Description
a)親GPCRと比較して特定の立体構造において増大した安定性を有する、該親GPCRのGPCR変異体を提供する工程、

c)前記1つ又は複数の試験化合物の各々が、特定の立体構造を備えた前記GPCR変異体に結合するか否かを測定する工程、及び
d)前記特定の立体構造を備えた前記GPCR変異体に結合する試験化合物を単離する工程
を含む方法を提供する。

親GPCRと比較して増大した安定性を有するGPCR変異体は、以下に記載する方法によって、並びにPCT出願WO 2008/114020号及びPCT/GB2008/004032号に開示されている方法の任意のものによって提供されてよい。
増大した安定性を有するGタンパク質共役型受容体(GPCR)変異体を選択するための方法は、
(a)親GPCRの1つ又は複数の変異体を提供する工程、
(b)親GPCRが特定の立体構造を備えている際に前記GPCRに結合するリガンドを選択する工程、
(c)前記選択したリガンドの結合に関する親GPCRの安定性と比較して、前記GPCR変異体又はその各々が前記リガンド結合に関して増大した安定性を有するか否かを測定する工程、並びに
(d)前記選択したリガンドの結合に関して、親GPCRと比較して増大した安定性を有する変異体を選択する工程
を含む。
(a)親GPCRの1つ又は複数の変異体を提供する工程、

上記のように選択したGPCR変異体を調製するための方法は、
(a)上述したGPCR変異体を選択する方法を実施する工程、
(b)増大した安定性について選択された1つ又は複数のGPCR変異体中の1つ又は複数の変異したアミノ酸残基の位置を同定する工程、及び
(c)同定した位置の1つ又は複数に変異を含むGPCR変異体を合成する工程
を含む。
主細胞においてポリヌクレオチドからポリペプチドを発現させるための分子生物学的方法が、参照によって本明細書に取り込む「Molecular cloning, a laboratory manual”, third edition, Sambrook, J. &Russell, D.W. (eds), Cold Spring Harbor Laboratory Press, Cold Spring Harbor, NY 」に例示されているように、当該技術分野においてよく知られている。
実施例1から3に示し、且つ上述したように、熱安定化変異は、シチメンチョウβ1アドレナリン受容体、ヒトアデノシン受容体、ラットニューロテンシン受容体、及びヒトムスカリン受容体の配列全体に広く分布する。図17は、ヒトβ−2ARの配列を有するこれらの配列のアラインメントを提供し、熱安定化変異が前記配列に位置する際に、全部で70のうち11例において、2つの配列が同じ位置に変異を含有している(図17において星印で示す)。かくして、1つ又は複数の安定化変異が1つのGPCRで同定されると、増大した安定性を有する更なるGPCRを、GPCRのアミノ酸配列アラインメント及び対応する1つ又は複数の位置で1つ又は複数の変異を作製することによって産生されてよい。この概念は、シチメンチョウβ1−m23の6つの熱安定化変異がヒトβ2受容体に直接転用した図26に明確に例示されている。結果として得られた変異体であるβ2−m23は、ヒトβ2受容体よりも12℃高いTmを有していた。

本発明者は、増大した安定性を有するGPCR変異体における1つ又は複数の変異が存在する構造モチーフの同定が、増大した安定性を有する更なるGPCR変異体を製造するのに有用であろうと考えた。
a.第一の親GPCRと比較して増大した安定性を有する第一の親GPCRの1つ又は複数の変異体を提供する工程、
b.1つ又は複数の変異体が第一の親GPCRと比較して少なくとも1つの異なるアミノ酸残基を有する、1つ又は複数の構造モチーフを構造膜タンパク質モデルにおいて同定する工程、及び
c.第二の親GPCRにおける1つ又は複数の対応する構造モチーフを規定するアミノ酸配列における1つ又は複数の変異を作製し、第二の親GPCRと比較して増大した安定性を有する第二の親GPCRの1つ又は複数の変異を提供する工程
を含む。
(I)特定の立体構造を備えた第二の親GPCRに結合するものであるリガンドを選択する工程、

ヒトゲノムによってコードされる500超の非嗅覚Gタンパク質共役型受容体(GPCR)が存在し、その多くが潜在的な治療標的であると予測されているが、ファミリーの全てを代表するものとしてウシロドプシンのみの構造が利用可能である。GPCR構造決定における進歩の欠落には多数の理由が存在するが、本発明者は、これらの受容体の界面活性剤安定性を改善すること、及び同時にそれらを1つの好ましい立体構造に固定化することが、結晶化の可能性を大幅に改善するであろうと仮説を立てた。GPCR、すなわちβ−アドレナリン受容体の界面活性剤可溶化熱安定化変異体の単離のための一般的な戦略は、アラニンスキャニング変異誘発、それに続く受容体安定性のアッセイに基づいて開発した。試験した318の変異体のうち、15が安定性における測定可能な増大を示した。最初の変異部位の各々におけるアミノ酸の最適化の後に、最適に安定化した受容体を特定の変異を組み合わせることによって構築した。最も安定な受容体変異体であるβAR−m23は、6つの点変異を有し、天然のタンパク質よりも結合したアンタゴニストの存在下で21℃高いTmを生じさせ、βARm23はウシロドプシンと同程度に安定であった。加えて、βAR−m23は広範な界面活性剤において顕著により安定で、結晶化に理想的であり、リガンドの不在下においてアンタゴニスト立体構造で好適に存在した。
β1アドレナリン受容体の熱安定性を増大する単一変異の選択
シチメンチョウ赤血球由来のβARはよく特性決定されており、バキュロウイルス発現系を使用して昆虫細胞において高レベルで発現されるため、構造研究に理想的な対象である(非特許文献10、11)。βARの最も良好な過剰発現は、残基34−424を含有する受容体の切断型を使用して得られ(βAR34−424)(非特許文献9)、これが本研究の出発点として使用された。アラニンスキャニング変異誘発を使用して、変異した際に、受容体の熱安定性を変化させるβAR34−424におけるアミノ酸を規定した;アラニンが配列中に存在する場合には、ロイシン残基に変異させた。全部で318の変異をアミノ酸残基37−369、7つの膜貫通ドメインを含む領域、及びC末端の23アミノ酸残基に作製した;15アミノ酸残基における変異は、DNA鋳型における強力な二次構造によって得られなかった。各変異体の配列決定をして所望の変異のみが存在することを確認した後に、受容体を大腸菌において機能的に発現させ、安定性についてアッセイした。
最初に、βARの一次アミノ酸配列において互いに隣接する、熱安定性を改善した変異を組み合わせた。G67A及びR68Sという変異又はヘリックス5の末端の変異の異なる組み合わせ(Y227A、R229Q、V230A、及びA234L)を含有する構築物を発現させて、アッセイした;Tm値(結果示さず)は、βAR34−424のTmよりも1から3℃のみ高く、1つの変異は実際には僅かにより安定でないものであり、一次アミノ酸配列において互いに隣接する変異の組み合わせは大きく熱安定性を改善するものではないことが示唆された。続いて、構造において互いに離れていると予測された変異を組み合わせた。各種のプライマー混合物を使用してPCR反応を実施し、無作為な様式で5つの異なる変異を組み合わせ、次いで、熱安定性について試験した(表1)。これらの組み合わせの最も良好なものは、βAR34−424のTmと比較して10℃より大きくTmを増大させた。幾つかの場合では、個々の変異の連続的な包含でTmに対しては相加的な効果が明らかに存在した。これは一連の3つの変異体、m4−1、m4−7、及びm4−2において認められ、m4−1に対するV230Aの付加はTmを2℃増大させ、m4−7におけるD332Aという付加的な変異はTmをさらに3℃増大させる。Y227A及びM90Aを含有する変異体の全ては、10℃以上のTmの増大を示した。これらの2つの変異が共に、βAR34−424のTmを13℃まで増大させたが(m7−5)、アンタゴニスト結合全体はβAR34−424の50%であり、これらの変異体の発現低下が示唆された。m7−5に対するF338Mの添加は熱安定性を増大しなかったが、大腸菌における機能的な発現のレベルを増大させた。
6種の変異の効果を同定するためにβAR−m23及びβAR34−424について測定した3種の特徴的な活性は、アンタゴニスト結合の親和性、アゴニスト結合の相対的な効果、βAR−m23のGタンパク質への結合能であった。アンタゴニストである[3H]−ジヒドロアルプレノロール(図8)を使用した膜への飽和結合実験は、βAR−m23に対する結合の親和性(KD 6.5±0.2nM,n=2)がβAR34−424(KD 2.8±0.1nM,n=2)よりも僅かに低いことを示し、アンタゴニスト結合立体構造ではβARm23の構造に大きなゆらぎが存在することを示唆している。このことは、βAR−m23における変異のいずれも、リガンド結合に関与すると解されているアミノ酸に相当するものではないことと一致する。アンタゴニスト結合と対照的に、βAR−m23によるアゴニスト結合の効果は、βAR34−424よりも3桁弱いものである(図5)。アゴニストであるイソプレナリンの有効性は、天然のアゴニストであるノルエピネフリンよりも、βAR−m23及びβAR34−424において一貫して低く、当該2つの受容体のアゴニスト結合立体構造は類似している可能性を示唆する。しかしながら、βAR34−424と比較した際のβAR−m23におけるアゴニストの効力における大きな低減は、βAR−m23における6種の変異が、アンタゴニストが結合した立体構造に選択的に固定化されていることを示唆する。立体構造の予測からすると、このことは、回折に使用し得る品質の結晶の生産のために立体構造が均一なタンパク質集団を有することが重要であるため、熱安定性に予期せぬ利点を加える。
シチメンチョウβ−アドレナリン受容体の幾つかの異なる構築物を結晶化する以前の試みは失敗した。各種の条件で実験したにもかかわらず、天然の配列並びに幾つかの切断型及びループ欠失構築物の双方の使用によっては、長年に亘って、結晶が得られなかった。
物を使用して結晶が得ることが可能であったか否かを確認した。同様に又はおそらく非常に小さな結晶を得ることが可能であるかもしれないが、実際には、「野生型」(すなわち、変異誘発させる出発構造)は結晶を生じなかった。
材料
シチメンチョウ由来の切断型のβ1アドレナリン受容体(βAR34−424)(非特許文献9)は、DrTony Warne(MRC Laboratory of Molecular Biology,Cambridge,UK)によって快く提供された。34−424残基をコードするβAR構築物は、C116L変異を含有して発現を改善し(非特許文献11)、精製のために10ヒスチジンのC末端タグを含有している。1−[4,6−プロピル−3H]−ジヒドロアルプレノロール([3H]−DHA)はAmersham Bioscienceによって提供され、(+)L−ノルエピネフリン酒石酸水素塩、(−)イソプレナリン塩酸、(−)アルプレノロール酒石酸塩、及びs−プロプラノロール塩酸はSigmaからのものである。
βAR cDNAは、pRGIIIにライゲートし、MalE融合タンパク質としての大腸菌におけるβARの機能的な発現を可能にした(非特許文献16)。変異体は、QuikChange II方法(Stratagene)を用いて、発現プラスミドを鋳型として使用してPCRによって産生した。PCR反応物は、XL10−Gold ultracompetent cell(Stratagene)に形質転換し、個々のクローンを完全に配列決定して、所望の変異のみが存在していることを調べた。異なる変異は、以下の変異:Mut4、G67A、G068A、V230A、D322A、及びF327A;Mut6、R068S、Y227A、A234L、A282L、及びA334L;Mut7、M90V、I129V、Y227A、A282L、及びF338M;Mut10、R68S、M90V、V230A、F327A、及びA334Lを導入する全てのペアのプライマーを含めることによって、PCRによって無作為に組み合わせた。PCR混合物を形質転換して、クローンは配列決定して変異が確かに導入されていることを測定した。
βAR及び変異体の発現は、XL10細胞(Stratagene)において実施した。50mlのアンピシリン含有(100μg/ml)2×TY培地の培養物を、OD600=3まで浸透しながら37℃で増殖させ、次いで、0.4mM IPTGを用いて誘導した。誘導した培養物は、4時間に亘って25℃でインキュベートし、次いで、細胞を13,000×gで1分に亘って遠心分離することによって回収し(2mlの一定分量)、−20℃で保存した。アッセイに関しては、細胞を凍結融解(5サイクル)によって破砕し、500μlの緩衝液[20mM Tris pH8、0.4M NaCl、1mM EDTA、及びプロテアーゼインヒビター(Complete(商標)、Roche)]に再懸濁した。100μg/mlリソザイム及びDNアーゼ I(Sigma)を用いて4℃で1時間に亘ってインキュベートした後に、氷上で30分に亘ってサンプルを2%DDMに可溶化した。不溶性物質を遠心分離(15,000×g、2分、4℃)によって除去し、上清を放射リガンド結合アッセイに直接使用した。
登録コードが1GZM[14]であるロドプシン構造のpdbファイルは、Protein DateBankウェブサイト(www.pdb.org)からダウンロードし、PyMOLX11Hybrid(DeLano Scientific)プログラムに表示した。βARにおける熱安定変異についての、ロドプシンの対応するアミノ酸残基は、本発明者のよく知っている4つのGPCR、すなわち、ロドプシン、β1アドレナリン受容体、ニューロテンシン受容体、及びアデノシンA2a受容体の間のアラインメントに基づいてロドプシン構造に配置した(非特許文献19)。
増大した熱安定性を有するアデノシンA2a受容体(A2aR)の変異体
1.315の部位特異的変異体を、A2aRの2から316残基の間に作製した。
2.これらの変異体の全てを、加熱工程後のアゴニスト及びアンタゴニスト結合測定アッセイを使用して熱安定性についてアッセイした(図12に記載のリガンド(−)フォーマット)。

野生型のNTRは、ニューロテンシンが結合した状態で35℃のTmを有する。
安定化GPCR変異が存在する構造モチーフの同定
β2アドレナリン受容体の構造を決定した(非特許文献20及び21)。それはシチメンチョウβ1受容体と59%同一であったが、はっきりと異なる薬理学的プロフィールを有していた(非特許文献22及び23)。シチメンチョウβ1受容体の安定化変異が存在する構造モチーフを決定するために、本発明者は、ヒトβ2構造に変異をマッピングした(非特許文献21)。
GPCRの立体構造特異的結合パートナーの作製
親GPCRと比較して特定の立体構造で増大した安定性を有するGPCR変異体の作製は、結合パートナーのスクリーニングのために多くの利点を提供する。例えば、本発明の方法はスクリーニングに必要とされる物質の量を低減する。標準的なスクリーニングでは、GPCRは全細胞中又は全細胞からの膜中に存在し、これらは通常、化合物のライブラリーではなく隔離室における個々の化合物とのインキュベーションによってスクリーニングされる。かくして本発明は、化合物のスクリーニングを実施するために必要とされる時間に関する利点を提供する。GPCRを特定の立体構造に固定できることは、必要な薬理学的特性を有するリガンドを同定する可能性を高めるという利点を提供する。標準的な結合スクリーニングでは、GPCRは多くの異なる立体構造をとることが可能であり、結合化合物は各種の薬理学的タイプに亘って同定されるであろう。アッセイ形式を小型化できるため試薬の費用を低減でき、これは本発明の方法によって促進される。
化合物のスクリーニング
特定の立体構造で増大した安定性を有するGPCR変異体を固体表面に固定化し、コードされる化合物のライブラリーを含む緩衝液と共にインキュベートする。GPCR変異体とGPCR変異体に選択的に結合するライブラリーからの化合物との間の結合を可能にする適切な時間後、緩衝液を除去する。次に、GPCR変異体に特異的に結合しなかった化合物を除去するために数回の洗浄工程を実施する。読取りコード、タグ又は空間的位置(DNA又はRNAなど)を使用して、依然としてGPCR変異体に結合している又はGPCRから溶出した後の、結合低分子を同定する。その後立体構造特異的結合パートナーを単離する。
固定化した安定化GPCRを使用して、安定化GPCR、天然受容体又は受容体からのペプチドで免疫した動物からの血漿中に存在するような抗体の混合物から受容体に対する抗体を選択することが可能である。抗体は、免疫動物から採取したB細胞から得られる上清中から、又は免疫動物からのB細胞の不死化後に得られるハイブリドーマから、又はファージ粒子上で若しくはリボソームディスプレイなどのインビトロ発現系を介して発現されてよい組換え抗体ライブラリーから同定できるであろう。前記方法は、受容体の特定立体構造に対する抗体を選択するという利点を有する。受容体のアンタゴニスト又は基底状態に固定された安定化GPCRは、アンタゴニスト抗体を選択する確率を高め、一方、活性又はR*状態に固定された安定化GPCRは、活性化抗体を選択する確率を高めるであろう。標準的なスクリーニングでは、抗体は、多くの場合、受容体のペプチドエピトープに結合するが、アンタゴニスト又はアゴニスト特性を有さず、したがって治療薬としては有用でないGPCRに対するものが選択される。
β−アドレナリン受容体に対する抗体ファージ選択
要約
本発明者らは、ファージディスプレイを使用して抗体を作製するために安定化β−アドレナリン受容体(β−AR)を使用した。陽性ファージクローンはβ−ARに対する特異性を示し、選択した抗体遺伝子のサブクローニングは、抗−β−AR特異的scFv抗体の生産の成功をもたらした。
受容体−リガンド系の領域における抗体の治療適用は大きな潜在的可能性を有する。しかしながら、Gタンパク質共役受容体(GPCR)に対する抗体を作製する(インビトロ又はインビボ法のいずれかによって)際の主要な問題は、不均一な製剤における多数の立体構造の精製抗原を認識する抗体結合物質のプールを作製することよりもむしろ、均一な抗原製剤における特定の立体構造、すなわちアゴニスト立体構造又はアンタゴニスト立体構造の免疫原性認識に関係する。加えて、組換え受容体抗原は通常、細胞外ドメイン形態でのみ入手可能であり、このことは、受容体の他の部分に関与する任意の三次構造を排除する。この問題に対して提案される解決法は、標的抗原としての安定化GPCRの適用である。
典型的なファージディスプレイ戦略は、阻害性scFvクローンを同定するための工程においていくつかの段階を含む(図29)。最初の部分は、各種の方法(例えば、パニング選択、可溶性選択等)を使用してファージ抗体結合物質の集団を単離するための抗原に対するファージライブラリーの選択である。生じるこのファージ抗体結合物質の集団は、選択アウトプットと称される。特異的抗原結合物質を富化するためにこの工程を2から4回反復する。
安定化β−ARを抗原として使用し、且つSchofield et al.,2007(24)に記載されている「McCafferty」抗体ファージディスプレイライブラリーを使用して、2から3回の抗体選択を実施した。選択は、結合及び洗浄工程において20nMのリガンド(−)−アルプレノロールの存在下(A)又は不在下(B)にPBS中で実施した。さらに、β−ARタンパク質の取り扱いに関しては、全ての被覆、洗浄及びブロッキング緩衝液に0.1%の界面活性剤デシルマルトシド(Anatrace,Anagrade)を添加した。選択抗体の相対的な成功を、ポリクローナルファージELISA及びモノクローナルファージELISAを使用して測定した。
固定化戦略はHisタグ抗原を利用することに基づいた。第一の場合には、βAR−m23ではなく対照タンパク質を使用して固定化を行った。Ni−NTAプレートを含む各種の表面を最初の評価において使用した。すべてをNuncプレートへの標準的な受動吸収に対して比較した。すべての場合に、2回の選択後にポリクローナルファージELISAを使用して結果を評価した。
ライブラリーの選択、溶出及び回収は、Schofield et al(2007)に記載されているとおりとした。150μlの20μg/ml濃度のβ−ARをニッケルキレートプレート(Nunc)の2つの(24)ウエルにおいて+4℃で一晩被覆した。受容体を被覆緩衝液に希釈し、被覆緩衝液は、20mM Tris pH8、100mM NaCl、0.1%デシルマルトシド(dec−M)であり、選択Aについては20nMリガンドも含むものとした。翌日、ウエルをPBSで洗浄し、0.1%dec−Mを添加したPBS(PBS−M)中の3%Marvel乳タンパク質により室温で1時間ブロックした。被覆後、ウエルをPBSで洗浄し、2%Marvel/PBS中であらかじめブロックした100μlのファージライブラリーを添加して、室温で1時間インキュベートした。結合後、試料を、0.1%dec−Mを添加したPBS/0.1%Tween中で6回及び0.1%dec−Mを添加したPBS中で6回洗浄した。結合ファージをトリプシンで溶出した(24)。
ELISAのために、βARをアミノプレート(Nuncカタログ番号:436008)に共有結合的に固定化した。被覆及び洗浄緩衝液には20nMリガンド及び0.1%dec−Mを添加した。ELISAプレートを24μg/mlのβ−AR及び5μg/mlの2つの対照タンパク質(CD86及びNotch1)で一晩被覆した。翌日、ウエルをPBS−Mで洗浄し、ブロックした。2回の選択からのポリクローナルファージ50μl/ウエルを添加し(PBS−M中に最初の培養容積に対して0.1×の濃度で)、α−M13抗体(GE Healthcare、製品番号:27−9421−01)と共に室温で1時間インキュベートした。プレートを洗浄し、ユウロピウム標識抗マウス抗体(Perkin Elmer、製品番号:AD 0207)と共に室温で1時間インキュベートした。ウエルを洗浄し、50μl/ウエルの増強溶液(Perkin Elmer、製品番号:4001−0010)を添加して、10分間インキュベートした。結果を図30Aに示しており、2回の選択後の実験A及びBにおけるβ−AR結合ファージの特異的富化を示唆する。シグナルはファージ選択A(緩衝液中にリガンドが存在する)からの方が高かった。
このアッセイのために、選択Aからの2回目のファージの個々のクローンを採取し、回収して、PEGで沈殿させた。やはり、被覆、洗浄、ブロッキング及び抗体検出緩衝液に20nMリガンドと0.1%界面活性剤を添加した。Nuncアミノプレートのウエルは、β−ARにより室温で1時間半に亘って被覆した。被覆したウエルをPBSTで3回、PBSで3回洗浄し、PBS−Mでブロックして、50μl/ウエルの0.1×ファージを添加し、室温で1時間インキュベートした。ウエルをPBSで6回洗浄し、α−M13抗体と共に室温で1時間インキュベートした。PBSで6回洗浄した後、ウエルを抗マウスEu抗体と共に室温で1時間インキュベートした。次に、ウエルを洗浄し、増強溶液と共に10分間インキュベートした。69のスクリーニングしたクローンのうちで、20のクローンが陽性と検出された(図30B)。
モノクローナルELISAからの陽性ファージクローンのうちの17をβ−AR及び3つの無関係なタンパク質に対する結合特異性に関して試験した。これは、N1−EGF(R&D Systemsからの、ヒトFcドメインに融合したマウスNotch 1のEGFドメイン1〜12、カタログ番号1057−TK)を含む。その他の対照タンパク質は、Chapple et al 2006(25)に記載されている一過性発現系においてCD4融合体として発現される、マウスCD86の細胞外ドメイン及びマウスNotch 1の負の調節領域(それぞれCD86及びN1(NRR))を含む。このELISAでは、ニッケルキレートとアミノプレートの両方を比較のために使用した。やはり、β−ARタンパク質を含むウエルに関しては、被覆、洗浄、ブロッキング及び抗体検出緩衝液に20nMリガンドと0.1%界面活性剤を添加した。前の章でモノクローナルELISAに関して述べたようにウエルを洗浄し、抗体と共にインキュベートした。アッセイは、His及びアミノプレートの両方が使用可能であること、及び抗β−ARクローンの大部分が無関係なタンパク質とは交差反応しないことを示した(図31)。
サブクローニング、抗体一本鎖Fv(scFv)の発現及び精製は、Schofield et al(2007)に記載されているとおりとした。2回目のファージ集団の中の選択した抗体遺伝子(選択A)をpSANG10−3Fベクター(26)にサブクローニングし、BL21(DE3)細胞に形質転換した。96のコロニーを採取し、当該技術分野における標準的な方法を使用して96穴フォーマットでscFv抗体のペリプラズム発現を誘導した。scFvをペリプラズムから回収し、アミノプレート(Nunc)でのELISAのために使用した。洗浄し、ブロックしたウエルを50μl/ウエルのscFvと共に1時間インキュベートした。プレートを洗浄し、ユウロピウム標識抗FLAG抗体と共に室温で1時間インキュベートした。12のクローンが1000単位を超えるシグナルを生じ、陰性クローンについては50未満のバックグラウンドレベルであった(図32)。
β1−ARと化合物の相互作用の評価
方法及び結果
ストレプトアビジン被覆CM5チップを備え、作業緩衝液(20mM trisHCl、 150mM NaCl、1mM EDTA、1%DMSO、0.1%デシルマルトシド、pH7.8)で平衡化させたBiacore S51光学バイオセンサーを使用して10℃で結合試験を実施した。
β1AR36−M23を、EZ結合スルホ−NHS−LC−LCビオチン(Pierce、No.21338)を使用して最小限にビオチニル化した:前記ビオチンを受容体製剤に添加し、(100μMアルプレノロールでスパイクして)、4℃で3時間反応させた後、遊離ビオチンをカラムクロマトグラフィーによって除去した。
S51バイオセンサーは2つの反応スポットを同時に観測する能力を有するので、本発明者らは、他のストレプトアビジン被覆スポット上で8000RUの密度に受容体を捕捉した(図39)。
1.最小限のビオチニル化及びストレプトアビジンによる捕捉は、化合物の結合を測定するために使用可能である活性なβ1AR表面を生じた。
2.10℃で、β1AR表面は数日間に亘って活性なままであった。

薬剤探索における化合物のスクリーニングのためのアデノシンA2a StaRの使用
方法
A2a受容体において活性を有する化合物を同定するため、アンタゴニスト形態(Rant22と呼ぶ)で立体構造的に選択した熱安定化アデノシンA2a受容体(A2a StaR)を使用して、ライブラリーからの化合物をスクリーニングした。StaRは以前に開示されているように作製した(Magnani et al,Co−evolving stability and conformational homogeneity of the human adenosine A2a receptor.Proc Natl Acad Sci U S A.2008 Aug 5;105(31):10744−9)。Rant22 A2a受容体でトランスフェクトしたHEK293T細胞を、10%ウシ胎仔血清を添加したダルベッコ改変イーグル培地中、37℃、5%CO2にてT−175フラスコの単層で増殖させた。T−175表面から細胞を削り取って採取し、遠心分離によって収集した。
細胞ペレットを10mlの20mM HEPES、pH7.4プラスプロテアーゼインヒビターカクテル錠(Roche)に再懸濁し、Tissuemizerを使用して20,500rpmで30秒間に亘ってホモジナイズした。ホモジネートを200×gで15分間に亘って4℃で遠心分離した。
調製した膜の10μg分量を、[3H]ZM241385 3.7nM及び適切な量の非標識リガンドと共に室温で60分間インキュベートした。10mMから1μMの範囲の連続希釈を96穴フォーマットでスクリーニングした。フィルターGFCプレートを0.1%PEI中で60分間予浸した。Microbeta counterを3分/ウエルで使用した液体シンチレーション計数によって放射能を測定した。
結果
濃度応答曲線にフィットさせるためにGraphPadプリズムを使用してデータを解析した。化合物のIC50を、[3H]ZM241385の特異的結合の50%阻害を生じさせる濃度として計算した。図45及び以下の表Bに提示するデータは、試験した化合物がA2a受容体StaRへの結合を阻害することができ、このアッセイにおいて一連の活性を有していたことを明らかにする。このデータは、化合物スクリーニングのためのStaRの有用性を明らかにする。

- 前記ムスカリン受容体変異体が、図17に記載のラットニューロテンシン受容体の配列と少なくとも20%同一であるアミノ酸配列を有する、請求項94に記載の方法。
- 工程(a)で提供される前記GPCR変異体がムスカリン受容体変異体であって、前記受容体変異体が、その親受容体と比較して、図17に記載のヒトムスカリン受容体の番号付けによる以下の位置:Leu 65,Met 145,Leu 399,Ile 383及びMet 384のいずれかに対応する位置に異なるアミノ酸を有する構造モチーフ内に少なくとも1つの異なるアミノ酸残基を有する、請求項1から82のいずれか一項に記載の方法。
- GPCRの結合パートナーを生産する方法であって、請求項1から96のいずれか一項に記載の方法を実施することによって同定可能な結合パートナーを合成する工程を含む方法。

- 前記結合パートナーが抗体である、請求項100に記載の結合パートナー。
- 前記結合パートナーに対する前記GPCR変異体の結合親和性が、前記結合パートナーに対する前記親GPCRの結合親和性と実質的に同一であるか又はそれよりも大きい、請求項98から101のいずれか一項に記載の結合パートナー。
- 親GPCRのGPCR変異体を含むバイオセンサーであって、前記GPCR変異体が親GPCRと比較して特定の立体構造で増大した安定性を有し、且つ、標的物質が前記GPCR変異体に結合した際に検出可能なシグナルが生成される、バイオセンサー。
- 前記GPCR変異体が、請求項45から82のいずれか一項で規定されるように提供される、請求項103に記載のバイオセンサー。
- 前記GPCR変異体が、請求項83から96のいずれか一項で規定されるものである、請求項103又は104に記載のバイオセンサー。
- 前記検出可能なシグナルが、色の変化;蛍光;エバネッセンス;表面プラズモン共鳴;電気伝導度若しくは電荷分離;紫外、可視若しくは赤外吸収;発光;化学発光;電気化学発光;蛍光異方性;蛍光強度;蛍光寿命;蛍光偏光;蛍光エネルギー移動;分子量;電子スピン共鳴;核磁気共鳴;流体力学的容積若しくは半径;比重;シンチレーション;電界効果抵抗;電気インピーダンス;音響インピーダンス;量子消失;共鳴散乱;蛍光クエンチング;蛍光相関分光法;音響負荷;音響せん断波速度;結合力;又は界面応力のいずれかである、請求項103から105のいずれか一項に記載のバイオセンサー。
- 前記バイオセンサーが、水晶膜厚計バイオセンサー、エバネッセント波バイオセンサー、プレーナー導波路バイオセンサー、表面ラマンセンサー、又は表面プラズモン共鳴バイオセンサーなどのフローベースのバイオセンサーである、請求項103から106のいずれか一項に記載のバイオセンサー。
- 前記標的物質が、分子、生体分子、ペプチド、タンパク質、炭水化物、脂質、GPCRリガンド、合成分子、薬剤、薬剤代謝産物又は疾患バイオマーカーのいずれかである、請求項103から107のいずれか一項に記載のバイオセンサー。
