フィブロネクチン
フィブロネクチンは、選択的スプライシングに供される抗原であり、本明細書の他所に記載する通り、数々の代替的なフィブロネクチンのアイソフォームが知られている。エキストラドメイン−A(EDAまたはED−A)も、ED、エキストラタイプIIIリピートA(EIIIA)、またはEDIとして知られている。ヒトED−Aの配列は、Kornblihttら(1984年)、Nucleic Acids Res.、12巻、5853〜5868頁、およびPaolellaら(1988年)、Nucleic Acids Res.、16巻、3545〜3557頁によって公表されている。ヒトED−Aの配列はまた、受諾番号P02751の下に寄託されているアミノ酸配列のアミノ酸1631〜1720(フィブロネクチンIII型12;エキストラドメイン2)として、SwissProtデータベース上で入手できる。マウスED−Aの配列は、受諾番号P11276の下に寄託されているアミノ酸配列のアミノ酸1721〜1810(フィブロネクチンIII型13;エキストラドメイン2)として、SwissProtデータベース上で入手できる。
選択的スプライシングは、DNAの一次RNA転写物のスプライシングの様々なパターンが生じて様々なmRNAを生成することを指す。イントロンを切り出した後、選択により、mRNAを形成するのにどのエクソンが一緒にスプライシングされるかが決定され得る。選択的スプライシングにより、様々なエクソンおよび/または様々な数のエクソンを含む様々なアイソフォームの生成がもたらされる。例えば、1つのアイソフォームは、1つまたはそれ以上のエクソンに対応するさらなるアミノ酸配列を含むことができ、1つまたはそれ以上のエクソンは1つまたはそれ以上のドメインを含むことができる。
これは、相互に特異的に結合する1対の分子の1メンバーを記述するものである。特異的な結合対のメンバーは、天然由来でも、または全体的もしくは部分的に合成で生成されていてもよい。分子の対の1メンバーはその表面上、または孔上に、分子の対の他のメンバーの特定の空間および極性の組織化に結合し、ゆえに相補的である区域を有する。結合対のタイプの例として、抗原−抗体、ビオチン−アビジン、ホルモン−ホルモン受容体、受容体−リガンド、酵素−基質がある。本発明は、抗原−抗体タイプの反応に関する。
これは、天然でも、または部分的もしくは完全に合成で生成されたものでも、免疫グロブリンを記載するものである。この語は、抗体の抗原結合部位を含めた、任意のポリペプチドまたはタンパク質にも関する。本明細書において、本発明は天然の形態の抗体に関するものではなく、すなわち抗体は天然の環境におけるものではなく、天然の源から精製によって単離および入手することができ、またはそうでなければ遺伝的組換えによって、もしくは化学合成によって得ることができ、次いで、これらは後に記載する非天然のアミノ酸を含むことができることを理解されたい。抗体の抗原結合部位を含む抗体フラグメントには、それだけには限定されないが、Fab、Fab’、Fab’−SH、scFv、Fv、dAb、Fdなどの抗体分子、およびダイアボディが含まれる。
よび(ix)遺伝子融合によって構築される多価または多特異性のフラグメントである「ダイアボディ」(WO94/13804;Holligerら(1993年a)、Proc.Natl.Acad.Sci.USA、90巻、6444〜6448頁)がある。Fv、scFv、またはダイアボディ分子は、VHとVLドメインとを連結するジスルフィド架橋を組み込むことによって安定化することができる(Reiterら(1996年)、Nature Biotech、14巻、1239〜1245頁)。CH3ドメインに接続しているscFvを含むミニボディも作製することができる(Huら(1996年)、Cancer Res.、56巻(13)、3055〜61頁)。結合性フラグメントの他の例にはFab’およびFab’−SHがあり、Fab’は、抗体のヒンジ領域からのシステインを1つまたはそれ以上含めた、重鎖CH1ドメインのカルボキシル末端に数個の残基を加えることによりFabフラグメントと異なり、Fab’−SHは、定常領域のシステイン残基(複数可)が遊離のチオール基を保有するFab’フラグメントである。
これは、標的の抗原の全てまたは部分に結合し、相補的である分子の部分を記載するものである。抗体分子では、これは抗体の抗原結合部位と呼ばれ、標的の抗原の全てまたは部分に結合し、相補的である抗体の部分を含む。抗原が大型である場合、抗体は抗原の特定の部分にのみ結合し得、この部分をエピトープと呼ぶ。抗体の抗原結合部位は、1つまたはそれ以上の抗体の可変ドメインによって提供され得る。抗体の抗原結合部位は、抗体の軽鎖可変領域(VL)および抗体の重鎖可変領域(VH)を含むことができる。
これは、本発明において用いるための特異的な結合メンバー、またはこのような特異的な結合メンバーをコードする核酸が、概ね本発明にしたがう状態を述べることを指す。このように、特異的な結合メンバーである、本発明のVHおよび/またはVLドメインは、例えば、これらの天然の環境から、実質的に純粋な、もしくは均一な、または核酸の場合は、必要とされる機能を有するポリペプチドをコードする配列以外を起源とする核酸もしくは遺伝子がない、もしくは実質的にない形態において、単離および/または精製されて提供され得る。単離されたメンバーおよび単離された核酸は、天然の環境において、またはこのような調製がin vitroまたはin vivoで行われる組換えDNA技術による場合は、これらが調製される環境(例えば、細胞培養物)において、これらが一緒に見出される他のポリペプチドまたは核酸など、これらが天然に関連する物質がなく、または実質的にない。特異的な結合メンバーおよび核酸は、希釈剤またはアジュバントと調合し、さらに実際的な目的で単離することができ、例えば、メンバーは通常、免疫アッセイで用いるためのマイクロタイタープレートをコーティングするのに用いる場合はゼラチンもしくは他の担体と混合され、または診断もしくは治療において用いる場合は薬学的に許容される担体もしくは希釈剤と混合される。特異的な結合メンバーは、天然に、もしくは異種性の真核細胞(例えば、CHOもしくはNS0(ECACC 85110503))細胞の系によってグリコシル化されていてよく、またはこれらは(例えば、原核細胞における発現によって生成される場合は)非グリコシル化であってよい。
炎症性腸疾患は、結腸および小腸に罹患する一群の炎症状態である。IBDの主なタイプには、クローン病(CD)および潰瘍性大腸炎(UC)があり、IBDの他のタイプには、コラーゲン性大腸炎、リンパ球性大腸炎、虚血性大腸炎、空置性大腸炎、ベーチェット病、および不確定の大腸炎が含まれる。CDは、消化管の任意の部分に罹患し得るが、UCは結腸および直腸に限定されるのが典型的である。
材料と方法
マウスIBDモデル
DSSの投与を伴うIBDに対するマウスモデルは当技術分野において知られている。適切なマウスモデルは、例えば、Okayasuら(1990年)、Gastroenterology、98巻、694〜702頁にも記載されている。
ヨード安息香酸スクシンイミジル(SIB)を用いて125I−F8/IL10を調製した(ZalutskyおよびNarula(1988年)Cancer Research、48巻、1446〜1450頁;ZalutskyおよびNarula(1987年)Appl.Radiat.Isot.、38巻、1051〜1055頁;Chengら(2002年)J.Med.Chem.、45巻、3048〜3056頁)。簡潔に述べると、ヨウ素125(20μL〜2.0mCi)(Perkin Elmer、Waltham、MA)のアリコートを、1.5%酢酸(v:v)を含むメタノール(両方ともSigma−Aldrich)50μL中の酸化体としてNCS(Sigma−Aldrich、St.Louis、MO)10μgと一緒に、N−スクシンイミジル−3−(トリ−n−ブチルスタニル)安息香酸(C23H35NO4Sn;MW=508.23、Texas Biochemicals、Inc.College Station、TXによって合成されたもの)2.5μgと反応させた。周囲温度で15分インキュベートした後、硫酸水素ナトリウム(Sigma−Aldrich)10μg(還元剤)を加えることによって、反応溶液中の残余の酸化体をクエンチし、周囲温度でさらなる5分間インキュベートした。125I−標識したSIBを、周囲温度で20分間、pH8.0でインキュベートすることによって、F8−IL10抗体(およそ0.2mg、出発のモル比は中間体対抗体でおよそ2.5:1)にコンジュゲートさせた。放射標識した生成物(125I−F8/IL10)を、予め平衡にしたPD−10カラム(GE Healthcare、Little Chalfont、Buckinghamshire、UK)を用いて精製し(カラム上の潜在的な非特異的なタンパク質結合部位を、ウシ血清アルブミンを用いて飽和させた後、少なくとも3カラム容積のPBSでカラムをすすぐことにより)、PBS中に溶出した。放射化学収率およそ30%を得た。
非処置またはDSS処置したマウスを両方とも、投薬のおよそ2〜4日前(DSS開始後5〜7日目)、ヨウ化カリウム(KI)水(20mM)で処置して、in vivoで産生された任意の潜在的な非結合の遊離I−125の甲状腺取込みを阻止した。DSS処置開始後9日目、単一投与量の125I−F8/IL10を静脈内投与(IV)した。1群あたりの投与量および放射能を以下に概要する:
i)0群−マウス1匹あたりのおよその投与量:5mg/kg
0群−マウス1匹あたりのおよその放射能:7.5μCi(比放射能75uCi/mg)
ii)1群−マウス1匹あたりのおよその投与量:5mg/kg
1群−マウス1匹あたりのおよその放射能:10μCi(比放射能100uCi/mg)
iii)2群−マウス1匹あたりのおよその投与量:0.15mg/kg
2群−マウス1匹あたりのおよその放射能:12μCi(比放射能4490uCi/mg)
0群および2群のマウスからの結腸に対して結腸のオートラジオグラフィーを行った。結腸を収集し、その内容物を除去し、組織をリン光体イメージングスクリーン(Molecular Dynamics)に24時間曝露し、Storm860によって画像化した。結果を図2に示す。レーン1:0群(健常マウス)から注射6時間後に収集した結腸、レーン2:2群(DSS処置マウス)から注射6時間後に収集した結腸、レーン3:0群(健常マウス)から注射24時間後に収集した結腸、レーン4:2群(DSS処置マウス)から注射24時間後に収集した結腸。125I−F8/IL10のパッチ状の局在化が、DSS処置したマウスにおける結腸に沿って観察され、正常マウスでは観察されず、このモデルにおける不規則な結腸の炎症、および標的EDAの関連の発現プロファイルに一致した。
125I−F8/IL10の血清の当量濃度(ng eq./g)を、TCA(トリクロロ酢酸)沈澱性の(タンパク質関連の)放射能の測定値に基づいて推定した。TCA沈澱に対して、血清試料のアリコート(50μL)をマウス血清50μLと混合した後、20%TCA溶液100μLを加えた。試料混合物を10000gで5分間回してタンパク質を沈澱させた。上清中の合計およびTCA可溶性の放射能を決定した。所与の試料中のTCA沈澱性の放射能(cpm)、投薬溶液の比放射能(タンパク質1mgあたりのTCA沈澱性のcpm)、ならびに試料(tS)および投薬溶液(tD)の測定日を用い、式:[TCA沈澱性のcpm/EXP(−0.693/60.2*(tS−td))]/[比放射能(cpm/mg)*試料体積(mL)]を用いて、所与の試料中の試験物品の当量濃度(ng eq./mL)を算出した。
IL−10は炎症促進性サイトカインを低減することが知られているので、本発明者らは、IBDのマウスモデルにF8−IL10を投与すると、このモデルにおける血清サイトカイン応答に影響を及ぼすか否かを試験した。DSS処置を開始後3、6、および9日目、マウス1匹あたり200μgのF8/IL10、または対照の小型免疫タンパク質(F8−SIP)をIV投与した(n=10マウス/群)。この投与レジメンは、コラーゲン誘発性関節炎モデルにおける有効なレジメンと同じであった(Schwager Kら、Arthritis Research and Therapy、(2009年)11巻、R142頁)。対照群には、非疾患(レギュラーの水)および非処置疾患が含まれた(n=10マウス/群)。DSSの開始後10日目、局在化試験に対して上記に記載した通りに血液を採取し、血清を得た。血清を、MSD法のプラットホームおよびマウス7plexMSDキットを用い、製造元の指示に従って(Mesoscale Discovery、Gaithersburg、MD)、IL−1b、IL−12p40、IFNg、IL−6、KC、IL−10、およびTNFaのレベルに対して評価した。サイトカインのレベルを、図4および5において、血清1mlあたりのタンパク質のpgとして表す。
潰瘍性大腸炎に罹患している60歳女性患者、およびクローン病に罹患している42歳男性患者の結腸組織をOCT包埋した検体を凍結し、免疫組織化学的分析を行った。両方の検体をSIPフォーマットにおけるF8抗体、およびフォンウィルブランド因子で検査した。
結腸のオートラジオグラフィー
図2は、非疾患マウス(水)(レーン1および3)、または疾患マウス(DSS処置)(レーン2および4)の、放射標識したF8−IL10を投与し6時間後(レーン1および2)または24時間後(レーン3および4)いずれかの結腸のオートラジオグラフィーを示す。これは、F8−IL10は、正常の結腸よりも炎症を起こした結腸に多く蓄積することを実証している。F8−IL10が罹患している結腸に局在するパッチ状の外観は、不定のレベルの炎症およびこのモデルにおいて結腸の長さに沿って観察されるEDA発現に一致し、炎症でのF8−IL10の局在をさらに支持するものである。
図3は、正常(水)マウスと比べた疾患(DSS)マウスにおける、6、24、および96時間の時間点の、結腸および腸間膜リンパ節(L.N.)における125I−F8−IL10の蓄積を示す。図2同様、これらのデータは、DSS処置マウスでは、F8−IL10が結腸および関連のリンパ節を標的化することを実証している。またこれらの試験より、血清半減期はおよそ3.5時間であることが決定され、結腸におけるF8−IL10の半減期はおよそ35時間であり;10倍の増大は組織残留性の増強を示唆している。これは、F8−IL10は結腸の炎症の間、結腸および腸間膜リンパ節を標的化するだけでなく、一旦存在すると循環中よりも長期間残留することを示す。まとめると、これらのデータは、ヒトにおけるクローン病および潰瘍性大腸炎など、結腸における炎症の条件下では、F8−IL10はこれらの部位を優先的に標的化し、残留することを示唆している。
図4は、治療モダリティにおいてF8−IL10を投与すると、対照群に比べて、炎症性サイトカインであるIL−1β、IL12p40、IFNγ、およびIL−6の血清レベルの有意な低減をもたらすことを示すものである。
図6は、F8 SIP抗体は、新たに形成した血管を染色するが、潰瘍性大腸炎に罹患している患者における正常な血管は染色しないことを示す。(フォンウィルブランド因子は、正常な脈管構造のマーカーとしてルーチン的に用いられる。)

Claims (30)
- クローン病(CD)または潰瘍性大腸炎(UC)のような炎症性腸疾患(IBD)を処置する方法において用いるための、フィブロネクチンのエキストラドメイン−A(ED−A)アイソフォームに結合する、特異的な結合メンバー。
- CDまたはUCのようなIBDを処置するための薬物を製造するための、フィブロネクチンのED−Aアイソフォームに結合する特異的な結合メンバーの使用。
- フィブロネクチンのED−Aアイソフォームに結合する特異的な結合メンバーを含む治療有効量の薬物を患者に投与することを含む、患者におけるIBDを処置する方法。
- 特異的な結合メンバーはフィブロネクチンのED−Aに結合する、請求項1に記載の使用のための特異的な結合メンバー、請求項2に記載の使用、または請求項3に記載の方法。
