Abstract

本発明は、一般に、幹細胞の発生の分野に関する。特に、本発明は、多能性幹細胞の生成に関する。

本発明の態様は、外因性のベクターエレメントを本質的に含まない人工多能性幹細胞を調製するための新規な方法を提供することによって当技術分野における主要な欠陥を克服し、したがって、iPS細胞の適用の点から特有の利点を提供する。
I.導入
本発明は、GSK−3阻害剤、MEK阻害剤、およびTGF−β受容体阻害剤の存在下において、リプログラミングされた細胞を培養することによってエピソームリプログラミング効率および反応速度を改善するために細胞内シグナル伝達の阻害剤が使用されてもよいという驚くべき発見に部分的に基づく。MEK阻害剤およびTGF−β受容体阻害剤を含む化学反応混液を使用することによって、ヒト線維芽細胞のレトロウイルスリプログラミングが改善されたことが報告された(Linら、2009年)が、レトロウイルス(レンチウイルスを含む)リプログラミングは、レトロウイルスベクターエレメントのゲノム組み込みおよび組み込まれたベクターエレメントの持続的な導入遺伝子発現について、エピソームのリプログラミングとは基本的に異なる。たとえば、実施例において示されるように、これらの3つの阻害剤の組合せの存在下におけるエピソームのリプログラミングは、ベースラインを超える最小限の増強しか有していないMEK阻害剤およびTGF−β受容体阻害剤の存在下における効率と比較して、予想外に高度なリプログラミング効率をもたらした。
本明細書において使用される「初代細胞」は、ある細胞系に樹立されていないまたは固定されていない、生きている生物またはその子孫から直接得られる細胞を指す。「ヒト初代細胞」は、生きているヒト対象から得られる初代細胞を指す。
人工多能性幹細胞は、一般にiPS細胞またはiPSCと略され、非多能性細胞、典型的に、成体の体細胞から人工的に誘導される多能性幹細胞のタイプである。人工多能性幹細胞は、ある種の幹細胞遺伝子およびタンパク質の発現、クロマチンメチル化パターン、倍加時間、胚様体形成、奇形腫形成、生存可能なキメラ形成、ならびに潜在能および分化可能性(differentiability)の点からなどのように、多くの点において、胚性幹細胞などのような自然の多能性幹細胞と同一ではなくても、類似していると考えられるが、自然の多能性幹細胞とのそれらの関係の詳細な程度は、なお評価され続けている。
ヒト体細胞からの多能性幹細胞の誘導は、遺伝子をリプログラミングするために異所性の発現のためにレトロウイルスまたはレンチウイルスベクターを使用して達成されてきた。モロニーマウス白血病ウイルスなどのような組換えレトロウイルスは、安定した方法において宿主ゲノムの中に組み込まれる能力を有する。組換えレトロウイルスは、宿主ゲノムへの組み込みを可能にするリバーストランスクリプターゼを含有する。レンチウイルスは、レトロウイルスのサブクラスである。レンチウイルスは、非分裂および分裂細胞のゲノムに組み込まれる能力のおかげでベクターとして広く適応されている。ウイルスが細胞に入ったとき、RNAの形態をしたウイルスゲノムが逆転写されて、DNAを産生し、DNAは、次いで、ウイルスインテグラーゼ酵素によってランダムな位置でゲノムの中に挿入される。そのため、リプログラミングの成功のための現在の技術は、組み込みベースのウイルスアプローチに依存している。
ヒトヘルペスウイルス4(HHV−4)と呼ばれるエプスタインバーウイルス(EBV)は、ヘルペスファミリー(単純ヘルペスウイルスおよびサイトメガロウイルスを含む)のウイルスであり、ヒトにおいて最も一般的なウイルスの1つである。EBVは、そのゲノムを染色体外で維持し、効率的な複製および維持のために宿主細胞機構と共同で作用し(LindnerおよびSugden、2007年)、細胞分裂の間の細胞内でのその複製およびその保持のために2つの本質的な特徴にもっぱら依存する(Yatesら 1985年;Yatesら 1984年)。一般にoriPと呼ばれる1つのエレメントは、シスで存在し、複製起点として役立つ。他の因子、EBNA−1は、プラスミドDNAの複製および維持を促進するためにoriP内の配列に結合することによってトランスで機能する。非限定的な例として、本発明のある態様は、これらの2つの特徴を抽出し、従来のプラスミドに対して、これらの遺伝子の複製および発現の保持を促進するために、体細胞をリプログラミングするのに必要な遺伝子を往復させるためのベクターと関連してそれらを使用する。
ある態様では、EBVの複製開始点、OriPが使用されてもよい。OriPは、DNA複製が開始する部位にありまたはその近くにあり、family of repeats(FR)およびdyad symmetry(DS)として公知の、およそ1キロ塩基対離れた2つのシス作用性配列から構成される。
トランス作用因子の特定の例は、エプスタイン−バー核抗原1(EBNA−1)とすることができ、これは、それぞれの細胞分裂の間に、細胞染色体と無関係であるが、それと協力する複製および娘細胞へのEBVベースのベクターの正確な分配を促進するために、oriPのFRおよびDSまたはRep*に結合するDNA結合タンパク質である。
重要なことには、oriPベースのエピソームベクターの複製および維持は、不完全であり、それが細胞の中に導入される最初の2週間以内に細胞から急に(細胞分裂当たり25%)失われるが、プラスミドを保持する細胞は、それをそれほど頻繁に失わない(細胞分裂当たり3%)(LeightおよびSugden、2001年;NanboおよびSugden、2007年)。一度、プラスミドを持つ細胞についての選択を除いたら、プラスミドは、結果として生じる娘細胞内にその前者の存在のフットプリントを残すことなく、それらがすべてある期間にわたって排除されるまで、それぞれの細胞分裂の間に失われるであろう。本発明のある態様は、iPS細胞を生成するための遺伝子を送達するための現在のウイルス関連アプローチに対する代わりとして、oriPベースの系のこのフットプリントレスの特徴を使用する。他の染色体外ベクターもまた、宿主細胞の複製および増殖の間に失われるであろうし、これらもまた、本発明において利用することができる。
iPS細胞の生成では、誘導に使用される遺伝子が決定的である。以下の因子またはその組合せは、本発明において開示されるベクター系において使用することができる。ある態様では、SoxおよびOct(好ましくはOct3/4)をコードする核酸は、リプログラミングベクターの中に含まれるであろう。たとえば、リプログラミングベクターは、Sox2、Oct4、Nanog、および任意選択でLin−28をコードする発現カセットまたはSox2、Oct4、Klf4、および任意選択でc−mycをコードする発現カセットを含んでいてもよい。これらのリプログラミング因子をコードする核酸は、同じ発現カセット、異なる発現カセット、同じリプログラミングベクター、または異なるリプログラミングベクターにおいて含まれてもよい。
本発明のある態様では、リプログラミングプロセスの少なくとも一部の間に、細胞は、シグナル伝達カスケードに関与するシグナルトランスデューサーを阻害する1つ以上のシグナル伝達阻害剤の存在下において、たとえば、MEK阻害剤、GSK3阻害剤、TGF−β受容体阻害剤、MEK阻害剤およびGSK3阻害剤の両方、GSK3阻害剤およびTGF−β受容体阻害剤の両方、MEK阻害剤およびTGF−β受容体阻害剤の両方、3つの阻害剤すべての組合せ、または、これらの同じ経路内の他のシグナルトランスデューサーの阻害剤の存在において維持されてもよい。ある態様では、HA−100などのようなROCK阻害剤またはブレビスタチンなどのようなミオシンII阻害剤は、リプログラミングされた細胞および結果として生じるiPS細胞のクローン増殖を促進するために使用されてもよい。馴化ヒトES細胞培養培地などのような特定のリプログラミング培地または無血清限定N2B27培地(serum−free defined N2B27 medium)などのような既知組成培地(chemically defined medium)と組み合わせた高濃度のFGFもまた、リプログラミング効率を増加させるために使用されてもよい。
グリコーゲン合成酵素キナーゼ3(GSK−3)は、特定の細胞基質におけるあるセリンおよびトレオニンアミノ酸上へのリン酸分子の追加を媒介するセリン/トレオニンプロテインキナーゼである。GSK−3によるこれらの他のタンパク質のリン酸化は、普通、標的タンパク質(「基質」とも呼ばれる)を阻害する。述べられるように、GSK−3は、グリコーゲン合成酵素をリン酸化し、したがって不活性化することが公知である。それはまた、損傷DNAおよびWntシグナル伝達に対する細胞の応答のコントロールに関係する。GSK−3はまた、Hedgehog(Hh)経路においてCiをもリン酸化し、タンパク質分解のためにそれを不活性形態に向ける。グリコーゲン合成酵素に加えて、GSK−3は、他の多くの基質を有する。しかしながら、GSK−3は、それが、普通、基質を最初にリン酸化するために「プライミングキナーゼ」を必要とするという点で、キナーゼの中で珍しい。
マイトジェン活性化プロテインキナーゼキナーゼ(MAPK/ERKキナーゼもしくはMEK)またはMAPKカスケードのようなその関係するシグナル伝達経路の阻害剤を含むMEK阻害剤が、本発明のある態様において使用されてもよい。マイトジェン活性化プロテインキナーゼキナーゼ(sic)は、マイトジェン活性化プロテインキナーゼをリン酸化するキナーゼ酵素である。マイトジェン活性化プロテインキナーゼキナーゼはまた、MAP2Kとしても知られている。細胞外刺激は、MAPキナーゼ、MAPキナーゼキナーゼ(MEK、MKK、MEKK、またはMAP2K)、およびMAPキナーゼキナーゼキナーゼ(MKKKまたはMAP3K)から構成されるシグナル伝達カスケード(「MAPKカスケード」)を介してMAPキナーゼの活性化を導く。

多能性幹細胞、特にヒトES細胞およびiPS細胞は、細胞の脱離および解離に際してアポトーシスを受けやすく、細胞の脱離および解離は、クローンの単離または増殖および分化誘導にとって重要である。最近、ROCK関連シグナル伝達経路のための阻害剤であるRho関連キナーゼ(ROCK)阻害剤、たとえばRho特異的阻害剤、ROCK特異的阻害剤、またはミオシンII特異的阻害剤などのような小さなクラスの分子は、解離した多能性幹細胞のクローンの効率および生存を増加させることが分かった。本発明のある態様では、ROCK阻害剤は、多能性幹細胞の培養および継代そして/または幹細胞の分化のために使用されてもよい。そのため、ROCK阻害剤は、接着培養液または浮遊培養液などのような、多能性幹細胞が成長する、解離する、凝集物を形成する、または分化を受ける任意の細胞培養培地中に存在することができる。本明細書においてその他に述べられない限り、ブレビスタチンなどのようなミオシンII阻害剤は、ROCK阻害剤の実験的な使用のために代用することができる。
開始細胞および目的のリプログラミングされた細胞は、一般に、培養培地および培養条件について異なる必要条件を有する。細胞のリプログラミングが起こることをも可能にしながらこれを可能にするために、開始細胞の成長に適することが公知の培地の存在下においておよび培養条件下で、リプログラミング因子の導入後に、培養の少なくとも1つの初期段階を実行することは普通である。血清を有するフィーダー上での、リプログラミング培地の存在下におけるおよび多能性細胞に適することが公知の条件下でのまたは既知組成培地もしくはフィーダーフリーの条件を使用する培養の続く期間が、これに続く。適したフィーダーは、それらがリプログラミングされている細胞の成長を押しのけないように典型的に不活性化された初代または不死化線維芽細胞系を含む。リプログラミングのための十分な時間の後に、リプログラミングされた細胞は、増殖培地において、iPS細胞の選択の前またはその後に、iPS細胞の増殖のためにさらに培養されてもよい。そのような増殖培地は、上記に記載される1つ以上のシグナル伝達阻害剤を含むまたはこれらの阻害剤を本質的に含まない培養培地を含んでいてもよい。
本発明による培養条件は、使用される培地および幹細胞に依存して、適切に決定されるであろう。本発明のある態様による培地は、TeSR、BME、BGJb、CMRL 1066、Glasgow MEM、Improved MEM Zinc Option、IMDM、Medium 199、Eagle MEM、αMEM、DMEM、Ham、RPMI 1640、およびFischerの培地ならびにその任意の組合せのいずれかなどのような、その基本培地として、動物細胞を培養するために使用される培地を使用して調製することができるが、培地は、動物細胞を培養するためにそれを使用することができる限り、その培地に特に限定されない。
培養条件に依存して、多能性幹細胞は、分化細胞または未分化の細胞のコロニーを産生することができる。用語「分化する」は、発生経路に沿った細胞の進行を意味する。用語「分化した」は、他の細胞と比較した、発生経路に沿った細胞の進行を説明する相対的な用語である。たとえば、多能性細胞は、体のあらゆる細胞を誘発することができるが、造血細胞などのようなより分化した細胞は、より少ない細胞型を誘発するであろう。
C.細胞継代
本発明のある態様は、幹細胞を解離させるステップをさらに含むことができる。幹細胞解離は、任意の公知の手順を使用して実行することができる。これらの手順は、キレート剤(EDTAなど)、酵素(トリプシン、コラゲナーゼなど)、またはその他同種のものを用いる処理および機械的な解離(ピペット操作など)などのような操作を含む。幹細胞(複数可)は、解離後におよび/または前にROCK阻害剤を用いて処理することができる。たとえば、幹細胞(複数可)は、解離後にのみ処理することができる。
本発明のある態様では、1つ以上の染色体外遺伝エレメントが体細胞の中に導入された後に、細胞は、増殖のために培養されてもよく、(トランスフェクトされた細胞を濃縮するために、ポジティブ選択マーカーまたはスクリーニング可能なマーカーのようなベクターエレメントの存在について任意選択で選択されてもよく)、これらの遺伝エレメントは、これらの細胞においてリプログラミング因子を発現し、複製し、細胞分裂と共に分割するであろう。これらの発現されたリプログラミング因子は、体細胞ゲノムをリプログラミングして、自立多能性状態を確立し、その間にまたはベクターの存在についてのポジティブ選択を除いた後に、外因性の遺伝エレメントは、徐々に失われるであろう。これらの人工多能性幹細胞は、それらが、多能性胚性幹細胞と実質的に同一であると予想されるので、胚性幹細胞の特質に基づいて、これらの体細胞から誘導された子孫から選択することができる。さらなるネガティブセレクションステップは、リプログラミングベクターDNAの不在を試験するまたは選択マーカーを使用することによって、外因性の遺伝エレメントを本質的に含まないiPS細胞についての選択を速めるまたは助けるために利用することができる。
従来の研究からの生成に成功したiPSCは、以下の点において、自然に単離された多能性幹細胞(マウスおよびヒト胚性幹細胞、それぞれmESCおよびhESCなど)に著しく類似し、したがって、自然に単離された多能性幹細胞に対するiPSCの同一性、確実性、および多能性を確認した。したがって、本発明において開示される方法から生成される人工多能性幹細胞は、1つ以上の以下の胚性幹細胞の特質に基づいて選択することができる。
形態:iPSCは、ESCに形態学的に類似する。細胞はそれぞれ、円形、大きな核小体、およびわずかな細胞質を有していてもよい。iPSCのコロニーもまた、ESCに類似し得る。ヒトiPSCは、hESCに類似する鋭い縁の平らな、しっかりと詰まったコロニーを形成し、マウスiPSCは、mESCに類似するコロニー、hESCよりも平らではなく、より凝集したコロニーを形成する。ある実施形態では、本方法は、大きなヒトiPS細胞を生成してもよく、これらは、少なくとももしくは約1.5、1.6、1.7、1.8、1.9、2.0、2.1、2.2、2.3、2.4、2.5mmまたはその中で推論できる任意の範囲の直径を有していてもよく、非iPS細胞から容易に認識できてもよい。
ii.後成的なリプログラミング
プロモーター脱メチル化:メチル化は、DNA塩基に対するメチル基の移入、典型的に、CpG部位(近接するシトシン/グアニン配列)におけるシトシン分子に対するメチル基の移入である。遺伝子の広範囲のメチル化は、発現タンパク質の活性を妨げるまたは発現に干渉する酵素を動員することによって、発現に干渉する。したがって、遺伝子のメチル化は、転写を妨げることによって有効にそれをサイレンシングする。Oct−3/4、Rex1、およびNanogを含む、多能性関連遺伝子のプロモーターは、iPSCにおいて脱メチル化され、iPSCにおいて、それらのプロモーター活性ならびに多能性関連遺伝子の活性促進および発現を示してもよい。
本発明におけるoriPベースのベクターなどのようなリプログラミングベクターは、染色体外で複製し、数代後に宿主細胞においてその存在を失わせることができる。しかしながら、外因性のベクターエレメントを本質的に含まない子孫細胞についてのさらなる選択ステップは、このプロセスを容易にしてもよい。たとえば、子孫細胞の試料は、当技術分野において公知の外因性のベクターエレメントの存在または損失を試験するために抽出されてもよい(LeightおよびSugden、2001年)。
ある実施形態では、リプログラミングベクターは、細胞においてリプログラミング因子を発現するために、上記に記載されるこれらのリプログラミング因子をコードする核酸配列に加えて、さらなるエレメントを含むように構築することができる。これらの方法の1つの特徴は、染色体外複製ベクターの使用であり、これは、宿主細胞ゲノムの中に組み込まれないであろう、また、多数の複製の間に失われてもよい。これらのベクターの構成成分および送達の方法の詳細については、下記に開示される。
プラスミドもしくはリポソームベースの染色体外ベクター、たとえばoriPベースのベクターおよび/またはEBNA−1の誘導体をコードするベクターの使用は、DNAの大きな断片が細胞に導入され、染色体外で維持され、細胞周期当たり1回複製され、娘細胞に効率的に分割し、免疫応答を実質的に誘発しないことを可能にする。特に、oriPベースの発現ベクターの複製を担うウイルスタンパク質であるEBNA−1は、それが、MHCクラスI分子上のその抗原の提示に必要とされる処理を迂回するための効率的なメカニズムを発達させたので、細胞性免疫応答を誘発しない(Levitskayaら、1997年)。さらに、EBNA−1は、クローニング遺伝子の発現を増強するために、トランスで作用し、いくつかの細胞系において、100倍まで、クローニング遺伝子の発現を誘導することができる(Langle−Rouaultら、1998年;Evansら、1997年)。最後に、そのようなoriPベースの発現ベクターの製造は、低費用である。
ベクター中に含まれる真核生物発現カセットは、好ましくは、コード配列に操作可能に連結された真核生物転写プロモータータンパク質、介在配列を含むスプライスシグナル、および転写終結/ポリアデニル化配列を含有する(5’から3’方向に)。
i.プロモーター/エンハンサー
「プロモーター」は、転写の開始および速度がコントロールされる核酸配列の領域であるコントロール配列である。プロモーターは、核酸配列の特異的な転写を開始するために、RNAポリメラーゼおよび他の転写因子などのような調節タンパク質および分子が、結合してもよい遺伝エレメントを含有していてもよい。語句「作動可能に位置する」、「作動可能に連結された」、「コントロール下の」、および「転写コントロール下の」は、プロモーターが、その配列の転写開始および/または発現をコントロールするために、核酸配列に関して適正な機能的な場所にあるおよび/または方向づけをされていることを意味する。
特異的な開始シグナルもまた、コード配列の効率的な翻訳のために必要とされてもよい。これらのシグナルは、ATG開始コドンまたは近接する配列を含む。ATG開始コドンを含む外因性翻訳コントロールシグナルが提供される必要があってもよい。当業者はこれを容易に決定し、必要なシグナルを提供することができるであろう。開始コドンは、挿入物全体の翻訳を確実にするために、所望のコード配列のリーディングフレームとインフレームでなければならないことは周知である。外因性翻訳コントロールシグナルおよび開始コドンは、自然または合成のものであってもよい。発現の効率は、適切な転写エンハンサーの包含によって増強されてもよい。
iii.マルチクローニング部位
ベクターは、マルチクローニング部位(MCS)を含むことができ、これは、複数の制限酵素部位を含有する核酸領域であり、これらのいずれも、ベクターを消化するための標準的な組換え技術と共に使用することができる(たとえば、参照によって本明細書において組み込まれるCarbonelliら、1999年、Levensonら、1998年、およびCocea、1997年を参照されたい)。「制限酵素消化」は、核酸分子中の特異的な場所でのみ機能する酵素を用いる核酸分子の触媒性の切断を指す。これらの制限酵素の多くは、市販で入手可能である。そのような酵素の使用は、当業者らによって広く理解されている。しばしば、ベクターは、外因性の配列がベクターにライゲーションされるのを可能にするために、MCS内を切断する制限酵素を使用して、直線化されるまたは断片化される。「ライゲーション」は、2つの核酸断片の間にホスホジエステル結合を形成するプロセスを指し、これらは、互いに隣接していてもよいまたは隣接していなくてもよい。制限酵素およびライゲーション反応に関する技術は、組換え技術の当業者らに周知である。
ほとんどの転写真核生物RNA分子は、一次転写物からイントロンを除去するためにRNAスプライシングを受けるであろう。ゲノム真核生物配列を含有するベクターは、タンパク質発現のための転写の適切な処理を確実にするために、ドナーおよび/またはアクセプタースプライシング部位を必要としてもよい(たとえば、参照によって本明細書において組み込まれるChandlerら、1997年を参照されたい)。
本発明のベクターまたは構築物は、一般に、少なくとも1つの終結シグナルを含むであろう。「終結シグナル」または「ターミネーター」は、RNAポリメラーゼによるRNA転写の特異的な終結に関与するDNA配列から構成される。したがって、ある実施形態では、RNA転写物の産生を終了する終結シグナルが企図される。ターミネーターは、望ましいメッセージレベルを達成するためにin vivoにおいて必要であってもよい。
vi.ポリアデニル化シグナル
発現、特に真核生物の発現では、典型的に、転写物の適切なポリアデニル化を引き起こすためのポリアデニル化シグナルを含むであろう。ポリアデニル化シグナルの性質は、本発明の実施の成功に重大であると考えられず、任意のそのような配列が利用されてもよい。好ましい実施形態は、様々な標的細胞において十分に機能するのに好都合であり、そのことが公知であるSV40ポリアデニル化シグナルまたはウシ成長ホルモンポリアデニル化シグナルを含む。ポリアデニル化は、転写物の安定性を増加させてもよいまたは細胞質輸送を促進してもよい。
宿主細胞においてベクターを増殖させるために、ベクターは、複製が開始される特異的な核酸配列である、1つ以上の複製起点部位(多くの場合、「ori」と称される)、たとえば、上記に記載されるEBVのoriPに対応する核酸配列を含有していてもよい。その代わりに、上記に記載される他の染色体外で複製するウイルスの複製開始点または自己複製配列(ARS)を利用することができる。
viii.選択マーカーおよびスクリーニング可能なマーカー
本発明のある実施形態では、本発明の核酸構築物を含有する細胞は、発現ベクター中にマーカーを含むことによって、in vitroまたはin vivoにおいて同定されてもよい。そのようなマーカーは、細胞に同定可能な変化を与え、発現ベクターを含有する細胞の容易な同定を可能にするであろう。一般に、選択マーカーは、選択を可能にする特性を与えるものである。ポジティブ選択マーカーは、マーカーの存在がその選択を可能にするものであるが、ネガティブ選択マーカーは、その存在がその選択を妨げるものである。ポジティブ選択マーカーの例は、薬剤抵抗性マーカーである。
本発明を用いる体細胞へのリプログラミングベクターの導入は、本明細書において記載されるようにまたは当業者に公知であるように、細胞の形質転換のために、任意の核酸送達に適した方法を使用してもよい。そのような方法は、ex vivoトランスフェクションによって(Wilsonら、1989年、Nabelら、1989年);マイクロインジェクション(HarlandおよびWeintraub、1985年;参照によって本明細書において組み込まれる米国特許第5,789,215号)を含む注射によって(それぞれ参照によって本明細書において組み込まれる米国特許第5,994,624号、第5,981,274号、第5,945,100号、第5,780,448号、第5,736,524号、第5,702,932号、第5,656,610号、第5,589,466号、および第5,580,859号);エレクトロポレーションによって(参照によって本明細書において組み込まれる米国特許第5,384,253号;Tur−Kaspaら、1986年;Potterら、1984年);リン酸カルシウム沈殿によって(GrahamおよびVan Der Eb、1973年;ChenおよびOkayama、1987年;Rippeら、1990年);ポリエチレングリコールが後続するDEAEデキストランの使用によって(Gopal、1985年);直接的な音波ローディングによって(Fechheimerら、1987年);リポソーム媒介性のトランスフェクションによって(NicolauおよびSene、1982年;Fraleyら、1979年;Nicolauら、1987年;Wongら、1980年;Kanedaら、1989年;Katoら、1991年)ならびに受容体媒介性のトランスフェクション(WuおよびWu、1987年;WuおよびWu、1988年)によって;微粒子銃によって(それぞれ、参照によって本明細書において組み込まれるPCT出願WO94/09699および95/06128;米国特許第5,610,042号;第5,322,783号 第5,563,055号、第5,550,318号、第5,538,877号、および第5,538,880号);炭化ケイ素繊維を用いる撹拌によって(それぞれ参照によって本明細書において組み込まれるKaepplerら、1990年;米国特許第5,302,523号および第5,464,765号);乾燥/阻害媒介性のDNA取り込み(Potrykusら、1985年)によって、ならびにそのような方法の任意の組合せなどのように、DNAの直接的な送達を含むが、これらに限定されない。これらの技術などのような技術の適用を通して、細胞小器官(複数可)、細胞(複数可)、組織(複数可)、または生物(複数可)は、安定してまたは一時的に形質転換されてもよい。
本発明のある実施形態では、核酸は、たとえばリポソームなどのような脂質複合体中に封入されてもよい。リポソームは、リン脂質二重層膜および内部水性媒体によって特徴付けられる小胞性の構造物である。多重膜リポソームは、水性媒体によって分離される複数の脂質層を有する。多重膜リポソームは、リン脂質が過剰な水溶液中に懸濁される場合、自発的に形成される。脂質成分は、閉鎖した構造物の形成の前に自己再配列を受けて、脂質二分子膜の間に水および溶解した溶質を封入する(GhoshおよびBachhawat、1991年)。Lipofectamine(Gibco BRL)またはSuperfect(Qiagen)と複合体を形成した核酸もまた、企図される。使用されるリポソームの量は、リポソームの性質および使用される細胞次第で変動してもよく、たとえば、1〜10百万の細胞当たり約5〜約20μgベクターDNAが企図されてもよい。
本発明のある実施形態では、核酸は、エレクトロポレーションを介して、細胞小器官、細胞、組織、または生物の中に導入される。エレクトロポレーションは、細胞およびDNAの懸濁液の高電圧放電への曝露を含む。レシピエント細胞は、機械的な傷によって形質転換に対してより感受性にすることができる。さらに、使用されるベクターの量は、使用される細胞の性質次第で変動してもよく、たとえば、1〜10百万の細胞当たり約5〜約20μgベクターDNAが企図されてもよい。
本発明の他の実施形態では、核酸は、リン酸カルシウム沈殿を使用して、細胞に導入される。ヒトKB株細胞は、この技術を使用して、アデノウイルス5 DNA(GrahamおよびVan Der Eb、1973年)がトランスフェクトされた。さらに、このようにして、マウスL(A9)、マウスC127、CHO、CV−1、BHK、NIH3T3、およびHeLa細胞は、ネオマイシンマーカー遺伝子がトランスフェクトされ(ChenおよびOkayama、1987年)、ラット肝細胞は、様々なマーカー遺伝子がトランスフェクトされた(Rippeら、1990年)。
他の実施形態では、核酸は、ポリエチレングリコールが後続するDEAE−デキストランを使用して、細胞の中に送達される。このようにして、レポータープラスミドは、マウス骨髄腫細胞および赤白血病細胞の中に導入された(Gopal、1985年)。
本発明のさらなる実施形態は、直接的な音波ローディングによる核酸の導入を含む。LTK−線維芽細胞は、音波処理ローディングによってチミジンキナーゼ遺伝子がトランスフェクトされた(Fechheimerら、1987年)。
さらに、核酸は、受容体媒介性の送達ビヒクルを介して目的の細胞に送達されてもよい。これらは、標的細胞において起こるであろう受容体媒介性のエンドサイトーシスによる高分子の選択的な取り込みを利用する。様々な受容体の細胞型に特異的な分布を考慮して、この送達方法は、本発明に他の特異性を追加する。
vii 微粒子銃
微粒子銃技術は、少なくとも1つの細胞小器官、細胞、組織、または生物の中に核酸を導入するために使用することができる(米国特許第5,550,318号;米国特許第5,538,880号;米国特許第5,610,042号;およびPCT出願WO94/09699;それぞれ、参照によって本明細書において組み込まれる)。この方法は、DNAをコーティングする微粒子を高速度まで加速し、それらが細胞膜を貫通し、かつそれらを死滅させることなく細胞に入ることを可能にする能力に依存する(Kleinら、1987年)。当技術分野において公知の種々様々の微粒子銃技術があり、これらの多くは、本発明に適用可能である。
以下の実施例は、本発明の好ましい実施形態を示すために含まれる。続く実施例において開示される技術は、本発明の実施において十分に機能するように、発明者によって発見された技術を示し、したがって、その実施のための好ましいモードを構成すると考えることができることが、当業者らによって十分に理解されるはずである。しかしながら、当業者らは、本開示を考慮して、本発明の精神および範囲から逸脱することなく、多くの変化が、開示される特定の実施形態において成され、同様のまたは類似する結果をなお得ることができることを十分に理解するはずである。
小さな化学化合物を用いるヒト包皮線維芽細胞のエピソームリプログラミング
iPS細胞誘導に影響を与える公知の化学化合物のスクリーニングを通して、発明者らは、ヒト包皮線維芽細胞のエピソームリプログラミング効率を著しく改善したいくつかの化合物を同定した。BIX01294(B)は、G9aヒストンメチルトランスフェラーゼの選択的阻害剤である。BayK8644と組み合わせて、それは、Oct4およびKlf4のみが形質導入されたマウス胚性線維芽細胞のリプログラミングを可能にすることができた小分子として最初に同定された(Shiら、2008年)。PD0325901(P)は、マイトジェン活性化プロテインキナーゼキナーゼ(MAPK/ERKキナーゼまたはMEK)の阻害剤である。CHIR99021(C)は、GSK3βの最も選択的な阻害剤であり、一方、A−83−01(A)は、TGF−βI型受容体ALK5、アクチビン/Nodal受容体ALK4、およびnodal受容体ALK7の強力な阻害剤である。PD0325901およびCHIR99021の組合せ(2i)は、不応性株からのマウスES細胞の効率的な誘導を可能にした(Yingら、2008年)。2i条件において、重要でないが、マウスES細胞培養に決まって使用される白血病抑制因子(LIF)は、マウスES細胞のクローン形成能および誘導を促進した(Yingら、2008年)。この2i/LIF条件は、マウス神経幹細胞の真の多能性へのリプログラミングを促進することが示された(Silvaら、2008年)。興味深いことに、真のラットES細胞は、この2i/LIF条件において初期胚から直ちに誘導することができたが(Buehrら、2008年;Liら、2008年)、A−83−01の追加は、ラットiPS細胞の長期的な培養を保持するために必要とされることが示された(Liら、2009年)。PD0325901、CHIR99021、A−83−01、およびLIFの組合せもまた、レンチウイルス媒介性のリプログラミング培養由来の異型マウスES細胞様ヒトiPS細胞を選択し、安定化することができることが示された(Liら、2009年)が、リプログラミング、特にエピソームリプログラミングに対するこれらの阻害剤の組合せの効果は、立証されていない。
小分子を用いるフィーダーフリーエピソームリプログラミング
ヒトiPSCは、ヒト胚性幹細胞(ESC)に類似して、無限の増殖が可能であり、体のすべての細胞型に分化するための潜在能力を有する。これらの細胞は、したがって、基本的な生物学、疾患モデリング、薬剤開発、および移植療法における適用を有する。リプログラミング因子の決定されたセットの発現によって、iPSCは、異なる種の多くの細胞型から生成された(Takahashiら、2007年;Yuら、2007年;TakahashiおよびYamanaka、2006年;Liuら、2008年;Estebanら、2009年;Lohら、2009年;Sunら、2009年;Shimadaら、2010年)。iPSC生成のための初めの方法は、ゲノム組み込み型レトロウイルスまたはレンチウイルスベクターを利用した(Yuら、2007年;TakahashiおよびYamanaka、2006年)。これらのアプローチは、腫瘍形成性の挿入変異をもたらし得、iPSC分化の間の導入遺伝子発現の残留または再活性化は、iPSC誘導体の系列選択および機能性に影響を与え得た(Yuら、2007年;Okitaら、2007年)。これらの問題を克服するために、リプログラミング因子(プラスミド、小環状DNA、非組み込み型アデノウイルスベクター、およびタンパク質)を用いる繰り返しの処理、トランスポゾン、ならびにRNAウイルスベクター(Okitaら、2008年;Stadtfeldら、2008年;Fusakiら、2009年;Kajiら、2009年;Woltjenら、2009年;Zhouら、2009年;Jiaら、2010年)を含む様々な方法が、フットプリントフリーのiPSCを誘導するために開発された。しかしながら、これらの方法は、以下の1つ以上の制限を受ける:容認できない低リプログラミング効率;iPSCからのリプログラミング因子の重労働な除去;ウイルスパッケージ細胞またはフィーダー細胞の必要。したがって、多くのヒトドナー試料からのフットプリントフリーのiPSCのルーチン的な誘導および最終的に、臨床グレードのヒトiPSCの誘導を可能にするための単純で効率的なフィーダーフリーの方法を開発するための必要性がある。

Claims (40)
- iPS細胞の集団を作製するための方法であって、
a)1つ以上のリプログラミング因子を発現する染色体外遺伝エレメントを含む体細胞を得るステップと、
b)外部添加のGSK−3阻害剤、MEK阻害剤、およびTGF−β受容体阻害剤を含むリプログラミング条件において該体細胞およびその子孫細胞を培養し、それによってiPS細胞の集団を作製するステップと
を含む方法。 - 前記リプログラミング条件は、フィーダー細胞を本質的に含まない、請求項1に記載の方法。
- 前記リプログラミング条件は、マトリックス構成成分を含む、請求項1に記載の方法。
- 前記マトリックス構成成分は、Matrigel(商標)を含む、請求項3に記載の組成物。
- 前記体細胞は、ヒト細胞である、請求項1に記載の方法。
- 前記体細胞は、線維芽細胞、ケラチノサイト、造血細胞、間葉細胞、脂肪細胞、内皮細胞、神経細胞、筋細胞、乳房細胞、肝細胞、腎細胞、皮膚細胞、消化管細胞、卵丘細胞、腺細胞、または膵島細胞である、請求項1に記載の方法。
- 前記染色体外遺伝エレメントは、DNAを含む、請求項1に記載の方法。
- 前記染色体外遺伝エレメントは、RNAを含む、請求項1に記載の方法。
- 前記染色体外遺伝エレメントは、エピソームベクターとしてさらに定義される、請求項1に記載の方法。
- 前記エピソームベクターは、細菌エレメントを本質的に含まない、請求項9に記載の方法。
- 前記エピソームベクターは、リプログラミング因子の発現のための複製開始点および1つ以上の発現カセットを含み、該発現カセットの1つ以上は、染色体外鋳型を複製するために複製開始点に結合するトランス作用因子をコードするヌクレオチド配列をさらに含み、そして/または前記体細胞は、そのようなトランス作用因子を発現する、請求項9に記載の方法。
- 前記複製開始点は、リンパ向性ヘルペスウイルスの複製開始点であり、エプスタインバーウイルス(EBV)のoriPに対応する、請求項11に記載の方法。

- 前記リプログラミング因子は、形質転換が欠損しているmyc変異体またはホモログを含む、請求項15に記載の方法。
- 前記リプログラミング因子は、LMYC(NM_001033081)、N末端の41アミノ酸が欠失したMYC(dN2MYC)、またはアミノ酸136に変異を有するMYC(W136E)を含む、請求項16に記載の方法。
- 前記細胞は、少なくとも5日間、前記リプログラミング条件において培養される、請求項1に記載の方法。
- 前記細胞は、前記体細胞への前記染色体外遺伝エレメントの導入後の少なくとも約1日間〜5日間を含む期間、前記リプログラミング条件において培養される、請求項18に記載の方法。
- 前記細胞は、前記体細胞への前記染色体外遺伝エレメントの導入後の約1日間〜15日間を含む期間、前記リプログラミング条件において培養される、請求項19に記載の方法。
- 外部添加のGSK−3阻害剤、MEK阻害剤、およびTGF−β受容体阻害剤を本質的に含まない増殖培地を有する増殖条件において前記iPS細胞を培養するステップをさらに含む、請求項1に記載の方法。
- 前記増殖培地は、既知組成培地である、請求項21に記載の方法。
- 前記増殖培地は、TeSR培地またはmTeSR培地である、請求項22に記載の方法。
- 前記iPS細胞を選択するステップをさらに含む、請求項1に記載の方法。
