Abstract
【解決手段】本発明の非酵素的糖化反応抑制剤は、麦類の溶媒抽出物を有効成分として含有する。上記溶媒は、好ましくは、水、エタノール、含水エタノール、メタノール、含水メタノール、アセトン及び含水アセトンからなる群から選択される1種以上である。上記麦類は、好ましくは、小麦、大麦、ライ麦及びオーツ麦からなる群から選択される1種以上である。

Description
下記<エタノール抽出処理>又は<熱水抽出処理>に従い、各種抽出源から溶媒抽出物A−1〜A−7及びB−1〜B−5を得、それぞれ、各実施例及び比較例のサンプルとした。抽出源としては次の植物の種子(乾燥させた種子)を用いた。小麦、大麦、ライ麦、オーツ麦(以上、麦類)、アーモンド、大豆。小麦については、種子の全部(全粒穀物)及びその外皮部(小麦ふすま)の両方を抽出源とし、小麦以外の植物については、全粒穀物を抽出源とした。全粒穀物を抽出源とする場合は、抽出処理に先立って全粒穀物の粉砕処理を実施した。粉砕処理は、粉砕機(安井器械製、マルチビーズショッカー(登録商標))を用い、室温(25℃)下、2500rpmの条件で全粒穀物を10秒間粉砕する処理を3回行うことにより、実施した。小麦ふすまを抽出源とする場合は、粉砕処理は行わずにそのまま抽出処理に供した。上記小麦ふすまは、常法に従って調製した。
抽出源3gに抽出溶媒としてのエタノール15mlを加え、室温(25℃)で2時間振とうさせた後、室温(25℃)下、5000rpmの条件で5分間遠心分離して、その上清を回収し、この上清を減圧乾燥させて、エタノール抽出物を得た。
<熱水抽出処理>
抽出源3gに抽出溶媒としての純水15mlを加え、加えた純水を80℃まで加温して熱水とし、その80℃の熱水中に該抽出源を2時間静置(浸漬固定)させた後、5000rpmの条件で5分間遠心分離して、その上清を回収し、この上清を減圧乾燥させて、熱水抽出物を得た。

・実施例6:小麦ふすまの熱水抽出物(B−1)
・実施例7:大麦の熱水抽出物(B−2)
・実施例8:オーツ麦の熱水抽出物(B−3)
・比較例1:アーモンドのエタノール抽出物(A−6)
・比較例2:大豆のエタノール抽出物(A−7)
・比較例3:アーモンドの熱水抽出物(B−4)
・比較例4:大豆の熱水抽出物(B−5)
4mg/mlになるようにゼラチンに200mMリン酸緩衝液(pH7.4)を加え、室温で30〜60分間膨潤させ、次いで、50〜60℃の湯せんで溶解後、孔径0.22μmのフィルターでろ過した。また別途、60mMリボースを200mMリン酸緩衝液(pH7.4)で調製後、孔径0.22μmのフィルターでろ過した。こうして調製・精製されたゼラチンとリボースとを等量混合して混合系を得、該混合系に被験物質を最終濃度が5mg/mlになるよう添加し、被験物質無添加(対照)の混合系と共に、37℃で1週間静置した。被験物質としては、上記各種溶媒抽出物以外に、ポジティブコントロールとしてルテオリンを用いた。ルテオリンはエタノールで融解させ、最終濃度が50ppm(C−1)又は5ppm(C−2)になるよう上記混合系に添加した。1週間静置後の上記混合系(糖化ゼラチン)について、AGEsの一種であるNε−(カルボキシメチル)リジン(CML)抗体を用いた下記ELISA法によりCMLを定量し、CML生成度を評価した。結果を図1に示す。
CML生成度の評価対象である糖化ゼラチンをPBSで1,800倍、5,400倍希釈した。更にそのサンプルを100μlずつプレートの各ウェルに分注して室温2時間、又は4℃で一晩静置した。その後、サンプルを洗浄液で3回洗浄後、サンプルにブロッキング液200μlを分注し、室温で1時間静置し、次いで、サンプルを洗浄液で3回洗浄後、サンプルに1次抗体を100μlずつ分注し、室温で1時間静置し、次いで、サンプルを洗浄液で3回洗浄後、サンプルに洗浄液で10,000倍希釈した2次抗体を100μlずつ分注し、室温で1時間静置した。その後、サンプルを洗浄液で3回洗浄後、サンプルに発色剤を100μlずつ分注し、室温で発色させた。コントロールの発色のグラデーションがきれいに出たら、サンプルに反応停止液を100μlずつ分注して反応停止させ、492nmの吸光度を測定し、目的とするCMLを定量した。
・ゼラチン:Gelatin,from porcine skin type A (SIGMA G2500-100G)・リボース:D(-)-Ribose (和光純薬 180-01053)。
・ブロッキング液:Gelatin hydrolysate EnzymaticをPBSで希釈して0.5質量%ゼラチン溶液としたものを使用。
・一次抗体:CML:Anti Nε-(carboxylmethyl)lysine(736μg/ml:東海大学より分与)をブロッキング液で1472倍希釈して使用。
・二次抗体:Peroxidase-Labeled Antidoby to Mouse IgG(H+L)(KPL 074-1806)。バイアル(1mg)に水500μlを添加して廻しながら溶解させた後、更にグリセロール500μlを添加してピペッティングで混合し、濃度1mg/mlとしたものを使用。
・発色剤:次の手順で調製したものを使用。100mMクエン酸100ml及び100mMリン酸水素二ナトリウム200mlをそれぞれ別途調製し、前者に後者を添加しながらpHを5.0に調整し、クエン酸緩衝液を得る。室温下、このクエン酸緩衝液10mlとOPDタブレット(和光純薬 158-02151)1粒とを混合し、更に、発色試験の直前に、32%過酸化水素水6μl(関東化学18084-00)を添加し、目的とする発色剤を調製し、使用。
・反応停止液:36N H2SO4(和光純薬 199-15995)をイオン交換水で希釈し、2N H2SO4を調製し、使用。
・洗浄液:PBS(ダルベッコの組成:日水製薬 08190)+0.5%Tween 20 (ポリオキシエチレンソルビタンモノラウレート、東京化成工業 9005-64-5)を使用。
・反応プレート:NUNC MAXISORP(NUNC 439454)を使用。
Claims (3)
- 麦類の溶媒抽出物を有効成分として含有する非酵素的糖化反応抑制剤。
- 上記溶媒は、水、エタノール、含水エタノール、メタノール、含水メタノール、アセトン及び含水アセトンからなる群から選択される1種以上である請求項1記載の非酵素的糖化反応抑制剤。
- 上記麦類は、小麦、大麦、ライ麦及びオーツ麦からなる群から選択される1種以上である請求項1又は2記載の非酵素的糖化反応抑制剤。
