定義された特異性の抗体が、増大する数の多様な治療応用において使用されている。多数の方法が、ヒト治療用途に有用な抗体を取得するために用いられてきている。これらには、キメラ抗体およびヒト化抗体、ならびに、ライブラリー、例えば、ファージ提示ライブラリーから、またはトランスジェニック動物から選択された完全ヒト抗体が含まれる。バクテリオファージにおいて構築された免疫グロブリンライブラリーは、ナイーブな個体または特異的に免疫化された個体の抗体産生細胞に由来することができ、原則として、ヒト免疫グロブリンの重鎖および軽鎖の新たなかつ多様な対形成を含む可能性がある。この戦略は、内因性のレパートリーの限定はこうむらないが、発現した免疫グロブリン断片の相補性決定領域(CDR)が、細菌細胞において適正に合成されてフォールディングされることを必要とする。しかし、多くの抗原結合領域は、細菌細胞において融合タンパク質として正確にアセンブルすることが難しい。加えて、タンパク質は、正常な真核生物翻訳後修飾を受けないであろう。結果として、この方法は、取得することができる抗体特異性に対して様々な選択フィルターをかける。あるいは、完全ヒト抗体は、真核生物系におけるライブラリー、例えば、酵母提示、レトロウイルス提示、またはポックスウイルスなどのDNAウイルスにおける発現から単離することができる。例えば、その全体が参照により本明細書に組み入れられる、米国特許第7,858,559号(特許文献1)および米国特許出願公開第2013-028892号(特許文献2)を参照されたい。
本開示は、単離された膜内在性タンパク質(IMP)またはその断片を、関心対象の標的IMPに結合する抗体または抗体様分子のスクリーニング、選択、および特定における使用のために、天然コンフォメーションで発現させて提示するための組成物および方法を提供する。
本開示は、膜内在性タンパク質(IMP)、例えば、複数回貫通(IMP)を、ポックスウイルス、例えば、ワクシニアウイルスの細胞外エンベロープビリオン粒子(EEV)の表面上にコンフォメーションが無傷または天然の状態で、ポリペプチドセグメントであるEEV特異的膜結合タンパク質、例えばF13Lとの融合物として発現させて提示するための方法および組成物を提供する。
「1つの(a)」または「1つの(an)」実体という用語は、その実体の1つまたは複数を指し;例えば、「1つの結合分子(a binding molecule)」は、1つまたは複数の結合分子を表すように理解される。そのように、「1つの」(または「ある」)、「1つまたは複数の」、および「少なくとも1つの」という用語は、本明細書において互換的に用いることができる。

ZaudererのPCT公開番号第WO 00/028016号および米国特許第7,858,559号において開示されている三分子組換えは、関心対象のタンパク質を発現させるため、および、またはワクシニアウイルスにおいてライブラリーを作製するための、高効率、高力価の作製法である。三分子組換え法により、少なくとも90%の効率、および、少なくとも、直接ライゲーションによって得られるものよりも少なくとも2桁高い力価での組換えウイルスの生成が可能になる。
本開示は、膜内在性タンパク質(IMP)を、タンパク質が天然に発現している細胞において出現するであろうような、タンパク質の天然コンフォメーションにほぼ等しい、コンフォメーションが無傷の状態で発現させるための方法を提供する。本開示にしたがって、IMPを、ポックスウイルス、例えば、ワクシニアウイルスEEV上に発現しているポックスウイルスタンパク質との融合タンパク質として発現させる。本明細書において提供されるIMP融合タンパク質は、EEVの表面上で発現されて提示される場合に、結合分子のライブラリー、例えば、抗体提示ライブラリーをスクリーニングするための標的抗原として有用である。

IMPをコードするcDNAを、パルミトイル化EEV膜糖タンパク質をコードするワクシニアウイルスF13L遺伝子(SEQ ID NO: 1)に対してインフレームで、ワクシニアウイルス中への導入の目的で以前に記載されているpJEM1プラスミド中にクローニングした。pJEM1は、米国特許出願公開第2013/0288927号に記載されているp7.5/tkの誘導体であり、NcoIまたはBssHIIおよびBsiWIで消化した場合に、ワクシニアウイルスTK遺伝子およびワクシニアウイルスH5プロモーターと相同組み換えができる隣接領域を含有する。
BHK細胞に、CD20-F13L融合タンパク質(SEQ ID NO: 4)をコードするIMVまたは対照のウェスタンリザーブ(WR)ウイルスのいずれかを、1細胞あたり1ウイルスの感染多重度(MOI)で2日間感染させ、その後、EEVを含有する上清を収集して、破片を低速遠心分離によって除去した。プロテインG DYNABEADS(登録商標)(110μL)を磁石で取り出して、1 mLのPBS+20μgの精製抗CD20抗体をビーズに添加した。溶液を、室温で穏やかに回転しながら30〜60分間インキュベートして、抗体をプロテインGビーズにカップリングさせた。10μgの精製mIgG1アイソタイプ対照を、完全なブロッキングを確実にするために溶液に添加し、溶液を、室温で穏やかに回転しながらさらに10〜30分間インキュベートした。ビーズを磁石で取り出して、1 mLのPBSで1回洗浄し、110μLのPBSに再懸濁した。
BHK細胞に、CD20-F13L融合タンパク質(SEQ ID NO: 4)をコードするIMV、CD20-A56R融合タンパク質(SEQ ID NO: 13)をコードするIMV、タグ付加されていない(融合していない)CD20をコードするIMV、または対照のHER2-A56R細胞外ドメイン(ECD)(SEQ ID NO: 7)ウイルスのいずれかを、MOI=1で2日間感染させ、その後、EEVを含有する上清を収集して、破片を低速遠心分離によって除去した。ストレプトアビジンDYNABEADS(登録商標)(200μL)を磁石で取り出して、0.2 mLのPBS+20μgの精製ビオチン化抗CD20抗体またはビオチン化抗HER2をビーズに添加した。溶液を、室温で穏やかに回転しながら30分間インキュベートして、抗体をストレプトアビジンビーズにカップリングさせた。ビーズを磁石で取り出して、1 mLのPBSで1回洗浄し、200μLのPBSに再懸濁した。

- ポックスウイルスEEV特異的タンパク質またはその膜結合断片に融合した異種IMPまたはその断片を含む、組換えポックスウイルスEEVであって、該融合タンパク質が、EEV外側エンベロープ膜に位置しており、該IMPまたはその断片が、EEVの表面上にその天然コンフォメーションで提示される、前記組換えポックスウイルスEEV。
- 前記ポックスウイルスEEVがワクシニアウイルスEEVである、請求項67または68に記載の組換えポックスウイルスEEV。
- (a)請求項26および67〜69のいずれか一項に記載の組換えEEVを、固体支持体に付着させる工程;
(b)多数の抗原結合ドメインを提示する提示パッケージを含む、抗体提示ライブラリーを提供する工程;
(c)EEV上に発現したIMPに特異的に結合する抗原結合ドメインを提示する提示パッケージが、それに結合できるように、提示ライブラリーをEEVと接触させる工程;
(d)結合していない提示パッケージを除去する工程;および
(e)EEV上に発現したIMPに特異的な抗原結合ドメインを提示する提示パッケージを回収する工程
を含む、複数回貫通膜タンパク質に結合する抗体を選択する方法。 - 組換えEEVが、固体支持体への付着の前に不活性化される、請求項70に記載の方法。
