Abstract

本発明がより容易に理解されるように、まず或る特定の用語を以下に定義する。以下の用語及び他の用語に対する更なる定義は、本明細書を通して記載される。
キメラポリペプチド又は融合ポリペプチドを作製する(例えば、設計、操作する)手段を提供する組成物が本明細書に記載される。本明細書に記載されるペプチド(例えば、リンカー)配列は、融合タンパク質の適切な発現、フォールディング及び活性を可能にする。本発明は、とりわけ、新規ポリペプチド(例えば、リンカー)及びそれを含む融合タンパク質を提供する。幾つかの実施形態では、本発明は、本明細書に記載されるペプチド(例えば、リンカー、タグ)又は融合タンパク質をコードするポリヌクレオチド組成物を提供する。幾つかの実施形態では、本発明は、ペプチド(例えば、リンカー、タグ)又は融合タンパク質をコードするポリヌクレオチドを含む発現ベクターを提供する。他の実施形態では、本発明は、ペプチド(例えば、リンカー、タグ)又は融合タンパク質をコードするポリヌクレオチド及び/又は発現ベクターを含む細胞を提供する。本明細書に記載されているのは、ペプチドリンカーを含む新規組成物、ペプチドリンカーによって結合されたポリペプチドを含むポリペプチド組成物、並びに関連するポリヌクレオチド、ベクター、細胞及び医薬組成物である。幾つかの実施形態では、ペプチド(例えば、リンカー、タグ)は、1つ以上のポリペプチドに融合される。記載されるリンカー配列は、リンカーによって接続されたポリペプチドの発現及び活性(例えば、抗原結合)が永続的かつ最適であるように、目的の2つのペプチド/ポリペプチドを制御可能に結合する。ペプチドリンカー又はタグは、C末端、N末端、又はポリペプチド内のどこにでも融合して、所望の機能を達成し得る。
コンセンサス配列XYPXXXZXを含む本明細書に記載される新規キメラポリペプチドリンカーは、治療的処置に有用な融合タンパク質への組み込みに適した望ましい属性を併せ持つ。1つの態様においては、本発明は、8個〜20個のアミノ酸とコンセンサス配列XYPXXXZXとを含むリンカーを提供し、ここで、Xはグリシン(G)又はセリン(S)であり、Bは正電荷アミノ酸であり、Zはグリシン(G)又は負電荷アミノ酸である。本発明者らは、コンセンサス配列におけるアミノ酸残基の間隔と電荷の両方が、リンカー配列の抗体認識に加えて、リンカーの機能性に寄与することを発見した。

1つの態様においては、本発明は、第1のポリペプチドと、第2のポリペプチドと、本明細書に記載されるリンカーとを含む融合タンパク質を提供する。ポリペプチド組成物及び該ポリペプチド組成物をコードするポリヌクレオチドが本明細書に記載されており、ポリペプチド組成物は、本明細書に開示されるリンカー配列によって接続された第1及び第2のペプチド/ポリペプチドを含む。本発明者らは、驚くべきことに、本発明によるリンカーが、第1及び第2のペプチドの最適な柔軟性と長さの両方を提供して、立体障害を回避し、正しいフォールディングを可能にすることを見出した。
幾つかの態様において、融合タンパク質は抗原結合分子である。幾つかの実施形態では、第1のポリペプチドは軽鎖可変ドメインであり、第2のポリペプチドは重鎖可変ドメインである。幾つかの実施形態では、抗体の重鎖及び軽鎖可変領域を結合するための本明細書に記載されるリンカーの使用は、立体障害を回避し、抗原結合ドメインの正しいフォールディングを可能にする最適な柔軟性及び長さを可能にするという利点を提供する。第1及び第2のペプチドの適切な立体配座は、抗原の認識及び結合に不可欠である。
抗原結合分子は、CAR又はTCRのコンポーネントを形成し得て、CAR又はTCRに目的の標的を認識するように指示する役割を果たす場合がある。CAR又はTCRの文脈に関連して、本明細書で使用される場合、抗原結合分子は、CAR又はTCRを所望の標的に向け、その標的と会合するCAR又はTCRの任意のコンポーネントを意味する。特定の実施形態では、CAR又はTCRの抗原結合分子コンポーネントは、本明細書に記載されるリンカーによって結合された重鎖及び軽鎖の可変領域を含むscFvを含む。重鎖及び軽鎖の可変領域は、同じ抗体又は2つの異なる抗体に由来し得る。CAR又はTCRで使用される抗原結合分子は、目的の標的に結合することが知られているか又はそれが疑われる抗体に由来し得る。
1つの態様において、本発明は、本明細書に記載されるリンカーをコードするポリヌクレオチドを提供する。幾つかの実施形態では、本発明は、本明細書に記載される融合タンパク質をコードするポリヌクレオチドを提供する。本開示はまた、本明細書に記載されるリンカーを含む、抗体及びscFv等の抗原結合分子をコードするポリヌクレオチド、この配列を含む分子、並びにかかる分子を提示する細胞に関する。

幾つかの実施形態では、本発明は、本明細書に記載されるリンカー又は融合タンパク質をコードするポリヌクレオチドを含む組み換え細胞を提供する。幾つかの実施形態では、組み換え細胞は、本明細書に記載されるリンカー又は融合タンパク質をコードするポリヌクレオチドを含む発現ベクターを含む。幾つかの態様においては、本開示のポリヌクレオチド又はベクターを含む細胞が本明細書で提供される。幾つかの実施形態では、本開示は、本明細書に記載されるリンカー又は融合タンパク質をコードするポリヌクレオチドを含むin vitro細胞等の宿主細胞に関する。幾つかの実施形態では、本開示は、本明細書に開示される抗体又はその抗原結合分子をコードするポリヌクレオチドを含む宿主細胞、例えばin vitro細胞に関する。
GSTSGSGKPGSGEGSTKG(配列番号1)のリンカー配列を含むCARに対して生じた抗体クローン8及びクローン16の特異的結合を、完全長の配列番号1、及びN末端又はC末端のいずれかで切断され、N末端にビオチン部分又はC末端にビオチン部分を持つリジン残基のいずれかを含むバリアント(配列番号21〜配列番号30)のエピトープマッピングELISA実験に使用した。
GSTSGSGKPGSGEGSTKG(配列番号1)のリンカー配列、リンカー配列GGGGSGGGGSGGGGS(配列番号2)、又は抗CD19 scFvクローンFMC63を含むCARに対して生成された抗体パネルの特異的結合を、本明細書に記載されるように、それぞれKIP−1抗体、KIP−4抗体、及びKIP−3抗体からの例示的なリンカー配列の抗体結合プロファイルを決定するために使用した。このアッセイには、図1に記載される、リンカー配列KL1(配列番号9)、KL2(配列番号10)、KL3(配列番号11)、及び切断型Whitlowリンカー(配列番号17)を含むペプチドも含まれていた。プロテインG(Pierce)でプレコートされたプレートを使用して、抗体を96ウェルプレートフォーマットで捕捉した。プレートをPBSTバッファーで6回洗浄した後、標的ペプチドとインキュベートした。PBSTで更に6回洗浄し、抗体をストレプトアビジン−HRPと共に更にインキュベートした。PBSTで最後に6回洗浄して、シグナルを検出し、増強化学発光キット(GE Healthcare製のECL)及びVarioskan Flashプレートリーダー(Thermo Fisher)により定量した。図3に示す抗体プロファイルELISA実験の結果は、本発明によるリンカーの抗体結合の幅を実証している。
プロテインLを対照として使用するフローサイトメトリーによりCAR T細胞をアッセイし、配列番号1(FMC63 WT)又は配列番号2(FMC63 G4S)のリンカー配列を含む各CARコンストラクトの発現を確認した。これらの結果は、T細胞の表面でのCARコンストラクトの発現を確認する。図4に示すように、CAR T細胞をscFv FMC63及び24C1 scFvのコンテキストで生成した。KL2(配列番号10)、KL3(配列番号11)、KL4(配列番号7)、KL5(配列番号12)、KL6(配列番号8)、及びG4S2(配列番号18)のリンカーを使用して、scFvのVLドメインとVHドメインとを連結した。
本開示は、幾つかの核酸及びポリペプチド配列を含む。便宜上、下記表Bは、各配列をその適切な配列番号と関連付ける。
