幾つかの態様において、本発明の洗浄組成物は少なくとも1つのキレート剤を含む。好適なキレート剤は銅、鉄、及び/又はマンガンキレート剤、並びにこれらの混合物を含む。少なくとも1つのキレート剤を用いる態様において、本発明の洗浄組成物はこの洗浄組成物の重量に基づいて、約0.1%乃至約15%、更には約3.0%乃至約10%、のキレート剤を含む。
本発明の洗浄組成物は沈着補助剤を含むことができる。好適な沈着補助剤には、ポリエチレングリコール、ポリカルボン酸、ポリテレフタル酸のような防汚ポリマー、カオリナイト、モンモリロナイト、アタパルガイト、イライト、ベントナイト、ハロイサイトのような粘土、およびこれらの混合物が含まれる。
本明細書において、再付着防止剤が本発明の幾つかの態様に用いられる。幾つかの好適な態様において、非イオン界面活性剤が用いられる。例えば、洗浄組成物の態様において、非イオン界面活性剤は目的物の表面を修飾するために用いられ、特に汚れに関して、膜形成及びスポット形成を避け、輝きを増すために用いられる。これらの非イオン界面活性剤は汚れの再付着を防止するためにも用いる。幾つかの好適な態様において、再付着防止剤は当業者に知られている非イオン界面活性剤である。(例えば、EP2100949参照。)
本発明の洗浄組成物はまた、一つ以上の染料転移阻害剤を含むことができる。好適なポリマー性染料転移阻害剤には、ポリビニルピロリドン・ポリマー、ポリアミン−N−オキシド・ポリマー、N−ビニルピロリドンおよびN−ビニルイミダゾールのコポリマー、ポリビニルオキサゾリドンおよびポリビニルイミダゾール、またはこれらの混合物が含まれるが、これらに限定されない。対象洗浄組成物に存在する場合、前記染料阻害剤は洗浄組成物の重量で約0.0001%〜約10%、約0.01%〜約5%、またはさらに約0.1%〜約3%のレベルで存在することができる。
幾つかの態様において、ケイ酸塩を本発明の組成物に含むことができる。幾つかのそのような態様において、ケイ酸ナトリウム(例えば、二ケイ酸ナトリウム、メタケイ酸ナトリウム、及び結晶性フィロシリケート)を用いる。幾つかの態様において、ケイ酸は約1%乃至約20%のレベルで存在する。幾つかの好適な態様において、ケイ酸塩は組成物の重量により約5%乃至約15%含まれる。
幾つかの態様において、本発明の洗浄組成物は少なくとも1つの分散を含む。好適な水溶性有機物質は、ホモ又はコポリマー酸又はそれらの塩を含むがこれらに限定されない。ポリカルボン酸はお互いに離れた少なくとも2つのカルボキシルラジカルを含み、炭素原子は2を超えない。

本発明の幾つかの態様において、本発明の洗剤組成物に用いる酵素を安定化させる。幾つかの態様において、この酵素安定剤はオリゴサッカライド、ポリサッカライド、及びカルシウム塩等のアルカリ土類金属を含む無機二価金属塩を含む。酵素を安定化するための各種技術を本発明に用いることができる。例えば、幾つかの態様において、本明細書で用いる酵素は組成物中に亜鉛(II)、カルシウム(II)、及び/又はマグネシウム(II)イオン、並びに他の金属イオン(例えば、バナジウム(II)、スカンジウム(II)、鉄(II)、マンガン(II)、アルミニウム(II)、スズ(II)、コバルト(II)、銅(II)、ニッケル(II)及びアキソバナジウム(II))の水溶性ソースを存在させることにより安定化される。それらはイオンを酵素に提供する。塩化物及び硫酸塩も本発明の幾つかの態様に用いることができる。好適なオリゴサッカライソ及びポリサッカライド(例えば、デキストリン)の例は当業者に知られている(例えば、WO07/145964参照)。幾つかの態様において、可逆性プロテアーゼインヒビターも用いることができ、そのようなものとしてホウ酸含有化合物がある(例えば、ホウ酸塩、4−ホルミルフェニルボロン酸)。必要に応じてトリペプチドアルデヒドも更に安定化させるために用いることができる。

スブチリシン変異体の構築
スブチリシン変異体は既知の融合PCRにより調製した。(例えば、US Publn. No.2006/0252155参照。該文献を参照により本明細書に援用する。)表1−1は融合PCRで用いたプライマーの配列を提供する。
バチルススブチリス(Bacillus subtilis)におけるスブチリシン変異体の生成
バチルススブチリス(B.subtilis)形質転換体を10mlのTSB(トリプトン及びダイズブロス)培地中で37℃一晩育成してスブチリシン変異体を生成した。一晩培養物の250μLのアリコートを100ml振とうフラスコ中にある25mlのMOPSベース既成培地に移し、37℃で68時間育成した。この既成培地は、NH4Cl2、FeSO4、及びCaCl2が取り除かれ、3mMK2HPO4を用いている以外は当業者に知られているものである(例えば、Neidhardt et al., J Bacteriol,119:736−747,1974参照。)この培地は、60mM尿素、75g/Lグルコース、及び 1%ソイトンを補っている。微量元素は100倍ストックとして調製した。該ストックは1リットル当たり400mgのFeSO4.7H2O、100mgのMnSO4.H2O、100mgのZnSO4.7H2O、50mgのCuCl2.2H2O、100mgのCOCl2.6H2O、100mgのNaMoO4.2H2O、100mg のNa2B4O7.10H2O、10mlの1MCaCl2、及び10mlの0.5Mクエン酸ナトリウムを含む。所望のプロテアーゼは培養培地から単離された。
スブチリシンサンプルの精製度を決定するための解析方法
この実施例において、バチルススブチリス(B.subtilis)培養物から得られた組換えスブチリシンの精製度を決定するために用いる方法を説明する。ゲル電気泳動及び高圧液体クロマトグラフ(HPLC)において、それぞれ単一のバンド及びピークが検出されたときにプロテアーゼは精製されたと考えられる。
スブチリシン濃度の決定
この実施例において、スブチリシン濃度の決定に用いる方法を説明する。幾つかの実験において、計算された消滅係数(ε[イプシロン])を用いて280nmで消滅測定を行った。活性部位の力価は以下で説明するように、精製されたプロテアーゼ溶液におけるタンパク質濃度を決定するために用いた。
洗浄性能試験
この実施例において、スブチリシン変異体ER11及び商業的に利用されている食器用洗剤に用いられているGCI−P038対照スブチリシンの洗浄性能を評価するのに好適な方法を説明する。
この実施例において、スブチリシン変異体ER11及び商業的に利用されているGCI−P038対照シブチリシンの洗浄性能を測定するための方法を説明する。
1mm振幅(Thermo Electron Corporation)、及びシーリングテープ(Nunc No.236366)である。試薬は、5mMのHEPES、pH8.0又は5mMのMOPS、pH7緩衝液、中程度の硬度にするためにCa:Mgを3:1にした。(CaCl2:MgCl2・6H2O);1ガロン当たり15000グレイン(gpg)を6gpgに希釈したもの;1プレート当たり2つのBMI(血液/ミルク/インク)汚れ見本:CFTのEMPA−116BMコットン汚れ見本:予めすすいで穴をあけた見本を1ウェルあたり2つ:熱失活させたTIDE(商標)2X冷水用洗剤(プロテアーゼ活性がないことを確認したもの)。このアッセイにおいて、基質をプロテアーゼが加水分解して顔料及び不溶性の汚れを基質から剥がす。
得られた吸光度の値をブランク値(酵素なしの基質)で補正して、加水分解活性の測定値を提供する。各サンプル(変異体)のために、性能インデックス(PI)を計算した。性能インデックスは変異体の性能(実際の値)及び対照酵素の性能(理論値)を同じタンパク質濃度で比較する。更に、標準酵素のラングミュア方程式のパラメーターを用いて理論値を計算する。1よりおおきい性能インデックス(PI>1)は標準(例えば、野性型)と比較して、好ましい変異体であることを示し、1のPI(PI=1)は標準と同じ性能の変異体であることを示し、1より小さいPI(PI<1)は標準よりも悪い変異体を示す。したがって、PIで特定の環境に用いるのに、好ましいもの、好ましくないものを同定することができる。
液体洗濯洗剤組成物
この実施例において、液体洗濯洗剤組成物のための各種製剤を提供する。本発明の液体洗濯洗剤組成物を以下に示すように調製した。各製剤において、本発明のプロテアーゼ変異体が約0.0001乃至約10重量パーセントの濃度で含まれている。幾つかの代替的な態様において、必要に応じてこの製剤を基にして他の濃度を用いることができる。
手洗い用液体食器洗浄組成物

この実施例は各種タブレット洗剤製剤を提供する。本発明のタブレット洗剤組成物を標準的な12ヘッドロータリープレスを用いて13KN/cm2の圧力で顆粒食器洗浄組成物を圧縮して調製した。それぞれの製剤において、本発明のプロテアーゼ変異体が約0.0001乃至約10重量パーセントの濃度で含まれている。幾つかの代替的な態様において、必要に応じてこの製剤を基にして他の濃度を用いることができる。
液体硬表面洗浄洗剤
この実施例において、液体硬表面洗浄洗剤のための各種製剤を提供する。本発明の液体洗濯洗剤組成物を以下に示すように調製した。各製剤において、本発明のプロテアーゼ変異体が約0.0001乃至約10重量パーセントの濃度で含まれている。幾つかの代替的な態様において、必要に応じてこの製剤を基にして他の濃度を用いることができる。
Claims (11)
- 配列番号8で定義されるアミノ酸配列を含むスブチリシン変異体を含む洗浄組成物であって、前記洗浄組成物が洗濯洗剤及び手洗い用食器洗剤から選択される、洗浄組成物。
- 前記組成物が洗濯洗剤である、請求項1の洗浄組成物。
- 前記組成物が手洗い用食器洗剤である、請求項1の洗浄組成物。
- 前記組成物が液体洗剤である、請求項1の洗浄組成物。
- 前記組成物が、粉末、顆粒、タブレット、又はゲル洗剤である、請求項1の洗浄組成物。
- 前記組成物がリン酸塩を含まない、請求項1の洗浄組成物。
- 少なくとも1つの追加的な酵素を更に含む、請求項1の洗浄組成物。
- 前記少なくとも1つの追加的な酵素が、ヘミセルラーゼ、セルラーゼ、ペルオキシダーゼ、プロテアーゼ、メタロプロテアーゼ、キシラナーゼ、リパーゼ、フォスフォリパーゼ、エステラーゼ、ペルヒドロラーゼ、クチナーゼ、ペクチナーゼ、ペクチン酸リアーゼ、マンナナーゼ、ケラチナーゼ、リダクターゼ、オキシダーゼ、フェンロキシダーゼ、リポキシゲナーゼ、リグニナーゼ、プルラナーゼ、タンナーゼ、ペントサナーゼ、マラナーゼ、βグルカナーゼ、アラビノシダーゼ、ヒアルノニダーゼ、コンドロイチナーゼ、ラッカーゼ、及びアミラーゼ、並びにこれらの混合物から選択される、請求項7の洗浄組成物。
