Abstract
Description

死細胞および瀕死細胞からの(リボ)核タンパク質粒子の過剰放出は、二通りの機序によってループス病態を引き起こす可能性がある:(i)クロマチン/抗クロマチン複合体の沈着またはインサイチュー形成が腎炎を引き起こし、腎機能の低下を招く;および(ii)核タンパク質が、toll様受容体(TLR)7、8および9ならびにTLR非依存的経路を介して先天性免疫を活性化する。核タンパク質の放出は、SLEにおける自己抗体の強力な抗原として働いて、抗原受容体およびTLRの共関与(co-engagement)を通じてB細胞およびDC活性化の増幅をもたらす可能性がある。したがって、それを必要とする対象において、誘発抗原を除去するため、ならびに/または免疫刺激、免疫増幅、および免疫複合体媒介性疾患を弱めるための手段に対しては、需要が存在する。
本明細書において開示されるのは、第1のヌクレアーゼドメインと改変Fcドメインとを含むハイブリッド型ヌクレアーゼ分子であって、第1のヌクレアーゼドメインがFcドメインと機能的に接続されているハイブリッド型ヌクレアーゼ分子である。Fcドメインは、分子の細胞傷害作用が、非改変Fcドメインを有するハイブリッド型ヌクレアーゼ分子と比べて低下するように改変されている。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子は、Fcγ受容体、補体タンパク質、またはその両方に対する結合性が減少するように改変されたFcドメインを有する。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子の細胞傷害作用は、対照分子、例えば、改変Fcドメインを有しないハイブリッド型ヌクレアーゼ分子と比較して、少なくとも1分の1に、2分の1に、3分の1に、4分の1に、または5分の1に低下している。
全身性エリテマトーデス(SLE)は、自己の核タンパク質を対象とする高力価の自己抗体の存在を特徴とする多系統自己免疫疾患である。SLEにおける死細胞および死滅中の細胞の排除または処理の欠陥が、主としてリボ核タンパク質およびデオキシリボ核タンパク質(核タンパク質と略記される)の蓄積を通じて疾患につながるという有力な証拠がある。核タンパク質は3つの機序を通して障害を引き起こす:i)先天免疫系を活性化して炎症性サイトカインを産生させる;ii)循環血中の免疫複合体を生じさせる抗原としての役を果たす;および、iii)腎臓などの局所部位でインサイチュー複合体形成を生じさせる抗原としての役を果たす。本発明は、少なくとも一部には、細胞外核酸の消化にインビボ治療効果があるという発見に基づく。
ハイブリッド型ヌクレアーゼ分子
いくつかの態様において、本発明の組成物はハイブリッド型ヌクレアーゼ分子を含む。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子は、Fcドメインと機能的に連結されたヌクレアーゼドメインを含む。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子は、Fcドメインと連結されたヌクレアーゼドメインを含む。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子はヌクレアーゼタンパク質である。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子はヌクレアーゼポリヌクレオチドである。

ある態様において、本発明のハイブリッド型ヌクレアーゼ分子で使用されるFcドメインは、例えばアミノ酸突然変異(例えば、付加、欠失または置換)によって変更または改変される。本明細書で用いる場合、「Fcドメイン変異体」という用語は、Fcドメインが由来する野生型Fcと比較して少なくとも1つのアミノ酸改変、例えばアミノ酸置換を有するFcドメインのことを指す。例えば、FcドメインがヒトIgG1抗体に由来する場合、変異体は、ヒトIgG1 Fc領域の対応する位置に、野生型アミノ酸と比較して少なくとも1つのアミノ酸突然変異(例えば、置換)を含む。
いくつかの態様において、ハイブリッド型ヌクレアーゼ分子はリンカードメインを含む。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子は多数のリンカードメインを含む。いくつかの態様において、リンカードメインはポリペプチドリンカーである。ある局面においては、ポリペプチドリンカーを使用して1つまたは複数のFcドメインを1つまたは複数のヌクレアーゼドメインと融合させてハイブリッド型ヌクレアーゼ分子を形成させることが望ましい。
ある局面において、ハイブリッド型ヌクレアーゼ分子はヌクレアーゼドメインを含む。したがって、本発明のハイブリッド型ヌクレアーゼ分子は、典型的には、少なくとも1つのヌクレアーゼドメインおよび少なくとも1つの連結したFcドメインを含む。ある局面において、ハイブリッド型ヌクレアーゼ分子は多数のヌクレアーゼドメインを含む。
本発明のハイブリッド型ヌクレアーゼ分子は、主として、組換えDNA手法を用いた形質転換宿主細胞において作られる。それを行うためには、そのペプチドをコードする組換えDNA分子を調製する。そのようなDNA分子を調製する方法は当技術分野において周知である。例えば、それらのペプチドをコードする配列を、適した制限酵素を用いてDNAから切り出すことができる。または、ホスホルアミデート法などの化学合成手法を用いてDNA分子を合成することもできる。また、これらの手法の組み合わせを用いることもできる。
ある態様において、ハイブリッド型ヌクレアーゼ分子は単独で投与される。ある態様において、ハイブリッド型ヌクレアーゼ分子は、少なくとも1つの他の治療薬の投与の前に投与される。ある態様において、ハイブリッド型ヌクレアーゼ分子は、少なくとも1つの他の治療薬の投与と同時に投与される。ある態様において、ハイブリッド型ヌクレアーゼ分子は、少なくとも1つの他の治療薬の投与後に投与される。他の態様において、ハイブリッド型ヌクレアーゼ分子は、少なくとも1つの他の治療薬の投与の前に投与される。当業者には理解されるであろうが、いくつかの態様においては、ハイブリッド型ヌクレアーゼ分子を他の薬剤/化合物と組み合わせる。いくつかの態様において、ハイブリッド型ヌクレアーゼ分子および他の薬剤を同時に投与する。いくつかの態様においては、ハイブリッド型ヌクレアーゼ分子および他の薬剤を同時には投与せず、ハイブリッド型ヌクレアーゼ分子を、薬剤を投与する前または後に投与する。いくつかの態様において、対象は、同じ予防期間、障害発生時および/または治療期間中に、ハイブリッド型ヌクレアーゼ分子および他の薬剤の両方の投与を受ける。
キットは、本明細書に開示されたハイブリッド型ヌクレアーゼ分子および使用説明書を含みうる。キットは、適した容器内に、本明細書に開示されたハイブリッド型ヌクレアーゼ分子、1つまたは複数の対照、ならびに当技術分野において周知のさまざまな緩衝剤、試薬、酵素および他の標準的な成分を含みうる。
ハイブリッド型ヌクレアーゼ分子を、単酵素構造または多酵素構造の所望の構造および機能的活性が組み入れられるように、シャトリング(shuttling)およびドメイン交換のための適合性制限酵素部位を有するモジュール式カセットとして設計した。ハイブリッド型ヌクレアーゼ分子の種々の態様の模式的構造は図1に図示されている。代表的なハイブリッド型ヌクレアーゼ分子のヌクレオチド配列およびアミノ酸配列は表1に示されている。
COS-7細胞に対して、ハイブリッド型ヌクレアーゼ分子遺伝子のインサートを含む発現ベクターpDGを一過性にトランスフェクトした。トランスフェクションの前日に、細胞を、4ml DMEM(ThermoFisher/Mediatech cell gro)+10% FBS組織培地中にて、60mmディッシュ当たり4×10e5個で播種した。DMEM基本培地に4.5g/Lのグルコース、ピルビン酸ナトリウム、L-グルタミン4mM、および非必須アミノ酸を加えた。ウシ胎仔血清(Hyclone, Logan, UT ThermoFisher Scientific)を、最終容量比10%となるように培地に添加した。細胞を37℃、5% CO2にて一晩インキュベートしたところ、トランスフェクションの日の集密度はおよそ40〜80%となった。Qiagen(Valencia, CA)QIAprep miniprepキットを製造元の指示に従って用いてプラスミドDNAを調製し、50ulのEB緩衝液中に溶出させた。Nanodrop 1000(ThermoFisher Scientific, Wilmington DE)分光光度計を用いてDNA濃度を測定した。プラスミドDNAは、Polyfect(Qiagen, Valencia, CA)トランスフェクション試薬を製造元の指示に従って用い、60mmディッシュ当たり2.5ugのプラスミドDNA、および150ulの無血清DMEMトランスフェクション用カクテル中の15ulのpolyfect試薬を用いてトランスフェクトした。複合体の形成後に、反応物を血清およびすべての添加物を含む1mlの細胞増殖培地中に希釈して、3mlの新たなDMEM完全培地を含むプレートに滴下した。一過性トランスフェクション物を48〜72時間インキュベートした後、さらなる分析のために培養上清を採取した。
ハイブリッド型ヌクレアーゼ分子の安定した産生は、CMVプロモーターの制御下にあるヌクレアーゼ-Ig cDNAを含む選択可能かつ増幅可能なプラスミドpDGの、チャイニーズハムスター卵巣(CHO)細胞内へのエレクトロポレーションによって達成された。このpDGベクターは、プラスミドに対する選択圧を高めるために減弱化されたプロモーターを有する、DHFR選択マーカーをコードするpcDNA3の改変された型である。Qiagen maxiprepキットを用いてプラスミドDNAを調製し、精製されたプラスミドを唯一のAscI部位で線状化した後に、フェノール抽出およびエタノール沈殿を行った。サケ精子DNA(Sigma-Aldrich, St. Louis, Mo.)を担体DNAとして添加し、100μgずつのプラスミドおよび担体DNAを用いて、107個のCHO DG44細胞にエレクトロポレーションによってトランスフェクトした。本明細書中で以後「Excell 302完全」培地と称する、グルタミン(4mM)、ピルビン酸、組換えインスリン、ペニシリン-ストレプトマイシンおよび2×DMEM非必須アミノ酸(すべてLife Technologies, Gaithersburg, Md.から)を含むExcell 302培地(JRH Biosciences)中で、細胞を対数期になるまで増殖させた。トランスフェクトされていない細胞用の培地にも、HT(ヒポキサンチンおよびチミジンの100×溶液から希釈)(Invitrogen/Life Technologies)を含めた。選択下でのトランスフェクション用の培地には、50nMから1μMまでの範囲にわたるさまざまなレベルのメトトレキサート(Sigma-Aldrich)を選択剤として含めた。エレクトロポレーションは280ボルト、950マイクロファラッドで行った。トランスフェクトされた細胞を非選択培地中に一晩おいて回復させた後に、96ウェル平底プレート(Costar)中への選択的プレーティングを、細胞125個/ウェルから細胞2000個/ウェルまでのさまざまな系列希釈で行った。細胞クローニング用の培地は、50nMメトトレキサートを含むExcell 302完全培地とした。クローン増殖が十分になったところで、マスターウェルからの培養上清の系列希釈物を、-IgGサンドイッチELISAを用いることによってハイブリッド型ヌクレアーゼ分子の発現に関してスクリーニングした。手短に述べると、NUNC immulon IIプレートを、PBS中の7.5マイクログラム/mlのF(ab'2)ヤギ抗マウスIgG(KPL Labs, Gaithersburg, MD)または2ug/mlのヤギ抗ヒトもしくは抗マウスIgG(Jackson Immunoresearch, WestGrove PA)により、4℃で一晩かけてコーティングした。プレートをPBS/2〜3% BSA中でブロックし、培養上清の系列希釈物を室温で2〜3時間インキュベートした。プレートをPBS/0.05% Tween 20中で3回洗浄して、それぞれPBS/1.0% BSA中に1:3500とした西洋ワサビペルオキシダーゼ結合F(ab'2)ヤギ抗マウスIgG2a(Southern Biotechnologies)およびヤギ抗マウスIgG(KPL)を混合したものとともに、または1:2500とした西洋ワサビペルオキシダーゼ結合F(ab')2ヤギ抗ヒトIgG1(Jackson Immunoresearch, WestGrove, PA)中にて、室温で1〜2時間インキュベートした。プレートをPBS/0.05% Tween 20中で4回洗浄し、SureBlue Reserve、TMB基質(KPL Labs, Gaithersburg, MD)によって結合を検出した。等容量の1N HClの添加によって反応を停止させ、プレートの450nMでの読み取りをSpectramax Proプレートリーダー(Microdevices, Sunnyvale CA)で行った。融合タンパク質の産生が最も高度であったクローンをT25フラスコ内で、続いてT75フラスコ内で増やして、凍結のため、および融合タンパク質の産生規模拡大のために十分な数の細胞を得た。優れた上位4つのクローンからの培養物における産生レベルを、メトトレキサート含有培地中での累進増幅によってさらに高めた。DHFRプラスミドが増幅された細胞のみが生存しうるように、細胞の連続継代のたびに、Excell 302完全培地に含めるメトトレキサートの濃度を徐々に高めた。
マウスRNase 1を、ESTライブラリー(Dr. C. Raine, Albert Einstein School of Medicine, Bronx, NYによる。彼は本発明者らの研究室にMTAを交わさずにクローンを送ってきた)から完全長cDNAとして増幅した。用いた配列特異的な5'および3'プライマーは、公開配列によるものとした。クローンの配列を配列解析によって確かめた。Genebankアクセッション番号はNCBI geneID 19752である。完全長ヒトRNase 1は、ヒト膵臓全RNA由来のランダムプライマー法およびオリゴdTプライマー法によるcDNA(Ambion/Applied Biosystems, Austin, TX)から単離した。

マウスFcドメインおよびヒトFcドメイン(SEQ ID NO:40)の単離のために、以下の通りにマウスまたはヒトの組織からRNAを得た。マウス脾臓からRPMI培地中の単細胞浮遊液を作製した。または、Lymphocyte Separation Media(LSM)Organon Teknika(Durham, NC)を用いて新鮮な全血からヒトPBMCを単離した。バフィーコートを製造元の指示に従って採取して、細胞をPBS中で3回洗浄した後に用いた。遠心によって培地から細胞をペレット化し、2×107個の細胞をRNAの調製に用いた。QIAGEN RNAeasyキット(Valencia, Calif.)全RNA単離キットおよびQIAGEN QIAshredderカラムをキットに添付の製造元の指示に従って用いて、RNAを細胞から単離した。1マイクログラム(4μg)の全RNAを、逆転写によってcDNAを調製するためのテンプレートとして用いた。このRNA、300ngのランダムプライマーおよび500ngのOligo dT(12〜18)および1μlの25mM dNTPを混ぜ合わせ、80℃で5分間変性させ、その後に酵素を添加した。Superscript III逆転写酵素(Invitrogen, Life Technologies)を、酵素とともに提供された第二鎖緩衝液および0.1M DTTの存在下で総容量25μlのRNA+プライマー混合物に添加した。逆転写反応は50℃で1時間進行させた。cDNAをQIAquick(QIAGEN)PCR精製カラムを用いて精製し、40マイクロリットルのEB緩衝液中に溶出させた後に、PCR反応に用いた。

RSLV-124構築物(SEQ ID NO:106)のマウスにおけるインビボ安定性分析
4匹のマウス(C220、C221、C222、C223)に対して、ゼロ時点でRSLV-124の単回の注射による静脈内注射を行った。注射後のさまざまな時点で血液試料を収集して、RSLV-124タンパク質(野生型ヒトIgG1 Fcドメインと連結されたヒト野生型RNase(SEQ ID NO:106))の存在およびRNase酵素活性に関して分析した。マウス血清中のRSLV-124化合物を検出するために、マウス血清からヒトFcを捕捉して、その後にヒトRNaseの検出を行うELISAを開発した。マウス4匹の血液試料に対してELISAを実施したところ、150ugの単回静脈内注射から5分後の時点で、RSLV-124タンパク質の存在が38μg/ml〜55μg/mlで検出された(図2)。注射1日後には、RSLV-124の濃度は8μg/ml〜12μg/mlに急速に低下した。薬物の血中濃度は7日間の分析期間にわたって比較的安定に保たれ、薬物の血中レベルはおよそ5μg/mlであった。ELISAによってRSLV-124タンパク質を測定するために用いたのと同じ血液試料を用いて、薬物のRNase酵素活性を定量した。AmbionによるRNaseAlert QCシステム(カタログ番号AM1966)を若干の変更を加えた上で用いて、マウス血液試料中のRSLV-124タンパク質の酵素反応速度を測定した。ヒト抗Fcモノクローナル抗体を用いて薬物化合物をマウス血清からRNaseAlertアッセイプレート上に捕捉して、Ambionキットの指示の通りに蛍光の測定によって定量した。RSLV-124分子の相対蛍光単位(RFU)の分析により、注射5分後には80,000〜140,000 RFUが示された(図3)。RFUはタンパク質濃度に平行して急速に低下し、その後は第7日まで18,000〜40,000 RFUの間に比較的安定に保たれた。RNaseAlert QCシステムを用いて、既知の量のタンパク質を用いた標準曲線を作成し、この標準曲線から、血液試料中のRSLV-124のRFUを用いてRSLV-124のタンパク質濃度を外挿した。この分析から、RNase酵素活性アッセイを用いて算出した、7日間の実験にわたってマウス血液中に存在したタンパク質濃度が、ELISAを用いて測定した値に極めて類似していたことが明らかになった(図4)。これらの実験から、RSLV-124化合物が7日間にわたってマウス循環血中にてインビボで安定であり、その酵素活性を保つことが結論づけられた。このことは、酵素活性がほぼ100%、マウス循環血中で7日間にわたって保たれたことから、この化合物がマウスにおいてインビボで分解を受けないことを示唆する。Fc融合タンパク質は循環血中で分解を受けやすいことが多いため、この知見は、RNase-Fc融合タンパク質が有益な薬物として用いられることをさらに裏づけるものである。
RNaseAを過剰発現するマウス(RNase Tg)を作製した。このヌクレアーゼはRNase Tgマウスにおいて高レベルで発現される。本発明者らは、血清中のRNaseを定量するための一元放射拡散(SRED)法(図5)、およびはるかにより定量的なELISAを開発した(図6参照)。両アッセイとも、RNase TgにおけるRNase活性の有意な増加を示している。図6のRNaseレベルの定量は、野生型B6マウスと比較して、RNase TgにおいてRNaseが約10倍増加していたことを示した。本発明者らは、RNaseA TgをTLR7.1 Tgマウスと交配させて二重Tg(DTg)を作製した。TLR7.1マウスはTLR7のコピーを8〜16個有しており、悪性度が非常に高い急速進行性のループス様疾患を発症し、3カ月齢の時点で死亡し始め、生存期間中央値は6カ月である。予備分析において、本発明者らは、DTgマウスが改善の徴候を示すか否かを調べるために、DTgおよび同腹仔対照を3カ月齢まで育てた(bled)。図5に示されているように、DTgマウスは、それらの血清中のRNaseのレベルが非常に高かった(比活性993U/mgの本発明者らの標準物質に基づけば、13U/mlを上回るRNaseに相当)。図6に示されているように、TgおよびDTgマウスにおけるRNaseA濃度をELISAアッセイによっても測定した。RNase A TgマウスおよびTLR7.1×RNaseA Dtgマウスは、RNase Aの血清中濃度が1〜2ng/mlであった。

DTgとTLR7.1同腹仔対照との間には、生存に関して高度の有意差が認められた。図7に示されているように、10カ月の時点でTLR7.1マウスの61%が死亡したのに対して、DTgマウスは31%が死亡した。このデータは、RNaseAの過剰発現が強い治療効果を発揮したことを示している。TLR7.1マウスが早発性に死亡した理由は完全には明らかでないが、重症貧血、血小板減少および糸球体腎炎が役割を果たした可能性が考えられる。DTgマウスにおいて、赤血球数および血小板数がRNaseA発現によって良い影響を受けたか否かを明らかにするために、本発明者らは血算を行ったが、TLR7.1マウスとDTgマウスとの間に差は見いだされなかった。対照的に、DTgマウスでは腎臓の組織病理に有意な改善が認められた。本発明者らは、DTgマウスにおいてIgGおよびC3の沈着減少を観察した。メサンギウムにおける炎症を反映するPAS染色も、DTgマウスではTLR7.1同腹仔対照と比較して低下していた。本発明者らが今回、腎臓のマクロファージ浸潤を抗MAC-2(ガレクチン3)抗体(Lyoda et al. Nephrol Dial Transpiat 22: 3451, 2007)を用いて比較したところ、mac-2陽性細胞はDTgマウスの糸球体の方が非常に少なかった。各群当たり5匹ずつのマウスにおいてマウス1匹当たり20個の糸球体で算定したところ、単独TgおよびDTgに関する平均+/-SEはそれぞれ3.8+/-1.1および1.4+/-0.2であり、p=.05であった。加えて、本発明者らは糸球体係蹄サイズも定量し、DTgマウスにおける糸球体係蹄サイズの有意な減少を観察した(単独TgおよびDTgにおいてそれぞれ179+/-41および128+/-16.8um2、p=0.037)。以上を要約すると、TLR7.1×RNaseA DTgマウスはそれらの単独Tg TLR7.1同腹仔よりも長く生存し、かつそれらの腎臓における炎症および傷害はより軽度である。この知見は、このマウスモデルにおいてRNA免疫複合体を除去することによって、全体的な死亡率が有意に改善し、かつこのループス様病態に関連する腎臓の損傷および全体的な炎症が減少したことを指し示している。
TLR7.1 TgマウスおよびTLR7.1×RNaseA DTgマウスの脾臓におけるインターフェロン応答遺伝子(IRG)の分析により、DTgマウスにおいてIRF7遺伝子(インターフェロン調節因子7(UniProtKB P70434))の発現が有意に低いことが示された(p=0.03)。MX1(インターフェロン誘導性GTP結合タンパク質Mx1(UniProtKB P09922))およびVIG1(ラジカルS-アデノシルメチオニンドメイン含有タンパク質2(UniProtKB Q8CBB9))を含む他のいくつかのIRGは、Tgマウスと比較してDTgマウスの方が低値であったが、その差は有意ではなかった(図8)。定量的PCRを以下の通りに行った。RNeasy miniキット(Qiagen, Valencia, CA, USA)を用いてマウス脾臓から全RNAを単離し、Turbo DNA-free(Applied Biosystems, Foster City, CA, USA)を用いてDNaseを処理した上で、RNA-to-cDNAキット(Applied Biosystems)により、ランダムプライマーを用いて第一鎖cDNAを生成させた。単離されたRNAに関して、NanoDrop(Thermo Scientific, Waltham, MA, USA)を用いた測定では、260/280値は1.7〜2.0の間であった。cDNAを1ng/ul全RNAに相当するよう希釈し、1つの反応につき8ulを用いた。参照遺伝子(18s)および関心対象の遺伝子(GOI)用のプライマーを合成し(IDT, Coralville, Iowa, USA)、分子グレードの水(molecular grade water)を用いてqPCR用に適切な濃度に希釈した。プライマーのBLASTの結果から、参照遺伝子またはGOIのみに対する特異的な配列相同性が示されている。SensiMix SYBR low-ROX master mix(Bioline, London, UK)に対するテンプレートおよびプライマーの1:1混合物を用いて、ABI Fast7500システムで、2つずつの反応物(20ul)を進行させた。年齢を一致させた野生型B6マウスを各GOIに関する変化倍数を求めるためのベースラインとして用いる2-ddCT法を用いて、相対的定量の算出を行った。反応物に関する解離曲線から、各遺伝子に関して単一の融解ピークが示されている。標準曲線からは、各遺伝子に関して同程度の増幅効率が示され、テンプレート濃度はプライマーセットのそれぞれに関して直線範囲(linear dynamic range)内にあった。

図9は、RSLV 125〜129構築物からのCOSトランスフェクション上清についてのウエスタンブロットを示している。RSLV125、126、127、128または129を含む発現プラスミドを、Polyfectトランスフェクション試薬を用いてCOS7細胞にトランスフェクトして、上清を48時間後に採取した。RSLV 125および126に含まれる単一のヌクレアーゼ分子に加えて、RSLV 127、128および129によってコードされる、より複雑な多ヌクレアーゼ融合タンパク質も、COS細胞の一過性トランスフェクト物から発現させた。ウエスタンブロット分析を、一過性トランスフェクタントからの上清に対して行った。図9に示された分子は、ヒトRNase1にヒトSSSIgG1ヒンジおよびIg G1 P238S-P331S Fcドメインを融合させたものを含有するか、またはヒトRNase1(野生型)をヒトIgG1のSSSヒンジ-(P238S-331S)CH2-CH3 Fcドメインに融合させて、その後にリンカードメインをプロテアーゼ切断から保護するためのN結合型グリコシル化部位を含有する新規リンカー、および分子のカルボキシ末端に突然変異型アレルG105R-A114F型のヒトDNase1が続くものを含む。加えて、RSLV 127は、突然変異型-Ig尾部のアミノ末端に上記のヒトDNase1突然変異体を、カルボキシ末端にRNase 1 WTをコードする。COS上清を72時間後に採取し、0.5mlの試料を、100ulのプロテインA-アガロースビーズにより、4℃で一晩かけて免疫沈降させた。プロテインAビーズを遠心し、PBS中で2回洗浄した後に、SDS-PAGEローディング緩衝液中に、NuPAGEゲル用には還元性または非還元性LDS試料用緩衝液(Invitrogen, Carlsbad, CA)中に再懸濁させた。試料を製造元の指示に従って加熱し、プロテインAビーズを遠心してペレット化させて、試料用緩衝液を5〜12% NuPAGE勾配ゲル上にローディングした。試料の電気泳動を150ボルトで1.5〜2時間行い、ニトロセルロースメンブレンへのゲルのブロッティングを30mAmpで1時間行った。ウエスタンブロットをTBS/5%脱脂乳中に一晩おいてブロックした。ブロットを1:2500のHRP(西洋ワサビペルオキシダーゼ)結合ヤギ抗ヒトIgG(Fc特異的、Jackson Immunoresearch)とともに室温で1.5時間インキュベートし、PBS/0.5% Tween20で5回またはそれ以上洗浄した上で、ECL試薬を用いてブロットを現像した。その結果は、ヌクレアーゼFc融合タンパク質の構築が成功し、タンパク質がCOS細胞から容易に発現されたことを実証している。その上、これらのヌクレアーゼFc融合タンパク質に関する還元性および非還元性プロフィールの分析により、DNA構築物が適切な分子量のタンパク質をコードすることが裏づけられている。非還元性SDS-PAGEでのパターンは、タンパク質のジスルフィド結合特性が予想された構築物の挙動に一致することを裏づけている。
図10は、実施例10による、RSLVをトランスフェクトしたCOS上清からのプロテインA精製タンパク質のアリコートを比較したSRED分析を示している。2%アガロースゲルを蒸留水で調製した。ポリ-IC(Sigma)を蒸留水に3mg/mlで溶解させた。ゲルプレートは以下の通りに調製した:1.5mlの反応緩衝液(0.2M Tris-HCl pH 7.0、40mM EDTAおよび0.1mg/mlの臭化エチジウム)、1mlのPoly-ICおよび0.5mlの水をチューブ内に入れ、50℃に5分間維持した。3mlのアガロース(50℃に保った)をチューブに添加した。この混合物を直ちにガラスプレート上に注いだ。ゲルへの穿孔によってサンプリングウェルを作製した。対照、血清試料、またはアフィニティー精製したRSLVタンパク質の各2μlをウェル中にローディングし、湿潤チャンバー内でゲルを37℃で4時間インキュベートした。続いてゲルを、緩衝液(20mM酢酸ナトリウムpH5.2、20mg/ml臭化エチジウム)中にて氷上で30分間インキュベートし、UV下で読み取った。ゲルの写真は、UVトランスイルミネーター上で、臭化エチジウム用フィルターを装着したKodakデジタルカメラDC290システムを用いて撮影し、Kodak MolecularImagingソフトウェアを用いて解析した。RNase酵素活性アッセイによる結果は、構築物がすべて、触媒活性のあるRNaseモイエティーを含有することを指し示している。
図11は、実施例10におけるRSLV融合プラスミドをトランスフェクトしたCOS7上清からのプロテインA精製タンパク質に対して行ったDNaseヌクレアーゼ活性アッセイによる結果を示している。図11は5つのパネル(11a、1lb、11c)を示しており、各ゲルパネルは、量を漸減させた表記の融合タンパク質による消化パターンおよび1マイクログラムのプラスミドDNAを示している。各タンパク質を、酵素500ngから4ngまでの2倍刻みでヌクレアーゼ非含有水中に系列希釈した。各試料に対して、1ugのPDGプラスミドDNAを添加し、37度で30分間インキュベートした。各試料の2分の1を、1.2%のTAEアガロースゲルを用いる100ボルトで30分間のアガロースゲル電気泳動に供した。図11cは、市販のDNase 1(Biolabs, Inc.)を用いたゲル中DNase酵素活性アッセイの結果を示している。最も右のレーンはDNA単独で酵素を伴わない陰性対照であり、その左側にあるレーンは別の陰性対照であって、これはRNase活性を有するがDNase活性は有しないRNase-Ig分子のものであり、予想された通り、どちらの場合にもプラスミドDNAは無傷のままであり、消化されなかった。これらの結果は、市販のDNase1酵素が高活性であり、試験した濃度の大半でDNAのすべてを消化したことを実証している。図11aおよび11bにおける結果は、4種類のヌクレアーゼFc融合構築物のDNase活性を示している。図11a中の上のパネルはDNase-Ig融合タンパク質がプラスミドDNAを消化する能力を示しており、ゲルから見てとれるように、この酵素は試験したすべての濃度でDNAのすべてを、市販のDNase 1と同じ活性で、またはそれを上回る活性で消化した。図11aの下のパネルにあるのは、DNaseをFcのアミノ末端に有する二重特異性ヌクレアーゼFc融合タンパク質(SED ID 65〜66)であり、これも強固なDNase酵素活性を有するが、図11aの上のパネル中のDNase-Igよりは幾分弱かった。図11bの上のパネルは別の二重特異性ヌクレアーゼのDNase酵素活性を示しており、このFc融合タンパク質は、特別に作られたNLGリンカーを介してFcのC末端でFcとつながれたDNaseを有する(SEQ ID 67〜68)。データによって見てとれるように、この酵素も強固なDNase酵素活性を有し、ここで検討した他の二重特異性ヌクレアーゼよりも活性が高いように思われる。図11bの下のパネルは、RNaseモジュールをFcとつなぐ(G4S)4リンカーを持たない、別の二重特異性ヌクレアーゼ分子(SEQ ID 69〜70)のDNase酵素活性を示している。この二重特異性ヌクレアーゼも良好なDNase活性を有するが、この実験で示した他の2つの二重特異性ヌクレアーゼFc融合タンパク質よりも幾分弱いように思われる。このデータは、二重特異性ヌクレアーゼのすべてが良好なDNase活性を有することを示唆しており、このことは、これに関連した他の者の以前の取り組みを考えれば予想外である(Dwyer et al. JBC; Vol 271, No. 14; pp 9738-9743)。その上、構築物におけるDNaseの位置、ならびにDNaseをFcとつなぐリンカーの長さおよび組成は、二重特異性ヌクレアーゼFc融合タンパク質という状況で高活性のDNase酵素を作り出す上で決定的に重要である。
図12〜13は、組換えRNase A(Ambion)、RSLV 125、RSLV 126、hRNase WT-SCCH-WThIgG1、およびhRNaseG88D-SCCH-(P238S/K322S/P331S)hIgG1のRNase酵素活性を比較した速度論的蛍光酵素活性アッセイによる結果を示している。二価mRNase-Ig融合タンパク質の機能的特徴をさらに明確にするために、本発明者らは種々のヌクレアーゼ融合タンパク質の酵素反応速度をRNase Alert Substrate(Ambion/IDT)を用いて調べ、Biotek Synergy2マイクロプレートリーダーを用いて蛍光を定量した。データは、Gen5ソフトウェア(Biotek Instruments, Inc., Winooski, Vermont)を用いて解析した。時間の関数としての相対蛍光単位を、製造元の指示に従って37Cでインキュベートし、酵素濃度を10pg/ulから始めて低下させ、0.67倍刻みで0.1pg/ulまで系列希釈していく、45分間の実験の過程を通じて毎分アッセイした。各試料には、固定濃度のRNase Alert基質(200nM)を1×RNase Alert反応緩衝液中に含めた。
図14〜15は、野生型または突然変異型(SCC、P238S、P331Sを含む)のIgG Fcドメインを有するRNaseIg融合タンパク質の、ヒト単球細胞株THP1の生存に対する影響を分析したインビトロ試験の結果を示している。THP1細胞をRPMI/10% FBS中で対数増殖下に維持し、その後にアッセイ用に採取した。細胞傷害作用アッセイに用いる前の、細胞の生存度は98%を上回った。THP1細胞を96ウェルプレートに1×10e6個/mlまたは1ウェル当たり100,000細胞でプレーティングした。ハイブリッド型ヌクレアーゼタンパク質を、連続ウェルに対して、1つの反応ごとに5マイクログラム/mlで始めて0.01マイクログラム/mlの融合タンパク質で終える2倍系列希釈の系列を用いて添加した。この実験では、野生型IgG1 Fcを有するRNase-Fc融合タンパク質(wtRNasewtIgG)を、Fc受容体に対する結合性および内部移行が有意に低下している突然変異型Fc(P238S、P331S)を有するRNase-Fc(mtRNasemgIgG)と、培養THP1細胞における細胞傷害作用を誘導する能力に関して比較した。反応物を96ウェルプレート中にて37℃、5% CO2下で3日間インキュベートし、その後に細胞の採取および分析を行った。3日後に、細胞を1000rpmでの遠心分離によって採取し、PBS/2% FBSで洗浄して、FITCアネキシンVアポトーシス検出キット試薬(#556547、Becton Dickinson/Pharmingen)とともに製造元の指示に従ってインキュベートした。細胞を、キットとともに供給された冷却した結合緩衝液100マイクロリットルで洗浄した上で、アネキシンV-FITC/ヨウ化プロピジウム(PI)を100ul結合緩衝液中に1:100で添加した。試料を氷上で20分間インキュベートし、その後に400μlの追加の結合緩衝液を各試料に添加した。染色した試料を、FACS Canto(Becton Dickinson)を用いるフローサイトメトリーによって分析し、Flowjoソフトウェア(Treestar,Ashland,OR)を用いてデータを解析した。
RSLV132添加により、3人のSLE患者由来の血清に壊死細胞抽出物(NCE)を加えたもので形成された免疫複合体を用いて刺激したヒト末梢血単核細胞からのインターフェロン-αの誘導が消失した(図16)。ループス患者の免疫複合体に含まれるRNAと結合してそれを分解するRSLV-132の能力を測定するために、インビトロバイオアッセイを開発した。この実験は、ループス患者由来の自己抗体および培養ヒト細胞(U937)由来のNCEを用いた、インビトロでの免疫複合体の形成を伴う。ループス患者血清をNCEと合わせると、インターフェロンの極めて強力な誘導物質である免疫複合体(IC)の形成が起こるが、正常ヒト血清はインターフェロンの産生を刺激しない。ICを、レポーター細胞としての正常ヒト末梢血単核細胞(PBMC)とともにインキュベートする。レポーター細胞によるインターフェロンの産生は、インターフェロン-α ELISAを用いて測定する。レポーター細胞は、正常志願者からFicoll密度勾配遠心分離によって入手した。ループス患者または健康正常志願者の血清は、University of Washington Institutional Rreview Board #HSD No. 3971に準拠して入手し、血清を1/1000に希釈した上で、上記の通りに培養U937細胞から得た壊死細胞抽出物(NCE)に対して10%(v/v)で添加した。希釈したループス患者または正常志願者の血清をNCEとともに室温で15分間インキュベートし、その結果生じたICを、さまざまな用量のRSLV-132、RSLV-124または野生型RNaseとともに、または伴わずに15分間インキュベートし、続いて500U/mLのUniversal IFNの存在下で正常PBMCとともに20時間インキュベートして、その後にPBMC培養物から分泌されたIFNの量を測定した。血清は、疾患活動度が軽度から活動性までの範囲でさまざまである3人の異なるループス患者から入手した。NCEを、ループス患者血清または健康正常志願者血清のいずれかとともに室温で15分間インキュベートし、その後にPBMCとの20時間のインキュベーションを行った。IFN-αは、IFN-αをヒトIFNαに対するマウスMAb(MMHA-11)[PBL Biomedical Laboratories、product #2112-1]を用いて捕捉し、IFNαに対するウサギポリクローナル抗体[PBL Biomedical Laboratories、product #31101-1]で検出した後に、抗ウサギHRP[Jackson Immuno Research、product #711-035-152]およびTMB基質を用いて発色させるELISAによって定量した。場合によっては、PBMCへのNCEの添加の前に、NCEに対して、被験体(RSLV-124もしくはRSLV-132)を0.16、0.5、1.6および5.0ug/mLの濃度で添加するか、またはRNaseを0.05、0.16、0.5および1.6ug/mLの濃度(等モル)で添加した。ループス患者血清がPBMCからのIFNの産生を刺激する能力は、5.0ug/mLのRSLV-124の添加によっておよそ50%低下した。この阻害は、等モル量のRNaseのそれを反映していた。同じ濃度のhuRSLV-132の添加は、IFNを阻害する効果がRSLV-124と同等であるかまたはより上回り、5.0ug/mLのhuRSLV-132の添加により、IFN産生はほぼ完全に消失した。NCEと合わせると、ループス患者の抗RNA/DNA抗体は、新たに単離したPBMCからのIFNを強力に誘導する。正常志願者由来の血清は、レポーター細胞からのIFN産生を誘導するという、これと同じ能力を有しない。このデータは、ループス患者血清中の循環している自己抗体が、おそらくTLR7およびその後のIFN産生を誘発するであろう免疫複合体を形成しうることを指し示している。IFNの厳密なタイプおよびサブタイプは分析しなかった。このデータは、RSLV-132がその分子標的である、ループス患者のICと会合したRNAと結合して、それを強力に分解し、それによって、PBMCによるIFNの刺激を妨げたことを指し示している(図16)。このアッセイでは、RSLV-132はRSLV-124よりも活性が高いように思われる。
RSLV-132がマウス循環血中のRNAと結合してそれを分解する能力を評価するために、インターフェロン経路の強力な活性化物質であるRNA模倣体、ポリイノシン:ポリシチジン酸(ポリI:C)を用いて薬力学モデルを開発した。ポリ(I:C)は、一方の鎖がイノシン酸の重合体であって、もう一方がシチジン酸の重合体である、ミスマッチの二本鎖RNAである。これは、B細胞、マクロファージおよび樹状細胞の膜で発現されるtoll様受容体3(TLR3)と相互作用することが知られている。ポリ(I:C)はInvitrogenから入手可能である。ポリI:Cの効果は、投与後のマウス脾臓におけるインターフェロン刺激遺伝子の発現レベルを測定することによって定量することができる。第0日に、3カ月齢のB6マウス10匹に対して、RSLV-132(マウス1匹当たり250ug)または対照としての静脈内免疫グロブリン(IVIG)(Privigen,Behring)(マウス1匹当たり250ug)のいずれかを、どちらも腹腔内注射によって投与した。RSLV-132またはIVIGの注射の20時間後に、ポリ(I:C)を、マウス1匹当たり200ugでマウスに腹腔内注射した。2時間後にマウスをCO2曝露によって屠殺して、脾臓をRNAlater(Qiagen)中に収集した上で、インターフェロン刺激遺伝子(ISG)の発現に関する後の試験のために-80Cで貯蔵した。脾臓試料を、qPCRによる、Ifit1(テトラトリコペプチド反復配列1を有するインターフェロン誘導性タンパク質(UniProt Q64282))、Irf7(インターフェロン調節因子7(UniProt P70434))およびMx1遺伝子を含むISGの発現の試験に供した。これらの実験による結果は、RSLV-132の腹腔内注射により、循環血中のポリ(I:C)と結合してこのRNA模倣体を効果的に分解することのできるRSLV-132の血清中濃度がもたらされ、それによってインターフェロン経路およびモニターした3つのISGの刺激が効果的に妨げられることを実証している(図17)。
RSLV-132およびRSLV-133をCHO細胞において一過性に発現させ、プロテイン-Aを用いて精製した。これらのRNase Fc融合タンパク質のRNase活性を、AmbionによるRNaseAlert QCキット(カタログ番号AM 1966)を用いて定量した。さまざまな量のRNase Fc融合タンパク質を用い、その結果を経時的な相対蛍光単位(RFU)として図18に示している。この結果は、RSLV-132が高活性のRNase酵素であり、RSLV-124などの他のRNase Fc融合構築物および野生型RNaseと比べて高いRNase活性を有することを実証している。例えば、同じ量(400pM)のRSLV132およびRSLV-124を用いると、RSLV-132はRSLV-124と比べて2倍を上回るRFU(80,000対35,000)を生じる。加えて、2つの生産ロットを、4Cでのそれらの安定性に関して試験した。RSLV-132.1は4Cで8週間貯蔵した後にこの実験を行い、RSLV-132.2は-80Cで貯蔵した上で試験の直前に解凍させたところ、このタンパク質が4Cで最長2カ月まで安定であることが実証された。薬物の安定性および増大した触媒活性は、治療状況において有効性の向上をもたらす可能性がある。
RNase Fc融合タンパク質がインビトロでFc受容体と結合する能力を検討するために、RSLV124(野生型Fcドメイン)およびRSLV-132(突然変異型Fcドメイン;P238S/P331S)を、Fcを保有するヒト骨髄性細胞株THP1とともにインキュベートして、細胞に対する特異的結合を蛍光活性化細胞選別(FACS)分析によって定量した。RLSV-124およびRSLV-132を、Invitrogenによるalexa fluor色素AF-647(カタログ番号A20006)を用いて蛍光標識した。結合しなかった色素を除去するためにRNase Fc融合タンパク質を透析した後に、さまざまな量の標識タンパク質をTHP1細胞とともに1時間インキュベートし、結合しなかったRNase Fc融合タンパク質を除去するために細胞を強力に洗浄した上で、特異的に結合したタンパク質を、平均蛍光強度を測定するFACSによって定量した。図22における結果は、突然変異型Fcドメインを有するRSLV-132タンパク質は、野生型Fcドメインを有するRSLV-124よりもFc受容体に対する結合性が有意に低く、Fc受容体に対する結合性が4分の1未満に低下することを実証している。この知見は、突然変異型Fcドメイン(P238S/P331S)を有するRNase Fc融合タンパク質は細胞傷害作用が有意に減少しているという本発明者らの以前の知見と一致する。

Palmouth, MA)のPyrotellカブトガニアメーバ様細胞溶解物(Limulus Amebocyte Lysate)(LAL)キットなどの市販のキットを用いて、溶液をエンドトキシン混入に関して検査する。続いて分子を、生物活性に関する種々のインビトロアッセイを用いてアッセイする。

- DNaseの活性が対照DNase分子の活性とほぼ等しい、請求項24記載のハイブリッド型ヌクレアーゼ分子。
- 前記請求項のいずれか一項記載のハイブリッド型ヌクレアーゼ分子と、薬学的に許容される担体とを含む、組成物。
- 請求項1記載のハイブリッド型ヌクレアーゼ分子をコードする、核酸分子。
- 請求項28記載の核酸分子を含む、組換え発現ベクター。
- 請求項29記載の組換え発現ベクターによって形質転換された、宿主細胞。
- 以下の段階を含む、請求項1記載のハイブリッド型ヌクレアーゼ分子を作製する方法:
ハイブリッド型ヌクレアーゼ分子をコードする核酸配列を含む宿主細胞を用意する段階;および
ハイブリッド型ヌクレアーゼ分子が発現される条件下で該宿主細胞を維持する段階。 - 請求項1記載のハイブリッド型ヌクレアーゼ分子の有効量を対象に投与する段階を含む、異常な免疫応答と関連のある病状を治療または予防するための方法。
- 病状が自己免疫疾患である、請求項32記載の方法。
- 自己免疫疾患が、インスリン依存性真性糖尿病、多発性硬化症、実験的自己免疫性脳脊髄炎、関節リウマチ、実験的自己免疫性関節炎、重症筋無力症、甲状腺炎、実験型ブドウ膜網膜炎、橋本甲状腺炎、原発性粘液水腫、甲状腺中毒症、悪性貧血、自己免疫性萎縮性胃炎、アジソン病、早発閉経、男性不妊症、若年型糖尿病、グッドパスチャー症候群、尋常性天疱瘡、類天疱瘡、交感性眼炎、水晶体起因性ブドウ膜炎、自己免疫性溶血性貧血、特発性白血球減少症、原発性胆汁性肝硬変、活動性慢性肝炎Hbs-ve、特発性肝硬変、潰瘍性大腸炎、シェーグレン症候群、強皮症、ヴェーゲナー肉芽腫症、多発性筋炎、皮膚筋炎、円板状エリテマトーデス、全身性エリテマトーデス(SLE)、および結合組織病からなる群より選択される、請求項33記載の方法。
- 自己免疫疾患がSLEである、請求項34記載の方法。
- ヌクレアーゼを含有する組成物を対象に、RNA、DNA、またはRNAとDNAの両方を含有する免疫複合体を分解するのに有効な量で投与する段階を含む、SLEを治療する方法であって、
該組成物が、薬学的に許容される担体と、SEQ ID NO:62、64、66、68、70、78、80、82、84、86、92、94、96、または98に記載のアミノ酸配列を含むハイブリッド型ヌクレアーゼ分子とを含む、
前記方法。
