Abstract
Description
本発明の他の実施例において、上記のメンブレンはニトロセルロース、ナイロン、ポリフッ化ビニリデン(PVDF)、ガラスおよびプラスチックより構成される群から選択されることを特徴とする。
DMSOに5mg/mlで溶かしたSulfo NHS−LC−LC−Biotin(Thermo、USA)溶液100ulをPBSに4mg/ml濃度のBovine serum albumin(BSA)溶液1mlにゆっくり入れた。ホイルで光を遮断して4O/N放置して反応させる。4にO/Nで2回PBSでdialysisを進めてbiotin表紙BSAを製造した。希釈されたbiotin表紙BSA溶液を縦5cm、横16cmで切ったニトロセルロースメンブレン(NC−メンブレン)に25個の横ラインで溶液を奔注した。ビオチン表紙BSAに横ラインでコーティングされたNC membraneはDry chamberでRT O/N乾燥した。乾燥されたメンブレンをプラスチック支持体に接着させて横に1.5mm間隔で切った後、長方形のプラスチック反応容器(plastic well)に2つずつ両面テープを用いて固定した。メンブレンがコーティングされたプラスチック反応容器に0.4mlの0.5%BSAが含有されたPBS溶液を入れた後、1時間攪拌した。反応容器の溶液を捨てて、Streptavidin−Alkaline Phosphatase(Streptavidin−AP)(Promeg、USA)溶液0.4mlを入れて30分間攪拌した。Streptavidin−AP溶液を捨てて、洗浄液(50mM トリス、0.2M NaCl、0.05% Tween20)0.4mlを奔注して、常温で5分間攪拌した後除去する。この過程を2回追加して付着されていないstreptavidin−APを完璧に除去した。0.2mg/mlのbromochlorophenyl phosphateと0.3mg/mlのnitroblue tetrazoliumが含まれた発色溶液400uLを検体反応容器に入れて常温で攪拌させながら、発色反応をさせた。20分の後、溶液を除去して400ulの蒸留水を入れて、洗浄して溶液を除去した後乾燥させる。
それぞれ異なるアレルゲンとタンパク質が溶かされている溶液43種類を表1のように、二枚のNCメンブレン(A,B)に分けて実施例1のような方法でラインを引いて、アレルゲンを固定化した。アレルゲンが固定されたNC−メンブレンは室温の乾燥台に夜中乾燥させた後、プラスチック支持体に接着させて横に1.5mm間隔で切断してストリップを製造する。Aメンブレンで製造したストリップ一つとBメンブレンで製造されたストリップ一つを一つのプラスチック反応容器の中に並んで付着した。
それぞれ異なるアレルゲンとタンパク質が溶かされている溶液を表2のように4枚のNC−メンブレン(A、B、C、D)にそれぞれ分けて実施例1と同じ方法でラインを引いてマーカーを固定化した。アレルゲンが固定されたNC−メンブレンは室温の乾燥台に夜中乾燥させた後、プラスチック支持体に接着させて横に1.5mm間隔で切断してストリップを製造する。Aメンブレン、Bメンブレン、Cメンブレン、そしてDメンブレンから切られた切片を一つのプラスチックストリップに付着した(図2)。

Claims (11)
- ライン型ストリップを並列に配置した多重並列式ライン型バイオチップ
- 上記のバイオチップは並列に配置された複数個のライン型ストリップ(strip);および
上記のストリップを固定する容器;を含むことを特徴とする請求項1に記載のバイオチップ。 - 上記のストリップは支持体の上にメンブレン(membrane)をコーティングすることを特徴とする請求項2に記載のバイオチップ。
- 上記のメンブレンはマーカーを含むことを特徴とする請求項3に記載のバイオチップ。
- 上記のメンブレンはニトロセルロース、ナイロン、ポリフッ化ビニリデン(PVDF)、ガラスおよびプラスチックより構成される群から選択されることを特徴とする請求項3に記載のバイオチップ。
- 上記のマーカーはタンパク質、抗原、抗体、DNA、RNA、PNA、薬物(drug)、化合物(chemical)およびアプタマーより構成される群から選択されることを特徴とする請求項4に記載のバイオチップ。
- 請求項1の多重診断用の並列式ライン型バイオチップを用いる診断方法で、マーカーに生物学的試料を接触させて、存在する物質の濃度を測定する診断方法。
- 上記のマーカーはタンパク質、抗原、抗体、DNA、RNA、PNA、薬物(drug)、化合物(chemical)およびアプタマーより構成される群から選択されることを特徴とする請求項7に記載の方法。
- 上記の生物学的試料は組織、細胞、全血、血清、血漿、唾液、脳脊髄液および尿より構成される群から選択されることを特徴とする請求項7に記載の方法。
- 上記の物質は免疫グロブリンE(IgE)、自己抗体、サイトカイン、タンパク質、薬物、化合物、DNA、およびRNAより構成される群から選択されることを特徴とする請求項7に記載の方法。
- 多重診断用の並列式ライン型診断キットを分析するための請求項1に記載のリーダー機。
