Abstract
Description
L−バリンの生産性が向上した微生物突然変異株を得るために、下記の方法を用いて微生物の突然変異を誘導した。
<活性化培地>
牛肉エキス 1%,ポリペプトン1 %,塩化ナトリウム 0.5%,酵母エキス 1%,寒天 2%,pH7.2
<種培地>
ブドウ糖 5%,バクトペプトン 1%,塩化ナトリウム 0.25%,酵母エキス1 %,尿素 0.4%,pH7.2
<最小培地>
ブドウ糖 1.0%,硫酸アンモニウム 0.4%,硫酸マグネシウム 0.04%,リン酸二水素カリウム 0.1%,尿素 0.1%,チアミン 0.001%,ビオチン 200μg/L,寒天 2%,pH7.0
実施例1で得られた高濃度のABA、AHV、TA及びNVに共通耐性を有する突然変異株であるコリネバクテリウムグルタミカムCA08−0072(Corynebacterium glutamicum CA08−0072, KCCM11201P)を、そのL−バリン生産性を調べるために、下記の方法で培養した。
<生産培地>
ブドウ糖 5%,硫酸アンモニウム 2%,リン酸二水素カリウム 0.1%,硫酸マグネシウム7水和物 0.05%,CSL(コーンスティープリカー) 2.0%,ビオチン 200μg/L,pH7.2
Claims (6)
- L−バリンを生産するコリネバクテリウムグルタミカム(Corynebacterium glutmicum)の突然変異株KCCM11201P。
- 前記突然変異株が、L−バリン、L−イソロイシン及びその誘導体に対して耐性を有する
請求項1に記載の突然変異株KCCM11201P。 - 前記L−イソロイシン誘導体がα−アミノ酪酸(ABA)であり、L−バリン誘導体がα−ヒドロキシバリン(AHV)、チアゾールアラニン(TA)及びノルバリン(NV)からなる群から選択される
請求項2に記載の変異株KCCM11201P。 - 請求項1に記載のKCCM11201Pを培養培地で培養するステップを含む
L−バリンの製造方法。 - 前記KCCM11201Pの培養培地からL−バリンを回収するステップをさらに含む
請求項4に記載の製造方法。 - 前記培養が、培養温度20〜45℃、好気的条件下で10〜160時間行われる
