Abstract
Description
の2−シアノフェニルボロン酸エステルを製造するプロセスであって、
以下のステップ:
(a)式(IV):
の2−ハロゲンベンゾニトリルから、式(III):
の2−シアノフェニルマグネシウムハライド塩化リチウム複合体への変換、
(b)ステップ(a)で得られた化合物から、式(II)の化合物を得る変換
を含むプロセスに関する。
の2−シアノフェニルボロン酸エステルへのさらなる変換ステップ(c4)を含むことができる。
の2−シアノフェニルボロン酸エステルへのさらなる変換ステップ(c8)を含むことができる。
X1−B(OR1)(OR2)
[式中、
X1は、ハロゲンおよびORから選択され(ここでRは直鎖状または分岐鎖状のC1−C6アルキル基であり)、並びに
R1およびR2は、独立して直鎖状または分岐鎖状のC1−C6アルキル基であるか、または一緒になって置換基を有してもよい炭素原子を1〜6個含む環を形成する]
の化合物を用いて行うことができる。
の2−シアノフェニルマグネシウムハライド塩化リチウム複合体の製造を、式(IV):
の2−ハロゲンベンゾニトリルの変換によって可能とする。
の2−シアノフェニルボロン酸エステルに変換する。この好ましい態様は、式(V)の2−シアノフェニルボロン酸エステルを固形状として単離し、かつ、高い化学純度で製造することを可能とする。生成物の高い化学純度は実際、ステップ(a)の−25℃および−15℃の温度範囲のおかげで達成され、また式(I)の2−シアノフェニルボロン酸の有機または無機塩の製造にわたり行われる精製のおかげで達成され、これによって生成物を油状ではなくて固形状で単離し、化学純度を著しく高めることが可能となる。

Claims (23)
- ステップ(c1)で得られた化合物から、式(I)の2−シアノフェニルボロン酸の有機または無機塩へのさらなる変換ステップ(c2)を含む、請求項2のプロセス。
- ステップ(b)で得られた化合物から、式(I)の2−シアノフェニルボロン酸の有機または無機塩へのさらなる直接変換ステップ(c6)を含む、請求項1のプロセス。
- ステップ(c2)または(c6)で得られた化合物から、式(I)の2−シアノフェニルボロン酸へのさらなる変換ステップ(c3)を含む、請求項3または4のプロセス。
- ステップ(a)のXが臭素である、請求項1〜8のいずれか一つのプロセス。
- ステップ(a)が、塩化リチウム複合体グリニャール試薬によって行われる、請求項1〜9のいずれか一つのプロセス。
- グリニャール試薬が、イソプロピルマグネシウムクロリド−塩化リチウム複合体およびt−ブチルマグネシウムクロリド−塩化リチウム複合体から選択される、請求項10のプロセス。
- ステップ(a)が、−25℃および−15℃の間の温度で行われる、請求項1〜11のいずれか一つのプロセス。
- ステップ(b)が、式:
X1−B(OR1)(OR2)
[式中、
X1はハロゲンおよびORから選択され(ここで、Rは直鎖状または分岐鎖状のC1−C6アルキル基であり)、並びに
R1およびR2は、独立して直鎖状または分岐鎖状のC1−C6アルキル基であるか、または一緒になって置換基を有してもよい炭素原子を1〜6個含む環を形成する]
の化合物を用いて行われる、請求項1〜12のいずれか一つのプロセス。 - X1が塩素、OMeおよびOiPrから選択され、並びに
R1およびR2がメチルおよびイソプロピルから選択されるか、または置換基を有しない炭素原子を3個含む環を形成する、
請求項13のプロセス。 - ステップ(c2)が、有機または無機塩基を含むpHが9よりも高い水溶液を用いて式(I)の化合物の有機溶液が抽出されることで式(I)の2−シアノフェニルボロン酸の有機または無機塩を生成して得ることを含む、請求項3のプロセス。
- 水溶液のpHが10および14の間である、請求項15のプロセス。
- ステップ(a)および/またはステップ(b)および/またはステップ(c2)が、トルエンまたはトルエンおよびTHFの混合物中で行われる、請求項1〜16のいずれか一つのプロセス。
- ステップ(a)が、−25℃および−15℃の間の温度範囲で行われる、請求項1のプロセスであって、
プロセスが、ステップ(b)で得られた化合物から、式(I):
次いでそれを、ステップ(c8)またはステップ(c3)および(c4)によって式(V):
の2−シアノフェニルボロン酸エステルに変換することを含む、
請求項1のプロセス。 - 式(I)の2−シアノフェニルボロン酸あるいは式(II)または式(V)のそのエステルの製造あるいはペランパネルまたはE2040の製造のための、請求項1の式(III)の2−シアノフェニルマグネシウムハライド塩化リチウム複合体の使用。
- ナトリウム塩、カリウム塩およびマグネシウム塩から選択される、請求項21の塩。
- 式(I)の2−シアノフェニルボロン酸あるいは式(II)または式(V)のそのエステルの製造あるいはペランパネルまたはE2040の製造のための、請求項21または22の式(I)のシアノフェニルボロン酸の有機または無機塩の使用。
