実施例1-抗PCSK9抗体の製造
ヒトPCSK9タンパク質を免疫原として抗PCSK9抗体を生成させた。ヒトイムノグロブリン遺伝子組換えマウス技術によって全ヒト抗体の開発と製造を行い、初めてAbgenix(XenonマウスおよびMedarex(HuMab「mouse」); Lonbergら、1994、ネーチャー 368: 856-859; LonbergおよびHuszar、1995、Internal Rev. Immunol. 13:65-93; HardingおよびLonberg,、1995、Ann. N.Y. Acad. Sci. 764:536-546)に記載された。
(工程1)免疫原AであるHis標識PCSK9(PCSK9-His)の製造
標準の分子生物学クローン技術(SambrookおよびRussell、1989、「モレキュラー・クローニング:研究室マニュアル」、ニューヨーク:コールド・スプリング・ハーバー研究所出版社、第二版)によって、配列番号62の全長タンパク質配列(UniProtKB ID Q8NBP7)に相応するヒトPCSK9のコード配列(hPCSK9、配列番号61)をHisタグとともに、pCpCベクター(Invitrogen、#V044-50)にクローンした。ポリエチレンイミン(PEI,Polysciences)およびプラスミドによって、HEK293細胞(Invitrogen)に対して一過性形質移入を行い、37℃でFreeStyle 293発現培地(Invitrogen)において増幅を行った。4日間増幅した後、培地を回収して遠心で細胞成分を除去した。培養上清に組み換えられたHisタグされたPCSK9が含有された。組み換えられたHisタグされたPCSK9を含有する培養上清液に最終濃度が10 mMになるようにイミダゾールを入れた。サンプルに0.22ミクロンの無菌フィルターを通させてNi-NTAアフィニティークロマトグラフィーを行った。緩衝液A[20mMイミダゾール含有1×PBS、pH7.2)および緩衝液B [(緩衝液A、0.1%(v/v)Trition X100,0.1%(v/v)Triton X114]でサンプルを平衡化した後、緩衝液C[1×PBS(pH7.2)、250mMイミダゾール]でHis標識PCSK9タンパク質(PCSK9-His)をNi-NTAアフィニティークロマトグラフィーカラムから溶離させた。Superdex 200カラム(GEヘルスケア)でさらに溶離液を精製して高純度のHis標識組換えヒトPCSK9タンパク質を得た。サンプルを4℃で1×PBS(pH7.2)で一晩透析させた。透析後、精製されたPCSK9-Hisに0.22ミクロンの無菌フィルターを通させ、分けて-80℃で保存した。
一連の体外同定方法によって、His標識hPCSK9免疫原の生物活性を評価したところ、免疫原がPCSK9の生物活性を示し、かつ投与量に依存してHepG2細胞のLDLに対する取り込みを抑制したことがわかった。生産された2バッチのHis標識hPCSK9免疫原はHepG2のLDL取り込みの抑制において、相当する生物活性を示した(図1を参照する)。この2バッチの生物活性を有するHis標識hPCSK9免疫原はマウス免疫に使用された。
ヒトPCSK9のコード配列(hPCSK9、配列番号61)をpcDNA3.1ベクター(Invitrogen)にサブクローンし、得られたプラスミドを1.0um金コロイド弾(Bio-RAD)に包埋した後、Heliosジーンガンの説明書に従い、Heliosジーンガン(Bio-rad No.165-2431)によって免疫を行った。
ヒトIg FcはマウスのFc受容体と相互作用しないため、hFcを含有する免疫原がマウスにおいて引き起こす免疫応答が弱く、モノクローナル抗体の製造の効率が低くなる。マウスのゲノムにヒトイムノグロブリン(Ig)可変領域をコードする遺伝子およびラットIg定常領域の遺伝子を導入することによって、Harbour H2L2遺伝子組換えマウスを作り、マウスにhV-rCを含むキメラIgを含有させ、同時にマウスIgの発現が不活性化された(WO2010/070263A1)。Harbour H2L2遺伝子組換えマウスは野生型マウス(たとえばBalb/c)に相当する免疫応答および抗体力価を示すことができる。
ハイブリドーマ細胞由来の抗体の濃度が約1-10ug/mLと低く、かつ抗体の濃度が幅広く変わる。さらに、FBSおよび培地の成分は分析に干渉する可能性がある。そのため、小規模の抗体の生産と精製を行う必要がある(1〜5 mg)。
(A部分)ELISAによる抗PCSK9抗体と組換えPCSK9およびPCSK9D374Yタンパク質の結合の検出
ELISAによって実施例2で得られた精製された抗PCSK9抗体と組換えヒトPCSK9タンパク質(免疫原A)、PCSK9D374YおよびカニクイザルPCSK9タンパク質の結合を分析した。
野生型PCSK9およびその突然変異型PCSK9D374YはいずれもLDLRと結合し、野生型と比べ、突然変異型PCSK9D374YはLDLRに対してより高い結合親和力を有する。受容体配位子結合試験によって、抗PCSK9抗体のPCSK9およびPCSK9D374YとLDLRの結合を遮断する能力を検出した。
トリプターゼで対数期のHepG2細胞(ATCC)を消化し、遠心して10%(w/w)牛胎児血清が補充されたMEM培地(Invitrogen、カタログ番号11095-080)に再懸濁させた。細胞密度を3×105個細胞/mLに調整し、そして細胞を24ウェルプレートの各ウェルに入れた。プレートを6時間インキュベートしてHepG2細胞を付着させた後、培地を吸い取り、10%(w/w)LPDS(Kalen生物医薬有限責任公司、880100-2)が補充されたMEM培地を添加して一晩インキュベートした。2日目に、実施例2で得られた0.4ug/mLのPCSK9D374Yと抗PCSK9抗体を混合して24ウェルプレートに入れ、それを37℃で4時間インキュベートした。10%LPDSが添加されたMEM培地を捨て、PBSでプレートを2回洗浄した。ベルセン溶液(Invitrogen)でプレートからHepG2細胞を分離させて収集した。細胞をPBSで2回洗浄し、細胞数を測定し、かつPBSで細胞密度を2×106個細胞/mLに調整した。細胞懸濁液にブロッキング緩衝液[1%(w/w)FBS PBS]を入れ、そして96ウェルFACSプレート(FACS Calibur,BD)の各ウェルに100uLずつ細胞懸濁液を入れ、当該96ウェルFACSプレートを氷上に15分間インキュベートした後、遠心した。ブロッキング緩衝液を吸い取り、各ウェルに100μLずつ1ug /μLのLDLR抗体(Progen、61009)を入れ、氷上で15分間インキュベートした。FACS緩衝液[1%(w/w)BSA HBSS]でプレートを2回洗浄し、各ウェルに100uLずつAlexa-488とカップリングされた抗ウサギ二次抗体(モレキュラープローブス社、A-11034)を入れ、氷上で15分間インキュベートした。FACS緩衝液でプレートを3回洗浄し、かつ最後の洗浄後100uLのFACS緩衝液に再懸濁させた。FACS Calibur(BD)によって平均蛍光強度(MFI)を検出し、結果は図7および表13に示す。IgG対照はラットIgGで、表中の値は細胞群の平均蛍光強度。結果では、抗PCSK9抗体は投与量に依存してPCSK9D374Yで誘導されたHepG2細胞におけるLDLR分解を抑制したことが示された。
トリプターゼで対数期のHepG2細胞(ATCC)を消化し、遠心して10%(w/w)牛胎児血清が補充されたMEM培地(Invitrogen、カタログ番号11095-080)に再懸濁させた。細胞密度を5×105個細胞/mLに調整し、細胞をPDLで被覆された96ウェルプレート(Millipore、A-003-E)に入れた。プレートを6時間インキュベートしてHepG2細胞を付着させた後、培地を吸い取り、10%(w/w)LPDS(Kalen生物医薬有限責任公司、880100-2)が補充されたMEM培地を添加して一晩インキュベートした。10ug/mLのBODIPY<登録商標> FL LDL(Invitrogen,L3483)、0.5ug/mLのPCSK9D374Yおよび実施例2で得られた抗PCSK9抗体を混合して96ウェルプレートに入れ、それを6時間インキュベートした。培地を吸い取り、PBSでプレートを2回洗浄し、M5プレートリーダー(モレキュラーデバイス)によって520nm/485nmにおける蛍光強度を測定した。結果は図8および表14と15に示すように、抗PCSK9抗体が投与量に依存してPCSK9D374Yが介するLDL取り込みに対する抑制を遮断することが示された。
全RNAの分離:実施例2で得られたハイブリドーマクローンの上清液を同定した(すなわち生物活性の検証と測定、実施例3〜6)後、遠心して5×107個のハイブリドーマ細胞を収集した。1mLのTrizolを細胞の沈殿に入れ、混合して1.5mL遠心管に移し、室温で5分間インキュベートした。サンプルに0.2mLのクロロホルムを入れ、15秒ボルテックスした。2分間置いた後、混合物を4℃、12000gで5分間遠心した。上清液を収集し、新しい1.5mL遠心管に移し、0.5mLのイソプロパノールを入れ、軽く混合し、サンプルを室温で10分間インキュベートした。サンプルを4℃、12000gで15分間遠心した。上清液を吸い出し、沈殿物を1mLの75%(v/v)エタノールで洗浄した。混合物を4℃、12000gで5分間遠心し、上清液を捨て、沈殿物を自然乾燥した。沈殿物にDEPCで処理された水(55℃水浴10分間)を入れて全RNAを得た。
(工程1)プラスミドの構築と製造:実施例7で得られた抗PCSK9抗体の重鎖と軽鎖可変領域の配列。抗PCSK9抗体の重鎖可変領域の配列をシグナルペプチドおよびヒト重鎖IgG1定常領域を含有する発現ベクターにサブクローンした。抗PCSK9抗体の軽鎖可変領域の配列をシグナルペプチドおよびヒト抗体の軽鎖κ定常領域を含有する発現ベクターにサブクローンした。配列決定によって組換えプラスミドを検証・確認した(配列決定方法は実施例7と同様である)。キット(MACHEREY-NAGEL)によってアルカリ溶解を行うことによって、組換えプラスミドの純度と質量を向上させ、かつ0.22uMフィルター(Millpore)でプラスミドをろ過した。精製されたプラスミドを形質移入に使用した。
解離定数はOcted red 96(Fortiebio)によって測定された。詳細な操作および方法はメーカーによって提供される装置説明書に従って行われる。つまり、AHCセンサー(抗ヒトFcセンサー、Fortiebio)によって親和力の測定を行った。抗PCSK9抗体を0.1%(w/w)BSAおよび0.02%(v/v)Tween20を含有するPBS緩衝液(pH7.4)で10ug/mLに希釈し、かつAHCセンサーでインキュベートした。5つの異なる濃度の組換えhPCSK9-His(100nMまたは400nMの開始濃度から2倍の連続希釈されたもの)を抗体が入ったAHCセンサーと30℃で3分間インキュベートした。さらに反応混合物を0.1%(v/w)BSAおよび0.02%(v/v)Tween20を含有するPBS緩衝液(pH7.4)で30℃で5分間インキュベートした。Octet Red 96によってリアルタイムで抗PCSK9と免疫原Aの結合と解離シグナルを記録した。Octet Userソフトによって親和力、結合および解離定数を確認し、結果を表28に示す。
抗ヒトFc IgGの流動セル1と2への固定化:HBS-EP+(10mM HEPES、150mM NaCl、3mM EDTA、0.05%P2O、pH 7.4)を流動緩衝液として使用し、かつ固定化ウィザードテンプレート(immobilization wizard template)で抗ヒトFc IgGの固定化を行った。新しく混合された50mmol/L NHSと200mmol/L EDCでSシリーズCM5センサーチップの流動セル1と2を活性化させた。10mM NaAC(pH4.5)に希釈された20μg/mLの抗ヒトFc IgGを活性化された流動セル1と2に注射した。残った活性カップリング部位を1Mエタノールアミンでブロッキングした。
SPF条件において9〜10週齢のC57BL/6マウスを飼育した。マウスをランダムに5つの群に4匹ずつ分けた。1日目に、マウスの腹膜内に30mg/kg、3mg/kgまたは0.3mg/kgの被験抗PCSK9抗体(たとえば139G1C8または96F8C6)、対照IgGまたは溶媒を施用した。2日目に、すべてのマウスの静脉内に組換えヒトPCSK9タンパク質を施用した(30ug/マウス)。PCSK9注射の1時間後、マウスを殺処分してその肝臓を得た。マウスLDLR(mLDLR)に対する抗体およびマウスβ-アクチン(mβ-アクチン)に対する抗体で肝臓分解物のウェスタンブロット解析を行った。密度測定法によってタンパク質バンドを定量し、mLDLRとmβ-アクチンの比をプロットした。結果は図23AとBに示すように、抗PCSK9抗体が体内でPCSK9が介するLDLR分解を抑制したことが示された。
サンプル緩衝液で全ヒト抗PCSK9抗体を1mg/mLに調整し、最終体積は約700uLであった。パラメーターは以下の通りに設定した(VP-DSC)。開始温度30℃、最終温度100℃、走査速度50℃/時間で、再走査回数は0で、プレスキャンは恒温で3分間で、ポストスキャンは恒温で0分間で、後サイクルは恒温25℃で、ろ過時間は25秒で、フィードバックモード/ゲインは無しで、細胞充填パラメーターは35℃であった。得られたタンパク質-緩衝液の熱分析図の結果は、相応する緩衝液-緩衝液走査値を引くことによって処理した後、ベースラインをトレースラインにフィットした。OriginTM 7.0ソフトによって熱分析図で観察された各ピークの最大値におけるTmsを記録した。結果を図24に示す。
全ヒト抗PCSK9抗体の凍結・解凍安定性は以下のように同定された。100uLの各抗PCSK9抗体の少量の冷凍原液を室温で解凍した。完全に解凍すると、サンプルを-80℃の冷蔵庫で快速に冷凍し、かつ-80℃で少なくとも2時間維持した後、さらに室温で解凍した。サンプルに対して同様の冷凍/解凍のサイクルを3回行った。目視で沈殿の状況をチェックした。3回の冷凍/解凍のサイクル後、サンプルから20uLの少量試料を取ってサイズ排除クロマトグラフィー(SEC)分析を行った。HPLC-SECによって特徴付け、冷凍/解凍のサイクルの前と後の全ヒト抗PCSK9抗体の安定性を分析した。図25に示された結果から、3回の冷凍/解凍のサイクル後、単量体のIgGはテストされた各抗PCSK9抗体の95%以上を占めたことが示された。
4℃において、10mgのIgGを14000gの遠心ろ過器(Amicon Ultra-0.5mL 30K)で>100mg/mLまで濃縮することによって、全ヒト抗PCSK9抗体の溶解度を同定した。2mLまたはそれ以上のIgGを遠心ろ過器に入れ、4℃で14000gで濃縮させた。遠心の設定時間は2 min、3 min、5 min、8 min、15 minおよび20 min、毎回に20uL分けて収集管に入れ、分光光度計によってA280において濃度を検出した。濃度が100mg/mLに達した時、遠心を止めた。HPLC-SECの同定に関し、6uLの濃縮サンプルをHPLC-SECカラムに注射し、ピーク面積に基づいて単量体および多量体の百分率を確認した。図26に示された結果から、テストされたすべての全ヒト抗PCSK9抗体が100mg/mL超の溶解度を有し、かつテストされたすべての全ヒト抗PCSK9抗体に対して単量体IgGが95%超であったことが示された。
(工程1)CDR突然変異誘発ライブラリーの構築と検証
1.ファージ発現ベクターの構築とscFvディスプレイの検証:実施例6の方法によって、抗PCSK9抗体98F8C6の重鎖および軽鎖の可変領域の配列を得た。オーバーラップPCRによって重鎖可変領域、リンカーおよび軽鎖可変領域をコードする核酸をアセンブルし、scFvを生成させた。PCR産物をファージベクターに連結した後、TG1細胞に導入した。アンピシリン選択プレートからの陽性クローンを選択し、そのファージ発現ベクターを回収して配列決定を行った。

3.ライブラリーの構築:オーバーラップPCRによってランダム突然変異ライブラリーを構築した。PCR産物を制限酵素SfiIで50℃で2時間消化した後、T4リガーゼで16℃で一晩SfiIで消化されたファージに連結した。各ライブラリーをTG1大腸菌に電気的形質転換した。
平衡条件において、ビオチン化された可溶性hPCSK9を溶液相における抗原とし、CDR突然変異ScFvファージライブラリーに対してパンニングした。まず、2%のMPBS(2%牛乳を含有するPBS溶液)で使用される遠心管およびストレプトアビジン磁気ビーズを室温で2時間ブロッキングした。2%のMPBSにおいて、ビーズでライブラリーを予め消耗した後、ブロッキングされた管の中でビオチン-hPCSK9と混合し、ローテーターで室温で2時間インキュベートした。1回目のパンニングにおいて、使用された抗原濃度は10nMで、そして後の回で毎回10倍で減少した。ScFv-抗原-ビオチン複合体をストレプトアビジン磁気ビーズと混合して15分間インキュベートした。磁石でビーズを収集し、1.5の微量遠心管に移し、それぞれMPBST(2%牛乳、0.5%ツイン20のPBS溶液)、PBST(0.5%ツイン20のPBS溶液)、MPBS(2%牛乳のPBS)およびPBSで5回洗浄した。37℃において、1mLのトリプターゼ(PBS中10ug/mL)でビーズから結合したファージを30分間溶離させた。磁石でビーズを管の一側に吸い、溶離されたファージを含有する溶液を4mLのTG1(A600=0.6)を含有する管に移し、パンニングライブラリーの滴定に使用した。
(工程3)親和力が増加した結合バリアントのELISA選別
3回または4回のパンニング後、500個超の単一クローンが選ばれた。IPTGによってScFvバリアントの発現を誘導し、かつ強いELISAシグナルを有するクローンをシークエンシングした。
濃縮クローンからのVHおよびVL配列をIgG発現ベクターにクローンし、相応するリーダー配列と定常領域の間に挿入した。一過性形質移入によって得られたベクターをHEK293細胞に導入した。実施例8に記載のように、5〜7日後、タンパク質Aカラムで親和性精製によって培養上清液におけるIgGを精製した。親和力が向上したバリアントのVH配列は表30に示した。これらのバリアントの軽鎖配列は98F8C6抗体の軽鎖のもの、たとえば、それぞれ配列番号38〜40のLCDR 1〜3、または配列番号37の軽鎖可変領域、またはそのコード配列の配列番号58と同様である。

- 1つの重鎖可変領域および1つの軽鎖可変領域を含み、前記の重鎖可変領域は配列番号1、9、17、25、33、41および91〜108からなる群から選ばれる配列と少なくとも95%の相同性を有するポリペプチド配列を持ち、前記の軽鎖可変領域は配列番号5、13、21、29、37および45からなる群から選ばれる配列とそれぞれ少なくとも95%の相同性を有するポリペプチド配列を持つ、請求項1〜3のいずれかに記載の単離されたモノクローナル抗体または抗原結合断片。
- 請求項1〜4のいずれかに記載の単離されたモノクローナル抗体または抗原結合断片であって、
a.配列番号33および91〜108からなる群から選ばれるポリペプチド配列を有する重鎖可変領域、および配列番号37のポリペプチド配列を有する軽鎖可変領域、
b.配列番号1のポリペプチド配列を有する重鎖可変領域、および配列番号5のポリペプチド配列を有する軽鎖可変領域、
c.配列番号9のポリペプチド配列を有する重鎖可変領域、および配列番号13のポリペプチド配列を有する軽鎖可変領域、
d.配列番号17のポリペプチド配列を有する重鎖可変領域、および配列番号21のポリペプチド配列を有する軽鎖可変領域、
e.配列番号25のポリペプチド配列を有する重鎖可変領域、および配列番号29のポリペプチド配列を有する軽鎖可変領域、あるいは
f.配列番号41のポリペプチド配列を有する重鎖可変領域、および配列番号45のポリペプチド配列を有する軽鎖可変領域、
を含む抗体または抗原結合断片。 - キメラのものである、請求項1〜5のいずれかに記載の単離されたモノクローナル抗体または抗原結合断片。
- ヒトのものである、請求項1〜6のいずれかに記載の単離されたモノクローナル抗体または抗原結合断片。
- ヒト重鎖IgG1定常領域およびヒト抗体の軽鎖κ定常領域を含む、請求項7に記載の単離されたモノクローナル抗体または抗原結合断片。
- 単離された請求項1〜8のいずれかに記載のモノクローナル抗体または抗原結合断片をコードする核酸。
- 請求項9に記載の単離された核酸を含むベクター。
- 請求項9に記載の核酸を含む宿主細胞。
- 請求項1〜8のいずれかに記載の単離されたモノクローナル抗体または抗原結合断片と、薬学的に許容される担体とを含む薬物組成物。
- 必要な被験者においてPCSK9とLDLRとの結合を遮断するか、あるいはLDL取り込みを増加する方法であって、前記被験者に請求項12に記載の薬物組成物を施用する工程を含む方法。
- 必要な被験者において脂質疾患、障害または病症、代謝性疾患、障害または病症、炎症性疾患、障害または病症、感染性疾患、障害または病症を治療する方法であって、前記被験者に請求項12の薬物組成物を施用する工程を含む方法。
- 必要な被験者において脂質異常、代謝性疾患または炎症性疾患を治療する方法であって、前記被験者に請求項12の薬物組成物を施用する工程を含み、前記の脂質異常、代謝性疾患または炎症性疾患は高脂血症、原発性高脂血症、脂質代謝異常、混合性脂質代謝異常、ホモ型家族性高コレステロール血症および敗血症からなる群から選ばれる方法。
- 請求項1〜8のいずれかに記載のモノクローナル抗体またはその抗原結合断片を製造する方法であって、所定の条件においてモノクローナル抗体または抗原結合断片をコードする核酸を含む細胞を培養することによって、モノクローナル抗体または抗原結合断片を生成させる工程と、前記細胞または細胞培養物から抗体または抗原結合断片を回収する工程とを含む方法。
- 請求項1〜8のいずれかに記載のモノクローナル抗体または抗原結合断片を含む薬物組成物を製造する方法であって、前記モノクローナル抗体または抗原結合断片を薬学的に許容される担体と組み合わせることによって前記薬物組成物を得る工程を含む方法。
