNaCl、6.3mMのEDTA、pH7.2、22g)に続いて、PBS(5mMのリン酸カリウム、200mMのNaCl、pH7、35g)により、逐次的に洗浄した。濾液とすすぎ液との組合せを濃縮し、分子量カットオフを30kDとする遠心分離機フィルターを使用して、緩衝液交換し、次いで、保管のための製剤成分により希釈した。
本実施例は、DARを約2へと強化するのに使用される方法に加えて、vc−MMAEを、活性化可能抗CD71抗体である、抗CD71 TF02.13−2011へとコンジュゲートさせて、各抗CD71 TF02.13−2011分子につき、2つずつのMMAE分子による薬物ロードを含有する、抗CD71 TF02.13−2011−vc−MMAEを創出するのに使用される方法について概括する。
抗CD71 TF02.13−2011−vc−MMAE E2:抗CD71 TF02.13−2011(20.3mg/mL)の溶液を、1457gのPBSE(125mMのリン酸カリウム、150mMのNaCl、6.3mMのEDTA、pH7.7)により希釈し、4℃へと冷却した。冷却された溶液に、TCEP(WFI中に10mM、30.2mL、0.30ミリモル、1.5当量)を添加し、4℃において、15時間にわたり維持した。還元された抗体に、vc−MMAE(1.056g、0.80ミリモル、4当量)の、DMSO(300g)中溶液を、冷却しながら添加した後、DMSOすすぎ液(65g)を添加し、この後、溶液を、室温へと暖めた。3.5時間後、N−アセチルシステイン(0.262g、1.60ミリモル、8当量)を添加して、過剰なvc−MMAEをクエンチングし、反応混合物を、精製まで、2〜8℃において保管した。
生成物の一部は、UVによる、280nmにおけるモニタリングを伴う、pH7.1の硫酸アンモニウム/リン酸緩衝液による勾配を使用する、GE Life Sciences(アルキルSepharose)から市販されているHIC精製カラムなど、市販のHIC精製カラム上において精製した。Pellicon3 Ultracel 30kD膜を使用して、回収されたDAR2生成物プール(約9.5g、30%の収率)を、pH6.0において、TFFにより、緩衝液と緩衝液交換し、次いで、保管のための製剤成分により希釈した。
本明細書において概括された、標準的な緩衝液及び手順を使用して、HICにより精製されていない、実施例25に記載された方法から得られたコンジュゲート生成物を、HICカラムにかけた。図18Aに示されている通り、薬物ロードを0〜8とする、多数の分子種を同定した。
