Claims (16)
- 対象のタンパク質および一以上の混入物を含む混合物から対象のタンパク質を精製する方法であって、対象のタンパク質が抗体であり、混合物が、哺乳類培養細胞であり、以下を含む方法:

- 抗体がモノクローナル抗体である、請求項1または2記載の方法。
- モノクローナル抗体がヒト抗体、ヒト化抗体およびキメラ抗体からなる群から選択される、請求項3記載の方法。

- 第1の洗浄溶液のpHが約9.6である、請求項5記載の方法。
- 第1の洗浄溶液のpHが約10.4である、請求項5記載の方法。
- 第1の洗浄溶液の後に、第1のクロマトグラフィーマトリックスを第2の洗浄溶液と接触させることをさらに含む、請求項1から8のいずれかに記載の方法、ここで第2の洗浄溶液はpHが少なくとも9.0であり、アルギニンまたはアルギニン誘導体を含まない。
- 第2の洗浄溶液の後に、第1のクロマトグラフィーマトリックスを第3の洗浄溶液と接触させることをさらに含む、請求項9記載の方法、ここで第3の洗浄溶液はpHが約6と約7の間であり、アルギニンまたはアルギニン誘導体を含まない。

- 混合物を一以上のさらなるクロマトグラフィーマトリックスにさらす、請求項1から12のいずれかに記載の方法。
- 一以上のさらなるクロマトグラフィーマトリックスが、イオン交換クロマトグラフィー(例えば、陰イオン交換クロマトグラフィーまたは陽イオン交換クロマトグラフィー)、疎水性相互作用クロマトグラフィー、およびミックスモードクロマトグラフィーから選択される、請求項13記載の方法。
- 混合物が、回収された培養細胞液、培養細胞上清、および条件培養細胞の上清、細胞溶解物、および清澄化したバルクから選択される、請求項1から14のいずれかに記載の方法。
