ことを特徴とするリガンドである。

‐配列GFTFNTYA(配列番号14)によって規定されるVH−CDR1、

(1)マウス免疫ストラテジー
注射は全て腹膜内注射でマウス(Balb/c)に行われた。各免疫ストラテジーに5匹のマウスを使用した。
完全フロイントアジュバントと共に100μgの精製ヒツジFSHを用いて最初の注射(0日目)を行った。その後、次の順序で数回のブースター注射を行った。
‐21日目および44日目:不完全フロイントアジュバントと100μgの精製ヒツジFSH、
‐134日目および204日目:不完全フロイントアジュバントと50μgのヒツジFSH βサブユニット、
‐217日目、218日目および219日目:アジュバント無しの30μgのヒツジFSH βサブユニット、
‐220日目:融合。
RD Biotechが販売するFastElysaアイソタイピングキット(参照番号RDB3255)を製造業者の推奨に従って使用してCA5抗体およびCH10抗体の
アイソタイピングを行った。
CNCM I−4801ハイブリドーマおよびCNCM I−4802ハイブリドーマによってそれぞれ分泌されるCA5抗体およびCH10抗体の重鎖可変部分(VH)と軽鎖可変部分(VL)のヌクレオチド配列を以下のプロトコルに従ってそれらのメッセンジャーRNA(mRNA)から決定した。
(a)scFv抗体断片の構築
CA5抗体およびCH10抗体に由来する一本鎖可変断片(scFv)の合成遺伝子がATG:バイオシンセティクスGmbH(ドイツ)によって合成された。
細菌培養物
50μg/mlのアンピシリンを含む5mlの2×YT培地に前培養物を37℃で一晩にわたって調製した。翌日、500μlのこの前培養物を500mlの同じ培地に接種し、1.4のOD600nmが得られるまで150RPM、37℃で培養した。0.1mMのIPTGを添加することでscFvの合成を150RPM、16℃で16時間にわったって誘導した。
その培養培地を4500g、4℃で30分間にわたって遠心分離した。調製の後の部分は4℃で実施した。細菌のペリプラズムを抽出するため、沈殿物を10mlのTES(0.2Mトリス、pH8、0.5M EDTA、0.5Mショ糖)に再懸濁し、30分間にわたってインキュベートし、その後で4分の1に希釈された15mlのTESをそれに添加し、続いて30分間にわたってさらにインキュベートした。その細菌抽出物を10000gで30分間にわたって遠心分離した。上清をPBSに対して一晩にわたって透析した。その透析された上清をscFvの精製のためにすぐに処理するか、または使用するまで−20℃で貯蔵した。
前記ペリプラズムを5000g、4℃で20分間にわたって遠心分離した。その上清をHIS−Select(登録商標)ニッケルアフィニティーゲル(シグマ・アルドリッチ、ミズーリ州、米国)と共に4℃で1時間にわたって撹拌しながらインキュベートした。0に近いOD280nmが得られるまで、0.3MのNaClを含むpH8の0.05Mリン酸ナトリウム緩衝液でそのゲルを洗浄し、その次にそのゲルに添加された20mMのイミダゾールを含む同じ緩衝液で洗浄した。その後で0.3MのNaClと250mMのイミダゾールを含むpH8の0.05Mリン酸ナトリウム緩衝液でscFvを溶出した。その溶離液をPBSに対して一晩にわたって透析した。その溶離液は−20℃で貯蔵されている。
その精製されたscFvを15%ポリアクリルアミドゲル上での電気泳動によってクマシーブルーによる染色後に分析し、且つ、Sephadex(商標)75 10/300
GLカラム(参照番号17−5174−01、GEヘルスケア、ドイツ)上での排除クロマトグラフィーによって分析した。
前記抗体およびそれらのscFvの特異性をELISA法により検討した。評価される各ホルモンはpH9.6の0.1M炭酸ナトリウム緩衝液中に10μg/mlの濃度で調製され、ELISAプレート上でウェル当たり100μlの割合で分配された。吸着時間は+4℃で18時間であった。洗浄を5回行った後に0.1%ツイーンおよび1%BSAを添加した100μlのPBSでウェルを37℃で45分間にわたって処理し、その後、各抗体またはscFvを100μl/ウェルの割合で分配し、37℃で1時間にわたってインキュベートした。評価される各ホルモンに対して、抗体については10〜250μg/mlの範囲、scFvについては10〜150または200μg/mlの範囲に応じた様々な濃度でそれらの抗体およびscFvを分配した。
‐Aが最大光学密度の50%(ED50)を生じる濃度であるとする。別のホルモンで得られた曲線に基づき、
‐BがAを規定するために使用された光学密度値と同じ光学密度値に対応する濃度である
とする。
交差反応のパーセンテージはA割るBかける100、つまり[(A/B)×100]に等しい。
表17はヒツジFSHのαサブユニットとβサブユニット(s.u.)およびヒトFSHのβサブユニットとのCA5抗体の交差反応のパーセンテージを示している。
ヒツジLHおよびブタLHに関して中程度の親和性(それぞれ1.95μMおよび2.47μMのKd)を示している。hCGおよびeCGに関して推定されたKd(3.14〜4.07μMのKd)はこれらのホルモンに対するCA5 scFvの低めの親和性を示している。
表22はヒツジFSHのαサブユニットとβサブユニット(s.u.)およびヒトFSHのβサブユニットとのCH10抗体の交差反応のパーセンテージを示している。
FSHの生理活性に対する本発明のリガンドの増強作用の証明が、FSH単独か、またはFSH/モノクローナル抗体(MAb)複合体のどちらかで刺激された様々な細胞種または細胞株を用いて得られた生物学的応答を比較することにより行われた。
最初にヒツジFSH(oFSH)の生理活性に対するCA5 MAbおよびCH10 MAbの増強作用の特徴がウシFSH受容体を内在的に発現するウシ顆粒膜細胞上で解析された。
様々な種のFSHに対する前記MAbの増強作用がヒトFSH受容体を安定的に発現するHEK293細胞上で測定された。この系により、37℃で1時間にわたるFSH単独か、またはFSH/MAb複合体による刺激の後のFSH受容体の活性化に続くcAMP産生を測定することが可能になった。
培地中で2時間の休止の後、それらの細胞を37℃で1時間にわたって刺激した。培養上清を回収し、ELISAキット(バイオメディカルテクノロジーズ社、マサチューセッツ州、米国、BT−730)を使用してアッセイした。結果はエンドポイントにおけるcAMPの分泌量を表している。Prismソフトウェア(GraphPadソフトウェア社、サンディエゴ、カリフォルニア州、米国、バージョン5)を使用してそれらの結果を分析した。
様々な種のFSHに対する前記MAbの増強作用が、ヒトFSH受容体およびGloSensor(商標)ベクター(プロメガ、フランス)を安定的に発現するHEK293細胞上でリアルタイムに測定された。この細胞系により、アゴニスト(FSHのみ、またはFSH/モノクローナル抗体複合体)でのFSH受容体の刺激の後のcAMP産生をリアルタイムでモニターすることが可能になった。GloSensor(商標)タンパク質へのcAMPの結合の後、GloSensor(商標)基質(プロメガ、フランス、参照番号E1291)が加水分解され、それによりPolarStar Optimaリーダー(BMG Labtech、ドイツ)によって測定され、且つ、RLU(相対発光単位)で表される発光が放射された。この安定株はフランス、37380、ヌジリー、ヴァル・ド・ロワールにあるINRA(フランス国立農学研究所)センターのシグナル伝達システムのバイオロジーおよびバイオインフォマティクスチームによって開発され、これらのアッセイのために利用可能とさせてもらった。
、表示されている濃度を得るために11μlのリガンド(FSHのみ、またはFSH/モノクローナル抗体複合体)をそのプレートに添加した。その後、放射される発光を約1時間30分にわたって測定した。
図3は、単独の、またはCA5抗体と複合体化されたヒツジFSHの0nM、0.1nM、0.3nM、1nMおよび3nMの濃度で得られた、時間(分)の関数として相対発光単位で表されるcAMP産生カイネティクス曲線を示している。FSHを含まない単独の前記抗体によって「刺激」された細胞は応答を示さず、発光シグナルが基底レベルに留まっていることが観察されている(曲線3A)。単独のCA5抗体はHEK293細胞が発現するヒトFSH受容体に対してアゴニスト作用もアンタゴニスト作用も及ぼしていない。逆に、oFSHと複合体化されたCA5モノクローナル抗体(6nMの最終濃度)は前記ホルモンの刺激活性を非常に大幅に、且つ、顕著に増強している。oFSHの0.1nMおよび0.3nMの濃度における最大細胞応答の350%および330%の増加(曲線BおよびC)、およびoFSHの1nMおよび3nMの濃度におけるそれぞれ230%および140%の増加(曲線DおよびE)がこのように観察されている。細胞応答の飽和およびこの事例では11000RLUでの発光シグナルの飽和のため、oFSHの濃度が高くなるほど(1nMおよび3nM)増強作用の大きさが小さくなる。GraphPad
Prismによって測定されたEC50値はoFSHについては2.36×10-9Mであり、且つ、oFSH/CA5複合体については2.83×10-10Mであり、前記FSHがCA5抗体と複合体化したときの1単位のLogEC50の増加(10-8.6から10-9.54まで)を反映している。oFSH/CA5と比較したoFSHのそれらの二本の曲線の間の差は検査されたoFSH用量の全てについて非常に有意である(p<0.001)。
Hで200%の増強作用が観察され、そして最も高い0.3nMのhFSHの濃度で170%の増強作用が観察された。GraphPad PrismによるEC50の計算によってhFSHについては5.86×10-10Mの値、およびhFSH/CA5複合体については1.36×10-10Mの値が示された。これは前記FSHがCA5抗体と複合体化したときの0.63のLogEC50の増加(10-9.23から10-9.86まで)を反映している。CA5 scFvの増強作用も0.01nMのhFSHの生理活性に対して検討した。36nMのscFv濃度で160%という有意な増加(p<0.01)が得られた(図5)。この増加は二価のCA5完全抗体の増加と類似していた。この結果はその一価断片がCA5抗体と同じFSH活性増強特性を有していることを意味している。
CH10抗体の調節作用をヒツジFSH(oFSH)およびヒトFSH(hFSH)(ゴナールF、セローノ・ラボラトリー)に対して検討した。
インビトロで特徴解析された後、各モノクローナル抗体の増強作用をFSHの生理活性に対するそれらの抗体の作用についてはメスのラットにおいて、およびそれらが同じく認識するLH/CGの生理活性に対するそれらの抗体の作用についてはオスのラットにおいてインビボで特徴解析した。
類の調製物に対して評価した。
CA5抗体およびCH10抗体およびそれらのscFvの増強作用をヒツジFSHに対して検討し、且つ、ヒトの生殖において使用されているヒトFSHの様々な調製物であるゴナールFとピュレゴン(それぞれメルクセローノ・ラボラトリーとメルクシェリングプラウ・ラボラトリーに由来する組換えFSH)、およびフォスティモンとメノピュール(それぞれラボラトワール・ジェニエーブルとメルクシェリングプラウによって販売されている抽出型FSH)に対して検討した。
図8AはヒツジFSHの生理活性に対するCA5抗体の作用の代表例を示している。各バッチは5匹のメスから構成された。単独で注射されたCA5抗体で処置されたバッチは生理食塩水を受領した対照バッチのメスと同じ平均卵巣重量を示した(それぞれ25.6mgと29.15mg)。従来のホルモン処理(3.5IUのhCG+0.5μgのoF
SH)を受けたバッチは対照バッチよりも有意に高い(p<0.05)85.7mgという平均卵巣重量を生じた。CA5抗体と予め複合体化されたヒツジFSHで処置されたバッチは198mgという平均重量を、すなわち従来のホルモン処理を受けたバッチと比較して非常に有意な(p<0.0001)FSH生理活性の231%の増加を示した。ヒツジFSHの活性に対するCA5のインビボ増強作用を数回の実験において分析した。それらの実験の結果の全てが表27に示されている。3回の実験について記録された前記ホルモン/CA5複合体を用いる刺激の後の平均卵巣重量の増加は非常に有意である(p<0.0001)。
scFvが前記ホルモンから独立して、且つ、時間をおいて単独で注射されてもそのscFvの増強作用をインビボで立ち上げることができることを示しているので重要である。前記ホルモン/scFv複合体は処置動物においてインビボで形成される可能性がある。
ヒツジFSHの活性に対するCH10のインビボ増強作用を数回の実験の間に分析した。それらの実験の結果の全てが図10Aに示されている。hCG+oFSH混合物で処置されたメスのラットと前記ホルモン/CH10複合体で処置されたメスのラットとの間での応答についての3回の実験の比較から145%という有意な増加(p<0.01)が示されている。記録された平均卵巣重量は前記ホルモン/CH10複合体で処置されたメスのラット(n=13)において100gの体重当たり126.6±9.6mgであるのに対し、hCG+oFSH混合物で処置されたメス(n=12)では87.55±3.724mgであった。卵巣応答について単独のCH10抗体の効果は観察されなかった。
scFvとhFSHの独立した腹膜内注射によって得られた。CH10 scFvを前記ホルモンと別々に注射するにしても、または前記ホルモンとプレインキュベートして注射するにしても、そのscFvの増強作用はこのようにインビボで立ち上げられ得る。
ヒツジLHは非常に高価であるため、これらの生物学的アッセイは、容易に入手でき、非常に純粋であり、且つ、安価な形態であるhCGを用いて実施された。前記抗体の作用を抽出型ヒトhCG(ヒト絨毛性ゴナドトロピン)の2種類の調製物、すなわち一方の生殖補助医療処置の背景でのヒトの生殖に使用されるENDO5000(シェリングプラウ・ラボラトリー)と他方の獣医学で使用されるコルロン(MSDラボラトリー)に対して検討した。
‐それぞれ53匹および56匹という数の動物について、SVの平均重量はhCGであるコルロンで処置されたラットでは28.8mg/100gであり、且つ、前記複合体で処置されたラットでは46.7mg/100gであり(162%の増加)(図12B)、
‐それぞれ26匹および30匹という数の動物について、SVの平均重量はhCGであるENDO5000で処置されたラットでは24.64mg/100gであり、且つ、前記複合体で処置されたラットでは48.13mg/100gであった(195%の増加)(図12C)。
た対照動物と比較して精嚢重量の変化を示していないことが観察されている。したがって、前記ホルモンに複合体化していないCH10は標的器官に対して特異的作用を持たない。
‐それぞれ34匹および35匹という数の動物について、SVの平均重量は前記複合体で処置されたラットでは50.04mg/100gであったのに対し、hCGであるコルロンで処置されたラットでは29.6mg/100gであり(169%の増加)(図13B)、
‐それぞれ13匹および15匹という数の動物について、SVの平均重量はhCGであるENDO5000で処置されたラットでは24.64mg/100gであり、且つ、前記複合体で処置されたラットでは51.39mg/100gであった(208%の増加)(図13C)。
げっ歯類動物(小動物)においてCA5モノクローナル抗体およびCH10モノクローナル抗体の増強作用をインビボで実証し、特徴解析した後、多産でより大型の家畜、すなわち雌羊においてFSHの活性に対する各抗体の作用を研究することを目的とした。

CA5抗体(IgG)の増強作用を2つのプロトコル(プロトコル1および2)において発情期中および発情期の終わりに評価した。
‐「CA5抗体」バッチ(n=6)は前記スポンジの撤去の4日前に2mg、その撤去前に1mg、およびそのスポンジの撤去時に1mgの3回の精製CA5の筋肉内注射を受け、
‐「対照」バッチ(n=5)は前記スポンジの撤去の24時間前に生理食塩水の筋肉内注射を受け、
‐「FSH」バッチ(n=5)は前記スポンジの撤去の24時間前に100μgのブタFSH(pFSH)の筋肉内注射を受け、そのスポンジの撤去の12時間前に90μgの筋肉内注射を受けた。
れ、それに続いて周期の最後まで黄体期を通して維持される分泌の著しい増加が観察されている。平均プロゲステロン値は10日目ではCA5バッチ、FSHバッチおよび血清Φバッチについてそれぞれ1.38ng/ml、0.92ng/ml、および0.52ng/mlであり、15日目では2.58ng/ml、1.7ng/mlおよび1.18ng/mlである。対応のあるノンパラメトリックt検定(ウィルコクソン検定)を用いてそれらの3本の曲線の比較を行った。そうすると、CA5バッチの曲線はFSHバッチの曲線よりも上にあり、且つ、有意に異なっており(p<0.01)、同様に対照バッチの曲線とも有意に異なっている(p<0.001)。FSHバッチの曲線は対照バッチの曲線よりも上にあり、且つ、有意に異なっている(p<0.001)。
‐「CA5抗体」バッチ(n=7)は前記スポンジの撤去の24時間前に2mgのCA5の単回筋肉内注射を受け、
‐「対照」バッチ(n=9)は前記スポンジの撤去の24時間前に生理食塩水の筋肉内注射を受け、
‐「FSH」バッチ(n=11)は前記スポンジの撤去の24時間前に100μgのブタFSH(pFSH)の筋肉内注射を受け、そのスポンジの撤去の12時間前に90μgの筋肉内注射を受けた。
(1)刺激を受けたメスの100%において排卵を誘導することができ、
(2)FSH処理後に観察される分泌よりもずっと多いプロゲステロン分泌、および早くも4日目という非常に速いプロゲステロン分泌の成立を有する、より良好な品質の黄体を発生させることができる
という、FSHを用いる従来の処置よりも良好な結果が生じた。FSH処理と比較して追加的なCA5のこの特性の影響力は非常に重要であることが強調されるべきである。これは、血漿中プロゲステロン濃度が胚発生、特にその初期において重要な因子であるからである。
ラットおよび雌羊においてCA5モノクローナル抗体の増強作用を実証し、特徴解析した後、より大型の動物、すなわち若雌牛において内在性ゴナドトロピンの活性に対するその抗体の作用を研究することを目的とした。このため、前記抗体の効力を評価するためにその抗体単独の注射のみを含む処置をプリム・ホルスタイン種の若雌牛において開発した。これらの若雌牛はそれ以前にどのような卵巣刺激も受けておらず、それらの動物はゴナドトロピンを用いる排卵刺激のためのホルモン処理をその抗体の注射の前に受けていなかった。
‐ 2回の11mgの精製CA5抗体の筋肉内注射を受けた「CA5」バッチ(n=4):前記移植錠の留置から24時間後に1回目の注射が行われ、前記プロゲストゲン移植錠の撤去の24時間前に2回目の注射が行われた。
‐ 2回の生理食塩水の筋肉内注射を受けた「対照」バッチ(n=3):前記移植錠の留置から24時間後に1回目の注射が行われ、前記プロゲストゲン移植錠の撤去の24時間前に2回目の注射が行われた。

ラット、雌羊および若雌牛においてCA5モノクローナル抗体のインビボ増強作用を実証し、特徴解析した後、そのモノクローナル抗体の増強作用をヒトに近い種、すなわちカニクイザル(Macaca fascicularis)において検討した。このため、ヒトFSHおよび処置されるサルの内在性ホルモンに対する前記抗体の増強作用を評価することを目的として少なくとも36か月齢の思春期のサルに対して試験を行った。
‐8日間にわたって25IUで処置された動物(「25IU×8」バッチ):25IUのヒトFSH(ゴナールF(登録商標)プレフィルドペン、メルクセローノ)の毎日の皮下注射が8日間にわたって行われた。
‐1日だけ25IUで処置された動物(「25IU×1」バッチ):25IUのヒトFSH(ゴナールF(登録商標)プレフィルドペン、メルクセローノ)の単回皮下注射が処置の1日目に行われた。
‐CA5+hFSHで処置された動物(「CA5+hFSH」バッチ):処置の1日目だけに25IUのヒトFSH(ゴナールF(登録商標)プレフィルドペン、メルクセローノ)の注射から20分後にCA5抗体(80μg)の単回皮下注射が行われた。
‐CA5単独で処置された動物(「CA5」バッチ):処置の1日目だけにCA5抗体単独(88.5μg)の単回皮下注射が行われ、外因性のホルモンの注射は無かった。
巣超音波検査を48時間毎に行った。
CA5抗体およびCH10抗体のそれぞれのエピトープが、天然型立体構造と非天然型立体構造を区別することを可能にするボロノイ図と様々なスコア関数進化的学習法による最適化を用いるタンパク質構造モデリングに基づくプロテインドッキングアルゴリズム(Bernauer et al., Bioinformatics 2007, 5:555, [14]; Bernauer et al., Bioinformatics 2008, 24:652, [15]; Bourquard et al., PLoS One 2011, 6:e18541, [16] および Bourquard et al., Sci. Reports 2015, 5:10760 [17])を用いて様々な種の性腺刺激ホルモンに対して決定された。
Nature 2005, 433:269) [18]が使用された。他のホルモン(hLH、oFSH、oLH、pFSHおよびpLH)については相同モデルを作製し、そしてドッキングのために使用した。
CA5抗体の3D構造は入手できないのでCA5の一価VH断片と一価VL断片の配列を使用して検討を行った。このために可変部分の相同モデルを作製した。VHモデルとVLモデルを異なる構造から別々に作製し、VHモデリングを支援するものとして働いたその構造からそれらのモデルの相対的配向を決定した。相同モデルのために使用した構造であるCA5 VH用の3OKKとCA5 VL用の3MBXはプロテインデータバンク(PDB)において入手可能である。
CH10抗体の3D構造は入手できないのでCH10の一価VH断片と一価VL断片の
配列を使用して検討を行った。このために可変部分の相同モデルを作製した。VHモデルとVLモデルを異なる構造から別々に作製し、VHモデリングを支援するものとして働いたその構造からそれらのモデルの相対的配向を決定した。相同モデルのために使用した構造であるCH10 VH用の4QNPとCH10 VL用の3D85はプロテインデータバンク(PDB)において入手可能である。
ヒツジFSHおよびヒトFSH(ゴナールF(登録商標)、メルクセローノ)の生物活性を増強する能力を評価するためにCA5抗体の様々な断片を構築した。「CA5 VL」と呼ばれる軽可変鎖だけを含む断片、「CA5 VH」と呼ばれる重可変鎖だけを含む断片、および本発明の実施例1第4段落に記載されているCA5 scFvのVH−VL配列(配列番号19および配列番号20)と比較して逆のVL−VH順で構築された「リバースCA5 scFv」を作製した。
CA5抗体から得られたCA5 VL断片およびリバースCA5 scFv断片をコードする合成遺伝子がATG:バイオシンセティクスGmbH(ドイツ)によって合成された。リバースCA5 scFvはCA5 scFvのCA5 VL−リンカー−CA5
scFvのCA5 VHという融合体(配列番号19)から構成される。各合成遺伝子はpSW1プラスミド[7]の配列を融合することにより設計されており、HindIII部位とPelBタンパク質をコードする配列の末端との間に含まれ、そして合成予定の目的のタンパク質の配列(配列番号44および配列番号48)にはXhoI制限部位が横付けされる。それらの配列はpSW1プラスミドのHindIII部位とXhoI部位の間に挿入されている。大腸菌での発現向けにコドンを最適化した。
本発明の実施例2において先に記載されたプロトコルに従い、ヒトFSH受容体とGlosensor(登録商標)システムが安定的に形質移入されたHEK293細胞株を用いてヒツジおよびヒトFSHの生理活性に対する「CA5 VL」断片、「CA5 VH」断片および「リバースCA5 scFv」断片のインビトロ作用を検討した。単独の、または混合物としての「CA5 VL」断片および「CA5 VH」断片をそれぞれ40nMで検査した。リバースCA5 scFvをちょうど基準CA5 scFvのように80nMの濃度で検査した。ヒツジFSHおよびヒトFSH(ゴナールF(登録商標))を0.1nMで検査した。
断片およびVL断片単独で完全scFvと同様にoFSHの生理活性に対して増強作用を及ぼすことができることを示している。それらの結果より、本発明の実施例7のモデルに記載されている、その増強作用におけるそれらの2本の可変鎖の関与が立証されている。リバースCA5 scFv(VL−VH)と同様にCA5 VH+CA5 VL混合物は元のCA5 scFv(VH−VL)と同じ増強作用を及ぼす。
インビトロで特徴解析された後、hFSHの生理活性に対する様々なCA5断片の作用を特徴解析するためにそれらの断片の増強作用がメスのラットにおいてインビボで特徴解析された。

Claims (16)
- FSHの生理活性、黄体形成ホルモン(LH)の生理活性、および絨毛性ゴナドトロピン(CG)の生理活性を増強する卵胞刺激ホルモン(FSH)リガンドであって、抗FSH βサブユニット抗体のパラトープを含むことを特徴とする前記リガンド。
- 重鎖可変ドメインが次のCDRを含み、
‐配列GFTFSDFY(配列番号9)によって規定されるVH−CDR1、
‐配列SRNKAKDYTT(配列番号10)によって規定されるVH−CDR2、
‐配列ARDARFAY(配列番号11)によって規定されるVH−CDR3、且つ
軽鎖可変ドメインが次のCDRを含む
‐配列QSLLYSSNQKNY(配列番号12)によって規定されるVL−CDR1、‐配列WASによって規定されるVL−CDR2、
‐配列QQYYSYPRT(配列番号13)によって規定されるVL−CDR3
ことを特徴とする、請求項1に記載のリガンド。 - 重鎖可変ドメインが次のCDRを含み、
‐配列GFTFNTYA(配列番号14)によって規定されるVH−CDR1、
‐配列IRSKSNNYAT(配列番号15)によって規定されるVH−CDR2、
‐配列VRQDYYGSSYFDY(配列番号16)によって規定されるVH−CDR3、且つ
軽鎖可変ドメインが次のCDRを含む
‐配列QSISDY(配列番号17)によって規定されるVL−CDR1、
‐配列YASによって規定されるVL−CDR2、
‐配列QNGHSFPYT(配列番号18)によって規定されるVL−CDR3
ことを特徴とする、請求項1に記載のリガンド。 - 前記リガンドがFab、Fab’、F(ab’)2、Fv、dsFv、scFv、ディアボディ、トリアボディ、テトラボディおよびナノボディからなる群より選択されることを特徴とする、請求項1から3の何れか一項に記載のリガンド。
- 前記リガンドがCNCM I−4801ハイブリドーマにより産生されるCA5モノクローナル抗体であることを特徴とする、請求項2に記載のリガンド。
- 前記リガンドがCNCM I−4802ハイブリドーマにより産生されるCH10モノクローナル抗体であることを特徴とする、請求項3に記載のリガンド。
- 前記scFvのペプチド配列が配列番号20または配列番号22の配列であることを特徴とする、請求項4に記載のリガンド。
- 医薬品として使用するための請求項1から7の何れか一項に記載のリガンド。
- 請求項1から7の何れか一項に記載のリガンドとFSHまたはその活性ペプチドとの複合体、
請求項1から7の何れか一項に記載のリガンドとLHまたは絨毛性ゴナドトロピン(CG)ホルモン、または前記ホルモンの活性ペプチドとの複合体
から選択されるリガンド−ゴナドトロピン複合体。 - 医薬品として使用するための請求項9に記載の複合体。
- 前記医薬品がメスの哺乳類動物において排卵または多排卵を誘導することを目的としているものである、請求項8に記載のリガンドまたは請求項10に記載の複合体。
- 前記医薬品がメスの哺乳類動物において循環内在性プロゲステロンレベルを上昇させることを目的としているものである、請求項8に記載のリガンドまたは請求項10に記載の複合体。
- メスの哺乳類動物において排卵または多排卵を誘導することを目的としている医薬組成物であって、請求項1から7の何れか一項に記載のリガンドおよび/または請求項9に記載の複合体および薬学的に許容可能な担体を含むことを特徴とする前記医薬組成物。
- FSH、LHおよび/またはCGも含むことを特徴とする請求項13に記載の医薬組成物。
- 哺乳類動物の不妊症または低受胎症の治療および/または予防に使用するための請求項1から7の何れか一項に記載のリガンドまたは請求項9に記載の複合体。
- 健康なメスの哺乳類動物における生殖を刺激するための請求項1から7の何れか一項に記載のリガンドまたは請求項9に記載の複合体の使用。
