Description
L‐ロイシンの生産能を有する微生物変異株を得るために下記のような方法を用いて微生物の変異を誘導した。
肉汁1%、ポリペプトン1%、塩化ナトリウム0.5%、酵母エキス1%、寒天2%、pH7.2
グルコース5%、バクトペプトン1%、塩化ナトリウム0.25%、酵母エキス1%、要素0.4%、pH7.2
グルコース1.0%、硫酸アンモニウム0.4%、硫酸マグネシウム0.04%、リン酸第1カリウム0.1%、尿素0.1%、チアミン0.001%、ビオチン200μg/L、寒天2%、pH7.0
前記実施例1で得られた高濃度NL耐性変異株であるコリネバクテリウム・グルタミカム KCJ‐24及びコリネバクテリウム・グルタミカム KCJ‐28のL‐ロイシン生産性を確認するために、下記のような方法で培養した。
グルコース5%、硫酸アンモニウム2%、第1リン酸カリウム0.1%、硫酸マグネシウム7水塩0.05%、CSL(トウモロコシ浸漬液)2.0%、ビオチン200μg/L、pH7.2
Claims (4)
- 受託番号KCCM11661P変異株及び受託番号KCCM11662P変異株からなる群から選択されたL‐ロイシン生産能を有するコリネバクテリウム・グルタミカム(Corynebacterium glutamicum)変異株。
- 前記変異株が、L‐ロイシン及びその誘導体に対して耐性を有するものである、請求項1に記載の変異株。
- 前記L‐ロイシン誘導体が、ノルロイシン(Norleucine、NL)である、請求項2に記載の変異株。
- 請求項1に記載の変異株を培養する段階及び前記変異株またはその培養物からL‐ロイシンを回収する段階を含むL‐ロイシンの製造方法。
