デングウイルス型1〜4(DEN−1からDEN−4)は蚊媒介性フラビウイルス病原体である。フラビウイルスゲノムは、5’非コード領域(5’−NC)、それに続くカプシドタンパク質(C)コード領域、それに続くプレメンブレン/メンブレンタンパク質(prM)コード領域、それに続くエンベロープタンパク質(E)コード領域、それに続く非構造タンパク質(NS1−NS2A−NS2B−NS3−NS4A−NS4B−NS5)をコードする領域、最後に3’非コード領域(3’NC)を含む。ウイルス構造タンパク質はC、prMおよびEであり、非構造タンパク質はNS1〜NS5である。構造タンパク質および非構造タンパク質は、単一のポリプロテインとして翻訳され、細胞内プロテアーゼおよびウイルスプロテアーゼによってプロセシングされる。
本願に開示のフラビウイルスキメラは、対応するPDK−53遺伝子を除去し、それをデング−1、デング−3またはデング−4ウイルス遺伝子または当分野で既知の他の遺伝子に置換する組換え操作を用いて、それに対する免疫が望まれるフラビウイルスの1つ以上の構造タンパク質遺伝子をPDK−53デングウイルスゲノム骨格にスプライシングすることにより、または当分野で知られている他の方法により製造することができる。

DEN−2 16681ウイルスのNS1−53−Glyは、今日までに配列解析されているダニ媒介性ウイルスを含むほぼ全てのフラビウイルスにおいて保存されているため、DEN−2 PDK−53ワクチンウイルスにおけるNS1−53突然変異は、このウイルスの弱毒表現型に重要である。DEN−4ワクチンウイルスはまた、NS1タンパク質の253位にアミノ酸突然変異を含むことができる。この位置は、DEN−4 PDK−48ワクチンウイルスにおいてはGlnからHisへの突然変異であり、デングウイルスの全4種の野生型血清型でGlnである。このGln残基は、フラビウイルス属の中で、デングウイルスに特有である。NS1タンパク質は、フラビウイルスに感染した細胞から分泌される糖タンパク質である。これは感染細胞の表面に存在し、NS1特異的抗体はウイルスに感染した個体の血清に存在する。NS1タンパク質で免疫された動物またはNS1特異的抗体で受動的に免疫された動物の防御が報告されている。NS1タンパク質は早期ウイルスRNA複製に関与すると考えられる。
本願に記載のワクチンウイルスの各々の診断である迅速遺伝子試験が本発明により提供される。本発明のこの実施形態は、ウイルス血症を発症したワクチン接種されたヒトの血清から単離されたウイルスの解析を促進し、また、候補ワクチンウイルスで免疫された非ヒト霊長類におけるウイルス血症の特徴解析を促進する。
本願の実施形態は、対象に対する、in vivo薬剤投与に適した生物学的に適合性の形態での組成物の投与を提供する。「in vivo投与に適した生物学的に適合性の形態」は、活性作用剤(例えば、実施形態の医薬化学物質、タンパク質、遺伝子)の投与される形態であって、活性作用剤の治療効果があらゆる毒性効果を上回る形態を意味する。薬学的に活性な量の医薬組成物の投与は、所望の結果を達成するのに必要な用量および期間での有効な量と定義される。例えば、薬学的に活性な量の化合物は、個体の病気の状態、年齢、性別、および体重、ならびに個体において所望の応答を誘発する抗体の性能等の要因に応じて変動し得る。投薬レジメンは、最適な治療的応答を提供するように調整され得る。
一実施形態において、本願に開示の組成物は対象に皮下または皮内投与することができる。
治療方法
本発明の一実施形態において、方法は、本願で企図される弱毒生および/またはキメラウイルス構築物の一価、二価、三価または四価製剤を用いたデングウイルス血清型に対する免疫応答の誘導を提供する。
以下の実施例は、本明細書に示される特定の実施形態を実証するために含められる。以下の実施例において開示された技術が、本明細書に開示される実施において十分に機能することが見出されている代表的な技術に従っており、よってその実施の好ましい形態を構成するとみなすことができることは、当業者には理解されるはずである。しかしながら、当業者は本開示に鑑みれば、開示されるある種の実施形態では、本願の趣旨および範囲から逸脱することなく、多くの変更が可能であり、依然として同様のまたは類似の結果を得られることを理解するはずである。
いくつかの例示的方法において、本願では「マスターウイルスシード(MVS)」と呼ぶものを作製するために使用される組成物が開示される。これらの組成物は、DEN−1、DEN−2、DEN−3、およびDEN−4等の1つ以上の弱毒生デングウイルスから誘導され得る。特定の方法では、組成物は、DENVax−1、DENVax−2、DENVax−3、およびDENVax−4と呼ばれる本願に開示される特定の構築物を含むがこれらに限定されない1つ以上の弱毒生デングウイルスから誘導され得る。他の例示的方法において、これらの組成物を製造し特徴解析するのに用いられる方策が提供される。さらに他の実施形態において、四価デングウイルス製剤およびこれらの製剤の遺伝子のおよび表現型の特徴解析が提供される。
デングウイルス(例えばDENVax)の連続的増幅および精製等の特定の手順を実施してプレマスターデングウイルスシードを作製した。まず、完全長組換えDENVax cDNAから転写されたウイルスRNAを産生の保証された細胞(例えばVero細胞)に導入することによりDENVaxウイルスを再誘導して、P1(継代1)ウイルスシードを得た。デング−1からデング−4の各々からの4つのP1ウイルスを、続いて増幅し、プラークを精製して候補プレマスターワクチンP7シードを得た(表1参照)。特定の試験を実施してデングウイルスの各継代を解析した。例えば、完全長ゲノム配列解析により、P2(継代2)シードウイルスの全4種が、それらの同種の祖先である、研究的に誘導されたリサーチグレードの候補ワクチンウイルスと遺伝子的に同一であることが実証された。元のプラーク表現型もP2ウイルスで維持されていた。デングウイルスの各血清型(例えばDENVax1〜4)についてP2シードから6つのプラーク精製ウイルス(P3A〜F)を単離し、各単離プラークを更に2回、直接プラーク精製した。各ウイルスの3回目のプラーク精製物(P5)をVero細胞で2度増幅し(P6A〜FおよびP7A〜F)、潜在的なプレマスターP7DENVaxシードを作成した(表1)。
いくつかの例示的方法において、マスターウイルスシード、ワーキングウイルスシードおよびバルクウイルスシードの組成物、ならびにそれらの遺伝子のおよび表現型の特徴解析が説明される。これらの組成物は、臨床材料の製造、そして究極的には市販のワクチン供給のために提供される。製造されたワクチンシードの組成物が、治験材料の製造に最適な純度および遺伝子安定性であるようにするため、製造工程には連続的プラーク精製およびフルゲノム配列解析を含めた。
いくつかの研究では、保証されたVero細胞中でプレマスターP7シードを増幅することにより4DENVaxのMVSを製造した。他の研究では、MVSを用いて細胞ファクトリーで大量のWVSを作製した。さらに、DENVaxのBVSストックをWVSから増幅し、ヒト治験用の四価薬剤製品混合物として製剤化した。製品リリースの品質管理は、MVS、WVS、およびBVSの全てについての同一性、感染力価、無菌性、マイコプラズマ、ならびにin vitroおよびin vivo外来物質(adventitious agent)試験を含むがこれらに限定されないいくつかの例示的方法で実施された。全てのシードが、血清型特異的RT−PCRアッセイを用いたウイルス同一性試験に合格し、これは、その血清型に対応する正の増幅および異種血清型の負の増幅を示した(データは示されていない)。マイコプラズマおよび外来物質は、MVS、WVS、またはBVSストックでは検出されなかった。
ある例示的な方法において、選択されたプレMVS(P7)からのMVSの作成後、上記で選択された株を生成し、個々のウイルスRNAを再度配列解析した。完全長ゲノム配列解析により、DNVax−1のMVSはそのプレマスターシードと同一であったが、WVSおよびその後のBVSは、E−483およびNS4B−108に2つの更なる置換を獲得したことが判明した(表2および3参照)。E−483のAla置換は、MVSでは遺伝子型の一部を代表したが、BVSでは主要な遺伝子型となった。DENVax−2およびDENVax−3はそれらのプレマスターシードのそれぞれと同一であった(表2および3)。DENVax−2 MVSはそのマスターシードと同一であり、WVSおよびBVSはNS4A−36およびNS4B−111に2つの更なる突然変異を有した。どちらの突然変異も、WVSでは部分的であり、BVSでは主要な遺伝子型であった。DENVax−3のMVSもやはりそのプレマスターシードと同一であったが、WVSおよびBVSはNS4A−23に更なるaa置換を含んでいた。DENVax−4 MVSは、MVSの製造中、NS2A−99位置に更なる(LysからLys/Arg混合遺伝子型への)アミノ酸突然変異を獲得した(表3)。そのWVSおよびBVSはNS2A−99Lys/Arg混合遺伝子型を維持し、BVSは更にNS4B−238Ser/Phe混合遺伝子型を有した。コンセンサス配列結果は、MVS、WVSおよびBVが、5’NCR−57、NS1−53、およびNS3−250位置の3つの弱毒化遺伝子決定要因を保持したことも裏付けた。TaqMAMAによる最低安定性弱毒化位置の解析では、5’NCR−57復帰率がMVSでは<0.7%から0.13%であり、WVSでは≦0.07%であり、BVSでは<0.07から0.21%であったことが実証された。5’NCR−57位置での3%の復帰は、ワクチンロットの受け入れについての最大許容率であると考えられた(表3)。
1つの例示的方法において、DENVax MVSのプラーク表現型を野生型デングウイルスと、およびこれらと同種のリサーチグレードキメラウイルスとVero細胞において比較した(図2)。DENVax−1、−2、および−3のMVSの全てが、それらの野生型ホモログよりも有意に小さく、かつそれらの同種リサーチグレードウイルスに非常によく似た(0.4mm差以内の)プラークをVero細胞内で形成した。また、DENVax−4 MVSは野生型DENV−4よりも有意に小さかったが、元の実験室由来のD2/4−Vキメラよりは僅かに大きかった(0.9mm差)。
別の例示的方法において、Vero細胞における温度感受性をDENVax MVSについて調査し、それらの同種野生型および元のリサーチグレードキメラワクチンウイルスと比較した。野生型(wt)DENV−3 16562は温度感受性ではなかった。wtデングウイルス血清型1およびデングウイルス血清型−4は、39℃で中程度に温度感受性であった(39℃での力価は、37℃よりも約1.0 log10pfu/ml低かった。図3)。wtデングウイルス血清型−2 16681は、調査したwtデングウイルスの中で最も温度感受性であり、39℃で力価が100分の1に低下した。DENVax−1、−2、および−3はそれらの元の同種リサーチグレードキメラワクチンウイルスと同程度の温度感受性であった(図2)。これらのDENVax株では、39℃での力価は2.0〜3.0 log10pfu/ml低下した。DENVax−4も温度感受性であり、力価の5分の1の低下を示した。しかし、元のリサーチグレードD2/4−Vは、力価の約10分の1の低下を示した。最終安定化DENVax−4 MVSはF127(および、これらの製剤を安定化することが知られている他の作用剤(FTA))を含み、これはデングウイルスの熱安定性を高めることが示された。DENVax−4 MVSにおけるF127の存在は、Vero細胞培養アッセイにおいて、ウイルスのあまりはっきりしない温度感受性に貢献するように思われる。別の実験において、F127の存在しないMVS誘導性DENVax−4株の温度感受性をさらに評価した。株からF127を除去するために、ウイルスRNAをDENVax−4バルクウイルス調製物から単離し、Vero細胞に導入した。このF127の存在しないDENVax−4ウイルスは、pi3日目、D2/4Vリサーチ株と同程度(力価が1.5 log10pfu/ml低下)の温度感受性のようであった(図3)。
リサーチグレードキメラワクチンウイルスはC6/36細胞において骨格DENV−2 PDK53ウイルスの弱毒表現型を維持したという知識の下、いくつかの例示的方法において、DENVax MVSをC6/36細胞で増殖させ、これらのin vitro弱毒表現型の維持について評価した。wtデングウイルスに比べ、DENVax−1、DENVax−2およびDENVax−4 MVSは、C6/36細胞においてpi6日目に有意な増殖低下(少なくとも3 log10pfu/mlの低下)を示した(図4)。DENVax−3 MSVも、wtDENV−3 16562と比べて増殖の低下を示したが、それほど顕著な低下ではなかった(1〜2 log10pfu/mlの低下)。しかしながら、DENVax−3シードロットのC6/36力価は元のリサーチグレードキメラD2/3−VワクチンウイルスのC6/36力価に類似していた(1 log10pfu/ml以内の差)。
いくつかの例示的方法において、DENVaxの感染率および播種率をそれらの親のwtデングウイルスと比較した。ある例示的な実験において、ネッタイシマカで経口感染実験を実施した。感染性の血液飼料を逆滴定してウイルス力価を測定し、各血清型について親のデングウイルスとDENVaxとの間でウイルス力価が類似している(1 log10pfu/ml未満の差)血液飼料を用いた実験のみを、比較のために表4に含めた。DENVax−1、DENVax−2、およびリサーチグレードD2 PDK−53−VV45Rは、経口給餌では蚊を感染させず、これはそれらの親ウイルスであるDENV−1 16007(44%感染)およびDEN−2 16681(43.3%感染)とは有意に異なっている(p<0.0001)。DENVax−1および−2によって感染した蚊は存在しなかったため、これら2つのワクチンウイルスについて播種の心配はほとんどあるいは全くなかった。DENVax−4は一部の蚊を経口給餌により感染させたが(55匹中2匹)、感染率は親wtウイルスのDENV−4 1036(50匹中8匹)よりも有意に低かった(p<0.05)。DENVax−3は、ウイルス力価5.2±0.02 log10pfu/mlの血液飼料を用いた2つの実験(表4)では蚊を全く感染させず、ウイルス力価6.0 log10pfu/mlの血液飼料を用いた別の実験では30匹中1匹の蚊だけが感染した(データは示されていない)。しかしながら、wtデングウイルス−3 16562も5.2 log10pfu/mlでの感染率は非常に低く(8%)、この率は、6.2 log10pfu/mlのより高い血液飼料ウイルス力価を用いた別の実験でも上昇しなかった(3%、蚊30匹中1匹が陽性、データは示されていない)。野生型(wt)デングウイルス−3およびデングウイルス−4はwtデングウイルス−1およびデングウイルス−2よりも有意に低い感染率であったが、感染した蚊における平均ウイルス力価は類似していた(3.1〜3.9 log10pfu/蚊)。対照的に、2匹の感染した蚊のDENVax−4力価は両方とも最低限(0.7 log10pfu/蚊)であり、これはwtデングウイルス血清型−4 1036に感染した蚊の力価(3.9±1.5pfu/蚊)の1000分の1であった。
元のリサーチグレードワクチンウイルスは、DVBD/CDCでセンター内に維持された新生ICRマウスにおける神経毒性に関して高度に弱毒化された。これらのマウスは全て、104pfuでの各ワクチンウイルスのic(脳内)攻撃を生き延びた。一方、wtデングウイルス血清型−2 16681ウイルスは、様々な実験においてこれらのCDC−ICRマウスの62.5%〜100%の死亡率をもたらした。いくつかの実験では、タコニック・ラブズ(Taconic Labs)から入手した市販のICRマウス(Taconic−ICR)を用いて新生マウスにおける神経毒性を調査した。新生Taconic−ICRマウスは、以前のCDC−ICRマウスよりも、デングウイルス血清型−2に対して有意に感染しやすいことが観察された。図5Aは104pfuのウイルスでic攻撃されたTaconic−ICR新生マウスおよびCDC−ICRコロニーにおけるwtデングウイルス血清型−2 16681の神経毒性をまとめたものである。Taconic−ICRマウス(32匹のマウスで100%死亡率、平均生存時間8.3±0.5日)は、以前のCDC−ICRマウス(72匹のマウスで91%致死率、平均生存時間14.6±2.3日)よりも、icデングウイルス血清型−2 16681攻撃に対し、より影響を受けやすかった。
WVSおよびBVSのプラーク表現型をMVS、wtデングウイルスおよびそれらの同種の実験室由来のリサーチグレードキメラとVero細胞において比較するために、ある調査を行った(図6)。各ワクチンウイルスにつき10個(ただしwtDENV−1、−3、および−4については減らした数)のプラークから平均プラークサイズを計算した。DENVax−1、−2、および−3のMVSウイルスはいずれも、Vero細胞において、それらのwtホモログよりも有意に小さく、同種リサーチグレードウイルスと非常に近い(0.4mm差以内)プラークを形成した。DENVax−4 MVSもwtDENV−4よりも有意に小さかったが、元の実験室由来のD2/4−Vキメラよりは僅かに(0.9mm)大きかった。DENVax−2を除き、DENVax−1、−3、−4のWVSおよびBVSはいずれも、それらのwtホモログから形成されるプラークよりも有意に小さいプラークサイズを維持した。DENVax−2 WVSおよびBVSは、Vero細胞においてwtDENV−2ウイルスのプラークに近いプラークを形成したが、LLC−MK2細胞で試験したところ、DENVax−2製造シードの全てが、wtDENV−2のものよりも幾分小さく(1.4±0.4)、実験室由来のD2 PDK−53−VV45Rに近い(1.0±0.3)プラークを形成した(図6)。
以前の研究により、リサーチグレードPDK−53ベースキメラワクチンウイルスは、C6/36細胞内で骨格DENV−2 PDK53ウイルスの弱毒表現型を維持することが実証された。いくつかの例示的方法において、DENVax MSV、WVS、およびBVSをC6/36細胞内で増殖させ、大規模製造後のこのin vitro弱毒化マーカーの維持について評価した。wtデングウイルスと比較して、DENVax−3を除き、製造されたシードはC6/36細胞内でpi6日目に顕著な増殖低下(少なくとも3 log10pfu/ml)を示した(図7)。DENVax−3シードもwtDENV−3 16562と比べて低下した増殖を示したが、それほど顕著な低下ではなかった(1〜2 log10pfu/ml)。しかし、DENVax−3シードロットの力価は元のリサーチグレードキメラD2/3−Vワクチンウイルスと同程度であった(1 log10pfu/ml差以内)。
新生ICRマウスにおける神経毒性を解析するために更なる実験を行った。頭蓋内への104PFUの用量で、wtDENV−2 16681およびD2 PDK−53−VV45Rに対する生存率は、ICRマウスではそれぞれ0%および18.8%であった(図9A)。しかし、CDC ICRマウスでは、wtDENV−2 16681に対しては約20%であり、D2 PDK−53−VV45Rに対しては100%であった。この研究では、104PFUの用量のDENVax−1およびDENVax−3 MVSはマウスに対して弱毒化されたが(生存率100%)、DENVax−2およびDENVax−4のMVSは、104PFUを超える用量で100%の死亡率をもたらした(図5A)。しかしながら、103PFU用量のウイルスで試験したところ、DENVax−2(生存率31.3%)およびDENVax−4(生存率81.3%)はwtデングウイルス血清型−2 16681(生存率0%)に対し低下した神経毒性を示し、これらの生存率は、それぞれリサーチグレードワクチン候補D2 PKD−53−VV45R(32.3%)およびD2/4−V(100%)と同程度であった(図9B)。wtDENV−1、−3、または−4は、この研究では比較のために含めなかったが、以前の研究はwtDENV−1 16007がCDC−ICRマウスにおいてic経路により弱毒化されたのに対し、wtDENV−3 16562およびDENV−4 1036は両方ともCDC−ICRマウスに対し高毒性(生存率0%)であったことを実証した。これら3つのwtDENVは、より感受性の高いTaconic ICRマウスでは同程度またはそれ以上の毒性を示すと考えられる。したがって、これらのwtデングウイルスをそれらの同種DENVax MVSとの比較のために含めることは無益であると考えられた。この研究は、全4種のDENVax MVSおよび元の実験室由来の候補ワクチンウイルスが、wtDENV−2 16681と同程度のマウス弱毒表現型を提示することを示した。
DENVaxの全シードロットを、同一性、無菌性、および望ましくない物質の不在について試験した。フルゲノム配列解析により、1つの余分なアミノ酸突然変異がDENVax−4 MVSに生じたが、他の3つのDENVax MVSは、それらのプレマスターシードのコンセンサスゲノム配列を保持していることが判明した。WVSロットでは、DENVax−3は1つの余分なアミノ酸突然変異を獲得し、他の3つの血清型は、それらのプレマスターシードに対し2つの余分なアミノ酸置換を蓄積した。4つのBVSロット全てのゲノム配列がそれらのWVSロットと同一であった。P2シードからプレマスター(P7)シードまでの全体で、僅か1つまたは2つの非サイレント変異が所与のシードに生じた。プレマスターシードとBCS(P10)シードとの間では、僅か1〜2個のヌクレオチド置換が見られ、それらはいずれもNS2A、4A、または4Bに生じた(残基E−483の保存されたグリシンおよびアラニンをもたらす単一ヌクレオチド変化は例外)。P2からBVS(P10)シードまで、合計で3〜8個のヌクレオチド置換が所与のDENVaxシードで同定され、これらの置換のうち僅か2〜4個がアミノ酸変化をもたらした。BVSにおけるサイレント変異はいずれも、ウイルス複製に影響を及ぼし得る5’−および3’−NCR領域内にはなかった。これらの結果は、DENVaxウイルスが、製造中、遺伝子的に高度に安定であることを示唆している。5’NCR、NS1−53、およびNS3−250に位置する定義された3つのDENV−2 PDK−53弱毒化位置は、BVSストックを生成するためにDENVaxを連続継代しても、コンセンサスゲノム配列において変化していなかった。5’−NCR−57位置の高感度TaqMAMAは、DENVaxのMVS、WVS、およびBVSにおける最少のまたは検出不能な復帰を示した。0.21%の最高の復帰率がDENVax−2 BVSにおいて確認された。P10等価BVSの復帰率(<0.07%〜0.21%)は、Vero細胞での連続継代後に他のワクチン候補に生じた復帰率よりも有意に低かった(P10までに4〜74%の復帰)。これは、DENVaxシードのこの大規模製造の方策が、候補ワクチンウイルスにおいて遺伝子安定性を維持し、弱毒化マーカーを保持することに関して成功することを示唆している。
ウイルスおよび細胞
DENV−1 16007、DENV−2 16681、DENV−3 16562、およびDENV−4 1034を野生型(wt)DENV対照とし、これらは4つの組換えDENVaxワクチン候補の親遺伝子型ウイルスであった。DENVax前駆リサーチグレードウイルスとして、D2/1−V、D2 PDK−53−VV45R、D2/3−V、およびD2/4−Vと称するものが、以前に調製および特徴解析された。ワクチン製造のためのマスターおよびワーキングセルバンクの作製に用いたVero(アフリカミドリザル腎臓)細胞は、世界保健機関(WHO)によってワクチン製造について特徴解析されている米国培養細胞系統保存機関(American Type Culture Collection:ATCC)CCL81細胞株(WCB−Vero細胞)を起源とした。
cGMP製造条件下で候補ワクチンウイルスを再誘導するため、以前に作出されたDENV感染性cDNAクローンである、完全ゲノム長ウイルスcDNAを含有するpD2−PDK−53−VV45R、pD2/1−V、pD2/4−V、およびin vitroライゲーションpD2/3−Vを用いて、以前に説明されたようなin vitro転写により新しいウイルスRNA転写産物を生成した。簡潔に述べると、XbaI直鎖化DENVゲノムcDNAをプロテイナーゼKで処理し、フェノール/クロロホルムで抽出し、エタノール中で沈殿させてあらゆる残留タンパク質を除去した。その後、転写前にRNaseフリーのTris−EDTAバッファーに懸濁させた。AmpliScribe T7高収率転写キット(エピセンターテクノロジーズ(Epicentre Technologies))を用いて、メーカーの推奨プロトコルにしたがってin vitro転写を実施した。RNA転写産物に5’末端Aキャップを付加するため、RNAのAキャップアナログであるm7G(5’)ppp(5’)A(ニューイングランドバイオラブズ(New England BioLabs))を、2時間の転写反応中に組み込ませた。続いて試料をDNaseIで処理してテンプレートcDNAを消化し、その後、低pHフェノール/クロロホルム抽出およびエタノール沈殿を行って残留DNAおよびタンパク質を除去した。精製されたRNA転写産物をRNaseフリーの水に懸濁させ、20μlのアリコートに分けて、細胞の形質転換の準備が整うまで−80℃で保管した。RNA転写産物の完全性および濃度をアガロースゲル電気泳動により解析した。各20μlアリコートは、エレクトロポレーションで0.4〜1×107個の産生保証Vero細胞を形質転換するのに十分なゲノム長のウイルスRNAを含んでいると推定された。
製造されたロットの最適な遺伝子安定性および安全性を確実にするように設計された方策によって、P1ウイルスシードを用いてDENVaxプレマスター、マスター、ワーキング、およびバルクウイルスシードロットを増殖させた。この方策は、3回の連続したプラーク精製、および、継続的なシード製造用の最適なクローンウイルス集団を選択するための様々な継代レベルでのウイルスの遺伝子解析を含んでいた(表1)。簡潔に述べると、形質転換した細胞から回収したP1シードを、0.001のMOIでのVero細胞の感染により一度増幅させ、P2シードを生成した。P2シードストックのアリコートをプラーク表現型および完全ウイルスゲノム配列解析により評価した。遺伝子が確認されたP2ストックを、下記のプラーク滴定の節で説明するようなオーバーレイ培地を伴うVero細胞単層にプレーティングし、十分に単離されたプラークを形成した。ニュートラルレッドで可視化した後、ワクチンウイルスの4種の血清型の各々から6つの別個のプラークを単離し(プラーククローンA〜F)、0.5mlの培地と混合した(継代P3)。
MVS、WVS、およびBVSシードを、同一性、無菌性、および検出可能な外来物質について試験した。各ワクチンストックの同一性は、DENVax血清型特異的プライマーを用いたRT−PCRにより確認した。増幅されたcDNAフラグメントは、4種のDENVax血清型の各々の同定を可能にするE/NS1キメラジャンクション部位を含んでいた。各シードは、ウイルスの同一性および異種DENVax血清型による交差汚染の不在を確認するために、全4種の血清型特異的RT−PCR反応において試験した。無菌性試験はUSP71(米国薬局方、セクション71)に準拠して実施した。マイコプラズマ試験を実施した。
Vero細胞を用いたプラークアッセイまたはイムノフォーカスアッセイによりウイルス力価を測定した。プラークアッセイは、以前に説明されたように、コンフルエントVero細胞の6ウェルプレートにおいて二重アガロースオーバーレイで実施し、これらはDENVaxシードのプラーク表現型を評価するのにも利用した。正確な比較のため、全てのウイルスのプラークサイズを測定し、同じ実験内で比較した。pi9日目にニュートラルレッドで可視化した後、平均プラークサイズ計算のために最大10ウェルの単離したプラークを各ウイルスにつき測定した。wtDENV−1、−3、および−4については、プラークサイズが大きく、しばしば10ウェル単離プラークの測定が不可能であり、測定されたプラークの数は少なくなった。
MVSおよびWVSの完全長ゲノムを配列解析した(下記参照)。簡潔に述べると、QIAmpウイルスRNAキット(キアゲン(Qiagen))を使用してウイルスRNAをDENVaxシードから抽出し、Titan One Tube RT−PCRキット(ロシュ・アプライド・サイエンス社(Roche Applied Science,Inc.))を使用してゲノム全体をカバーする重複したcDNAフラグメントを増幅した。増幅したcDNAフラグメントをゲルで精製した後、BigDye Terminator v3.1サイクルシークエンシングキット(アプライドバイオシステムズ(Applied Biosystems))を用いて、フォワードおよびリバースプライマーにより配列解析した。BigDye XTerminator Purificationキット(アプライドバイオシステムズ)を用いてシークエンス反応をきれいにし、DVBD/CDCで3130xl Geneticアナライザー(アプライドバイオシステムズ)にかけた。ゲノム解析および比較にはLasergene SeqManソフトウェア(DNAスター社(DNAStar,Inc))を用いた。
TaqMAMAは、弱毒化の5’NC−57位置における復帰レベルのより精密な評価を可能にするために開発された高感度の定量的単一ヌクレオチド多型性アッセイであり、この研究のために更に最適化された。MVSおよびWVSから抽出されたウイルスRNAは、wtまたはワクチン5’NC−57領域に特異的なプライマー/Taqmanプローブの両対を用いたTaqMAMAにより解析された。DENV−2のwt配列およびワクチン配列を検出するのに使用したフォワードプライマーは、それぞれD2−41−GCおよびD2−40−TTであった。各ウイルスについて、各フォワードプライマーの3’末端ヌクレオチドは特異的5’NCR−57ヌクレオチドと一致していたが、各プライマーの3’末端ヌクレオチドに隣接するヌクレオチドは、ミスマッチ効果を高めるため、DENV−2ウイルスゲノム配列とは異なっていた。リバースプライマーのCD−207とTaqman(登録商標)プローブのCD−169Fは、wtおよびワクチンセットの両方とも同一であった。プライマーおよびプローブの配列、ならびにサイクル条件は以前に説明された。リアルタイムRT−PCRを、iQ5またはCFX−95システム(バイオラッド)で、BioRad iScript RT−PCR(プローブ用)キットを用いて、5μlのウイルスRNAテンプレート、0.4uMの各プライマー、および0.2uMのプローブを含有する25μl反応で行った。各試料につき、各wt特異的およびワクチン特異的反応についてトリプリケート反応を行った。各ウイルス血清型について用意した標準曲線に対してゲノムコピー数を決定した。ここで、RNA標準は各遺伝子型特異的cDNAのnt1〜2670を含むプラスミドから誘導された転写産物であった。さらに、全ての実験で最少の交差反応性を確実にするため、異種遺伝子型プライマー/プローブセットにより各RNA標準を試験することによりアッセイの特異性を確認した。結果を、復帰を示したウイルスゲノムの割合として報告した。以前、このアッセイのインプットRNAレベルを制限する高い交差反応性バックグラウンドのために、当初の検出感度は約0.1%の復帰(識別能)であった。以降、改善されたリアルタイムPCR装置および反応キットを用いてアッセイは更に最適化されて、交差反応性バックグラウンドは遥かに高レベル(7〜8 log10コピー)のRNAテンプレートインプットに大幅に低減された。この最適化は、0.01〜0.07%の復帰率にまで、検出感度のかなりの向上をもたらした。
4つのDENVax MVSストックおよびwtDENV−1、−2、−3、および−4ウイルスの複製表現型をC6/36蚊細胞(Aedes albopictus)で評価した。6ウェルプレートで増殖させたC6/36細胞を、0.001のMOIの各ウイルスにデュプリケートで感染させ、28℃、5%CO2のインキュベーター中、4ml/ウェルの2%FBS含有DMEM培地でインキュベートした。各ウイルスについてpi6日目に培養上清の小アリコートを回収し、40%のFBSを含有する同じ体積の培地と混合し、ウイルスプラーク滴定の準備が整うまで−80℃で保管した。
研究に用いたネッタイシマカは、タイ王国メーソート近郊の村(16’N,33’E)に由来する2002年に確立されたコロニーに由来した。幼虫から変態後、成虫の蚊を28℃、16:8(明:暗)の光周期で、10%スクロース溶液を自由摂取で提供することにより維持した。5〜7日齢のメスの蚊を感染性血液飼料供給または胸腔内(IT)接種に使用した。後述するような経口感染用のウイルス血液飼料を作るため、新たに培養したDENVaxおよびwtDENVのアリコートを回収後直ちに(凍結解凍サイクルを経ずに)使用した。残りのウイルス上清にはFBSを最終濃度20%で添加し、アリコートを後のウイルスプラーク滴定およびIT接種実験のために−80℃で保管した。これらの実験用に新たに調製したDENVaxシードをプレマスターシードからVero細胞内で増幅し、これらはDENVax MVS等価物とみなした。
タイミング調整した妊娠メスICRマウスをタコニックラブズから入手し、各腹の子マウスのおおよその出産時期を決定するために毎日数回モニタリングした。所与の実験において、出生の約12〜24時間後、各ウイルスにつき8匹の子マウス二腹分(n=16)を、30ゲージ針を用いて20μlの希釈剤中103〜104pfuのウイルスで頭蓋内(ic)攻撃した。攻撃後、動物を少なくとも毎日3回、少なくとも32日間モニタリングした。疾患の最初の徴候が出たとき(被毛粗剛、屈背、体重減少、異常な動き、まひ、または無気力)、動物をイソフルランガスでの致死麻酔により安楽死させ、その後、頸椎脱臼させた。感染後安楽死までの日数は、動物の「疾患/病的状態までの時間」または「生存時間」を表した。動物実験はDVBD/CDC IACUCに認可された動物プロトコルに従って実施された。
