Abstract
Description
ラブドウイルスファミリーの一員である、水疱性口内炎ウイルス(VSV)は、非分節、マイナス鎖、一本鎖RNAゲノムを有する。ウイルスの11kbゲノムは、ウイルスの5つの構造タンパク質をコードする5つの遺伝子:複製されたRNAをカプシドに包むために化学量論的な量で必要である、ヌクレオカプシドタンパク質(N);RNA依存性RNAポリメラーゼ(L)のコファクターである、リンタンパク質(P);マトリックスタンパク質(M)および接着糖タンパク質(G)を有する(例えば、Gallione et al., 1981 J. Virol., 39:529−535; Rose and Gallione, 1981, J. Virol., 39:519−528; 米国特許第6,033,886号;米国特許第6,168,943号を参照)。
水疱性口内炎ウイルス(VSV)は、免疫原性組成物および/または上で記載されるような治療用タンパク質をコードする遺伝子の送達における使用のために、VSVを魅力的なベクターにする多くの特性を有するので、哺乳類細胞培養からの改良された純度の組換えVSVを産生する精製プロセスのために、当技術分野において現実的に必要とされている。以下に記載される組成物およびプロセスは、その必要性を検討する。以下の実施例3−8において示されるように、哺乳類細胞培養からVSVを精製する改良されたプロセス(例えば、図1を参照)、およびそれにより精製されたVSVが記載される。
哺乳類細胞培養におけるVSVの産生は当業者に周知であり、組換えVSVにより細胞培養(宿主細胞)を感染させること、細胞培養においてVSVを増殖すること、および適切な時間に細胞培養を採取することを一般に含んでいる。VSVは培地中へ宿主細胞から分泌されるので、VSV産物は細胞培養液から採取される。
本明細書において記載されるVSVを感染させた哺乳類細胞培養の細胞培養液からVSVを精製する新規プロセスは、特定の精製工程を含む。図1におけるフローチャートは全般的な精製スキームの概要を述べ、(a)一次清澄化、(b)二次清澄化、(c)陰イオン交換膜吸着、(d)タンジェンシャルフロー濾過および(e)濾過の工程を含んでいる。さらに特に、そのような工程は、(a)低速遠心分離による細胞培養液の清澄化、(b)0.2〜0.45μmフィルターを通した濾過による上清のさらなる清澄化、(c)VSVの濾過溶液の陰イオン交換膜吸着材での精製、(d)緩衝液交換するおよびタンジェンシャルフロー濾過(TFF)によるVSVの濃縮および(e)0.2〜0.22μmフィルターを介するVSV未透過残留液の最終濾過を含む。特定の他の実施形態において、上記の精製プロセスの工程(a)〜(e)は、室温で行なわれる。以下に定義されるように、「室温」は、約15℃および25℃のまたはその間の温度または複数の温度である。したがって例えば、工程(a)〜(e)を行なうための適温は、少なくとも15、16、17、18、19、20、21、22、23、24および25℃を含む温度、またはその間の端数の温度を含んでいる。1つの特定の実施形態において、精製プロセスの工程(a)〜(e)は、20℃で行なわれる。
特定の実施形態において、VSVを感染させた哺乳類細胞培養の細胞培養液は、低速遠心分離によって(またはあるいは深層濾過によって)清澄化され、上清中に回収されたVSVは、細胞培養液の「一次(または1°)清澄化」と本明細書において呼ばれる。特定の実施形態において、細胞培養液の一次清澄化は室温で行なわれる。
遠心分離(または深層濾過)による一次清澄化後に、VSV上清(または濾液)は、0.2〜0.25μmフィルターを介する濾過(または精密濾過)によってさらに清澄化され(2°)、VSVの回収は濾過溶液中にある。1つの特定の実施形態において、精密濾過は、上で定義されるように室温で行なわれる。濾過/精密濾過培地は、製品の様々な材料および方法において利用可能であり、当業者に公知である。例示的な精密濾過フィルターユニットは、Millipore・Millex(登録商標)−GVフィルターユニット(Millipore;ビレリカ、マサチューセッツ)、Millipore・Millex(登録商標)−GPフィルターユニット、Pall Supor(登録商標)フィルターユニット(ポール社;イーストヒルズ、ニューヨーク)、Sartorius・Sartobran(商標)フィルターユニット(Sartorius;エッジウッド、ニューヨーク)およびSartorius・ザルトポア(商標)2フィルターユニットを含むが、これらに限定されない。特定の実施形態において、これらの濾過ユニットは、0.2〜0.45μmの間のサイズのフィルターを有する。これらのフィルターは、少なくとも0.2、0.25、0.3、0.35、0.4および0.45μmの孔、ならびにその間の端数の孔径を有するフィルターを含んでいる。1つの特定の実施形態において、フィルターは0.2μmSartorius・Sartobran(商標)フィルターユニットである。濾過されたVSVは、濾過溶液中で回収される。
VSV産物が清澄化によって回収されたならば(すなわち上で記載される1°および2°)、VSVは陰イオン交換膜吸着材でさらに精製される。膜吸着材の材料は当業者に周知であり、Sartorius(エッジウッド、ニューヨーク)、ポール社(イーストヒルズ、ニューヨーク)およびシグマ・アルドリッチ社(セントルイス、ミズーリ)のような供給業者から利用可能である。例示的な陰イオン交換膜吸着材は、Sartobind(商標)Q膜吸着材(Sartorius)およびMustang(商標)Q膜吸着材(ポール社)を含むが、これらに限定されない。1つの特定の実施形態において、陰イオン交換膜吸着材は、ポール・Mustang(商標)Q膜吸着材である。一般的に、通常のイオン交換クロマトグラフィーからの公知の方法および緩衝液は、膜吸着材クロマトグラフィーに直接適用することができ、当業者に公知である。特定の実施形態において、陰イオン交換膜吸着材クロマトグラフィーは、上で定義されるように室温で行なわれる。
陰イオン交換膜吸着材クロマトグラフィーによるVSV精製に続いて、VSVはさらにタンジェンシャルフロー濾過(TFF)により精製される。一般的に、TFFは、液体溶液(または懸濁物)中の成分を分離するために膜を使用する圧力駆動プロセスであり、液体(供給フロー)は膜の表面に沿って接線方向に(タンジェンシャルに)ポンプで送り込まれ、加圧力は(膜の)濾液側へ膜を通して液の「一部」を押し出す役目をする。特定の実施形態において、TFFは室温で行なわれる。このプロセスにおいて、緩衝液は交換され、VSVは濃縮される。1つの実施形態において、TFFは、陰イオン交換膜吸着工程から回収されるVSVの少なくとも5倍の濃縮、続いて少なくとも1回の緩衝液交換を含む。別の実施形態において、TFFは陰イオン交換膜吸着工程から回収されるVSVの少なくとも5〜10倍の濃縮、続いて少なくとも5、または少なくとも6回の緩衝液交換を含む。さらに他の実施形態は、陰イオン交換膜吸着工程から回収されるVSVの濃縮に続いて、少なくとも2、少なくとも3、少なくとも4、少なくとも5、または少なくとも6回の緩衝液交換を含んでいる。
精製の最終プロセスの工程は、TFFからのVSV未透過残留液の最終精密濾過であり、精密濾過を介する二次清澄化について上で記載されるように、未透過残留液は0.2〜0.25のμmフィルターを通して濾過され、実施例7において以下にさらに記載される。例えば、そのような濾過セットは、サイズ0.20、0.21、0.22、0.23、0.24もしくは0.25μm、またはその間の端数のサイズのフィルターを利用してもよい。
本明細書において記載されるように、改良された純度のVSVは、上で記載される新規精製方法の利用によって哺乳類細胞培養から得られる。「改良された純度」によって、精製VSVが細胞培養タンパク質および核酸の混入物を少なくとも90.0%不含であることが意味される。他の実施形態において、改良された純度のVSVは、細胞培養タンパク質および核酸の混入物を99.0%不含である。1つの特定の実施形態において、改良された純度のVSVは、細胞培養タンパク質および核酸の混入物を99.8%不含である。
特定の実施形態において、免疫原性組成物は、本明細書において記載される精製プロセスに従って精製された遺伝子操作VSVの免疫原性用量を含む。例えば特定の実施形態において、免疫原性組成物は、本明細書において記載される精製プロセスに従って精製された組換えVSVを含み、VSVは、複製には必須でないVSVゲノムの領域中へ挿入された、またはその領域を置換する1つまたは複数の外来のRNA配列を含む。上記のセクションIII中に記載されている組換えVSVの実施形態のいずれかは、これらの免疫原性組成物において利用することができる。したがって特定の実施形態において、精製VSV免疫原性組成物は、哺乳類被験体(例えばヒト)に対する投与のために製剤化される。
以下の実施例は、ほかに詳細に記載された場合以外は、当業者に周知のルーチンの標準技術を使用して実行された。以下の実施例は例示的目的のために提供され、本明細書において記載される組成物およびプロセスの範囲を限定するいかなる手段で解釈されるべきでない。実施例1および2は、例示される3つのVSVコンストラクトすべてに関する。実施例3〜9は、具体的にはコンストラクトVSVIN N4CT9−gag1に言及する。実施例10〜11は、具体的にはコンストラクトVSVIN N4CT1−gag1に言及する。実施例12は、具体的にはコンストラクトVSVNJ N4CT1−gag1を言及する。以下の実施例における精製組換えVSV( Indiana血清型;rVSVIN)は、VSVゲノムの第1の位置でのHIVのgag遺伝子(gag1)、およびVSVゲノムの第4の位置へシャッフルされたN遺伝子(N4)を含む。1つのコンストラクトにおいて、VSVは、短縮細胞質尾部(「CT9」)を持つG遺伝子を有し、このコンストラクトは「VSVIN N4CT9−gag1」と呼ばれた。別のコンストラクトにおいて、VSVは、短縮した細胞質尾部(「CT1」)を持つG遺伝子を有し、このコンストラクトは「VSVIN N4CT1−gag1」と呼ばれた。他の実施例において、以下の実施例における精製組換えVSV(New Jersey血清型;rVSVNJ)は、VSVゲノムの第1の位置でのHIVのgag遺伝子(gag1)、VSVゲノムの第4の位置へシャッフルされたN遺伝子(N4)、および短縮した細胞質尾部(「CT1」)を持つG遺伝子を含み、このコンストラクトは「VSVNJ N4CT1−gag1」と呼ばれた。これらのコンストラクトおよび変異は、国際特許公開番号WO 2005/098009中に詳細に定義されており、参照することにより本明細書に組み入れられる。
以下の分析は、実施例2〜12において以下に記載される精製プロセスを評価するために利用された。
VSVの実験稼動を、ベロ細胞(アフリカサル腎細胞)マイクロキャリアー培養を使用して、10リットルのバイオリアクター中で産生した。使用されるベロ細胞は、cGMPマスターセルバンクから得られた。ベロ細胞を、7.5グラム乾燥ビーズ/リットルの密度でサイトデックス(Cytodex)(商標)Iマイクロキャリアー(アマシャム・バイオサイエンス社;ピスカタウェイ、New Jersey)上で増殖した。バイオリアクター培養のための作動体積は5.5〜6.5リットルであった。接種のために、ベロ細胞を、およそ2リットルの全体積中でサイトデックス(商標)Iマイクロキャリアーと組み合わせた。培養の目標播種密度は5×105細胞/mlであった。マイクロキャリアーへの細胞接着を促進するために、この減少された体積で、2時間の断続的撹拌サイクルを行なった。培養を40rpmで5分間撹拌し、次に0rpmで20分間沈降させる、完全なサイクルを4回行なった。
バイオリアクターから細胞培養を採取した後に、細胞/細胞残屑および他の微粒子不純物は、「産物回収」として公知のプロセスにおいて除去された。VSVはベロ細胞から培養液中へ分泌されたので、VSVは清澄化培養液中に回収された。したがって、VSV細胞培養液上清(例えば10Lのバイオリアクターの稼動から約4.0〜4.5L)は、深層濾過または低速遠心分離のいずれかによって清澄化された。
実施例3において上で記載される一次清澄化後に、濁度レベルを減少させるために、上清(または濾液)をさらに処理した(二次清澄化)。いくつかの滅菌精密濾過フィルター(0.2〜0.25μm)を評価し(表3)、それらはMillipore・Millex(登録商標)−GVフィルターユニット(Millipore;ビレリカ、マサチューセッツ)、Millipore・Millex(登録商標)−GPフィルターユニット、Pall Supor(登録商標)フィルターユニット(ポール社;イーストヒルズ、ニューヨーク)、Sartorius・Sartobran(商標)フィルターユニット(Sartorius;エッジウッド、ニューヨーク)およびSartorius・ザルトポア(商標)2フィルターユニットを含んだ。最適なフィルターは限定されたVSV結合能力を有するか、またはVSV結合能力を全く持たないべきであり、さらにできるだけ微粒子汚染を除去するべきである。
実施例4中で記載されている二次清澄化工程に続いて、VSV濾液の精製はいくつかのクロマトグラフィー樹脂を使用して、検査/スクリーニングされた。VSV粒子が混入タンパク質と比較して大きいサイズなので、大きな孔径を備えた樹脂のみが評価され、それらはUNOスフェア(UNOsphere)(商標)Q陰イオン交換樹脂(バイオ・ラッド・ラボラトリーズ社)、UNOスフェア(商標)S陽イオン交換樹脂(バイオ・ラッド・ラボラトリーズ社)、CHTセラミックハイドロキシアパタイトタイプI樹脂(バイオ・ラッド・ラボラトリーズ社)、CFTセラミックフルオロアパタイトタイプI樹脂(バイオ・ラッド・ラボラトリーズ社)およびCST I混合モード樹脂(mixed mode resin)(GEヘルスケア社)を含んだ。比較のために、2つの親和性樹脂も評価された(Matrex(商標)CELLUFINE(登録商標)サルフェイト樹脂(Millipore)およびヘパリンセファロース樹脂(GEヘルスケア社))。実験は室温でバッチモードによって行なわれた。異なる洗浄条件および溶出条件からのサンプルを回収し、SDS−PAGEおよびプラーク分析によって分析した。
実施例5中に上で記載されているように、二次清澄化工程(すなわち0.2μm Sartobran(商標)フィルター)からの濾液が、UNOスフェア(商標)Q樹脂、UNOスフェア(商標)S樹脂、CHT I樹脂、CFT I樹脂、CST I樹脂、CELLUFINE(登録商標)樹脂またはヘパリンセファロース樹脂により精製される場合、VSV回収は比較的低い。したがって、実施例4からのVSV濾液の精製を、2つの陰イオン交換膜吸着材の、Sartobind(商標)Q膜吸着材(Sartorius;エッジウッド、ニューヨーク)およびMustang(商標)Q膜吸着材(ポール社;イーストヒルズ、ニューヨーク)を使用してさらに評価した。
膜吸着材精製のためのVSV供給液(出発物質)は、タンジェンシャルフロー濾過(TFF)分離(750kDaの分子量カットオフを有するTFF膜を使用して)からの未透過残留液であった。次に、TFF分離からのVSV未透過残留液(それらはVSVおよび不純物/混入物のタンパク質およびDNAを含む)は、4℃または−70℃のいずれかで保存された。

Mustang(商標)Q膜吸着材を用いたVSV精製
Mustang(商標)Q膜吸着材もまたVSV精製手段として研究された。Mustang(商標)Q吸着材のための操作条件は最適化され、以下に記載された。
ローディング体積1は培養体積に基づいた。
VSV力価2:VSV供給液の力価は6.9×106/mLであった。
Mustang(商標)Qフィルター膜体積は0.35mlであった。
FT1〜FT4はそれぞれフロースルー1〜4である。
VSV濃縮および緩衝液交換は、タンジェンシャルフロー濾過(TFF)限外濾過/ダイアフィルトレーション(UF/DF)システムを使用して行なわれた。Mustang(商標)Q膜吸着材からのVSV溶出プールは、高い(0.7M NaCl)塩濃度の10mM HEPES緩衝液中にあり、さらに微量の不純物を有していた。したがって、UF/DF工程は、残存不純物を除去し、適切な産物処方緩衝液中で最終的なVSV産物を産生するのに必要であった。

(g)5倍濃縮およびPBS+4%のショ糖(10mMリン酸カリウム、138mM NaCl、pH7.2)の中へ6回のダイアフィルトレーション;ここで、p1は入口圧力であり、p2は出口圧力であり、TMPは膜間圧であり、LMHはリットル/m2時間である。
VSV精製プロセスにおける最終工程は、上で記載されるTFF精製された物質の最終濾過であった。1分あたり100mlの流速で、0.2μm(0.45/0.2μm)Sartorius・Sartobran(商標)フィルターユニット(Sartorius;エッジウッド、ニューヨーク)を、VSV産物の最小損失で、可能性のあるバイオバーデンを除去するために使用した。緩衝液は、10mMリン酸カリウム(pH7.1〜7.3)、138mM NaClおよび7.5%ショ糖であった。
実施例9:VSVIN N4CT9−gag1コンストラクトのためのVSVスケールアップ精製
4回の4.5Lスケールアップ稼動(すなわち細胞培養体積)が行なわれた。これらのスケールアップ稼動の要約は、表10および表11、および図6において以下に示される。SDS−PAGE(データ不掲載)、総タンパク質(表11)、ウイルス力価(表10および図6)およびSE−HPLC(データ不掲載)を含む分析が行なわれた。一貫したプロセスの性能(すなわちVSV産物品質)および不純物除去は、図1において示されるプロセスを使用して、スケールアップ稼動の各々において達成された。
VSVスケールアップ精製プロセスの分析
本明細書において記載される新規VSV精製プロセスを開発する目的(例えば、図1を参照)は、精製されたVSV産物を高回収で高純度VSVを産生することであった。以下の分析は、4回の4.5Lの細胞培養スケールアップ稼動に基づくVSV精製および安定性を記載する。
VSVinN4CT1−gag1コンストラクトのための精製プロセス開発は、細胞培養液(<106pfu/ml)中の低い産物力価で最初に挑戦され、低い産物力価および最終精製バルク濃縮物中の高いDNA混入をもたらした。しかしながら、VSVINN4CT9−gag1のための実施例9において記載されるような精製プロセスは、このVSVコンストラクトに首尾よく適用され、10Lスケール(細胞培養体積で)へスケールアップした。高品質VSV産物はこの精製プロセスを介して産生された。
Mustang(商標)Q吸着工程は、VSVINN4CT9−gag1精製プロセスにおいて記載されるのと同一の緩衝液および操作条件を使用して行なわれた。要約すると、細胞および残屑は、遠心分離によって最初に除去された。1:9比率(v/v)で10×ショ糖・リン酸塩・グルタミン酸塩(SPG)の追加、および10mM HEPES、0.465M NaCl、pH7.5、2%のショ糖による2倍の稀釈物後に、溶液を0.2μmフィルターを通してポンプで送り込んだ。濾液をあらかじめ平衡化されたポール・Mustang(商標)Q膜吸着材(0.35mlカプセル体積)の中へロードし、フロースルーおよび洗浄液(FT&W)のプールを回収した。VSV産物は、VSVINN4CT9−gag1プロセスにおいて記載されるものと同一の溶出条件を使用して、溶出プール中に回収された。高品質のウイルス産物が得られた(データ不掲載)。しかしながら、ウイルス産物の3分の1はFT&Wプール中に観察され(表16)、Mustang(商標)Q溶出プール中のウイルス力価は、バイオリアクターにおける低いウイルス産生力価のために非常に低かった(VSVINN4CT1−gag1について4.6×105pfu/ml対VSVINN4CT9−gag1について>1.0×107pfu/ml)。
上で開発されたMustang(商標)Q条件を使用して、3回の確認稼動が、小スケールで、同一の供給物質により完了した。平衡化緩衝液は、10mM HEPES、0.29M NaCl、pH7.5、2%ショ糖であり;一方溶出緩衝液は、10mM HEPES、0.65M NaCl、pH7.0、2%ショ糖であった。同一の操作条件(同一の流速、同一のローディング体積および同一の溶出量)はすべての稼動について維持された。実験結果を表20で要約する。非常に一貫したプロセス性能が、力価分析結果から計算される産物回収に基づいて達成された。
マイクロキャリアーの除去後のVSVINN4CT1−gag1含有細胞培養液を、出発物質として全体の精製プロセスのために使用した。そのプロセスは、低速遠心分離による細胞培養液の清澄化および上清中のVSVの回収;0.45/0.2μmフィルターを通した上清の濾過および濾過溶液中のVSVの回収;陰イオン交換膜吸着材上へのVSV濾過溶液のローディング、溶出プール中のVSV産物の回収;750kDa分子量カットオフ膜を使用するタンジェンシャルフロー濾過(TFF)による回収VSVの精製および未透過残留液中のVSVの回収、および最終的に0.2μmフィルターを通したVSV未透過残留液の濾過および濾過溶液中のVSVの回収を含んだ。3回の6Lスケールアップ/一貫性稼動(CR)および1回の10L稼動(TTR)が、成功裡に完了した。実験条件を表21において要約する。
VSVINN4CT9−gag1のための実施例9において記載されるような精製プロセスは、このVSVコンストラクトへうまく適用され、10Lスケール(細胞培養体積で)へスケールアップした。高品質VSV産物はこの精製プロセスを介して産生された。
Mustang(商標)Q工程は、細胞培養液およびVSVINN4CT9−gag1精製プロセス中に記載されているものと同一の緩衝液および操作条件を使用して行なわれた。要約すると、細胞および残屑は遠心分離によって除去された。1×ショ糖・リン酸塩・グルタミン酸塩(SPG)の追加によって条件付けられ、10mM HEPES、0.465M NaCl、pH7.5、2%のショ糖により2倍希釈された後、上清を0.2μmフィルターを通してポンプで送り込んだ。濾液をあらかじめ平衡化されたMustang(商標)Q膜吸着材にロードし、フロースルーおよび洗浄液のプールを回収した。VSV産物は、VSVINN4CT9−gag1精製プロセス中に記載されるような溶出緩衝液および操作条件の使用により得られた。10mM HEPES、1.0M NaCl、pH7.5、2%ショ糖を使用する再生からのプールも回収された。最終的に、Mustang(商標)Qを1.0M NaOH溶液により洗浄した。高いVSV結合能力が、実験において観察された。しかしながら、産物は溶出プール中にほとんど回収されなかった(データ不掲載)。ウイルス産物は、再生プール中に優勢に検出され、ウイルス産物の2分の1以上は全く回収されなかった(参照、表24)。高レベルのDNA混入物は、溶出プールおよび再生プールの両方において検出された。Mustang(商標)Qのための結合および溶出の条件は、この新しいコンストラクトが、産物回収を増加させ、同時に残存DNAレベルを減少させるようにさらに最適化された。
VSV結合緩衝液条件のスクリーニングは、表25において示されるような全因子実験デザインを使用して、TMAE(商標)樹脂上で行なわれた。供給液は、異なるローディング緩衝液条件へ調整された細胞培養上清だった。フロースルー中のVSVを、ウエスタンブロット分析を使用してモニタリングした。
細胞および細胞残屑を、20〜24℃30分間5000rpmでの遠心分離によってVSVINN4CT9−gag1を含む細胞培養液から除去した。9対1の比率で10×SPGストック溶液に混合した後、上清を0.45/0.2μmフィルターを通してポンプで送り込んだ。濾液を4ml/分の流速でTMAEカラム(2ml)の供給液として使用した。カラムを、10mM HEPES、0.145M NaCl、2%のショ糖、pH7.4によりあらかじめ平衡化した。10カラム体積(CV)の平衡化緩衝液によりカラムをチェイスした後、結合された物質を、30CVで10mM HEPES、1.5M NaCl、2%ショ糖、pH7.5への直線的勾配によってカラムから溶出した。異なる溶出プールを回収した。
VSV結合条件のスクリーニングを、表27において示されるような全因子実験デザインを使用して、Mustang(商標)Q膜吸着材上で行なった。供給液は、異なるローディング緩衝液条件へ調整された細胞培養上清であった。異なるMustang(商標)Q溶出プールをSDS−PAGE分析を使用して解析した(図示せず)。
0.28Mの最終NaCl濃度に達するように、VSVINN4CT9−gag1を含む細胞培養上清を、HEPESストック溶液(10mM HEPES、0.415M NaCl、2%ショ糖、pH7.25)により2倍希釈した。調製された溶液を、実験の供給液として使用した。この研究のための実験デザインを表28において要約する。Mustang(商標)Q MAへ結合させた後に、VSVを異なるNaCl濃度での溶出緩衝液により溶出した。
高レベルのDNA除去および高いプロセス収率を達成するためのMustang(商標)Q工程におけるさらなる最適化は、よりよいVSV結合条件および溶出条件をもたらした。実験デザインを表29に示す。VSVINN4CT9−gag1における最初のスクリーニング研究および以前の経験に基づいて、6.5から7.0までのローディングpHおよび溶出pHを選択した。ローディング緩衝液および溶出緩衝液中の異なったNaCl濃度もまた最初のスクリーニング研究の結果から選択された。すべての実験における流速は7.0ml/分で維持され、20カプセル体積(CV)/分であった。
プロセス収率は、プラーク分析により決定されるVSV産物力価に基づいて計算された。結果は予測プロフィールで要約される(図示せず)。プロセス収率を最大限にするために、Mustang(商標)Q吸着において以下の緩衝液条件が同定された:
ローディング緩衝液中のNaCl濃度:0.26〜0.30M
ローディング緩衝液pH:7.0〜7.5
溶出緩衝液中のNaCl濃度:0.55〜0.65
溶出緩衝液pH:6.5〜7.0
異なる結合条件でMustang(商標)Q溶出プール中の残存DNAレベルは、等高線図において報告された(図示せず)。Mustang(商標)Q緩衝液条件はDNA除去の結果に基づいて、さらに改良された:
ローディング緩衝液中のNaCl濃度:0.27〜0.29M
ローディング緩衝液pH:7.0〜7.5
溶出緩衝液中のNaCl濃度:0.55〜0.65
溶出緩衝液pH:6.5〜7.0
Mustang(商標)Q溶出プール中のVSV生成物純度をSDS−PAGEのデンシトメトリーにより推定した(図示せず)。異なる結合緩衝液条件および異なる溶出緩衝液条件で、産物の純度を解析した。デンシトメトリー分析の変動を考慮して、すべての場合についてVSV産物純度で有意差はなかった。高品質VSV産物がすべての実験において産生された。
Mustang(商標)Q膜クロマトグラフィーの結合緩衝液および溶出緩衝液の条件は、表30において示されるような開発された条件内でおよびその条件外で、より多くの操作点の実験を行なうことにより確定された。平衡化/溶出緩衝液は、様々な量のNaClをともなう、10mM HEPES、2%のショ糖および異なるpH条件であった。同一の操作条件(同一の流速、同一のローディング体積および同一の溶出量)はすべての稼動において維持された。表30において示されるように、一貫したプロセス性能が観察された:SDS−PAGE分析に基づく高品質VSV産物が産生され;許容されるレベルの残存DNA除去はすべてのMustang(商標)Q溶出プールにおいて達成され;類似したプロセス収率が同様に得られた。
全体の精製プロセスは、VSVINN4CT9−gag1を含む細胞培養液を使用して行なった。このプロセスは、低速遠心分離による細胞培養液の清澄化および上清中のVSVの回収;0.45/0.2μmフィルターを通した上清の濾過および濾過溶液中のVSVの回収;陰イオン交換膜吸着材上へのVSV濾液のロード、陰イオン交換膜吸着材からのVSVの溶出およびVSV産物の回収;750kDa分子量カットオフ中空糸膜を使用するタンジェンシャルフロー濾過(TFF)による回収VSVの精製および未透過残留液中のVSVの回収、ならびに0.2μmフィルターを通したVSV未透過残留液の濾過および濾過溶液中のVSVの回収を含む。1回の10L稼動および2回の6L稼動が「一貫性稼動」としてワイス社で完了し、2回の10Lの稼動が技術移転稼動としてヘノジェン(Henogen)社で遂行された。実験条件を表31において要約する。
Claims (18)
- VSVに感染させた哺乳類細胞培養の細胞培養液から水疱性口内炎ウイルス(VSV)を精製するプロセスであって、
(a)低速遠心分離による細胞培養液の清澄化および上清中のVSVの回収と;
(b)0.2〜0.45μmフィルターによる工程(a)の上清の濾過および濾過溶液中のVSVの回収と;
(c)第1のpH緩衝塩溶液により平衡化された陰イオン交換膜吸着材上への工程(b)のVSV濾過溶液のロード、第2のpH緩衝塩溶液による陰イオン交換膜吸着材からのVSVの溶出、および溶出VSV画分の回収と;
(d)300kDa〜1,000kDaの分子量カットオフを有する膜を使用するタンジェンシャルフロー濾過(TFF)による工程(c)で回収されたVSVの精製および未透過残留液中のVSVの回収と、
(e)0.2〜0.22μmフィルターによる工程(d)からのVSV未透過残留液の濾過および濾過溶液中のVSVの回収と
を含むプロセス。 - 工程(e)で回収される前記VSVが、細胞培養タンパク質および核酸混入物を少なくとも90.0%〜約99.0%不含である、請求項1に記載のプロセス。
- 哺乳類の細胞が、ヒト胚腎(HEK)細胞、HEK 293細胞、チャイニーズハムスター卵巣(CHO)細胞、ベイビーハムスター腎(BHK)細胞、アフリカミドリザル腎(AGMK)細胞およびAGMKベロ細胞から選択される、請求項1または2に記載のプロセス。
- 前記低速遠心分離が、4,400×g〜8,000×gである、請求項1〜3のいずれか一項に記載のプロセス。
- 工程(c)における前記第1のpH緩衝塩溶液または第2のpH緩衝塩溶液が、
(a)NaClまたはKClから独立して選択される塩を含むことと;
(b)100mM〜400mMのイオン強度を有する塩を含むことと;
(c)pKa6.0〜8.5の緩衝液を含むことと;
(d)pH6.5〜8.0のpHを有することと;
(e)リン酸緩衝液、N−2−ヒドロキシエチルピペラジン−N’−2−エタンスルホン酸(HEPES)緩衝液またはトリス(ヒドロキシメチル)アミノメタン(トリス)からなる群から選択された緩衝液を含むことと;
(f)1.5%〜5%の濃度でショ糖を含むことと
からなる群から選択される1つまたは複数の特性により独立して特徴づけられる、請求項1〜4のいずれか一項に記載のプロセス。 - 前記第2のpH緩衝塩溶液中の塩がNaClであり、前記VSVが単一工程において第2のpH緩衝塩溶液を加えることにより膜吸着材から溶出され、NaClの単一工程溶出濃度が500mM〜750mMである、請求項5に記載のプロセス。
- 前記第2のpH緩衝塩溶液が、溶出流速10カプセル体積/分(CV/分)〜30CV/分を有する、請求項6に記載のプロセス。
- 前記第2のpH緩衝塩溶液中の塩がNaClであり、前記第2のpH緩衝塩溶液中のNaClのイオン強度が、10CV/分〜30のCV/分の溶出流速で直線的に1mM〜750mMに増加される、請求項5に記載のプロセス。
- 前記TFF膜が、
(a)300kDa分子量カットオフと;
(b)750kDa分子量カットオフと;
(c)中空糸膜モジュールと
からなる群から選択される1つまたは複数の特性により特徴づけられる、請求項1〜8のいずれか一項に記載のプロセス。 - 前記TFFが、工程(c)から回収されたVSVを少なくとも5倍濃縮し、続いて少なくとも1回、または少なくとも5回緩衝液を交換することを含む、請求項1〜9のいずれか一項に記載のプロセス。
- 緩衝液交換において使用される前記緩衝液が、
(a)リン酸緩衝液と;
(b)HEPES緩衝液と;
(c)トリス緩衝液と;
(d)5mM〜15mMの濃度およびpH7.2〜7.5の緩衝液と;
(e)100mM〜200mM NaClおよび3.5%〜4.5%のショ糖をさらに含む緩衝液と
からなる群から選択される1つまたは複数の特性により特徴づけられる、請求項10に記載のプロセス。 - 処理工程(a)〜(e)が、15℃〜25℃のまたはその間の温度または複数の温度で行なわれる、請求項1〜11のいずれか一項に記載のプロセス。
- 工程(a)における細胞培養液の清澄化が1.0μm〜4.5μm深層濾過モジュールによって行なわれ、低速遠心分離が工程(a)から省かれる、請求項1〜12のいずれか一項に記載のプロセス。
- 精製されたVSVが医薬組成物中へ処方される、請求項1〜13のいずれか一項に記載のプロセス。
- 請求項1〜14のいずれか一項に記載のプロセスに従って精製されるVSV。
- (a)VSV Indiana血清型、VSV New Jersey血清型、VSV San Juan血清型、VSV Isfahan血清型、VSV Glasgow血清型およびVSV Chandipura血清型からなる群から選択される血清型と;
(b)VSVの病原性を弱毒化する少なくとも1つの、または少なくとも2つの変異を含むゲノム配列と;
(c)前記弱毒変異が、温度感受性(ts)変異、点変異、遺伝子シャッフリング変異、G−ステム変異、非細胞変性のM遺伝子変異、アンビセンスRNA変異、短縮G遺伝子変異、G遺伝子挿入変異および遺伝子欠失変異からなる群から選択される(b)のゲノム配列と;
