Abstract
【解決手段】多重プラスミドインフルエンザウイルス発現系において使用するための2方向性発現ベクター。さらに、発育鶏卵および/または細胞(例えばVero細胞および/もしくはMDCK)において増強された複製能力を有するインフルエンザウイルスを作製するための方法、ならびにさらに増強された複製特性を有するインフルエンザウイルス。さらに、改善された回収方法を含み、この方法において動物細胞(SF Vero細胞など)に上記プラスミドおよびベクターをエレクトロポレーションする。

81G)、PB1661(A661T)、PB2265(N265S)およびNP34(D34G)の中から選択される突然変異を含む)新規組換えウイルスの核酸ならびに、このようなアミノ酸置換を有するポリペプチドは本発明の特徴である。
多くの病原性インフルエンザウイルス株は組織培養において、不十分に増殖するだけで、弱毒生ウイルスワクチンの製造に好適な株(温度感受性、好冷および/または弱毒インフルエンザウイルスなど)は、商業的生産にかなうほど培養細胞中にて首尾よく増殖しなかった。本発明は、標準的な細胞培養条件下での増殖に適合していないインフルエンザウイルス株の増殖および回収を可能とする多重プラスミドトランスフェクション系を提供する。インフルエンザワクチンの開発および製造におけるさらなる課題は1種以上の循環インフルエンザ株が発育鶏卵では十分に複製することができないことである。本発明はHAおよびNAタンパク質の活性に影響を及ぼす複数のアミノ酸残基を発見し、そのような活性を調節できる特定のアミノ酸置換をつきとめた。本発明はHA受容体結合活性および/またはNA ノイラミニダーゼ活性の調節によって、卵および/または宿主細胞(Vero細胞もしくはMDCK細胞など)におけるインフルエンザの複製を高めることができることを開示する。特に本発明は卵および/または細胞におけるウイルスの複製を高めることができるHAおよび/またはNAにおけるアミノ酸置換の組み合わせを開示し、これらのアミノ酸置換が組換えインフルエンザウイルスの抗原性に重大な影響を与えないことを示す。従って、本発明はインフルエンザウイルスワクチンの製造を改善するための逆遺伝子技術の使用を提供する。
別途定義されない限り、全ての科学的および技術的用語は、当該分野において一般に用いられているのと同一の意味を有するものとして理解される。本発明の目的に関して、以下の用語を以下に定義する。
インフルエンザウイルスのゲノムは8つのセグメントからなる線状(−)鎖リボ核酸(RNA)からなり、これらは免疫原性赤血球凝集素(HA)およびノイラミニダーゼ(NA)タンパク質および6つの内部核ポリペプチド:ヌクレオカプシド核タンパク質(NP);マトリックスタンパク質(M);非構造タンパク質(NS);3つの RNAポリメラーゼ(PA、PB1、PB2) タンパク質をコードする。複製の間にゲノムウイルスRNAは宿主細胞の核内において(+)鎖メッセンジャーRNAおよび(−)鎖ゲノムcRNAに転写される。8つのゲノムセグメントの各々はリボ核タンパク質複合体にパッケージされる。このリボ核タンパク質複合体はRNAに加えて、NPおよびポリメラーゼ複合体(PB1、PB2、およびPA)を含む。
発現されるべきインフルエンザウイルスのゲノムセグメントを、mRNA合成を導くために適切な転写制御配列(プロモータ)に機能的に結合する。種々のプロモータは、インフルエンザウイルスのゲノムセグメントの転写を調節するために発現ベクターに用いることに適している。特定の実施形態において、例えばベクターがプラスミドpAD3000である場合、サイトメガロウイルス(CMV)DNA依存性RNAポリメラーゼII(Pol II)プロモータを利用する。必要な場合には、例えば条件発現を調節するために、特定の条件下、または特定の組織もしくは細胞においてRNA転写を引き起こす他のプロモータを代わりに用いることができる。多数のウイルスおよび哺乳動物(ヒトなど)のプロモータが利用可能であるか、または意図される特定の用途により単離することができる。例えば、動物およびヒトのウイルスのゲノムから得た代替的なプロモータとしては、アデノウイルス(アデノウイルス2型など)、パピローマウイルス、B型肝炎ウイルス、ポリオーマウイルスおよびサルウイルス40(SV40)などのプロモータ、ならびに種々のレトロウイルスのプロモータが挙げられる。哺乳動物のプロモータとしては、アクチンプロモータ、免疫グロブリンプロモータ、熱ショックプロモータなどが挙げられる。さらに、バクテリオファージプロモータを同種のRNAポリメラーゼと共に用いることができる(例えばT7プロモータ)。
最も一般的には、インフルエンザウイルスタンパク質をコードするゲノムセグメントは、機能的ウイルスタンパク質への翻訳を含む、その発現に必要ななんらかのさらなる配列を含む。別の状況において、ミニ遺伝子またはウイルスタンパク質(HAもしくはNAタンパク質など)をコードする他の人工構築物を用いることができる。この場合においては、異種性コード配列の効率的な翻訳を補助する特定の開始シグナルを含むことがしばしば望ましい。これらのシグナルは、例えばATG開始コドンおよび隣接配列を含むことができる。全挿入物の翻訳を確実にするために、開始コドンをウイルスタンパク質に関して正しいリーディング・フレーム内に挿入する。外来性転写エレメントおよび開始コドンは、天然と合成の両方のさまざまな起源のものでありうる。発現の効率は用いる細胞系に適切なエンハンサーを含めることによって増強することができる。
歴史的に、インフルエンザウイルスワクチンは発育鶏卵において、関連株についての経験的予測に基づいて選択したウイルス株を用いて作製していた。ごく近年、認可された弱毒温度感受性主株との関連において、選択された赤血球凝集素およびノイラミニダーゼ抗原を含むリアソータントウイルスが作製された。鶏卵において継代を重ねたウイルス培養の後、インフルエンザウイルスを回収し、そして必要に応じて、ホルムアルデヒドおよび/またはβ−プロピオラクトンなどを用いて不活性化する。しかし、この方法によるインフルエンザワクチンの製造は多くの重大な欠点を有する。鶏卵から残存する汚染物質は高い抗原性、発熱性のものであり、しばしば投与の際に重大な副作用を生じる。さらに重大なことには、製造用に用意される株は、インフルエンザワクチンの製造および不活性化のための時間を得るために、典型的に次のインフルエンザ時期の数ヶ月前には選択および供給しなければならない。組換えおよびリアソータントワクチンを細胞培養にて製造する試みは、ワクチン製造用に認可された何れの株も標準的な細胞培養条件下にて効率的に増殖することができないことによって妨げられていた。
1つの態様において、本発明はウイルスの好ましい主要ドナー株におけるts表現型の基礎をなす突然変異の決定に基づく。MDV株ゲノムにおける単一のヌクレオチド変化の機能的重要性を決定するために、A/AA/6/60系統内の非常に関連のある株に由来するリアソータントウイルスを温度感受性について調べた。2つの親株の同質遺伝子的性質によって、ts表現型における単一のヌクレオチド変化を評価することができる。従って、PB1、PB2およびNP内の特定のアミノ酸残基にヌクレオチドレベルでMDV-Aのts表現型に関する遺伝子基盤が与えられる。
本発明はまた、発育鶏卵および/または宿主細胞におけるインフルエンザウイルスの複製能力を増加するために、HAおよび/またはNAに少なくとも1つのアミノ酸残基置換を導入する方法を提供する。本発明はさらに、HAおよび/またはNAが置換されていないインフルエンザウイルスと比べた場合、発育鶏卵および/または宿主細胞において増加された複製能力を有するインフルエンザウイルス変異体(本明細書中では「複製増強された変異体」という)を提供する。本発明方法を宿主細胞におけるインフルエンザウイルスの複製を増強するために利用することができ、また複製増強された変異体が鶏卵および/または宿主細胞において増強された複製を有し得るということが特に意図される。インフルエンザウイルスの複製に好適な宿主細胞としては、例えばVero細胞、Per.C6細胞、BHK細胞、MDCK細胞、293細胞およびCOS細胞(293T細胞、COS7細胞を含む)が挙げられる。
典型的に、ウイルス増殖は、一般に宿主細胞が培養される培地組成物中で成し遂げられる。インフルエンザウイルスの複製に好適な宿主細胞としては、Vero細胞、Per.C6細胞、BHK細胞、MDCK細胞、293細胞およびCOS細胞(293T細胞COS7細胞を含む)が挙げられる。一般に上記細胞系統の2つ(例えばMDCK細胞と293T細胞またはCOS細胞のいずれか)を例えば1:1の割合で含む共培養物を用いて、複製効率を改良する。典型的に、細胞を制御された湿度および中性に緩衝されたpH(例えばpH 7.0〜7.2)を維持するのに好適なCO2濃度の下、血清(例えば10%胎児ウシ血清)を添加した標準的な市販の培養培地(ダルベッコ改変イーグル培地など)または、無血清培地中で培養する。必要に応じて、培地は細菌の増殖を防ぐための抗生物質(ペニシリン、ストレプトマイシンなど)および/またはさらなる栄養分(L-グルタミン、ピルビン酸ナトリウム、非必須アミノ酸など)、好ましい増殖特性を促進するためのさらなる栄養補給物(例えばトリプシン、β-メルカプトエタノール)などを含む。
当該分野において周知である真核細胞に異種性核酸を導入するための方法(例えばリン酸カルシウム共沈殿、エレクトロポレーション、マイクロインジェクション、リポフェクションおよびポリアミントランスフェクション試薬を用いたトランスフェクションなど)により、宿主細胞にインフルエンザゲノムセグメントを含むベクターを導入(例えばトランスフェクト)する。例えば、ベクター(プラスミドなど)を製造者の指示に従ってポリアミントランスフェクション試薬 TransIT-LT1(Mirus)を用いて宿主細胞(COS細胞、293T細胞またはCOS細胞もしくは293T細胞とMDCK細胞との組合せ)にトランスフェクトすることができる。総量200 μl中において、160 μlの培地(好ましくは無血清培地)に希釈したおよそ2 μlのTransIT-LT1を用いて、宿主細胞の集団におよそ1 μg の各ベクターを導入する。DNA:トランスフェクション試薬混合物を室温にて45分間インキュベートし、その後、800 μlの培地を加える。トランスフェクション混合物を宿主細胞に加え、そしてこの細胞を上記のように培養する。このように、細胞培養における組換えまたはリアソータントウイルスの作製のために、8つのゲノムセグメント(PB2、PB1、PA、NP、M、NS、HAおよびNA)をそれぞれ含むベクターをおよそ20 μlのTransIT-LT1と混合し、そして宿主細胞にトランスフェクトする。必要に応じて、血清含有培地をトランスフェクションの前に無血清培地(例えばOpti-MEM I)に代え、4〜6時間インキュベートする。
典型的にウイルスを、感染(トランスフェクト)細胞が増殖していた培養培地から回収する。典型的に、粗製培地をインフルエンザウイルスを濃縮する前に清澄させる。一般的方法としては、濾過、限外濾過、硫酸バリウムへの吸着および溶出、ならびに遠心分離が挙げられる。例えば、感染させた培養物の粗製培地を初めに、例えば1000〜2000×gにて、細胞の残骸および他の大きな粒子物質を除去するのに十分な時間(例えば10〜30分間)遠心分離することによって清澄させることができる。あるいは、培地を0.8 μm酢酸セルロースフィルターを通して濾過し、インタクトな細胞および他の大きな粒子物質を除去する。必要に応じて、次に清澄した培地上清を例えば15,000×gにておよそ3〜5時間遠心分離し、インフルエンザウイルスをペレットにする。適切な緩衝液(STE(0.01 M Tris-HCl;0.15 M NaCl;0.0001 M EDTA)またはリン酸緩衝生理食塩水(PBS)(pH 7.4)など)にウイルスペレットを再懸濁した後、ウイルスをスクロース(60%〜12%)または酒石酸カリウム(50%〜10%)の密度勾配遠心分離によって濃縮する。連続的または段階的勾配(例えば12%ずつ4段階からなる12%〜60%のスクロース勾配)のいずれかが好適である。回収のためにウイルスが目に見えるバンドに濃縮するのに十分なスピード、および時間で勾配を遠心分離する。あるいは、最大規模の商業的用途のために、ウイルスを連続的方法で操作するゾーン遠心分離ローターを用いて、密度勾配により溶出する。組織培養からインフルエンザウイルスを抽出することを当業者にすべて示すのに十分なさらなる詳細は、例えばNicholsonら(eds) Textbook of Influenza pp. 324-332におけるFurminger. Vaccine Production;Cohen & Shafferman(eds) Novel Strategies in Design and Production of Vaccine s pp. 141-151におけるMertenら(1996) Production of influenza virus in cell cultures for vaccine preparationおよび米国特許第5,690,937号に提供される。必要な場合には、回収したウイルスを安定剤であるスクロース-リン酸-グルタミン酸(SPG)の存在下において−80℃で保存できる。
本発明の組み換えおよびリアソータントウイルスを、1つ以上のインフルエンザウイルス株に特異的な免疫応答を刺激するために適切なキャリアまたは賦形剤に入れて予防的に投与することができる。典型的に、キャリアまたは賦形剤は製薬上許容できるキャリアまたは賦形剤(滅菌水,水溶性生理食塩水、水溶性緩衝生理食塩水、水溶性ブドウ糖溶液、水溶性グリセロール溶液、エタノール、未感染の鶏卵に由来する尿膜液(すなわち、正常尿膜液「NAF」)またはそれらの組み合わせなど)である。無菌性、pH、等張性、および安定性を確実にする上記溶液の調製を当該分野において確立されたプロトコルにより行う。一般に、キャリアまたは賦形剤をアレルギーおよび他の望ましくない効果を最小限にするように、および特定の投与経路(皮下、筋肉内、鼻腔内など)に適するように選択する。
本発明のベクターおよびベクター系の使用を容易にするために、インフルエンザウイルスのパッケージングおよび感染に有用な、ベクターのいずれか(共通インフルエンザウイルスプラスミド、変異体インフルエンザポリペプチドプラスミド、インフルエンザポリペプチドライブラリープラスミドなど)およびさらなる構成要素(緩衝液、細胞、培養培地など)がキットの形態でパッケージされうる。典型的に、キットは上記構成要素に加えて本発明方法をおこなうための解説書、パッケージング物質、および容器などを含みうるさらなる物質を含む。

プラスミドpHW2000(Hoffmannら(2000) A DNA transfection system for generation of influenza A virus from eight plasmids Proc Natl Acad Sci USA 97:6108-6113)を改変して、ウシ成長ホルモン(BGH)ポリアデニル化シグナルとシミアンウイルス40(SV40)に由来するポリアデニル化シグナル配列を置換した。

好冷A型インフルエンザウイルス株A/AA/6/60変異体は一般に、経鼻投与されるA型インフルエンザワクチン製造のための主要ドナーウイルスとして用いられていた。この株は本発明との関連において、典型的な主要ドナーウイルス(MDV)である。便宜上、この株A/AA/6/60の変異体を本明細書中においてはMDV-Aと示す。MDV-AウイルスRNAをRNeasy mini kit (Qiagen)を用いて抽出し、8つの対応するcDNA断片を表1に挙げたプライマーを用いてRT-PCRによって増幅した。
Madin-Darbyイヌ腎臓(MDCK)細胞およびヒトCOS7細胞を、10%胎児ウシ血清(FBS)を含む改変イーグル培地(MEM)中に維持した。ヒト胎児腎臓細胞(293T)を5% FBSを含むOpti-MEM I(Life Technologies)中に維持する。MDCK細胞とCOS7細胞または293T細胞のいずれかを1:1の割合で6ウェルプレートにて共培養し、この細胞を、およそ80%コンフルエントな状態にてトランスフェクションに用いた。293T細胞およびCOS7細胞は高いトランスフェクション効率を有するが、インフルエンザウイルスの複製を許容しない。MDCK細胞との共培養により組換えウイルスの効率的な複製を確実にする。トランスフェクションの前に、血清含有培地を無血清培地(Opti-MEM I)に代え、4〜6時間インキュベートした。プラスミドDNAのトランスフェクションを、1 μgの8つのプラスミドDNA (PB2、PB1、PA、NP、M、NS、HAおよびNA)の各々のと160 μlのOpti-MEM Iに希釈した20 μlのTransIT-LT1を全量200 μlにて混合し、TransIT-LT1(Mirus)を用いて行った。DNA:トランスフェクション試薬混合物を45分間、室温にてインキュベートし、その後800 μlのOpti-MEM Iを加えた。次にトランスフェクション混合物を共培養したMDCK細胞/293T細胞またはMDCK細胞/COS7細胞に加えた。トランスフェクトした細胞を35℃または33℃にて、6時間〜24時間(例えば一晩)インキュベートし、そしてトランスフェクション混合物を各ウェルにおいて1 mlのOpti-MEM Iと代えた。35℃または33℃にて24時間インキュベーションした後、1μg/mlのTPCK-トリプシンを含有する1mlのOpti-MEM Iを各ウェルに加え、さらに12時間インキュベートした。次に回収したウイルスをコンフルエントな状態のMDCK細胞にて増幅するか、発育鶏卵において直接的に増幅した。12ウェルプレートに入れたMDCK細胞を室温にて1時間、0.2 mlのトランスフェクション混合物で感染させ、次に混合物を取り除き、1μg/mlのTPCK-トリプシンを含有する2mlのOpti-MEM Iと代えた。細胞を35℃または33℃にて、3〜4日間インキュベートした。増幅したウイルスをSPG安定剤の存在下にて-80℃で保存するか、またはプラーク精製し、MDCK細胞または発育鶏卵にて増幅した。
4つのMDV-Aポリメラーゼタンパク質(PB2、PB1、PAおよびNP)の機能的活性を、EGFPレポーター遺伝子をコードするインフルエンザウイルスのミニゲノムを複製するそれらの能力によって分析した。1組8つの発現プラスミド(例えば表4を参照のこと)(Hoffmannら(2001) Eight plasmid rescue system for influenza A virus;Options for the control of influenza International Congress Series 1219:1007-1013)は、A/PR/8/34株(H1N1)のcDNAと増強された緑色蛍光タンパク質(EGFP, pHW72-EGFP)をコードするレポーター遺伝子を含むインフルエンザウイルスミニゲノムを含んでいた。
pAD3000にクローン化した8つのMDV-Aゲノムセグメントの各々が、MDV-Aに由来する単一遺伝子セグメントと共に、対照であるA/PR/8/34株に由来する相補的な7つのセグメントをコトランスフェクトすることによるリアソータントの実験において機能的に発現することを示す。相補的な対照セグメントとの組合せにおいて8つ全ての単一ゲノムセグメントプラスミドは感染性リアソータントウイルスを生じた。これは感染したMDCK細胞において細胞変性効果を生じ、8つ全てのプラスミドが機能的MDV-Aタンパク質をコードしていることを示す。
上記手順に続いて、8つのMDV-Aプラスミド(組換え体)、または6つのMDV-A内部遺伝子、およびA/PR/8/34に由来するHAとNAを含むプラスミド(6:2リアソータント)のいずれかを用いてトランスフェクションして3日後、トランスフェクトした培養上清を用いて新しいMDCK細胞を感染させ、そしてこの感染細胞を1μg/mlのTPCK-トリプシンの存在下において33℃にて3日間インキュベートした。感染したMDCK細胞における組換えウイルスの細胞質効果を顕微鏡を使用して観察した。ウイルス赤血球凝集素の発現を標準的な赤血球凝集アッセイ(HA)を用いて調べた。HAアッセイを50μlの連続的に2倍希釈した培養上清と50μlの1%トリ赤血球とを96ウェルプレートにおいて混合することによって行った。トランスフェクトした8つのMDV-Aプラスミドまたは6:2リアソータントウイルスのいずれかに由来する増幅したウイルスについて、およそ1:254〜1:1024のHAタイターを検出した。Dr. E. Hoffmanより入手した8つのA/PR/8/34プラスミドを用いたトランスフェクション反応をポジティブコントロールとして用いた。感染性インフルエンザウイルスを表5に示すような3つのトランスフェクション反応により作製した。
MDV-Aインフルエンザウイルスワクチン株は、ワクチン製造に関する様々な表現型(弱毒生ワクチン:好冷性(ca)、温度感受性(ts)および弱毒性(att)など)を有する。MDV-A株と非ts病原性wt A/AA/6/60株との配列比較よって、これら2つの株の間で最小17ヌクレオチドの違いが明らかとなった(表6)。MDV-A配列における複数の変化は、GeneBankデータベースにおいて利用可能である全てのA型インフルエンザウイルスと比べて、この株特有のことである。これは1つ以上のこれらのアミノ酸置換がatt、caおよびts表現型に機能的に関連することを示唆している。PB2821における単一のアミノ酸変化は、MDV-Aのts表現型の決定因子としてこれまでに報告されていた、たった1つのヌクレオチド位置だけであった(Subbaraoら(1995) Addition of Temperature-Sensitive Missense Mutations into the PB2 Gene of Influenza A Transfectant Viruses Can Effect an Increase in Temperature Senstivity and Attenuation and Permits the Rational Design of a Genetically Engineered Live Influenza Avirus Vaccine J. Virol. 69:5969-5977)。
MDV-A ts表現型への各wt 遺伝子セグメントの影響を、組換え体、すなわち単一遺伝子リアソータントを作製することによって調べた(表7)。rMDV-Aへのwt PB2の導入によって、38℃においてのみ非tsであったウイルスを生じたが、しかし39℃においてはtsのままであった。33℃と比較して、38℃および39℃におけるウイルスタイターの減少は、MDCK細胞を用いたプラークアッセイによって測定されるように、それぞれ0.6 log10および2.7 log10であった。wt PB1遺伝子セグメントを含むリアソータントは、38℃と39℃の両方においてプラークを形成するその能力についていうと、非tsであった。しかし、この組換え体のプラークサイズは上昇した温度によって影響を受け、rWtと比べて、39℃において顕著に縮小した。rMDV-Aへのwt NP遺伝子セグメントの導入によって、38℃においても非tsであったウイルスを生じたが、wt PB2組換え体と対照的に、wt NP遺伝子セグメントを含むウイルスは39℃においてプラークを形成しなかった。rMDV-Aへのwt PA、MまたはNS遺伝子セグメントの独立した導入は、ts表現型を変化させなかった。以上より3つの遺伝子セグメントが、この表現型の維持において最小限の機能を有したことを示す。
rWtとrMDV-AのPB1、PB2およびNP遺伝子セグメントの間の特異的な差異を体系的に処理し、ts表現型に重要な機能を果たすこれらの変化を同定した。rMDV-AのNP遺伝子はヌクレオチド146においてのみrWt NPと異なった(G34D, 表6)。rMDV-AのPB2遺伝子は3つの部位でrWtと異なったが、ヌクレオチド821のみがアミノ酸変化を生じ(N265S, 表6)、PB2遺伝子セグメントに配置されているts遺伝子座をおそらく示した。MDV-AのPB1遺伝子は6つのヌクレオチド位置においてwt PB1と異なる。その内4つは翻訳領域の変化であった(表6)。各wtアミノ酸残基の置換をrMDV-AのPB1遺伝子セグメントに個々に置換することによって、ts表現型におけるそれらの機能を調べた。1395G(Glu-457)および2005G(Ala)は、MDV-Aのts表現型に影響しなかった。1195A(Lys-391)および1766A(Glu-581)の各々は、38℃においてts表現型にわずかな減少を生じたが、39℃においては影響がなかった(表8)。これらのデータは1195Aおよび1766Aが、PB1遺伝子セグメント内の有望なts遺伝子座であったことを示した。しかし、1195Aと1766Aとの両方の組合せはwt PB1に類似したts表現型を生じなかった(表6)。1395Aではなく2005GのPB1-1195A/1766Aへの付加によって、39℃におけるウイルスのts表現型がさらに減少した。このことは2005AがまたMDV-AのPB1セグメントによって特定されるts表現型の発現において機能を有したことを示す。
MDV-A ウイルスおよび上記の1つ以上のMDV-A由来セグメントを有するリアソータントウイルスによって示される温度感受性および弱毒表現型に加えて、MDV-Aウイルスは、MDCK細胞における増殖と相対的にPer.C6細胞における減少した増殖によって示されるような宿主細胞の抑制を示した。MDV-AおよびMDV-Aに由来するPB1およびPB2セグメントを有するリアソータントウイルスはMDCK細胞におけるそれらの増殖と相対的にPer.C6細胞において顕著に減少した増殖を示した(図20 AおよびBに示す)。
MDV-AのPB1、PB2およびNP遺伝子セグメント中に同定された5つのアミノ酸がMDV-Aのtsおよびatt表現型を再び生じるか否かを決定するために、PB1-391E、581G、661T、PB2-265S、NP-34Gを多岐にわたる野生型ウイルス株(A/PR/8/34;「PR8」)に導入した。得られたウイルスは38℃において1.9 log10減少のウイルスタイター、および39℃において4.6 log10減少(これはrMDV-Aの値と非常に近似した)を示した(図11)。
典型的なB型インフルエンザ主要ドナー株(MDV-B)であるインフルエンザB/Ann Arbor/1/66の好冷変異体(ca/Master Ann Arbor/1/66 P1 Aviron 10/2/97)に由来するウイルスRNAを、感染した発育鶏卵に由来する100 μlの尿膜液よりRNeasyキット(Qiagen, Valencia, CA)を使用して抽出し、そしてこのRNAを40 μlのH2Oに溶出した。各反応に1 μlの抽出したRNAを用いて、ゲノムセグメントのRT-PCRを提供されたプロトコルの従ってOne Step RT-PCRキット(Qiagen, Valencia, CA)を用いて行った。このRT反応を50℃にて50分間、続いて94℃にて15分間行った。PCRを、94℃にて1分間、54℃にて1分間、および72℃にて3分間を25サイクル行った。P遺伝子をBsmBI部位を有するセグメント特異的プライマーを用いて増幅し、2つの断片形成を生じた(表12)。
PCR断片を単離し、BsmBI(またはNPについてはBsaI)を用いて消化し、そして上記のようにpAD3000(ネガティブセンスvRNAおよびポジティブmRNAの転写を可能とするpHW2000の派生物)のBsmBI部位に挿入した。得られたプラスミドの2〜4つを配列決定し、RT-PCR断片の直接的な配列決定に基づいてMDV-Bのコンセンサス配列と比較した。コンセンサス配列と異なるアミノ酸変化を生じるヌクレオチド置換を有するプラスミドを、プラスミドのクローニングによるか、またはQuikchangeキット(Stratagene, La Jolla, CA)を利用して「修復」した。得られたB/Ann Arbor/1/66プラスミドを、pAB121-PB1、PAB122-PB2、PAB123-PA、PAB124-HA、PAB125-NP、PAB126-NA、PAB127-M、およびpAB128-NSと称した。この2方向性の転写系を用いて、全てのウイルスRNAおよびタンパク質を細胞内に産生し、そして感染性B型インフルエンザウイルスの産生を生じる(図4)。
ヘルパーウイルスの存在しない細胞培養系でB型インフルエンザを増殖する試みにて直面する障害を克服するために、本発明は組換えおよびリアソータントB型インフルエンザウイルス株を作製するための新規ベクターおよびプロトコルを提供する。B型インフルエンザウイルスの回収に用いられるベクター系は、A型インフルエンザウイルス作製のために開発されたものに基づく(Hoffmannら(2000) A DNA transfection system for generation of influenza A virus from eight plasmids Proc Natl Acad Sci USA 97:6108-6113;Hoffmann & Webster(2000) Unidirectional RNA polymerse I- polymerse II transcription system for the generation of influenza A virus from eight plasmids J Gen Virol 81:2843-7)。293T細胞またはCOS-7細胞(高いトランスフェクション効率とpolI活性とを有する霊長類の細胞)をMDCK細胞(インフルエンザウイルスの増殖を許容する)と共培養した。293T細胞を5% FBSを含有するOptiMEM I-AB培地中に維持し、COS-7細胞を10% FBSを含有するDMEM I-AB 培地中に維持した。MDCK細胞を、抗生物質および抗真菌剤を添加した1×MEM、10 % FBS中に維持した。ウイルスゲノムベクターを用いたトランスフェクションの前に、細胞をPBSまたはFBSを含まない5 mlの培地を用いて1回洗浄した。10 mlのトリプシン-EDTAを75 cm2 フラスコ中のコンフルエントな細胞に加えた(MDCK細胞を20〜45分間インキュベートし、293T細胞を1分間インキュベートした)。この細胞を遠心分離し、10 mlのOptiMEM I-AB中に再懸濁した。次に、1 mlの懸濁した各細胞系統を18 mlのOptiMEM I-ABに希釈し、混合した。次に、細胞を1ウェルあたり3 mlずつ6ウェルプレートに分けた。6〜24時間後、1 μgの各プラスミドを1.5 ml エッペンドルフチューブ中にてOptiMEM I-AB と混合した(プラスミド(x μlプラスミド + x μl OptiMEM I-AB + x μl TransIT-LT1 = 200 μl);1 μgのプラスミドDNAにつき2 μlのTransIT-LT1)。この混合物を室温にて45分間インキュベートした。次いで、800 μlのOptiMEM I-ABを加えた。培地を細胞から取り除き、そしてトランスフェクション混合物を33℃にて6〜15時間、細胞に加えた。トランスフェクション混合物を細胞から徐々に取り除き、1 mlのOptiMEM I-ABを加え、そして細胞を33℃にて24時間、インキュベートした。トランスフェクションから48時間後、1 μg/mlのTPCK-トリプシンを含有する1 mlのOptiMEM I-ABを細胞に加えた。トランスフェクションから96時間後、1 μg/mlのTPCK-トリプシンを含有する1 mlのOptiMEM I-ABを細胞に加えた。
MDCK細胞での続く継代の後に、感染細胞の上清のRT-PCRを用いて、産生したウイルスの出所の確かさを確認した。8つ全てのセグメントについて、セグメント特異的プライマー(表12)を用いてRT-PCRを行った。図5Aに示すように、全てのセグメントについてPCR産物を得た。PB1、HA、およびNSセグメントのPCR産物の直接的配列決定によって、分析した4つのヌクレオチドがプラスミドpAB121-PB1、PAB124-HA、およびpAB128-NSに見出されるものと同一であることが明らかとなった。これらの結果は、産生されたウイルスが特定のプラスミドより産生されたことを裏付け、また(ネガティブコントロールに加えて)親ウイルスによる研究室汚染の可能性を除外する(図5B)。
MDV-Bウイルスは2つの特徴的な表現型:温度感受性(ts)および好冷性(ca)を示す。tsは、33℃と比べて37℃におけるウイルスタイターが2 log(またはそれ以上)異なるという定義によって規定され、caは33℃と比べて25 ℃におけるウイルス増殖が2 log未満の差であることによって規定される。トリ腎臓プライマリー(PCK)細胞を親ウイルスMDV-Bまたはプラスミドに由来するトランスフェクトウイルスに感染させ、3つの温度におけるウイルス増殖を測定した。
B型インフルエンザの主要系統を表す複数の異なる株のHAおよびNAセグメントを、本質的に上記のように増幅し、そしてpAD3000にクローン化した。プライマーをHAおよびNAセグメントの同時RT-PCR増幅のために最適化した。セグメント4(HA)およびセグメント6(NB/NA)の非コード領域を示すvRNAの終末領域の比較によって、B型インフルエンザウイルスのHA遺伝子とNA遺伝子との間で、5’末端の20終末ヌクレオチドと3’末端の15ヌクレオチドが同一であることが明らかとなった。RT-PCR用のプライマー対(下線部の配列はB型インフルエンザウイルス特異的である) Bm-NAb-1:TAT TCG TCT CAG GGA GCA GAA GCA GAG CA(配列番号87); Bm-NAb-1557R:ATA TCG TCT CGT ATT AGT AGT AAC AAG AGC ATT TT(配列番号88)を合成し、種々のB型インフルエンザ株よりHAおよびNA遺伝子を同時増幅するために用いた(図8)。B/Victoria/504/2000、B/Hawaii/10/2001、およびB/Hong Kong/330/2001のHAおよびNAのPCR断片を単離し、BsmBIを用いて消化し、そしてpAD3000に挿入した。これらの結果は、B型インフルエンザ主要系統を表す複数の様々な野生型ウイルスよりB型インフルエンザのHAおよびNA遺伝子を含むプラスミドを効率的に作製するための上記プライマーの適応性を示した。このRT-PCR産物を配列決定および/または発現プラスミドへのクローニングのために用いることができる。
MDV-Bウイルス(ca B/Ann Arbor/1/66)はヒトにおいては弱毒化され、フェレットにおいては弱毒表現型を示し、そして細胞培養においては好冷および温度感受性表現型を示す。MDV-Bの内部遺伝子の推定アミノ酸配列をBLAST検索アルゴリズムを用いてロスアラモスのインフルエンザデータベース(ワールドワイドウェブ:flu.lanl.gov)の配列と比較した。他の株には存在しないMDV-B特有の8つのアミノ酸を同定した(表17)。PB1、BM2、NS1、およびNS2をコードするゲノムセグメントは特有の置換残基を示さない。PAおよびM1タンパク質はそれぞれ2つ、そしてNPタンパク質は4つの特有の置換アミノ酸を有する(表17)。1つの置換アミノ酸はPB2の位置630に見出される(さらなる株 B/Harbin/7/94(AF170572)はまた位置630にアルギニン残基を有する)。
これまでにVero細胞より組換えA型インフルエンザを回収できることが示唆されている(Fodorら(1999) Rescue of influenza A virus from recombinant DNA J. Virol. 73:9679-82;Hoffmannら(2002) Eight-plasmid system for rapid generation of influenza virus vaccine Vaccine 20:3165-3170)。報告された方法は脂質試薬を用いることを必要とし、たった1つの株、つまりA型インフルエンザ(A/WSN/33およびA/PR/8/34)の高い複製能力を有する研究用の株について記述されているのみであり、ワクチン製造に好適な弱毒生ウイルスの製造における用途を限定する。本発明は、エレクトロポレーションを用いてVero細胞より組換えインフルエンザウイルスを回収するための新規方法を提供する。これらの方法はA型インフルエンザウイルス株およびB型インフルエンザウイルス株の両方の製造に好適なものであり、鼻腔内ワクチン製剤などの投与に好適な弱毒生ワクチンの調製を容易にする無血清条件下にて増殖したVero細胞より好冷、温度感受性、弱毒ウイルスなどの回収を可能とする。ウイルス株における幅広い適用性に加えて、エレクトロポレーションは、細胞培養基のための増殖培地以外のさらなる薬剤を必要とせず、従って、望ましくない汚染の可能性を減少させる。特に、本方法は無血清条件下での増殖に適合したVero細胞(病原体が存在しないことが認められ、ワクチン製造に好適なVero細胞単離物など)を用いて組換えおよびリアソータントウイルスを作製することに有効である。この特性により、細胞基質へのDNAの商業的導入に適切な方法としてエレクトロポレーションを選択することを支持する。
上記考察のとおり、ウイルスゲノムの8つのセグメントの各々をコードするプラスミドを細胞にエレクトロポレーションすることによって、SF Vero細胞よりインフルエンザウイルスを回収できる。この方法を用いて、野生型インフルエンザ株のHAおよびNAとMDV株(例えば好冷MDV株またはPR8)のPB1、PB2、PA、NP、NS、およびMからなる6:2 ウイルスを作製することができる。一部の野生型HAおよびNAセグメントのために、SF Vero細胞における回収が不十分となる。エレクトロポレーションしたSF Vero細胞とトリ胚腎臓(CEK)細胞との共培養がプラスミドの回収効率を改善することが見出された。例えば、SF Vero細胞のエレクトロポレーションを行ってA/Panama 6:2 ウイルスを回収した場合、試験した30個の卵(1日あたり5個の卵、エレクトロポレーションの2〜7日後)のうちどれも検出可能なHAタイターを有さなかった。しかし、同じ試料を同様にエレクトロポレーションしたSF Vero細胞を、CEK細胞と共培養した場合、30個のうち27個の卵が検出可能なHA タイターを有し(90%効率)、これらのタイターは100またはそれ以上であった。さらに、この改善された回収効率はMDV Aについてもみられる。さらに、A/Sendai(SF cero細胞より回収することが困難である別の6:2ウイルス)は共培養方法によって回収した。
本発明のベクターはまた、遺伝子送達系として、また遺伝子治療のために用いることができる。このような用途については、組換えインフルエンザウイルス(外来性タンパク質を発現する組換えA型インフルエンザウイルスまたはB型インフルエンザウイルスなど)を作製するために望ましい。例えば、B型インフルエンザウイルスのセグメント7はスプライシングされないために、異種性核酸配列を挿入することに都合の良い遺伝子エレメントを提供する。mRNAはM1およびBM2タンパク質をコードする2つのオプーンリーディング・フレームを有する2つのシストロンを含む。BM2またはM1のオプーンリーディング・フレームは、目的の異種性配列(増強された緑色蛍光タンパク質(EGFP)をコードする遺伝子など)に置換される。本発明のプラスミドを基盤とするベクター系を用いて、M1-EGFPおよびBM2のオプーンリーディング・フレームをコードするcDNAを2つの異なるプラスミドにクローン化する。オプーンリーディング・フレームは、複製および転写に必要とされるシグナルを含む、セグメント7の非コード領域に隣接する。あるいは、2つのプラスミドを、1つはM1 ORFを含み、そしてもう1つはEGFP-BM2を含み、構築する。得られた9つのプラスミドのコトランスフェクションによって、異種性遺伝子配列を含む組換えB型インフルエンザウイルスの産生を生じる。同様にEGFPがA型インフルエンザのNS1 セグメントより発現されうる。
典型的な好ましい実施形態において、弱毒、好冷生インフルエンザA/AA/6/60株はA型インフルエンザ FluMistTM ワクチンの主要ドナーウイルス(MDV-A)である。MDV-Aの6つの内部遺伝子は好冷(ca)温度感受性(ts)および弱毒(att)表現型を各ワクチン株に与える。逆遺伝子学を用いて、多重アミノ酸が3つの遺伝子セグメント:PB1-K391E、E581G、A661T、PB2-N265S、およびNP-D34Gに分離したことを示す。これらはMDV-Aのtsおよびatt表現型の発現を制御する。6:2ワクチン株のプラスミド回収によって、古典的な再集合技術よりも、効率的なインフルエンザワクチンの製造を可能とする。
ウイルス株、細胞および抗体:野生型(wt) A型インフルエンザウイルス株、A/Fujina/411/02(A/Fujian)、A/Sendai-H/F4962/02(A/Sendai)およびA/Wyoming/03/03(A/Wyoming)をCenter for Disease Control(Atlanta, GA)から得て、MDCK細胞または発育鶏卵(卵)にて一度増幅した。バクテリオファージT7 RNAポリメラーゼ(MVA-T7)を発現する改変したワクシニアウイルスAnkara株をCEK細胞において増殖した。HEp-2細胞、COS-7細胞およびMDCK細胞(American Type Culture Collections(ATCC)より得た)を5%胎児ウシ血清(FBS)を含有する最小必須培地(MEM)中に維持した。A/Ann Arbor/6/60、A/Sendai-H/F4962/02およびA/Wyoming/03/03に対するポリクローナル抗血清をトリにおいて産生した。A型インフルエンザのNPタンパク質に対するモノクローナル抗体をBioDesign(Saco, MI)より得た。
野生型(wt) A型インフルエンザウイルス株、A/Fujian/411/02、A/Sendai-H/F4962/02およびA/Wyoming/03/03をCenter for Disease Control (Atlanta, GA)より得て、MDCK細胞または発育鶏卵において一度増幅した。表20に示すように、A/Fujianは細胞培養において3回継代したのみであったが、A/SendaiおよびA/Wyomingは卵において11回継代した。これらの3つの株のHAおよびNAの配列を、これらのウイルスから抽出したvRNAを用いたRT-PCR産物の配列決定によって決定した。これら3つのH3N2株のHAおよびNAの配列の差異を表1に挙げる。A/SendaiはそのHA1アミノ酸配列においてはA/Fujianと同一であったが、NA配列においては位置119、146および347の3つのアミノ酸が異なっていた。A/WyomingはA/Fujianと同一のNA配列を有したが、A/FujianおよびA/SendaiのHA1とは4つのアミノ酸が異なっていた。さらに、A/SendaiとA/Wyomingの両方はHA2のGly-150に代えてGlu-150を有した。MDCK細胞において一度増幅した後、wt A/FujianのHA1における183残基がHis-183からLeu-183に変化し、His-183を有するwt A/Fujianウイルスを単離することが困難となった。これはHis-183を有するウイルスが、in vitroにおける増殖優位性を有したことを示す。
A/Fujianは、A/SendaiとNAにおける3つのアミノ酸(E119Q、Q136KおよびH347Y)だけ異なり(表20)、1つ以上のこれらの変化によりA/Fujianよりも高いタイターまで発育鶏卵におけるA/Sendaiの複製を可能にしたと推測された。G、D、AまたはV残基によるE119の置換が、減少したノイラミニダーゼ活性を生ずる様々な抗ノイラミニダーゼ薬剤耐性株について報告されている。NAにおけるE119Qまたは他の2つのいずれかの変化がA/FujianのNA活性および発育鶏卵における複製能力に影響を及ぼすか否かを決定するために、単一または二重の置換突然変異をA/Fujian NA発現プラスミドに導入し、そしてトランスフェクトしたHEp-2細胞のNA活性を測定した。さらに、A/Fujian NAに突然変異を有する組換え6:2組換えウイルスをまた回収し、MDCK細胞および卵におけるその増殖を比較した(表21)。A/Fujian(E119Q136H147)はA/Sendai(Q119K136Y147)よりもおよそ80%高いNA活性を有した。単一のQ119突然変異は66%のNA活性を有し、Y347変化はNA活性に最小限の影響を及ぼしたが、K136のみでは25%の活性を有した。二重突然変異(K136Y347、Q119Y347、およびQ119K136)はA/Fujianのそれぞれ、29%、52%よび25%のレベルに減少したNA活性を有した。これらのデータは、上記3つのNA残基がK136>Q119>Y347の順序でNA活性に影響を及ぼしたことを示した。
A/Fujianとは異なるA/Wyoming/03/03における4つのHA1残基の変化を、ウイルス複製におけるそれらの役割について調べた。単一または多重の置換突然変異をA/Fujian HA cDNAに導入し、改変したHAプラスミドをA/Fujian NAのどちらかと共にMDV-Aに導入した。全ての6:2リアソータントウイルス変異体はMDCK細胞において十分に複製したが、発育鶏卵においては別の様式で増殖した(表33)。A/Fujian HA(T128G186S219V226)を有する6:2リアソータントは、卵において複製することができなかった。単一のT128A変化は、卵におけるウイルス増殖を改善しなかった。しかし、G186VまたはV226Iの単一の変化によって、卵における増加したウイルス複製を生じた。HAにおけるG186VおよびV226Iの二重の変化によって、卵において効率的に複製した。残基128および/または219におけるさらなる置換は、ウイルスの複製を顕著に増加させなかった。このように、G186VおよびV226Iの最小2つの変化によって、6:2 A/Fujianの発育鶏卵における効率的な増殖を可能とした。
6:2 A/Fujian株が発育鶏卵における増殖に適応できるか否かを決定するために、ウイルスをMDCK細胞において増幅し、その後卵において継代した(表23)。3.0 log10PFUのウイルスを卵に接種した場合、2.0 log10PFU/ml未満のウイルスを、回収した尿膜液中に検出した。この材料の継代後には、感染性ウイルスを回収することができなかった。第2の継代実験の間に、発育鶏卵に接種されるウイルスの量を5.9 log10PFUまで増加した。回収した尿膜液(FJ-EP1)において3.9 log10PFU/mlのタイターを検出し、そして卵においてさらに継代したものは6.2 log10PFU/mlのウイルスタイターまで増加した(FJ-EP2)。卵においてさらに継代したもの(FJ-EP3)は、ウイルスタイターが8.2 log10PFU/mlまで増加した。FJ-EP2ウイルスの配列分析によって、HA RNAセグメントのヌクレオチド625におけるAからUへの突然変異が明らかとなった。この突然変異はHAタンパク質におけるH183L変化を生じた。さらなる分析によって、この変化がMDCK細胞におけるウイルス増幅の間にも生じることが明らかとなった。H183L突然変異はまた、上記のようにMDCKおよび卵におけるwt A/Fujain HAの複製の間にwt A/Fujain HAにも見出された。V226A置換を生じるHAのヌクレオチド754におけるUからCへのさらなる突然変異が、FJ-EP3の増幅されたウイルスに見出された(表23)。NAセグメントに変化は検出されなかった。
上記研究から、A/FujianのNA活性を減少させるNA変化は、このウイルスが卵にて増殖するのに十分であることを示した。一方、HA変化(G186VおよびV226IまたはH183LおよびV226A)が受容体結合親和性を増加させ、A/Fujianの高いNA活性を補うことができる。A/FujianのHAタンパク質における変化がその受容体結合能を増加させるか否かを決定するために、HA-V186I226変化を有する6:2 A/FujianおよびHA-L183A226変化を含む卵に適合した6:2 A/Fujianの吸着を、6:2 A/Fujian, A/Sendai、およびA/Wyomingと比較した。各ウイルスを4℃または33℃において30分間、感染効率1.0にてMDCK細胞に吸着させ、接種物を取り除き、そして感染細胞を3回洗浄するか、または洗浄しなかった。33℃における6時間のインキュベーションの後、感染細胞の割合を抗NP抗体を用いた免疫蛍光分析によって測定した。図36に示すように、吸着を4℃にて行った場合、6:2 A/FujianおよびA/Sendaiには26〜27%の細胞が感染したが、大半のウイルスは洗浄工程によって容易に除去された。33℃にて、洗浄することによって、6:2 A/Fujianウイルスの感染を非常に減少させた(37.8%に対して6.2%)が、6:2 A/Sendaiの感染には顕著な効果がなかった(51.7%に対して42.8%)。対照的に、HA-V186I226またはHA-L183A226を有する6:2 A/Wyoming、A/Fujianは、細胞を4℃または33℃にて吸着させるとか、洗浄工程の有無にかかわらず、同様の感染効率を有した。これらのデータは、A/FujianおよびA/Sendai HAが、4℃において結合したウイルスは細胞より容易に洗い流すことができるような低い結合親和性を有することを示した。結合およびウイルス侵入速度は33℃において速かったため、洗浄工程は6:2 A/Sendaiウイルス感染に最小限度の影響を与えた。しかし、大半の結合6:2 A/Fujianは、その高いNA活性が33℃において効率的なウイルス結合を妨げるために(データは示さない)、同様の条件で洗い流された。
改変HAおよびNA残基を有するウイルスが、ウイルス抗原性に影響するか否かを調べるために、赤血球凝集阻害アッセイ(HAI)をフェレット抗A/Wyoming血清および抗A/Sendai血清を用いて行った(表24)。抗A/Wyomingフェレット血清または抗A/Sendaiフェレット血清は6:2 A/FujianウイルスまたはA/Sendaiウイルスのいずれかを用いて測定した場合、同様のHAIタイターを有した。6:2 A/Wyomingウイルスを用いた場合、やや高いHAIタイターを検出した。おそらくこれは、赤血球上の細胞受容体に対するA/Wyoming HAの堅い結合によるものである。2つの改変したウイルス(A/FujianHA-V186I226およびA/Fujian HA-L183A226)は、いずれかの血清で測定した場合、A/Wyomingと類似したHAIタイターを有した。これらの結果は、A/SendaiとA/Wyomingとの間のアミノ酸の差異および、本研究にて作製された改変HAウイルスが、ウイルス抗原性を変化させなかったことを示した。
Claims (15)
- リアソータントインフルエンザウイルスの回収方法であって、該方法は
(i)リボ核タンパク質複合体を形成でき、ヘルパーウイルスの非存在下でウイルス粒子を組み立てることができるように、Vero細胞内にてゲノムまたはアンチゲノムvRNAセグメント、核タンパク質、およびRNA依存性ポリメラーゼを発現するポリヌクレオチドベクターを用いて前記Vero細胞にエレクトロポレーションすること、
ここで、該ポリヌクレオチドベクターの少なくとも1つが、インフルエンザウイルスタンパク質のみをコードするインフルエンザウイルスのゲノムセグメントに対応するウイルスcDNAを含み、該cDNAがRNAポリメラーゼI(polI)プロモータエレメントと的確な3’末端を有するvRNAまたはcRNAの合成調節エレメントとの間に挿入されており、該エレメントがRNAポリメラーゼ II(polII)プロモータとポリアデニル化シグナルとの間に挿入されている、
(ii)該エレクトロポレーションしたVero細胞を別の細胞型と、ウイルス複製が許容される条件下で共培養すること、および
(iii)少なくとも90%の回収効率で、インフルエンザウイルスを回収すること、
を含む、前記方法。 - 前記Vero細胞が、無血清Vero細胞である、請求項1に記載の方法。
- 前記インフルエンザウイルスが好冷ウイルスである、請求項1または2に記載の方法。
- 前記インフルエンザウイルスが弱毒ウイルスである、請求項1〜3のいずれか1項に記載の方法。
- 前記ベクターが一組のプラスミドであり、該一組のプラスミドにおける異なるプラスミドの数が8個である、請求項1〜4のいずれか1項に記載の方法。
- 前記ベクターが一組のプラスミドであり、該一組のプラスミドにおける異なるプラスミドの数が12個である、請求項1〜4のいずれか1項に記載の方法。
- 前記別の細胞型がCEK細胞である、請求項1〜6のいずれか1項に記載の方法。
- 前記別の細胞型がMDCK細胞である、請求項1〜6のいずれか1項に記載の方法。
- 前記インフルエンザウイルスがA型インフルエンザウイルスである、請求項1〜8のいずれか1項に記載の方法。
- 前記ベクターがA/PR/8/34に由来する少なくとも一のvRNAセグメントを発現する、請求項9に記載の方法。
- 前記ベクターがMDV-Aに由来する少なくとも一のvRNAセグメントを発現する、請求項9に記載の方法。
- MDV-AがA/Ann Arbor/6/60である、請求項11に記載の方法。
- 前記インフルエンザウイルスがB型インフルエンザウイルスである、請求項1〜8のいずれか1項に記載の方法。
- 前記ベクターがMDV-Bに由来する少なくとも一のvRNAセグメントを発現する、請求項13に記載の方法。
- MDV-BがB/Ann Arbor/1/66である、請求項14に記載の方法。
