Abstract
【解決手段】溶解した向骨性のラジウム−223塩を含み、安定化するアルカリ土類金属カチオンアナログ担体として1つのカチオンまたはいくつかのカチオンの組合せを含むかまたは含まず、コロイドの沈殿および/または生成を防止する薬剤を含むかまたは含まず、さらに製薬上許容しうる担体およびアジュバントを含むことを特徴とするインビボ投与のための生理的に許容しうる製剤。

α粒子として放出されたエネルギーの割合:≧96%
β粒子として放出されたエネルギーの割合:≦ 3%
ガンマ線(総計( 0.3 MeV)の一部はまた崩壊中に放射され、ガンマ線分光法を使用して試料中の同位元素の定性および定量を行なうために用いることができる。例えば、ラジウム−223は154.19KeV(5.59% 同位体存在比(abundance))において特性ガンマ線ピークを有し、ラドン−219は401.78keV(6.6%)においてピークを有し、ビスマス−211は351.0keV(12.8%)においてピークを有する(Ekstrom 等, 1989)。これらを用いてインビボで娘核種同位元素に再分配が生じるかを測定することができる。また、223Raは13.6%同位体存在比について269.41KeVのピークを有するが、これは219Rnの9.9%同位体存在比について271.23KeVのピークと区別することは困難である。
227Acおよび227Thは、27年前に製造した231Pa供給源(試料はRadiochemistry Group, Department of Chemistry, University of Oslo, Norwayにより提供された)からf−元素選択的抽出クロマトグラフィー樹脂を用いて単離した。精製した227Acおよび227Thは次いで別のf−元素選択的抽出クロマトグラフィー樹脂に吸着させ、223Raのカウ(cow)として用いた。後者の材料は、225Acを基とする213Bi用ジェネレーターの構築のためにWu 等 (1997)により用いられている。
実施例1に記載するよう製造したラジウム−223の生体内分布を検討した。
骨試料のラジウム−223とビスマス−211の間に放射性同位元素の滞留に差が存在するかを調べるために、平衡にある223Raと娘放射性核種を用いた骨対標準溶液についてのガンマ線分光データを調査した。
223Raが骨に取り込まれた後、核反跳または拡散過程のいずれかによる娘放射性核種の放出の可能性を調査するために、注射後6時間に屠殺した5匹の動物および3日後に屠殺した5匹の動物からの大腿骨を検査した。
ヒトの患者においてしばしば観察されているものときわめて類似している実験的転移パターンを有する動物モデルが開発されている(Engebraaten と Fodstad, 1999)。これらのモデルの1つはヌードラットに心臓注射したMT−1細胞からなり、動物の後肢麻痺が一貫して進行することにより特徴づけられる。化学療法用のシスプラチンまたはドキソルビシンを用いた治療(腫瘍細胞の接種後7日)は、生存率を改善できなかった。腫瘍に冒された動物からの脊椎の解剖および顕微鏡的検査は、大量の腫瘍細胞が正常な骨髄細胞と置き換わり、脊椎の骨部分を侵食していることを明らかにした。
Claims (9)
- 溶解した向骨性のラジウム−223塩を含み、安定化するアルカリ土類金属カチオンアナログ担体として1つのカチオンまたはいくつかのカチオンの組合せを含むかまたは含まず、コロイドの沈殿および/または生成を防止する薬剤を含むかまたは含まず、さらに製薬上許容しうる担体およびアジュバントを含むことを特徴とするインビボ投与のための生理的に許容しうる製剤。
- 安定化するアルカリ土類金属カチオンとして作用する上記カチオンがマグネシウム、カルシウムおよびストロンチウムよりなる群から選択されることを特徴とする請求項1に記載の製剤。
- コロイドの沈殿および/または生成を防止する薬剤がカルボン酸またはカルボン酸の組合せであることを特徴とする請求項1または2に記載の製剤。
- 上記カルボン酸がシュウ酸、オキサロ酢酸、酒石酸、コハク酸、リンゴ酸およびマロン酸よりなる群から選択されることを特徴とする請求項3に記載の製剤。
- 注射、注入もしくは摂取またはその組合せのために調製されることを特徴とする請求項1〜4のいずれかに記載の製剤。
- 罹病した骨および骨の表面を治療する医薬活性製剤を製造するためのラジウム−223の使用。
- 製剤が、他の治療的に有効な構成要素、例えばビスホスホン酸を含む化学療法、外科的療法、外部ビーム照射、向骨性の放射性医薬を放射する低LET照射およびホルモン治療との併用治療において使用される請求項6に記載の使用。
- 溶解した向骨性のラジウム−223塩を含む製剤が骨および/または軟組織を冒す非悪性疾患および悪性疾患に関連する治療および/または緩和に使用される、請求項6または7に記載の使用。
- 悪性疾患が、前立腺癌、乳癌、腎臓および泌尿器癌、原発性骨癌、肺癌および多発性骨髄腫よりなる群から選択される、請求項8に記載の使用。
